ABSTRACT

Objective The correlation between blood system damage and splenomegaly in brucellosis patients and their effects on liver function were analyzed.Methods Data were retrospectively collected in patients with blood system damage diagnosed of brucellosis from 2005 to 2016 at the Department of Infectious Disease of Harbin Medical University First Affiliated Hospital.At the same time,test results of the selected patients liver function,ferritin,D-dimer and abdominal ultrasonography were collected.According to splenomegaly or not,patients were divided into splenomegaly and no-splenomegaly,and any difference in hematopoietic damage caused by splenomegaly or not was analyzed.Results Of the 210 patients (101 cases of splenomegaly,109 cases without splenomegaly),170 were male (80.95%),40 were women (19.05%),and age was (39.65 ± 10.79) years.The patients with abnormal blood system were 103 cases (49.05%);in splenomegaly group there were 58 cases of blood system damage;in no-splenomegaly group there were 45 cases of blood system damage,and there was a statistically significant difference between the two groups (x2 =5.465,P < 0.05);151 cases of brucellosis had elevated aminotransferase (71.90%),but transaminase elevated or not in different intervals of age groups was not statistically different between liver function and age (x2 =10.192,P > 0.05).Ferritin increased in 26 cases (12.38%).D-dimer increased in 22 cases (10.48%).There were 93 patients with splenomegaly and transaminase elevation,and 58 patients with non-splenomegaly and transaminase elevation.There were significant differences between the two groups (x2 =39.204,P < 0.05).Conclusion Brucellosis can cause blood system damage,probably caused by splenomegaly;and patients with brucellosis are often accompanied by elevated transaminases.

ABSTRACT

Objective To evaluate the appearances of ultrasound in bacterial endophthalmitis. Methods Thirty patients with endo-ophthalmitis (in 30 eyes) were examined by ultrasound and pathology, and their appearances were compared. Results All appearance in 30 patients (in 30 eyes) with endo-phthalmitis showed the turbidly of punctiform, fine particle, mass in ultrasound and bacterium, leukocytic infihrate,the patch of tissue in pathology. And two ways was correspond to diagnosis endophthalmitis. Conclusions Ultrasound can be used as an effective method to diagnose endophthalmitis.

ABSTRACT

Objective To explore the regulatory effects of thioredoxin on chemosensitivity of gastric cancer cells. Methods RT-PCR was employed to clone the full-length cDNA of thioredoxin (Trx) from human drug-resistant gastric cancer cell line SGC7901/VCR. The sense and antisense eukaryotic expression vectors corresponding Trx were constructed and transduced into gastric cancer cells SGC7901 and SGC7901/VCR by lipofectamine, respectively. MTT assay was used to evaluate the sensitivity of gene-transfected gastric cancer cells and control cells to anti-cancer drugs in vitro. Results The results indicated that SGC7901 cells transfected with Trx sense vector showed significantly decreased sensitivity to vincristine, cisplatin and adriamycin, while SGC7901/VCR transfected with Trx antisense vector showed significantly increased sensitivity to these drugs. Conclusion All these data suggested that Trx might participate in the formation of multidrug resistance of gastric cancer cells and alteration of Trx could regulate chemosensitivity of gastric cancer cells.

ABSTRACT

The aim of this study was to construct antisense of ZNRD1 encoding gene, and to transfect it into SGC7901/VCR cells, to seek measures to overcome multidrug resistance of gastric cancer cells. ZNRD1 cDNA amplified by PCR was confirmed by DNA sequencing, and then inserted into the multiple cloning site of the expressing vector pcDNA3 1 + with molecular cloning technique. The recombinant vector was identified by endonuclease digestion. Antisense recombinant vector was transfected into SGC7901/VCR cells using lipofectamine. Flow cytometric analysis was used to detect adriamycin (ADM) accumulation in SGC7901/VCR cells. The results showed that a fragment was obtained by PCR, and its sequence was consistent with ZNRD1 cDNA reported in the literature. After antisense recombinant vector having been transfected into SGC7901/VCR cell, ADM concentration in such cells was increased. The above results indicated that the antisense vector ZNRD1 could enhance the intracellular accumulation of ADM in SGC7901/VCR cells, which might be of potential treatment value.
