ABSTRACT

OBJECTIVE@#To compare the posterior cruciate ligament(PCL) index with six different measurement methods, and analyze and verify its clinical diagnostic value in anterior cruciate ligament (ACL) injury.@*METHODS@#The Magnetic resonance imaging (MRI) data of 225 knee joints in our hospital from May 2018 to March 2022 were retrospectively analyzed, aged from 18 to 60 years old, with a median of 32 years old. On the sagittal MRI images of 114 patients with ACL injury and 111 patients with intact ACL, Measure the straight-line distance (A) between the femoral attachment point and the tibial attachment point of the PCL on the MRI sagittal image and the maximum vertical distance (B) between the straight line and the arcuate mark point of the PCL on the sagittal image, calculate the PCL index and evaluate the diagnostic value of the PCL index for ACL injury.@*RESULTS@#The PCL index of the ACL normal group and the ACL injury group were statistically described. There was no significant difference in PCL index 1, 2, 3 and 6 between the two groups(P>0.05). The difference of PCL index 4 and 5 between the two groups was statistically significant (P<0.001). This study only found that the PCL index 2, 6 in the ACL normal group had a negative correlation with the patient's age (correlation coefficient=-0.213, -0.819;P<0.05), and the PCL index 5 in the ACL injury group was significantly correlated with the patient's body mass index(BMI)had a negative correlation (correlation coefficient=-0.277, P<0.05).@*CONCLUSION@#The change of PCL index is helpful for the diagnosis of ACL injury, PCL index 4 and 5 can be used as effective reference indexes for diagnosing ACL injury in clinic.

ABSTRACT

Disintegration time is a key parameter that affects the palatability and compliance of oral soluble films. At present, there is no standard method to determine the disintegration time of oral soluble films. In this study, we compared the six methods (pharmacopoeial disintegration method, petri dish method, sponge surface method, slide frame and ball method, partially immersed into liquid (without weight attached) and partially immersed into liquid (with weight attached)) to determine the in vitro disintegration time of oral soluble films with different thickness, and evaluated the correlation with the in vivo disintegration time. The results showed that the repeatability and correlation of pharmacopoeial disintegration method and the partially immersed into liquid method (with weight attached) were excellent, with the endpoint of disintegration testing easy to determine. Partially immersed into liquid method (with weight attached), properly simulating the physiological condition in oral cavity, showed strong operability, good repeatability and in vitro-in vivo correlation, and was suitable for in vitro disintegration evaluation of oral soluble film dosage form. The adult sensory evaluation study was a research-based clinical trial conducted with informed consent from all subjects in accordance with the ethical requirements of Good Clinical Practice.

ABSTRACT

Licorzine granules are common preparations for children zinc deficiency. Considering the long course of treatment, the taste of licorzine granules may become a main factor affecting medication adherence. To date there have been no taste evaluation research into licorzine granules yet. In this study, both sensory evaluation and electronic tongue method were utilized to optimize licorzine granules formulations, evaluate the tastes of licorzine, excipients, optimized formulation in vivo and in vitro. As the results show, bitterness and astringency are the main unpleasant tastes generating from licorzine. Xanthan gum is the main taste-masking excipient, lowering down the bitterness and astringency of licorzine by at least one grade. Good correlation exists between the results of sensory evaluation and electronic tongue method, and an integrated combination of the two helps to obtain objective and rational research conclusions. The adult sensory evaluation study was a research-based clinical trial conducted with informed consent from all subjects in accordance with the ethical requirements of Good Clinical Practice.

ABSTRACT

As a novel intervention, whole body periodic acceleration (WBPA) shows a variety of benefits to the cardiovascular system, such as reducing pulmonary arteria hypertension (PAH), improving coronary flow reserve (CFR) and increasing blood perfusion of organs as well as tissues. The underlying mechanisms of WBPA may lie in the increase of blood flow rate and endothelium shear stress, activation of phosphoinositide 3-kinase/serine threonine kinase (PI3K-Akt) and mitogen extracellular signal regulated kinases/extracellular signal regulated kinase (MEK-ERK1/2) signal pathway and the release of nitric oxide.

