ABSTRACT

Objective To establish a method for determining five isoquinoline alkaloids (including berberine,jatrorrhizine,jatrorrhizind,coptisine and palmatine) in Erhuang Xiaoyan tablets simultaneously.Methods Determination by reversed phase high performance liquid chromatography (RP-HPLC).Column:YMC-Pack ODS-AM (4.6 mm× 250.0 mm,5 μm);mobile phase:elution with 0.1% (v/v) formic acid aqueous solution-acetonitrile mobile phase gradient;flow rate:0.4 mL/min;injection volume:10 μL;detection wavelength:345 nm;column temperature:27 ℃.Results Five of isoquinoline alkaloids in Erhuang Xiaoyan tablets were separated perfectly.The good linear relationships were obtained in the following ranges,1.526 4-5.342 4μg (r=0.999 9) for berberine,0.403 2-1.411 2 μg (r=0.999 6) for palmatine,0.309 7-1.083 9 μg (r=0.999 8) for coptisine,0.111 2-0.389 2 μg (r=0.999 8) for jatrorrhizine and 0.162 2-0.567 7 μg (r=0.999 6) for epiberberine.The recoveries were 98.69%,98.66%,98.67%,98.67% and 98.67%,respectively.Conclusion The method possesses high feasibility,strong specificity,high accuracy and good reproducibility,which can be used for the quality control of Erhuang Xiaoyan tablets.

ABSTRACT

Objective To investigate the relationship between PCSK9 gene polymorphism with lipid metabolism and CHD risk among population within Tianjin, China. Methods Patients from Tianjin Chest Hospital were selected (n=778) and divided into coronary heart disease (CHD) group (n=502) and control group (n=276). Immediately after admission, tri?glyceride (TG), high density lipoprotein (HDL-C), low density lipoprotein (LDL-C), apolipoprotein A (ApoA 1) and apolipo?protein B (ApoB) were tested and genotype TT,TC and CC were determined. Results In CHD group, LDL-C level is higher in TC genotype individuals than that in TT genotype individuals with statistical significance (P0.05). The total population allele frequencies were T 96.1%(1 495/1 556), C 3.9%(61/1 556);genotype frequencies were TT 92.2%(717/778), TC 7.8%(61/778). Multivariate Logistic regression analysis revealed that ApoA1 and HDL-C were inversely associated with coronary heart disease, therefore are protective factors for coronary heart disease. Conclusion The study found that TC genotype is correlated with serum LDL-C increase in group CHD, and the incidence of allele C showed no obvious connection with CHD.

ABSTRACT

Objective To investigate expression and role of protease catenin-1(PN-1), thrombin(thrombin), protease activated receptor-1(PAR-1) in rats brain edema tissue after intracerebral hemorrhage(ICH).Methods Adult male SD rats 80 were randomly divided into sham group,ICH group after number, 40 in each group, ICH group autologous arterial method of making a rat model of experimental ICH in the right caudate nucleus unit injection.The degree of neurological dysfunction between 2 groups was evaluated at 12 h,24 h and 120 h after post-operation.Observed the morphology of brain cells by HE staining.Changes of PN-1,thrombin, PAR-1 index in rat brain tissue at 3,6,10,12,24,48 and 120 h were detected by Western blot.Results Neurological dysfunction score ICH rats after modeling 12,48,120 h were significantly lower than the sham group(P<0.05);ICH in rats after modeling 3,6,10,12,24,48 and 120 h of brain tissue PN-1, thrombin, PAR-1 compared with sham-operated group were significantly increased (P<0.05); ICH rat brain tissue PN-1, thrombin, PAR-1 appeared in 12 h after modeling shwed a gradual downward trend (P<0.05). Conclusion ICH hematoma surrounding brain tissue in rats PN-1 has the effect of inhibiting thrombin and PAR-1 overexpression, cause nerve damage.
