ABSTRACT

BACKGROUND@#Gastric cancer (GC) is one of the most globally prevalent cancers in the world. The pathogenesis of GC has not been fully elucidated, and there still lacks effective targeted therapeutics. The influence of altered kinesin superfamily protein 22 (KIF22) expression in GC progression is still unclearly. The aim of this study was to investigate the KIF22 effects on GC and related mechanisms.@*METHODS@#Gastric carcinoma tissues and matching non-cancerous tissues were collected from patients with GC who have accepted a radical gastrectomy in Lanzhou University Second Hospital from May 2013 to December 2014. The expression of KIF22 was examined in GC of 67 patients and 20 para-carcinoma tissues by immunochemical staining. The relationship between the expression of KIF22 and clinicopathologic characteristics was next investigated in the remaining 52 patients except for 15 patients who did not complete follow-up for 5 years. Cell viability was performed via 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide (MTT) test and colony formation assay in the MGC-803 and BGC-823 GC cells. Cell scratch and trans-well invasion assay was performed to assess migration ability in the MGC-803 and BGC-823 GC cells. Gene set enrichment analysis (GSEA) pathway enrichment analysis was performed to explore the potential functions. Cell cycle was detected by flow cytometry. In addition, the two GC cell lines were used to elucidate the underlying mechanism of KIF22 in GC in vitro via assessing the effects on mitogen-activated protein kinase and extracellular regulated protein kinases (MAPK/ERK) signal transduction pathway-related expressions by Western blotting assays. The differences were compared by t tests, one-way analysis of variance, and Chi-squared tests.@*RESULTS@#The study showed that KIF22 was up-regulated in GC, and KIF22 high expression was significantly related to differentiation degree (χ = 12.842, P = 0.002) and poorly overall survivals. GSEA pathway enrichment analysis showed that KIF22 was correlated with the cell cycle. Silence of KIF22 decreased the ability of the proliferation and migration in gastric cells, induced G1/S phase cell cycle arrest via regulating the MAPK-ERK pathways.@*CONCLUSIONS@#KIF22 protein level was negatively correlated with prognosis. KIF22 knockdown might inhibit proliferation and metastasis of GC cells via the MAPK-ERK signaling pathway.

ABSTRACT

AIM:To investigate the risk factors for malignant glaucoma occurrence after the surgery for primary angleclosure glaucoma,and to provide theoretical basis for the prevention and treatment of malignant glaucoma.METHODS:The clinical data of 767 cases (965 eyes) of primary angle-closure glaucoma patients treated in our hospital from June 2012 to June 2016 were retrospectively analyzed.Patients were divided into two groups according to the postoperative investigation,patients with malignant glaucoma were divided into observation group,and 50 eyes without malignant glaucoma were randomly selected into control group.The gender,age,preoperative high intraocular pressure,chamber angle structure,axial length,crystal thickness,anterior chamber depth,diabetes mellitus,and hypertension of the patients in two groups were investigated.The risk factors were analyzed by single factor analysis,and the independent risk factors were analyzed by multivariate logistic regression analysis.RESULTS:Malignant glaucoma occurred in 30 eyes after the surgery of primary angle closure glaucoma in 965 eyes with an incidence rate of 3.1%.The single factor analysis showed that age,preoperative high intraocular pressure,axial length,anterior chamber depth,lense thickness,complete closure of atrial angle were the risk factors of malignant glaucoma after primary angle closure glaucoma surgery,comparison between groups showed statistical significance (P < 0.05).Multivariate logistic regression analysis showed that age (OR=2.521,95% CI=1.434-8.876),preoperative continuous high intraocular pressure (OR=2.483,95% CI =2.123-11.543),axial length (OR=2.654,95% CI=1.547-12.678),complete closure of atrial angle (OR=3.212,95% CI=1.543-8.675) were the independent risk factors for malignant glaucoma after primary angle closure glaucoma surgery.CONCLUSION:The incidence rate of malignant glaucoma after primary angle-closure glaucoma surgery in our hospital is 3.1%.Age,preoperative high intraocular pressure,axialc length,complete closure of atrial angle can rise the risk of occurring malignant glaucoma,so special attention shall be payed for this kind of patients.

