ABSTRACT

Objective:This study aims to observe the effect of baicalein on the clonal formation of triple negative breast cancer MDA-MB-231 and MDA-MB-468 cells, and to explore the mediation role of Yes- related protein (YAP) in it. Method:MDA-MB-231 and MDA-MB-468 cells were treated with baicalein. Thiazole blue (MTT) colorimetric method was used to detect cell proliferation ability. Plate cloning experiments was used to detect the colony forming ability. Immunofluorescence method was used to detect the nuclear distribution of YAP, and Western blot test was used to detect the protein expression levels of YAP large tumor suppressor factor 1 (LATS1), YAP, phosphorylated Yes- related protein(p-YAP) and phosphorylated YAP large tumor suppressor factor 1 (p-LATS1). Result:Compared with the blank group, baicalein (40, 80, 160 μmol·L<sup>-1</sup>) significantly inhibited the proliferation ability of MDA-MB-468 and MDA-MB-231 cells (<italic>P</italic><0.05, <italic>P</italic><0.01), and the inhibitory effect was dose-dependent. The half inhibit concentration(IC<sub>50</sub>) of baicalein against MDA-MB-468 and MDA-MB-231 cells were (80.3±7.2),(70.4±6.5) μmol·L<sup>-1</sup>, respectively. Compared with blank group, baicalein (5, 10, 20 μmol·L<sup>-1</sup>) had no significant effect on the proliferation of MDA-MB-468 and MDA-MB-231 cells, and the difference was not statistically significant. Compared with the blank group, baicalein (5, 10, 20 μmol·L<sup>-1</sup>) significantly dose-dependently reduced the cell colony formation rates of MDA-MB-468 and MDA-MB-231 cells (<italic>P</italic><0.05, <italic>P</italic><0.01), and baicalein (10, 20 μmol·L<sup>-1</sup>) significantly inhibited the nuclear expression of YAP in MDA-MB-468 and MDA-MB-231 cells in a dose-dependent manner(<italic>P</italic><0.01). Also, baicalin (5, 10, 20 μmol·L<sup>-1</sup>) significantly up-regulated p-YAP and p-LATS1 protein expressions in MDA-MB-468 cells in a dose-dependent manner (<italic>P</italic><0.05, <italic>P</italic><0.01). Baicalein (10, 20 μmol·L<sup>-1</sup>) significantly up-regulated p-YAP and p-LATS1 protein expressions in MDA-MB-231 cells in a dose-dependent manner (<italic>P</italic><0.01). Conclusion:Baicalein can inhibit colony formation of triple negative breast cancer MDA-MB-468 and MDA-MB-231 cells by mediating the reduction of YAP entry into the nucleus.

ABSTRACT

Objective To systematically evaluate the clinical efficacy of relieving fever with sweet and warm drugs (Ganwen Chure Therapy) for noninfectious fever caused by vital-energy deficiency based on Meta-analysis. Methods Literature about RCTs of Ganwen Chure Therapy for noninfectious fever caused by vital-energy deficiency in CNKI, Wanfang Database, CBM, VIP, Pub Med, Cochrane Library, and Embase was retrieved by computers from establishment of database to April 2017. After two researchers independently conducted literature screening, cross-checking, data extraction, and literature quality evaluation, cumulative Meta-analysis was performed on the outcome indicators in order of publication time and sample size, and the trend of the results was tested. Then the quality of the literature based on GRADE was under an overall evaluation. Results Totally 27 articles were included in this study, involving 2599 patients. The results of cumulative Meta-analysis showed that the total effective rates of using Ganwen Chure Therapy only [OR=3.875, 95%CI (2.87, 5.24), Z=8.82, P=0.000 1]and Ganwen Chure Therapy combined with routine therapy [OR=5.791, 95%CI (3.55, 9.45), Z=7.03, P=0.000 1]were better than the routine therapy, with statistical significance. Trend test showed that there was variability in the timing of drug combination study, showing that the cumulative Meta-analysis results were not stable. Conclusion Ganwen Chure Therapy has certain efficacy for noninfectious fever caused by vital-energy deficiency. However, the overall quality of the included studies was low, with relatively high homogeneity. There are biases in publication, yet more high-quality clinical research is needed for further verification.

