ABSTRACT

Objective To explore the three ultrasonic technologies of two -dimensional ultrasound(2D -US),ultrasonic elastography(UE) and contrast -enhanced ultrasound(CEUS) for the measurement error in breast cancer and the correlation with the expression of ER,PR,VEGF.Methods 50 patients with breast cancer were meas-ured by 2D -US,UE,CEUS preoperatively, and the pathological specimen were measured postoperatively. Then used the immunohistochemistry to detect the expression of ER,PR,VEGF in tumor, and analyzed the correlation with the measurement errors. Results The results of differences between 2D -US,UE,CEUS and pathology were respectively as follows:( -0.59 ±-0.34)cm,( -0.20 ±-0.14)cm,( -0.40 ±-0.31)cm,and the differences were statistically significant(F =20.497,P <0.001).The positive expression rate of ER and PR was high if the difference between UE and 2D -US was less than or equal to 0.44cm.And the positive expression rate of VEGF was low if the difference between CEUS and 2D -US was less than or equal to 0.19cm.Three ultrasonic technologies in the measurement of breast cancer were different, the trend of difference between UE and 2D -US was smaller if the ER and PR were positively expression, and the trend of difference between CEUS and 2D -US was bigger if the VEGF was positively expression. Conclusion There is correlation between different immunohistochemical expression of breast cancer with measurement error in three different ultrasonic imaging technologies. The results suggest that the molecular pathology difference of breast cancer can impact on ultrasonic imaging, which contributes to know the reason and regulation of measurement error in different ultrasonic imaging technology.

ABSTRACT

Objective To investigate the effect of mixed-skin grafting with autologous microskin and allogenetic acellular dermal matrix microskin on wound healing in rats, and to make a further study on the related mechanism. Methods Wistar rats were served as a allogenetic acellular dermal matrix donor rats, and SD rats as acceptors with mould of full thickness skin defects on their back. The ninety SD rats were divided into 5 groups with 18 rats in each group.Group 1 was transplanted with autologous microskin, and group 2 with allogenetic acellular dermal matrix microskin.Groups 3,4 and 5 were grafted with mixed-skin ratio between autologous microskin and allogenetic acellular dermal matrix microskin 1 ∶ 1,1 ∶ 0.5 and 1 ∶ 0.25,repectively.The rate of wound healing was measured, wound samples collected, hematoxylin and eosin stain carried out, fibronectin (FN) and laminin (LN)detected, and intergroup comparison made,respectively,2,3 and 4 weeks after skin grafting. Results The wound healing rates and FN and LN expression of mixed-skin grafting groups were higher than those of the group with autologous microskin grafting. The group of 1 ∶ 0.25 obviously increased (P<0.05 or P<0.01).Conclusions The wound healing rate with mixed-skin grafting is higher than that with autologous microskin grafting. The best effect is achieved when the skin ratio between autologous microskin and allogenetic acellular dermal matrix microskin is 1 ∶ 0.25.It is possibly due to the increase of FN and LN on wound skin surface.

ABSTRACT

BACKGROUND: The preparation and antibacterial activity of metal ionic zirconium phosphates has been systemically investigated now, but the applications are limited owing to the discoloration or the low antibacterial activity. Here we prepared new antibacterial agents of quaternized zirconium phosphates by introducing quaternary ammonium salt bactericidal agent with high-effective, broad-spectrum and low-toxic into sodium zirconium phosphate through an ion-exchange method.OBJECTIVE: To explore the component structure and antibacterial activity of quaternized zirconium phosphates.DESIGN, TIME AND SETTING: An in vitro observational experiment was performed at Research Laboratory of Department of Chemistry, Jinan University from June to August 2009.MATERIALS: Quaternized zirconium phosphates were prepared by introducing dodecyl dimethyl benzyl ammonium chloride into sodium zirconium phosphate through an ion-exchange method.METHODS: The mol ratios of quaternary ammonium cations to cation exchange capacity of sodium zirconium phosphate in reaction solutions were 0.25: 1,0.5: 1, 1.0: 1, and 1.5 : 1, respectively, and four kinds of quaternized zirconium phosphates containing different contents of quaternary ammonium cations (QZrP-1, QZrP-2, QZrP-3, QZrP-4) were prepared through an ion-exchange method.MAIN OUTCOME MEASURES: The component structure and heat resistance of samples were measured by using an IR spectrometer, an elemental analyzer and a thermal analyzer, respectively. The minimum inhibitory concentrations (MICs) and minimal bactericidal concentrations (MICs) of the samples against Escherichia coli (E. co/i) and Staphylococci aureus (S. aureus) were estimated by a tube broth method.RESULTS: Quaternized zirconium phosphates were prepared, and the quaternary ammonium cation content increased with increasing the concentration of quaternary ammonium cations in reaction solution. The mass fraction of quaternary ammonium cations of QZrP-1, QZrP-2, QZrP-3, and QZrP-4 was 3.70%, 5.00%, 6.96%, and 10.01%, respectively. The onset temperatures of the decomposition for quaternary ammonium cations in quaternized zirconium phosphates were all higher than 345 °C, and they were preferable thermal stability. The antibacterial activity was higher when the quaternary ammonium cation content of quaternized zirconium phosphates increased. For quaternized zirconium phosphates QZrP-3 containing 6.96% mass fraction of quaternary ammonium cations, showed excellent antibacterial activity against E. coli and S. aureus.CONCLUSION: Quaternized zirconium phosphates QZrP-3 containing 6.96% mass fraction of quaternary ammonium cations, exhibited excellent thermal stability and antibacterial activity.