ABSTRACT

Mouthpart developmental histology of Whitmania pigra at different month of age were studied by paraffin section, HE staining combined alcian blue and periodic acid schifts reaction procedure (AB-PAS). The following results was obtained: Change ranges: oral width 0.6 mm (1-3 month), 1.2 mm (34 month); oral diameter 0.3 mm (1-3 month); 1.2 mm (34 month), the oral size reached maximum during 4-6 months and unchanged thereafter. Oral lip had a thin protective film located in the front of the mouthpart. The W. pigra possessed three jaws in oral cavity, the big one was in dorsum, the other two separated on both side of abdomen respectively. Jaws and muscular pharynx were interrelated closely. The jaws were composed by cuticle, epithelial layer, muscularis and jaw cavity from outside to inside. In the front of jaws had mastoid abdomen with function of secreting acidophilic granule from 2 month age. Oral cavity was composed by mucosa, submucosa and muscularis inside and outside. Oral cavity was rich of peristomial nerves. And pharynx was composed of mucosa, muscularis, adventitia from inside to outside. The folds height and width become heighten and thicken. Mucosa epithelium from complex flat epithelium changed into columnar epithelium, muscularis gradually developed into thickened along with growing. Muscular thickness reached maximum at 4 months. Mucous cells of W. pigra were classified into I-IV types based on different staining and two mainly morphological shapes (Tubular, Pear-shaped). Jaws, oral cavity, pharynx by AB-PAS staining showed little changes at different month of age. Mucous cells were few at 1 month age, and type II cells were increased rapidly in 2-3 month age in oral lip. Oral cavity contains more mucous gland cells type I. Under the muscularis there were connective tissues which distributed a few of mucous cells type II.

ABSTRACT

Objective To evaluate the clinical efficacy and safety of two glycolic acid-containing liquid preparations for the treatment of facial acne vulgaris.Methods Sixty patients with mild to moderate acne vulgaris were included in a randomized,single-blinded,split-face,controlled study.The left and right sides of each patient's face were randomly assigned to the test group (treated with one hydroxy acid-containing liquid preparation) and control group (treated with the other hydroxy acid-containing liquid preparation).In total,all the patients were given four times of treatment at week 0,2,6 and 10 with the concentration of hydroxy acid in both preparations being 20%,35%,50% and 70%,respectively.The number of inflammatory and noninflammatory acne lesions was counted,and the improvement in acne mark,scar and pores was evaluated by a 6-point grading scale before every treatment and 4 weeks after the last treatment.Results At 2,6,10,and 14 weeks after initiation of treatment,the clearance rate for inflammatory lesions was 20.4%,43.3%,66.9% and 80.2% respectively in the test group,21.6%,44.2%,68.1% and 79.8% respectively in the control group,while that for noninflammatory lesions was 18.5%,34.0%,41.3% and 59.6% respectively in the test group,17.8%,31.2%,44.9% and 57.1% respectively in the control group.The response rate was 80.0% in the test group and 78.3% in the control group at the end of the study.Adverse effects were observed in both groups,including temporary pain,erythema and formation of hoar frost,all of which well responded to symptomatic treatment.There were no significant differences in clinical efficacy or adverse effects between the two groups (both P > 0.05).Conclusion Both of the hydroxy acid-containing preparations are effective and safe for mild to moderate acne vulgaris.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the effects of hydrogen sulfide (H2S) on H2O2-stimulated primary neonatal rat cardiomyocytes and related mechanism.</p><p><b>METHODS</b>Primary neonatal rat cardiomyocytes were treated with various concentrations of H2O2 (10, 100, 1000 µmol/L) for 24 h to establish the oxidative stress-induced cell injury model after 3 days' conventional culture. In addition, different concentrations of NaHS (1, 10, 100 µmol/L) were added to cardiomyocytes in the absence and presence of 100 µmol/L H2O2 for 24 h. The viability of cardiomyocytes was measured by MTT assay. The SOD vitality was measured by xanthine oxidase method and MDA content was determined by thiobarbituric acid colorimetric method. LDH activity was measured by chemical colorimetric method. The percentage of apoptotic cells was assessed by flow cytometry (FCM). The mitochondrial membrane potential (MMP) was analyzed by rhodamine 123 (Rh123) staining and photofluorography. The level of reactive oxygen species (ROS) in cardiomyocytes was measured by DCFH-DA staining and photofluorography.</p><p><b>RESULTS</b>Cell viability and SOD vitality were significantly reduced while MDA content and LDH activity were significantly increased with increasing H2O2 concentrations. These effects could be partly reduced by cotreatment with H2O2 in a concentration-dependent manner (all P < 0.05). Compared with control group, the DCF fluorescence intensity significantly increased in the 100 µmol/L H2O2 group (P = 0.003), which could be attenuated by NaHS in a dose-dependent manner. Compared with control group, the MMP significantly decreased in the 100 µmol/L H2O2 group (P = 0.000), which could be partly reversed by cotreatment with NaHS in a dose-dependent manner. Moreover, H2O2 treatment also significantly reduced 100 µmol/L H2O2 induced apoptosis in a dose-dependent manner.</p><p><b>CONCLUSION</b>H2S protects primary neonatal rat cardiomyocytes against H2O2-induced oxidative stress injury through inhibition of H2O2 induced overproduction of ROS, dissipation of MMP and apoptosis.</p>

