ABSTRACT

Objective:To establish a HPLC method for determinating 9 components simultaneously in Swertia chirayita. Methods:By useing water Sunfire C18 column (4.6 mm× 250 mm,5 μm); Gradient elution was carried out with methanol-0.05% phosphoric acid solution as mobile phase. Setting the column temperature at 30 ℃, the flow rate at 1.0 ml/min, and the detection wavelength at 254 nm.Results:9 components showed good linear relationship within the injection quality range. The recovery rates of wertiamarin, Gentiopicroside, Angelica glycosides,Mangiferin, Isolysine, Gentianoside, Diol glycoside, 8-hydroxy-1,3,5 trimethoxyketone, and Daisy leaf gentinone were 95.38%, 92.41%, 95.14%, 91.87%, 92.24%, 92.51%, 95.08%, 91.72%, 95.74% ( n=6). Conclusion:The method is simple, efficient, sensitive, accurate, economical and practical, with repeatability and stability. It could provide reference for the quality control and comprehensive utilization of Swertia chirayita.

ABSTRACT

Objective: To establish the quality standard for Jiuwei Zhaxun pills. Methods: Fructus Chebulae, Flos Carthami and Herba Dracocephali Heterophylli were identified by TLC. HPLC was used to determine the content of gallic acid in Jiuwei Zhaxun pills. Results: TLC showed clear sports without any interference from the negative control. Gallic acid showed a good linear relationship (r=0. 999 7) within the range of 0. 028 6-0. 572 0 μg. The average recovery was 97. 74% with the RSD of 0. 95% (n=6). Conclusion:The methods are with strong specificity, high sensitivity and good reproducibility, which can be applied in the quality control of Jiuwei Zhaxun pills.

ABSTRACT

Objective To investigate the expression and significance of ADAMDEC1 in glioma.Methods We prospectively observed the expression of ADAMDEC1 in 77 glioma patients,of which 28 cases were grade Ⅳ glioma,26 were grade Ⅲ glioma (high-grade glioma);of which 23 cases were Grade Ⅱ glioma,5 were grade Ⅰ glioma (low-grade glioma).For 10 cases in the control group,brain tissue samples were collected from internal decompression patients with heavy traumatic brain injury.The expression of ADAMDEC1 in glioma tissue was detected immunohistochemistry,RT-PCR,and Western blot.With follow-up survey we explored the relationship between the survival time and the expression of ADAMDEC1 in the patients with high-grade gliomas and low-grade gliomas.Results Compared with the low-grade glioma group and the control group,qPCR,immunohistochemistry and Western blotting all showed that the protein and mRNA levels of ADAMDEC1 in high-grade glioma group were significantly increased,while ADAMDEC1 was expressed little in low-grade glioma group and not expressed in control group (P < 0.05).The survey results showed that the survival time of high-grade glioma patients was significantly shorter than low-grade glioma patients;however,the expression level of ADAMDEC1 in the highgrade glioma tissue was significantly higher than in low-grade glioma tissue (x2=5.031,P<0.05).Conclusion The expression of ADAMDEC1 is closely related to the malignant degree of glioma cells and the prognosis of glioma patients.Therefore,ADAMDEC1 can be used as an index to evaluate the malignance degree of glioma and the prognosis of glioma.

Objective:To establish a TLC identification method for three medicinal ingredients and a GC method for the content determination of metnthol in Baihua Dingchuan tablets. Methods:Citrus Reticulata, Ephedra Sinica and Schisandra Chinensis were i-dentified by TLC. GC was used to determine the content of menthol in Baihua Dingchuan tablets. The chromatographic conditions were as follows:a DM-5 capillary column(30 m × 0. 53 mm, 1. 5 μm) was adopted. The injection port temperature was 200℃. An FID was used as the detector and the temperature was 300℃. The temperature program was as follows:the initial column temperature was 80℃, and then rose to 120℃ at the rate of 8 ℃· min-1 , finally rose to 185 ℃ at the rate of 15 ℃· min-1 and maintained for 10 min. The carrier gas was nitrogen and the flow rate was 3. 0 ml·min-1 . The sample size was 1 μl. Split injection was performed with the diversion ratio of 1 :5. Results: The three traditional Chinese medicines could be identified by TLC. The content of menthol showed a good linear relationship within the range of 1. 484-74. 200μg · ml-1 ( r =0. 9998 ) by GC. The average recovery was 97. 76% RSD=0 . 58%(n=9). Conclusion:The methods are specific and simple with good reproducibility. They can be used for the quality control of Baihua Dingchuan tablets.

ABSTRACT

Objective To detect the change of brain activity under different depth of anesthesia (DOA)noninvasively. Methods The Lempel-Ziv complexity C(n) was used to analyze EEG and its four components (delta,theta, alpha, beta), which was recorded from SD rats under different DOA. The relationship between C(n) and DOA was studied. Results The C(n) of EEG will decrease while the depth of anesthesia increasing and vice versa. It can be used to detect the change of DOA sensitively. Compared with power spectrum, the change of C(n) is opposite to that of power spectru,. Only the C(n) of delta rhythm has obvious variations induced by the change of DOA, and the variations of delta is as similar as the EEG's. Conclusion The study shows that the desynchronized EEG is replaced by the synchronized EEG when rat goes into anesthesia state from awake, that is just the reason why complexity and power spectrum appear corresponding changes under different DOA. C(n) of delta rhythm dynamic change leads to the change of EEG, and the delta rhythm is the dominant rhythm during anesthesia for rats.

ABSTRACT

The classical cable function has been used to represent the response of peripheral nerves stimulated by external parallel electric field. Experiments show that peripheral nerves can be activated by perpendicular electric field induced by magnetic pulses, indicating that the activation mechanism needs to be thoroughly investigated. Several excitation properties of peripheral nerves in transverse-field were explored in the present paper. With a human's median nerve in vivo, stimulation threshold, excitation position and the relation between excitation threshold and fiber's radius were studied. The relation between stimulation threshold and stimulation duration was researched with sciatic nerves from toad in vitro. The experimental results verify that the modified cable function is much efficient than the classical cable function. The research will improve the nerve magnetic stimulating technique and be beneficial to further application.

ABSTRACT

A new nylon affinity matrix with L-tryptoph ane as ligand was prepared for adsorption of bovine γ-globulin. Effects of tem perature, ionic strength and pH on affinity adsorption of γ-globulin on affinity membrane were studied by batch method. The results show that the interaction between ligand and γ-globulin includes mainly electrostatic and hydrophobic interaction. The affinity adsorption at optimum condition obeys Langmuir adsorption model with maximum adsorption capacity and minimum nonspecific adsorption. The space location between protein and ligand and the configuration of proteins wi ll change when deviating from this condition.
