ABSTRACT

Aim To investigate the effect of TRPC5 gene on the inflammation of cliabetie cardiomyopathy.Methods The biological functions of TRPC5 and the correlation between TRPC5 gene and other genes were analyzed by bioinformatics.Studies were performed in TRPC5 knockout ( TRPC5 ) and C57 mice.Mice were randomly divided into blank control and T2DM model groups, and the model was established by intraperitoneal injection of STZ (n = 10).The myocardial injury was detected by HE and Masson staining.Hie level of serum IL-1(3, IL-2, IL-6, IFN-7 and creatine kinase was examined by ELISA.Gene and protein expressions of IL-1(3, IL-2, IL-6 and TRPC5 were analysed by RT-PCR and Western blot respectively.Results By constructing the PPI network and analyses.the TRPC5 gene was identified to internet with a variety inflammatory genes and involved in immunity.The result of pathologieal section showed less myocardial damage and infiltrated immune cells in TRPC5 mice than in C57BL/6J mice.RT-PCR and serum results showed a lower expression of inflammatory factors in myocardium and serum obtained from TRPC5 model mice than in those obtained from C57BL/6J model mice.Conclusions TRPC5 participates in the development of dilated cardiomyopathy by regulating cardio- myocyte inflammation.

ABSTRACT

Objective:To analyze the changes of bone mineral density (BMD), the characteristics of bone metabolic markers and related factors in patients with spinal cord injury (SCI). Methods:A total of 78 patients with SCI in our hospital from April, 2018 to May, 2020 were selected and divided into groups according to the injury courses. The people receiving physical examination in the same period were selected as control. BMD of proximal femur was measured by dual energy X-ray absorptiometry. Bone metabolic markers were detected. The correlation between BMD and clinical indicators was analyzed. Results:There was no difference in BMD between the patients within three months and the controls (P > 0.05). BMD at trochanter major, intertrochanteric and total hip in patients three to six months post injury was lower than that in the controls (|t| > 2.242, P < 0.05). The BMD at femoral neck, trochanter major, intertrochanteric and total hip in patients during six to twelve months post injury was lower than that in the controls (|t| > 2.026, P < 0.05). BMD at proximal femur in patients with twelve to 24 months post injury was lower than that in the controls (|t| > 2.189, P < 0.05). The decrease of BMD aggravated with the course of injury. There was no difference in BMD between paraplegia and quadriplegia, complete injury and incomplete injury (P > 0.05). Collagen type I C-terminal telopeptide (CTX) and N-terminal propeptide of type l precollagen (PINP) were higher than the reference range in the course of each injury, increased within three months post injury, reached the peak at three to six months post injury, and decreased to a steady state since seven months post injury. Vitamin D deficiency occurred in 87.5% of the patients. BMD at femoral neck and total hip was negatively correlated with the course of injury (|r| > 0.250, P < 0.05), and positively correlated with body mass index (r > 0.255, P < 0.05). Conclusion:The longer the duration of SCI, the more decrease of BMD. The early bone metabolism of patients with SCI is high conversion type. The rate of vitamin D deficiency in patients with SCI is quite high. It is necessary to detect and evaluate the bone metabolic markers combined with BMD at the early stage of SCI.

ABSTRACT

<p><b>OBJECTIVE</b>To observe the clinical effect of Qingzhen Decoction (QZD) on measles.</p><p><b>METHODS</b>Adopting the randomizing digital table, 62 patients with measles were assigned to two groups, 32 in the treated group and 30 in the control group. All patients were treated with routine therapy, but QZD was given to the treated group additionally for 5 days. Changes of clinical symptoms, blood routine and liver function before and after treatment were observed, and the medical cost was calculated.</p><p><b>RESULTS</b>After the 5-day treatment, the normalization rate of irritative cough in the treated and the control group was 88.9% (24/27) and 56.0% (14/25) respectively, that of conjunctival congestion was 90.0% (27/30) and 65.5% (19/29) respectively, showing significant differences between groups (P<0.05). The liver function normalization rate in the two groups was 28.6% (2/7) and 25.0% (2/8), and the average medical cost yen 740.7 and yen 749.3, respectively. The total effective rate in the two groups was 96.9% and 93.3% respectively, showing insignificant difference between groups (P>0.05).</p><p><b>CONCLUSION</b>QZD could actively improve the respiratory symptoms like irritative cough and the inflammatory symptoms of eye like conjunctival congestion in patients with measles.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the feasibility of selective inhibiting VEGF expression using VEGF short hairpin RNA (shRNA) interference, and observe the effects of VEGF gene silencing on NB4 cells growth.</p><p><b>METHODS</b>Three 19 bp reverse repeated motifs targeting exons 3, 4, 5 respectively of VEGF gene were synthesized and cloned into eukaryotic expression plasmid pGenesil-1 containing U6 shRNA promoter and termination signal of RNA polymerase. The recombinant plasmids pGenesil-VR1, pGenesil-VR2, pGenesil-VR3 and pGenesil-con (plasmid containing random DNA fragment) were transfected into NB4 cells respectively through lipofectamine reagent. The alteration of VEGF expression was examined by fluorescent real time RT-PCR and Western blot. The proliferation capacity of leukemia cells was measured by trypan blue exclusion, MTT assay, colony formation assay and cell cycles analysis.</p><p><b>RESULTS</b>Recombinant plasmids containing three shRNAs and random fragment were successfully constructed and transfected into NB4 cells respectively by liposome-mediated gene transfer method. shRNA in pGenesil-VR3 cells knocked down the expression of VEGF mRNA and protein dramatically in a sequence-specific manner when compared with that of pGenesil-VR1, Genesil-VR2 and pGenesil-con. The NB4 cells transfected with pGenesil-VR3 (NB4-VR3) had a more significant decrease in proliferation ability than NB4 and that transfected with pGenesil-con (NB4-con). The colony forming efficiencies of NB4-VR3, NB4-con and NB4 cell were (13.3 +/- 3.8)%, (21.3 +/- 6.4)% and (24.5 +/- 5.2)%, respectively (P < 0.05). Higher G(1) and lower S proportion were found in cell cycle distribution in comparison with the control groups by FCM.</p><p><b>CONCLUSIONS</b>The shRNA can efficiently suppress VEGF expression in NB4 cells. Selective VEGF gene silence can inhibit the malignant proliferation of leukemia cells.</p>