ABSTRACT

Objective To evaluate the effectiveness and safety of a sodium hyaluronate-containing dressing for skin wound healing after Q-switched laser therapy.Methods Sixty-six patients with facial pigmentary disorders were enrolled in this study.After treatment with Q-switched laser,the patients were randomly and equally divided into three groups to apply a sodium hyaluronate-containing dressing,a thermal spring water-containing facial mask and a distilled water-containing facial mask respectively.The dressing or facial masks were externally applied to the face for 15 minutes once immediately after the laser therapy,then,once every night for 14 consecutive days.Inflammatory responses,such as erythema,edema and burning sensation,were measured semi-quantitatively on day 1,3,7 and 14 separately after the laser therapy.The time for skin wound healing and degree of patients' satisfaction were compared among these groups by chi-square test and rank sum test,respectively.Results The sodium hyaluronate-containing dressing and thermal spring watercontaing facial mask were superior to the distilled water-containing facial mask in the improvement of erythema,edema and burning sensation after the laser therapy (all P < 0.05).Compared with the patients applying the thermal spring water-containg facial mask and those applying the distilled water-containing facial mask,those applying the sodium hyaluronate-containing dressing felt better,with significantly increased skin cleanliness,humidity,smoothness,confort degree and overall satisfaction (all P < 0.05).Conclusion The sodium hyaluronate-containing dressing is effective for improving acute inflammatory responses and increasing skin cleanliness,humidity and smoothness after laser therapy.

ABSTRACT

Objective To investigate the high-risk behaviors related to acquared immune deficiency syndrome/sexually transmitted disease (AIDS/STDs) infection among fishermen in Lv-si harbor,Jiangsu province.Methods A cross-sectional study was designed to investigate the research participants' demographic characteristics,high-risk behaviors that related to AIDS/STDs.Logistic regression was performed to measure the associations between potential risk factors and reported potential high-risk sexual behavior.Results 817 fishermen participated in the study and casual or commercial sex activities appeared to be the main high-risk behavior for AIDS/STDs infection in the target population.The rates of casual and commercial sex reported were 18.1% and 28.9% among fishermen.Risk factors associated with AIDS/STDs related high-risk behaviors among fishermen were high mobility (OR=1.516,P=0.038),higher lifetime sex frequency (OR=1.422,P=0.002)and unmarried status ( OR =7.527,P=0.014).Protective factors against high-risk behaviors were low intake of alcohol (OR=0.803,P=0.053),negative STD history (OR=0.268,P=0.001 ),age of initial sexual intercourse at or older than 22 years (OR =0.440,P=0.000) of age,as well as negative attitude toward multiple sexual parmers (OR=0.662,P=0.023 ) and legitimation for commercial sex (OR=0.612,P=0.007).Conclusion There were risk behaviors of AIDS/STDs in those infected fishmen.Casual and commercial sex were common high-risk behaviors.