ABSTRACT

BACKGROUND: Benefits of healthy people participating in physical activity are valued by governments, organizations and individuals, but the benefits of spinal cord injury (SCI) to physical activity are neglected. With the increase in the incidence of SCI, participating in physical activity to keep fit is recommended for SCI patients. OBJECTIVE: To review the relevant literatures on physical activity and health promotion of SCI at home and abroad, and systematically summarize the benefits of physical activity to SCI, so as to provide reference for participating in physical activity. METHODS: The first author recruited the Web of Science and the PubMed databases with the keywords of "spinal cord injury, physical activity, sport, exercise, health, fitness" in English and Chinese, respectively. RESULTS AND CONCLUSION: Physical activity has a positive effect on cardiovascular health, functional independence, quality of life, muscle strength, psychological and social adaptation in SCI patients, and the impact on bone density needs a further investigate. Physical activity holds a positive effect on the health of SCI patients, and it is hoped that SCI patients will actively carry out physical activity to improve or maintain health status. SCI patients exhibit individual differences, so in the use of physical activity to promote health, it is necessary to make individualized sports program to obtain satisfactory effectiveness.

ABSTRACT

The aim of this paper is to investigate the topical pharmacodynamics behavior of different lipophilic model drugs after treatment with essential oil from Zanthoxyli Pericarpium by using the cutaneous microdialysis technique, and then evaluate its in vivo transdermal penetration enhancing properties. Two traditional Chinese medicine active components, namely tetramethylpyrazine and puerarin, were chosen as lipophilic and hydrophilic model drugs, respectively. Firstly, the concentration difference method was employed to measure the in vitro recovery rate and loss of the microdialysis probe, and the in vivo recoveries of two model drugs were determined by using the retrodialysis method. Secondly, the skin pharmacodynamics behaviors of two model drugs were studied after treatment with different concentrations of the essential oil, and the well-established and standard penetration enhancer Azone was selected as a positive control. It was found that the recovery of microdialysis probe was equal to its loss for two model drugs, with no interaction between drugs in dialysis membranes. The retrodialysis studies revealed that the in vivo recovery of tetramethylpyrazine and puerarin were 59.17%, 19.85%, respectively. The skin pharmacodynamics studies showed that the essential oil could facilitate the transdermal absorption of tetramethylpyrazine in a concentration-dependent manner, and the enhancement ratio (ER) for 5% essential oil was 98.64, which was higher than that of the optimum concentration of Azone (3% Azone, ER=89.11). Meanwhile, the Zanthoxyli Pericarpium could effectively promote the transdermal permeation of the puerarin in a concentration-dependent manner. Hence, this study further confirmed that the Zanthoxyli Pericarpium had excellent penetration-enhancing activity as a natural transdermal penetration enhancer, providing data support for its application in traditional Chinese medicine external preparations.

ABSTRACT

To systematically evaluate the efficacy and safety of tripterygium glycosides (TG) combined with ACEI/ARB preparation in treating diabetic nephropathy stage IV. The computer retrievals were made in Cochrane Libarary, PubMed, Embase, SCI, Sinnomed, CNKI, Chinainfo and VIP, and hand retrievals were conducted for meeting and academic papers (updated to December 30, 2014), in order to collect randomized controlled trials and quasi-randomized control trials for TG combined with ACEI/ARB preparation in treating diabetic nephropathy stage IV and set the literature inclusion and elimination standards. Eligible literatures were included and evaluated according to standards in Cochrane Handbook. RevMan 5.3 and Stata 12.0 were used for a Meta-analysis. A total of 13 randomized controlled trials and quasi-randomized control trials involving 1119 patients with diabetic nephropathy were included. The Meta analysis result showed that compared with the control group, the combination group showed better effects in reducing the 24-hour urinary protein [MD = -0.84, 95% CI (-1.02, -0.66)], raising albumin [SMD = 0.98, 95% CI (0.81, 1.16)], the total efficiency [OR = 4.23, 95% CI (2.77, 6.46)] and the significant efficiency [OR = 5.35, 95% CI (2.70, 10.60)], with no statistical difference in Serum Creatinine between Both groups [MD = -0.82, 95% CI (-4.30, 2.66), P = 0.64]. However, the risk of adverse reactions increased by 7% [RD = 0.07, 95% CI (0.03, 0.12)]. The Egger's test showed no publication bias. Tripterygium Glycosides combined with ACEI/ARB in treating diabetic nephropathy stage IV is supper than the single administration of ACEI/ARB, with a good prospect in clinical application. Nevertheless, due to the small-size and low-quality samples in this study, more high-quality and large sample-size randomized controlled trials shall be conducted to verify the findings.