ABSTRACT

AIM:To study and compare the outcomes of coaxial 2. 2 mm phacoemulsiflcation with conventional coaxial 3 mm small-incision cataract surgery. METHODS: A randomized prospective study was conducted on 100 patients with age - related cataract:coaxial 2. 2 mm micro - incision cataract surgery was performed in 50 cases (50 eyes), and coaxial 3 mm small incision cataract surgery was performed in 50 cases ( 50 eyes) . Statistical analysis was takenwith the data of the two groups. Visual acuity, VF and QOL were compared at intervals of 1wk and 3mo after surgery. In addition, surgically induced astigmatism ( SIA ) was analyzed. Statistic analysis was taken by Student's t-test and Chi-square test. RESULTS:There was no significant difference on BCVA (t=-1. 366, -1. 688; P=0. 148, 0. 107) between these two groups. One week and 3mo after the surgery, SIA was (0. 46±0.29)D, (0. 43±0. 26)D in the 2. 2 group; and (1. 55±0. 59) D, (0. 89±0. 28) D, in 3. 0 group. The differences between these two groups were statistically significant ( t=-7. 348, -3. 788; P = 0. 000, 0. 000 ) There were no statistically significant differences on VF scores between two group, while it's got a better score in 2. 2 groups on vision adaptation. (t=-3. 348, P CONCLUSION:Coaxial 2. 2mm micro-incision cataract surgery could significantly reduce SIA and obtain morestable status of VF and QOL. This suggests that the coaxial 2. 2 phacoemulsification surgery implanted AkreosMI60 intraocular lens could get earliervisual rehabilitation postoperation.

ABSTRACT

Objective: To observe the changes in the anatomy of anterior segment before and after phacoemulsification for treatment of angle-closure glaucoma by Pentacam system, so as to assess the efficacy of phacoemulsification combined with artificial lens implantation(ALI) in treatment of early- and middle-stage angle-closure glaucoma. Methods: Forty-two patients with early- and middle-stage angle-closure glaucoma(44 eyes) were included in the present study. The central anterior chamber depth, anterior chamber volume and peripheral anterior chamber angle were examined by Pentacam system pre-operatively and 1 day, 1 week and 3 months post operatively. Meanwhile, A ultrasound was used to examine the anterior chamber depth before and 3 months after operation, and the results were compared with those by Pentacam system. Results: Pentacam system could clearly demonstrate the 3-dimensional structures of the anterior eye segment before and after operation. The central anterior chamber depth, anterior chamber volume and anterior chamber angle were increased significantly after operation(P<0.01); the intraocular pressure was significantly decreased(P<0.01); and the visual acuity was improved(P<0.01). The values of central anterior chamber depth and the dispersion coefficient obtained by Pentacam were smaller than those by A ultrasound. Conclusion: Pentacam system can directly demonstrate the increase of anterior chamber depth, anterior chamber volume, and anterior chamber angle after phacoemulsification combined with ALI in treatment of early- and middle-stage angle-closure glaucoma. It is a safe method for early examination after operation.

ABSTRACT

Objective: To evaluate the effect of subconjunctival injection of Bevacizumab (Avastin) on corneal neovascularization(CNV) in rabbits. Methods: Forty-eight New Zealand rabbits were randomly divided into three groups: normal control group (group A, 3 rabbits), neovascularization group (group B, 9 rabbits) and Bevacizumab treatment group (group C, 36 rabbits). Group C was further divided into group C1 (1 day small dose group), C2 (1 day high dose group), C3 (14 days small dose group) and C4 (14 days high dose group), with 9 rabbits each group. CNV model was made by suture in group B and C. Animals in group C were injected subconjunctivally with 0.1 ml or 0.2 ml Bevacizumab (25 mg/ml) in the left eyes. The growth of CNV was observed every day after operation and the neovascularization areas calculated. Expression of VEGF in the cornea was detected by immunohistochemistry on day 7, 14 and 28 after suture. VEGF content in the aqueous humor was determined by ELISA assay. Results: CNV growth in group C1 and C2 was inhibited significantly compared with that in group B on day 7, 14 and 28 (P<0.01). On day 28 the growth in group C3 and C4 was significantly inhibited compared with that in group B (P<0.01), that in group C1 was significantly inhibited than that in group C3, and that in group C2 was significantly inhibited compared with that in group C4 (P<0.01). VEGF levels in the cornea and aqueous humor in group B increased as time passed by, and they were positively associated with CNV area (P<0.01). On day 7, 14, and 28, the VEGF levels in the cornea and aqueous humor in C1 and C2 groups were significantly lower than that in group B (P<0.01). On day 28, those in group C3 and C4 were significantly lower than those in group B (P<0.01); those in group C1 were significantly lower than those in group C3 (P<0.01); and those in group C2 were significantly lower than those in group C4 (P<0.01). The CNV area and VEGF levels in the cornea and aqueou humor were similar between C1 and C2 groups and between C3 and C4 groups. Conclusion: Subconjunctival administration of Bevacizumab can effectively inhibit corneal neovascularization in experimental CNV model, and early administration has a better outcome than late administration. Bevacizumab may exert its effect by down-regulating VEGF in cornea and aqueous humor.