ABSTRACT

BACKGROUND: Transforming growth factor-β(TGF-β) is an essential factor for pathological scar formation. Smad protein group is the signal protein of lower reaches of TGF-β receptor. Asiaticoside can inhibit proliferation of fibroblasts and synthesis of collagen to reduce TGF-β expression in the scar.OBJECTIVE: To study the mechanism of asiaticoside on proliferation of scar fibroblasts and phosphorylated Smad2 and Smad7.DESIGN: Controlled study with observation, in which cell was taken as the object.SETTING: Department of burn and plastic surgery of a hospital affiliated to a university.PARTICIPANTS: The experiment was performed in Surgical Laboratory in Sun Yat-sen University from April 2002 to March 2003. The specimens were selected from 6 inpatients receiving plastic operation due to hyperplasic scar including 3 male and 3 female cases aged varied from 1 to 35 years. The hyperplasic scar fibroblasts were obtained generated from original culture in laboratory of surgical department.INTERVENTIONS: In the research, the experiment group and the control group were divided. In the experiment group, asiaticoside was applied on fibroblasts; in the control group, asiaticoside was not prescribed. The changes of every index were observed before and after medication.MAIN OUTCOME MEASURES: ① Effect of asiaticoside on expression of phosphorylated Smad2 and Smad7; ② Effect of asiaticoside on cell cycle and apoptosis.RESULTS: Asiaticoside inhibited scar fibroblasts entering M phrase from S phrase and reduced the content of phosphorylated Smad2 in fibroblasts, which did not present significant difference in two groups ( t = 1.53, P =0.08).The content of Smad7 in the cells was (50. 80 ± 22.40)% in the experiment group and (32.18 ± 17.84)% in the control group, which indicated significant difference ( t = 2. 17, P = 0. 024).CONCLUSION: Asiaticoside inhibits scar formation by Smad passage.

ABSTRACT

OBJECTIVETo investigate the bacterial epidemiology in our department in recent years, so as to provide assistance to the clinical management of burn patients.

METHODSA retrospective analysis was carried out with 345 bacterial cultures from burn wound and drug-sensitivity results in 784 burn patients during 1993 to 1999 in our department.

RESULTS(1) Among all the bacteria, gram negative (G(-)) bacilli accounted for 56.8%, while gram positive (G(+)) cocci and fungi in 3.8%. (2) Among all the G(+) cocci, 65.4% were Staphylococcus aureus, in which MRSA was identified in 53.9% during 1993 - 1999 and in 64.3% during 1998 - 1999. Pseudomonus aeruginosa accounted for 37.2% of all G(-) bacilli. (3) The 3rd generation of cephalosporins shew excellent anti-bacterial capabilities, but the bacterial resistance to them increased significantly. (4) MRSA was very sensitive to both vancomycin and norvancomycin with no report of antibiotic resistance to them.

CONCLUSIONG(-) bacilli were still predominant bacteria in our burn department when compared to G(+) cocci. The 3rd generation cephalosporins are the routine antibiotics for the present. But resistant bacteria are on the increase. There are also more and more MRSAs isolated from burn wounds. For this, vancomycin and norvancomycin should be preferably used.

ABSTRACT

@#ObjectiveTo explore the role of vascular endothelial growth factor(VEGF) in the growth and development of hypertrophic scar.MethodsThe burn wound samples of various stages were selected from transition of wound granulation tissue to scar and in long-persisting post-burn hypertrophic scar, and the concentrations of VEGF protein were detected using enzyme-linked immunosorbent essay (ELISA) method. ResultsThe tissue homogenate concentration of VEGF protein increases gradually from the wound granulation tissue to hypertrophic scar before it achieves summit concentration during 4 to 6 month. The concentration of VEGF degreases gradually after the maturation of hypertrophic scar. The high concentration of VEGF is synonymous with the large amount of capillary of the immature scar.ConclusionsThe abnormal expression of VEGF is related to the growth and development of hypertrophic scar and induces excessive and uncontrollable angiogenesis.
