ABSTRACT

Objective:To observe the effect of Jieyu Anmian prescription of Chinese medicine foot bath on gastrointestinal function in patients with digestive system tumor during perioperative period.Methods:A total of 130 patients with digestive system tumors were chosen from January 2019 to March 2020 for the first time visits to the first hospital of lanzhou university for surgical treatment, 15 cases lost follow-up, and the rest 115 cases of patients were divided into the experimental group (59 cases) and the control group (56 cases) according to random number table method. The control group using conventional care method, the experimental group was treated with Jieyu Anmian washing formula on the basis of routine nursing care for 10 days. During the foot bath, the recovery of gastrointestinal function, the time of the first exhaust and defecation, the length of stay, the cost of stay and the satisfaction of patients were compared between the two groups.Results:The recovery time of bowel sounds, anal exhaust time and first defecation time of the experimental group were all earlier than those of the control group (16.21±11.81) h, (19.64±12.40) h and (41.98 ±19.16) h, respectively, while those of the control group were (27.34±31.47) h, (35.81±18.26) h and (34.47±16.41) h, respectively. The differences were statistically significant ( t value was -2.41, -5.33, 2.17, P< 0.05). The results of the survey of patient satisfaction showed that the experimental group was superior to the control group, with 97.47 ± 4.37 in the experimental group and 94.19 ± 3.29 in the control group, with statistically significant differences ( t value was 3.89, P<0.01). The satisfaction of doctors in the experimental group was 97.41 ± 6.25 and 94.21±5.91 in the control group, with statistically significant difference ( t value was 3.95, P<0.05). The satisfaction of nurses in the experimental group was 97.58 ± 4.53 and 93.85±5.23 in the control group, and the difference was statistically significant ( t value was 4.12, P<0.05). There was no significant difference between the two groups in terms of length of stay and hospitalization expenses ( P>0.05). Conclusions:Jieyuanmian prescription of Chinese medicine foot bath can promote the recovery of gastrointestinal function, promote the exhaust and defecation, and improve the satisfaction of patients and nurses.

ABSTRACT

Objective To investigate the role of endogenous nitric oxide synthase (NOS) inhibitor asymmetric dimethylarginine (ADMA) in the contractile dysfunction of skeletal muscle in diabetic rats and on which the therapeutic effects of L-Arginine.Methods Type 2 diabetic rats were induced by high fat diet and a single intraperitoneal injection of streptozotocin (STZ,30 mg/kg),followed by high fat diet for 8 weeks.Specific twicth tension and specific tetanic tension of soleus (SOL) and extensor digitorum longus (EDL) isolated from control and diabetic rats were detected by electric stimulation to reflect contractile function of skeletal muscle.ADMA content of skeletal muscle was analyzed by enzyme linked immunosorbent assay (ELISA),and activities of dimethylarginie dimethylaminohydrolase (DDAH) and NOS,NO content were measured by colorimetry.The protein expression of ADMA synthetase protein arginine methyl transferase 1 (PRMT1) and ADMA hydrolase DDAH and NOS were determined detected by Western blotting.Oral glucose tolerance test and protein expressions of phosphorylated insulin receptor substrate 1 (p-IRS-1) and protein kinase B (p-Akt) as well as the membrane transportation of glucose transporter 4 (Glut4) were measured to reflect insulin resistance.Results In comparison with control rats,specific twicth tension and tetanic tension of SOL and EDL in diabetic rats were significantly decreased (P < 0.01),indicating the contractile dysfunction.Increased ADMA content (P < 0.05),decreased DDAH and NOS activities as well as NO content (P < 0.01) in comparison with up-regulated protein expression of PRMT1 and down-regulated protein expression of DDAH,endothelial NOS (eNOS) and neuronal NOS (nNOS) (P < 0.05) were observed in the skeletal muscle of diabetic rats compared to control rats,indicating that the pathway of PRMT1/ADMA/DDAH/ NOS/NO was disordered in the skeletal muscle of diabetic rats.Furthermore,the glucose tolerance,both IRS-1 and Akt protein phosphorylation as well as the membrane translocation of Glut4 were decreased (P < 0.05),indicating the insulin resistance in diabetic rats.Treatment with L-Arginine for 8 weeks not only significantly improved the contractile dysfunction but also reversed the disorder of ADMA signaling pathway and insulin resistance in skeletal muscle of diabetic rats compared to untreated diabetic rats.Conclusions The accumulation of endogenous NOS inhibitor ADMA contributes to the contractile dysfunction of skeletal muscle in diabetic rats,the underlying mechanism may be related to insulin resistance.

ABSTRACT

Ojective To investigate the direct effect of exogenous NOS inhibitor Nω-Nitro-L-arginine (L-NNA) on the erectile function of healthy male rats in order to clarify the mechanism of the erectile dysfunction of type 2 diabetic rats.Methods L-NNA (50 mg/kg) was administered by oral gavage to Sprague Dawley (SD) rats for 16 weeks to induce rat model of NOS inhibition.By the end of the experiment,corpora cavernosa was isolated from the rats under anesthetization and fixed in an organ chamber for the examination of relaxation function response to acetylcholine (ACh) to reflect erectile function.Enzymelinked immunosorbent assay (ELISA) was performed to determine the content of cyclic guanosine monophosphate (cGMP) in cavernosal tissue.NOS activity and nitric oxide (NO) content were measured by colorimetric method.Western blotting was employed to detect the protein expression of NOS and phosphodiesterase 5 (PDE5).The activity of the antioxidative enzyme superoxide dismutase (SOD) and content of the lipid peroxidative product malondialdehyde (MDA) were analyzed to reflect oxidative stress.Results In comparison with control group,the relaxation response to acetylcholine of corpus cavernosum was significantly impaired in L-NNA model rats,indicating penile erectile dysfunction.Either decreased contents of NO and cGMP or suppressed activity of NOS were observed in the corpus cavernosum of L-NNA model rats and in accompany with the down-regulation of endothelial NOS (eNOS) and neuronal NOS (nNOS) expression,up-regulation of inducible NOS (iNOS) and PDE5 expression as well as an increase of oxidative stress.Incubation of corpus cavernosum from control rats with L-NNA (1-10 μmol/L) ex vivo for 30 min could also inhibit the relaxation function responses to acetylcholine in a dose-dependent manner.Treatment with L-arginine ex vivo for 45 min could improve the impairments of relaxation function responses to acetylcholine of corpus cavernosum induced by L-NNA in vivo and ex vivo.Conclusions Exogenous NOS inhibitor L-NNA could induce penile erectile dysfunction in healthy male rats and the mechanism may be related to reducing NO content and increasing oxidative stress.

ABSTRACT

OBJECTIVE:To optimize the extraction technology of total lignans from pine needles of Cedrus deodara. METH-ODS:Using ethanol volume fraction,ratio of liquid to solid and extraction time as response factor,the content of total lignans as response value,the response surface methodology test was conducted based on Box-Behnken design to optimize the extraction tech-nology of total lignans from pine needles of C. deodara. RESULTS:The optimal extraction technology was as follows as ethanol volume fraction of 78%,ratio of liquid to solid 25∶1,extraction time 1.75 h. The content of total lignans from pine needles of C. deodara was 99.2 mg/g,which was close to predicted value 100.49 mg/g,with relative deviation of 0.65%. CONCLUSIONS:Op-timized extraction technology of pine needles of C. deodara by response surface methodology is feasible and stable,and has good predictability.
