ABSTRACT

Objective:Our study investigates the expression of Motor neuron and pancreatic homeobox 1 ( MNX1) in breast cancer tissues, and the effects of MNX1 on the proliferation, cell cycle, apoptosis, migration and invasion of MDA-MB-231 cells were studied by constructing the MNX1 knockdown MDA-MB-231 cell line. Methods:A retrospective study was conducted on 73 breast cancer tissues and adjacent normal tissues from 73 breast cancer patients in the First Affiliated Hospital of Guangxi Medical University. qRT-PCR was used to detect the relative expression of MNX1 in tissues and cells. Western blot detects the protein level of MNX1 in the tissue. We designed the shRNA MNX1 and constructed the MNX1 viral vector with low expression. The viral vector was further used to infect triple-negative breast cancer cells. The MNX1 with the best silencing effect was designed to silence the breast cancer cell line MDA-MB-231 as the silence group ( shMNX1), and the negative control group (Control) of lentivirus infection was designed to carry out follow-up cell function tests.CCK-8 method was used to detect cell proliferation ability. Transwell method was used to detect cell migration and invasion ability. Use flow cytometry to detect apoptosis. Each experiments at least 3 times independent experiments and measurement data with normal distribution were represented as the ( Mean± SD). The t-test was used for the comparison of two sample means. Results:The qRT-PCR showed that the expression level of MNX1 mRNA in breast cancer tissue was higher than that in adjacent tissues ( P<0.05). WB showed that the expression level of MNX1 protein in breast cancer tissue was significantly higher than that in adjacent tissues ( P<0.01) . CCK-8 experiment results showed that the OD (450 nm) of breast cancer cells in the silence group at 24, 48 and 72 h was lower than that of the negative control group ( P<0.05). The results showed that silencing the MNX1 gene can inhibit the proliferation of breast cancer cells MDA-MB-231. The results of Transwell migration experiment showed that the number of cells passing through the Transwell chamber in the silent group and the negative control group were 217.00±33.23 and 490.00±45.56, respectively, and the difference was significant ( P<0.05). The results of Transwell invasion experiment showed that the number of cells in the silent group and the negative control group passing through the Transwell chamber were (91.00±12.79)and (419.00±49.37), respectively, and the difference was statistically significant ( P<0.05). Our results show that silencing the MNX1 gene can inhibit the migration and invasion ability of breast cancer cell line MDA-MB-231. The results showed that the apoptosis rate of breast cancer cells in the silent group was (3.81±0.41)%, and the negative control group was (2.13±0.16)%. The apoptosis rate of breast cancer cells in the silent group was higher than that in the negative control group, and the result was statistically significant ( P<0.05). MNX1 promotes the apoptosis of triple-negative breast cancer cells. Conclusion:MNX1 is a new breast cancer gene, silencing the expression of MNX1 gene inhibits the proliferation, migration and invasion of breast cancer cells and promotes cell apoptosis, which provides a new regulatory mechanism and therapeutic target for breast cancer.

ABSTRACT

Objective:To investigate the attitudes and demands of medical students on medical education rituals in the context of the new era, to find out the existing problems, and to provide reference to the innovation and improvement of the rituals.Methods:An online questionnaire was conducted among medical students. SPSS 25.0 was used for descriptive statistical analysis and rank sum test of the data. Qualitative research methods such as in-depth interviews and panel discussions were used to comprehensively analyze the qualitative data.Results:"Post-00s" medical students, who are known as the aborigines of the Internet, are also looking forward to the return of traditional medical education rituals. Compared with junior students, senior students had lower awareness of the importance of rituals and lower willingness to participate in rituals, and the degree of professional identity arousal by the rituals was also lower ( P<0.05). Certain medical students indicated that they had never participated in medical education rituals, and some indicated that their specialties were not valued in the rituals. Conclusion:Medical education in the new era should revive traditional rituals and be adjusted to the changes in the background of the times and the characteristics of the student groups. The current ritual education fails to meet the needs of medical students at different stages, and its systematicness and continuity should be strengthened. The coverage of medical education rituals for medical students is incomprehensive and uneven, and it is necessary to further expand the coverage and appropriateness of rituals in medical-related specialties and fully consider the characteristics of each specialty in medical students.

ABSTRACT

To deeply analyze the existing problems in the medical humanities MOOCs, this study adopts qualitative research methods such as participatory observation, interview and special panel discussion from two different perspectives of teaching and learning. On one side, the study follows the course building process of two medical humanities MOOCs in a medical university in South China, and conducts interviews and panel discussions with teaching teams, MOOC producers, and medical education experts in order to understand the problems, challenges and related countermeasures and thoughts in the construction process. On the other side, students' learning feelings, gains and opinions on medical humanities MOOCs in the existing national excellent online open courses have been investigated. Findings revealed that there obvious differences between the attitudes of teaching and learning, which reflects the challenges of the medical humanities MOOCs at the micro level, including difficulties in realizing autonomous learning under effective monitoring and achieving important teaching achievements, excessive workload of teachers, etc.. Furthermore, the differences between teaching and learning have shed a light on medical humanistic education concept, teaching mode and teacher cultivation. Finally, the macro development trend of online and offline coordination and complementarity of medical humanities MOOCs, and the coexistence of mass and personalized curriculum is predicted.

ABSTRACT

Objective@#Explore the feasibility of POL intervention based on life skills in young students at high-risk of AIDS, and to provide reference for POL intervention.@*Methods@#Nine potential POLs were recruited from young students at high risk of AIDS in Xi’an, and received life-skill-based interventions from May to October in 2017. A unified approach was adopted before the intervention. The effect of intervention was evaluated at 1 month and 3 months after the intervention.@*Results@#The difference in POLs peer status and psychosocial ability scores was not significant after the intervention. The scores of persistence efficacy dimension of condom use efficacy before, 1-month and 3-month after the intervention were (10.56±1.88)(11.11±2.21)(12.89±2.09)(F=6.84, P<0.05) respectively. No significant changes were found in AIDS-related knowledge and behavior before and after the intervention. POL has increased from 108 students before intervention to 216 publicity coverage after three months intervention, however, the difference was not statistically significant. Feasibility analysis of the POL intervention showed that 8 of the POLs considered to be "very useful".@*Conclusion@#Life skill-based POL intervention for young students at high-risk of AIDS has a wide coverage and reasonable acceptance, as well as condom use adherence. The long-term effectiveness still needs to be verified.