ABSTRACT

<p><b>BACKGROUND</b>Intra-aortic balloon counterpulsation (IABP) is a mechanical cardiac-assist device that is used for cardiac support. There are no published reports about the use of IABP in elderly Chinese patients, especially for those over 80 years old. The aim of this study was to describe the clinical outcomes, influencing factors, and complications in patients ≥80 years old and requiring IABP.</p><p><b>METHODS</b>We performed a retrospective study of 134 consecutive patients who received IABP therapy. Based on age, we defined two groups; those ≥80 years old and those <80 years old.</p><p><b>RESULTS</b>The overall mortality was 41.8%. Patients ≥80 years old had higher mortality rates than those <80 years old (47.9% vs. 30.2%). Patients ≥80 years old had fewer successful revascularizations (45.8%) and more pulmonary infections (47.9%) than patients <80 years old (60.3% and 30.2%, respectively); these differences were statistically significant. The most common non-cardiac complication was pulmonary infection. Cardiogenic shock and pulmonary infection were risk factors for all-cause, in-hospital mortality, whereas revascularization success was a negative risk factor for the ≥80 years old patients.</p><p><b>CONCLUSIONS</b>IABP may be successfully and safely employed in patients ≥80 years old, having severe heart disease, with few complications. Patients ≥80 years old who need IABP therapy are less likely to have a successful revascularization and are more likely to develop pulmonary infections than patients <80 years old.</p>

ABSTRACT

Objective To observe the changes in expressions of apoptosis-promoting gene Bax, apoptosis-inhibiting gene Bcl-2, and cytochrome C in the renal tissue of diabetic rats. Methods Twenty-four male Sprague-Dawley rats were randomly divided into 2 groups (n=12): normal control group and diabetic group. Diabetic models were induced by single intraperitoneal injection of 2% streptozotocin (dissolved in pH 4. 4,0. 1 mol/L citric acid sodium buffer, 65 mg/kg). Normal control group was only injected with same volume of folie buffer. Animals were sacrificed at the 4th and 12th week, and body mass, 24-hour urine protein, blood glucose, blood urine nitrogen and serum creatinine were determined. The changes of the renal morphology were observed by H-E staining. Immunohistochemical method was used to investigate the expressions of Bax, Bcl-2 and cytochrome C protein. The apoptosis of renal cortex cells was determined by TUNEL method. Results Compared with normal control group, the 24-hour urine protein, blood glucose, blood urine nitrogen and serum creatinine were significantly increased in the diabetic group (P<0. 05, P<0. 01). The size of glomerulus was increased in diabetic rats during the 4th week; hyperplasia of renal glomerulus mesangial matrix, glomerular sclerosis, and vacuolar degeneration in renal tubular epithelial cells were observed during the 12th week. With disease progression in the diabetic group, the expressions of Bax and cytochrome C were increased and the expression of Bcl-2 was decreased. Apoptosis tests showed increased apoptotic cells in the 4th week, mostly in both the distant tubular epithelial cells; in the 12th week, apoptotic cells were seen in both the distant tubular and proximal tubules. Conclusion Renal expression of Bax and cytochrome C gradually increases with the progression of diabetes, inducing apoptosis of more cells and leading to renal dysfunction, which may partly contribute to the diabetic nephropathy in diabetic rats.

ABSTRACT

This study is to investigate the anti-tumor effect in vitro of methotrexate modified by LH-RH peptide (LH-RH-MTX). LH-RH receptors highly expressing MCF-7 human breast carcinoma cell line and lowly expressing K562 human erythroleukemia cell line were served as the tested cells. The cell proliferation inhibition rates of LH-RH-MTX were detected by MTT colorimetric assay. The effects of LH-RH-MTX on the cell cycle and apoptosis rates were detected by flow cytometry. The inhibition rate of LH-RH-MTX on MCF-7 cells was much higher than that on K562 cells, and the inhibition rate of LH-RH-MTX on MCF-7 cells was much higher than that of free MTX at the same concentration. The inhibition rate of LH-RH-MTX on rat bone marrow mononuclear cells was less than that of free MTX. The number of MCF-7 cells in S phase increased after administration of LH-RH-MTX. The apoptosis rate of LH-RH-MTX group significantly increased compared with that of the control group and MTX group. The relative expression of LHRHR mRNA of LH-RH-MTX group markedly decreased compared with that of the control group and MTX group. LH-RH-MTX is realizable to reduce drug side effects, increase the therapeutic index and achieve tumor-targeted therapy.