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the effect of a phospholipid-coated microbubble contrast agent for myocardium opacification in comparison with a albumin-coated microbubble contrast agent (Quanfuxian).</p><p><b>METHODS</b>In 10 dogs with single coronary artery stenosis involving the anterior descending branch or circumflex branch randomly received infusion of the two contrast agents through the femoral vein. The myocardial blood flow, heart rate and blood pressure were analyzed qualitatively and quantitatively. The concentration and the particle diameter of the two contrast agents were determined.</p><p><b>RESULTS</b>The concentration of the phospholipid-coated microbubbles was (1.06-/+0.22) x10(9)/ml, with a diameter of 3.04-/+0.34 microm, similar to the concentration and diameter of Quanfuxian ((1.31-/+0.33)x10(9)/ml and 2.88-/+0.58 microm, respectively, P>0.05). Both of the agents achieved grade three myocardium opacification, and produced no obvious effect on the heart rate and blood pressure. Quantitative analysis of myocardial opacification in terms of myocardial blood volume (A), blood velocity (beta), and blood flow (A x beta) revealed no significant difference between the two agents (P>0.05), and the parameters derived from the two agents showed good correlations (P<0.05, rA=0.809, r beta=0.932, rA.beta=0.925).</p><p><b>CONCLUSION</b>The phospholipid-coated microbubble contrast agent shows good effect for myocardial opacification without significant difference from Quanfuxian. Both of the agents are good ultrasound contrast agents for quantitative analysis of myocardium blood flow.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the value of CT perfusion in early diagnosis and management of superacute local cerebral infarction in rhesus monkeys.</p><p><b>METHOD</b>Acute local cerebral infarction was induced in the rhesus monkeys during digital subtraction angiography (DSA) by introduction of pale thrombus prepared from autologous blood into the M1 branch of the middle cerebral artery (MCA). Plain CT scan and CT perfusion scanning were performed at different time points before and after DSA operation, and the results were analyzed in conjunction with the pathologic changes.</p><p><b>RESULTS</b>Ischemic lesions were displayed on CT perfusion images, which showed local hypoperfusion, reduced cerebral blood flow and volume, and mean transit time delay in the compromised area. Local hypointense infarct area was identified in plain CT scan 24 h after the DSA operation, and the results were in good agreement with pathological examination during autopsy.</p><p><b>CONCLUSION</b>CT perfusion imaging of the brain can accurately capture the cerebral perfusion deficits in acute ischemic stroke before morphologic changes take place, and therefore provides good means for thrombolytic treatment evaluation of stroke.</p>

ABSTRACT

<p><b>OBJECTIVES</b>To explore the relation of ultrasonic findings to pathological features in cases of chronic viral hepatitis B and C.</p><p><b>METHODS</b>The ultrasonic and pathological findings were analyzed in 130 patients with chronic viral hepatitis B and 106 with chronic viral hepatitis C.</p><p><b>RESULTS</b>In patients with hepatitis B, the ultrasonic echo was thicker and more intensive and uneven cords were found. These findings were closely related to the pathological findings (P less than 0.001). In those with hepatitis C, the ultrasonic echo was slight and dense, which was also closely related to the pathological findings (P less than 0.001). In the patients complicated with fatty liver, the ultrasonic findings were also different (P less than 0.001).</p><p><b>CONCLUSION</b>Ultrasonography is helpful for differential diagnosis of hepatitis B and hepatitis C.</p>
