ABSTRACT

Objective To investigate the molecular mechanism of solanine-induced apoptosis of prostate cancer cells Du145 and LNCaP.Methods The effects of solanine on the viability of Du145 and LNCaP cells were evaluated by MTT assay.The generation of intracellular reactive oxygen species (ROS) and solanine-induced apoptosis were measured by flow cytometry.The protein levels of p38 and p-p38 expressions were examined by Western blot.Results Solanine significantly inhibited the viability of Du145 and LNCaP cells in a dose-dependent manner (P<0.01).The inhibition of solanine on cell viability was suppressed by the ROS scavenger NAC.ROS generation,apoptosis and phosphorylation of p38 were induced by treatment with solanine at 40 μmol/L for 24 h.The expression of p38 and solanine-induced apoptosis were suppressed by NAC and SB203580.Conclusion Solanine induces the apoptosis of human prostate cancer cell via the RO.S-p38 signaling pathway.

ABSTRACT

Objective To compare the efficacy of ureteroscopic lithotripsy(URL)combined with occlusion device and the supine and lithotomy position mini-invasive percutaneous nephrolithotomy(mPCNL)in the treatment of upper incarcerated ureteral stones. No difference could be found in age,sex and size of stones between the two groups. Method From Jan. 2014 to Dec. 2016 in our hospital,all cases of upper incarcerated ureteral stones were diviede into two groups:52 in ureteroscopic lithotripsy combined with occlusion device group and 45 in mini-inva-sive percutaneous nephrolithotomy group. Result The hospitalization and operation time in URL group were(5.5 ± 1.4)days and(53.3 ± 12.4)mins,which were significantly shorter than that in mPCNL group with(9.1 ± 3.2)days and(78.2 ± 14.1)mins,(P 0.05),stone clearance rate(100% vs 91.8,P > 0.05)and complication rate (4.4%vs 3.9%,P>0.05). Conclusion URL combined with occlusion device can obtain satifactory results as well as the supine and lithotomy position mPCNL in the treatment of upper incarcerated ureteral stones.

ABSTRACT

Objective To approach the effect of cathepsin K(CK) on the onset of osteoporosis,and to study the influence of Bushen Tongluo Prescription(BTP,with the actions of invigorating the kidney and dredging the meridians) on osteoclast CK in ovariectomized rats.Methods Thirty female SD rats were divided into three groups randomly: sham-operation group,model group and BTP(12.46g.kg-1.d-1) group.The bilateral ovaries in rats of the model group and BTP group were removed.Four weeks later,BTP group were given gastric gavage of BTP.On the 6th and 12th weeks,we detected the general bone density of rats,examined CK expressionin osteoclasts with Western blot and reverse transcription-polymerase chain reaction(RT-PCR) methods.ResultsThe in-vitro cultured cells had the phenotype feature of mature osteoclasts.The bone mineral density of ovariectomized rats was negatively correlated with CK expression in time-effect manner.BTP down-regulated the CK protein and CK mRNA expression(P
