ABSTRACT

Objective To develop a method for determination of gallic acid and ellagic acid in three kinds of Yitirifeili preparation. Methods HPLC method was used to determine the content of gallic acid and ellagic acid.The analysis was performed on Waters XBridge ShieldRP C18column(4.6 mm×250 mm,5 μm) with the mobile phase consisted of methanol-acetonitrile-0.2% phosphoric acid in gradient elution manner.The flow rate was 1.0 mL·min-1,the column temperature was 25 ℃,and the detection wavelength was 254 nm. Results The linear ranges of gallic acid and ellagic acid were 0.132-0.792 μg,0.108-0.648 μg,respectively.Both correlation coefficient were more than 0.999 9.The average recovery of gallic acid and ellagic acid were 98.11%(n=9),RSD=1.39%;99.32%(n=9),RSD=2.48%. Conclusion The method is accurate and reliable.It can be used as the quality control of Yitirifeili preparations.

ABSTRACT

Rosa rugosa Thunb, a deciduous shrub of the genus Rosa, has been widely used to treat stomach aches, diarrhoea, pain, and chronic inflammatory disease in eastern Asia. In recent years, our research team has extensively studied the Rosa rugosa flower extract, and specifically undertook pharmacological experiments which have optimized the extraction process. Our methods have yielded a standard extract enriched in phenolic compounds, named PRE. Herein, we expand our efforts and evaluated the anti-inflammatory activity of PRE on lipopolysaccharide (LPS)-induced inflammation in RAW 264.7 macrophages. PRE significantly inhibited production of nitric oxide (NO), prostaglandin E2 (PGE2), tumor necrosis factor (TNF)-a, interleukin (IL)-6, and interleukin 1β (IL-1β), as well as expression of their synthesizing enzymes, inducible nitric oxide synthase (iNOS) and cyclooxygenase2 (COX-2). Furthermore, PRE inhibited activity of mitogen-activated protein kinases (MAPK) as well as nuclear factor-kappa B (NF-κB) signaling pathway. Our findings are the first to explain the anti-inflammatory mechanism by PRE in LPS-stimulated macrophages. Given these results, we propose that PRE has therapeutic potential in the prevention of inflammatory disorders.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the effect of Abnormal Savda Munziq (ASMq) flavonoids on proliferation, apoptosis and apoptosis-related gene expression in human hepatoma (HepG2) cells in vitro and to probe the mechanism.</p><p><b>METHOD</b>The effects of ASMq flavonoids on proliferation, apoptosis and apoptosis-related gene expression of HepG2 cells were investigated respectively by MTT assay, gel electrophoresis, flow cytometry and RT-PCR.</p><p><b>RESULT</b>ASMq flavonoids significantly inhibited growth of HepG2 cells in vitro, arrested HepG2 in the sub-G, phase, induced cell apoptosis and significantly down-regulated expression level of Bcl-2 mRNA, and up-regulated expression of p53, p21, Bax gene mRNA expressions.</p><p><b>CONCLUSION</b>ASMq flavonoids has significantly regulative action on growth, apoptosis and apoptosis-related gene expression of cancer cells in vitro, which possibly are the important way to excert anticancer effect, and flavonoids are possibly a main active component of ASMq for exerting the anticancer effect.</p>