ABSTRACT

This study examined the effect of different b values on diffusion-weighted MR imaging (DWI) of human prostate by using single-shot spin echo echo planar imaging (SE-EP1) sequences,observed the normal appearances and measured apparent diffusion coefficient (ADC) values in anatomical regions of normal prostate. Twenty-four healthy volunteers (mean age: 32 y) were studied by using a 1.ST system with a phased array surface multicoil. Two kinds of single-shot SE-EPI sequence were used to perform DWI in the prostate in volunteers, with five b values being 0, 30, 300, 500 to 1000 s/mm2. The image quality with different imaging parameters was analyzed and the ADC values in anatomical regions of normal prostate were measured. DWI of prostate was successfully obtained in all volunteers. The images were of good quality, without artifacts containing pixels within the prostate. The contrast was good between the different anatomical regions of the prostatic gland, i.e.,the peripheral zone (PZ), which exhibited higher signal intensity, and the central gland (CG). Signal intensity contrast was related to the magnitude of b values. The ADC values in PZ and CG were (1.27±0.22)× 10-3 mm2/s and (1.01±0.17)× 10-3 mm2/s, respectively. The ADC values were found to he significantly higher in PZ than in CG (P<0.05, paired t-test). Significant differences were found between the slice-selecting component and both the read-out and phase-encoding components of the ADC values. It is concluded that SE-EPI is a suitable DWI sequence for human prostate. The contrast between PZ and CG is good when b values are low, while the diffusion and ADC values are accurate when b values are high. ADC values are higher in PZ than in CG in normal prostate. Diffusional anisotropy is present in normal prostatic tissue.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the protective effect and mechanism of Yimucao (Herba leonuri) injection against experimental cerebral ischemia.</p><p><b>METHODS</b>Mouse models of cerebral ischemia induced by bilateral carotid artery occlusion or potassium cyanide and rat models of middle cerebral artery occlusion (MCAO)-induced cerebral ischemia/reperfusion injury were established to evaluate the protective effect of Yimucao injection by measuring the changes in cerebral malondialdehyde (MDA) content and the activities of superoxide dismutase (SOD) and lactate dehydrogenase (LDH) after the injections.</p><p><b>RESULTS</b>Yimucao injection significantly lowered the cerebral index of mice with cerebral ischemia, prolonged the survival time of mice poisoned with potassium cyanide, resulting also in significantly decreased MDA content and increased activities of SOD and LDH in the brain tissue of rats after a 10-min cerebral ischemia followed by 30 min of reperfusion.</p><p><b>CONCLUSION</b>Yimucao injection provides protective effect against experimental cerebral ischemia.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the effect of Qinggan Huoxue recipe (QGHXR) in treating alcoholic liver disease (ALD).</p><p><b>METHODS</b>By adopting the multi-centered, randomized and controlled method, the patients were divided in to the QGHXR group (60 patients) treated orally by QGHXR, the XCHG group (30 patients) treated orally by Xiaochaihu granule and the control group (30 patients) treated orally by conventional therapy such as glucurolactone, vitamin C. The changes in symptoms, signs, liver function, blood lipid, liver fibrosis markers, cytokines, lipid superoxidation parameters and B-untrasonographic figure after treatment of the two groups were observed.</p><p><b>RESULTS</b>The total therapeutic efficacy of QGHXR, improvements in anorexia, nausea, vomiting and jaundice as well as effect in reducing blood levels of alanine transaminase (ALT), aspartate aminotransferase (AST) and triglyceride (TG) were superior in the QGHXR group to those in the other two groups (P < 0.01), and the effect in decreasing gamma-glutamyl transferase (GGT) and very low-density lipoprotein (VLDL) in the QGHXR group was more significant than that in the control group (P < 0.01). QGHXR also showed effects in lowering levels of liver fibrosis markers and cytokines, alleviating the anti-lipid superoxidation damage in liver, and could markedly improve the degree of fatty liver.</p><p><b>CONCLUSION</b>QGHXR shows obvious therapeutic effect in treating ALD, the mechanism could possibly be related with its effects in antagonizing lipid superoxidation, stabilizing hepatic cell membrane, adjusting the lipid metabolic disturbance of liver, regulating immune function, anti-liver fibrosis and promoting the intrahepatic metabolism of alcohol.</p>
