ABSTRACT

Exploratory experiment is the key to the experimental physiology,which is universally applicable training of innovative talents.More importantly,it is an important way to cultivate innovative talents of medicine.On the basis of the original multi-subject evaluation system,we have formed a comprehensive evaluation system through refining the scoring item for teachers and students,enriching the forms of student evaluation,and building the student self-assessment.This comprehensive evaluation system is set to promote the development of experimental physiology,especially the exploratory experiments.The practice of this evaluation system effectively improve the objectivity of the evaluation,provide a basis for reform of exploratory experiment,and then promote the training of innovative talents.

ABSTRACT

In this study,a series of new sildenafil analogues 11-27 possessing a guanidine group were synthesized to investigate their PDE5 inhibitory activity and selectivity using[~3H]cGMP SPA kit in vitro and efficacy in the rat model of erection.Most of the compounds showed potent activity against PDE5,and more importantly,several compounds exhibited higher PDE5 selectivity over PDE6 than that of sildenafil.Structure-activity relationship of these sildenafil analogues was also discussed.Within this series of compounds,compound 15(IC_(50) =1.7 nmol/L)not only exhibited more potent PDE5 inhibitory activity than that of sildenafil (IC_(50) = 6.5 nmol/L),but also showed functional efficacy in the rat model of erection.

ABSTRACT

AIM: To explore the mechanism of nicotine against the apoptosis induced by colchicines in rat cortical neurons.METHODS: Cortical neurons were cultured from newborn Sprague-Dawley(SD) rats(less than 12 h).The rate of apoptosis was measured by Hoechst33258 fluorescence staining in the neurons,and the activity of Akt473 was analyzed by assay kit Akt473.RESULTS: The apoptosis of cortical neurons can be induced by 0.1 ?mol/L colchicine.The phosphorlation of Akt 473 decreased significantly(1/3 times of the control group,P
