ABSTRACT

OBJECTIVE To prepare Xiongzhi shigao decoction soluble microneedles, characterize it and investigate its transdermal properties in vitro. METHODS Two-step centrifugal method was used to prepare Xiongzhi shigao decoction soluble microneedles. The formability and mechanical property of the microneedles were evaluated from aspects of stroma fluidity, microneedle formability, needle hardness, etc. The appearance, mechanical strength, dissolution performance, skin barrier recovery performance and drug loading of the prepared microneedles were characterized by using active components of the soluble microneedle (chlorogenic acid, ferulic acid, notopterol, imperatorin, ligustilide, isoimperatorin) as indicators. The in vitro transdermal performance was investigated by Franz diffusion cell. RESULTS The soluble microneedle tips of Xiongzhi shigao decoction prepared in this study were conical, evenly distributed and of the same thickness, with good mechanical properties; the tip of the needle could be almost completely dissolved after being penetrated into the skin of rats for 2 hours, and the skin barrier recovery performance was good; the drug loading of chlorogenic acid, ferulic acid, notopterol, imperatorin, ligustilide and isoimperatorin were (87.04±1.12), (67.69±1.23), (20.65±0.17), (35.00±0.11), (153.83±0.21) and (23.52±0.50) μg per patch respectively. The results of in vitro transdermal study showed that cumulative release rates of 6 active components in this microneedle after 72 hours were 36.94%, 56.72%, 19.36%, 57.98%, 11.06% and 35.19%, respectively. CONCLUSIONS Xiongzhi shigao decoction soluble microneedles are prepared successfully in this study and have good formability, mechanical properties and pliable backing, which can significantly promote the transdermal drug delivery.

ABSTRACT

OBJECTIVE To evaluate the qualit y of Xihuang pills ,and to screen the differential markers affecting its quality . METHODS Using muskone as internal reference ,the content of α-pinene and other 4 components were determined by quantitative analysis of multi -components by single marker (QAMS),and compared with the results of external standard method . The fingerprints of 13 batches of Xihuang pills were established by gas chromatography (GC)method. Cluster analysis (CA)and orthogonal partial least squares discriminant analysis (OPLS-DA) were performed by SPSS 25.0 software and SIMCA 14.1 software. The variable importance projection (VIP)value greater than 1 was used as the standard to screen differential markers affecting the quality of the samples . RESULTS The contents of α-pinene,octyl acetate and β-elemene measured by QAMS were 0- 0.628 4,0.378 0-2.679 4 and 0.320 9-0.815 4 mg/g,respectively. The contents of α-pinene,octyl acetate ,β-elemene and musk ketone measured by external standard method were 0.001 5-0.627 1,0.378 0-2.594 7,0.329 2-0.837 0 and 0.385 7-0.806 0 mg/g, respectively. The relative error of the content determination results of the two methods was less than 4%. There were 26 common peaks in 13 batches of Xihuang pills ,and 3 common peaks ,such as octyl acetate ,β-elemene and musk ketone ,were identified ; their similarities were 0.912-0.946. 13 batches of samples could be divided into two categories (S1-S2,S6-S10,S13 were clustered into one category and S 3-S5,S11-S12 were clustered into one category ). VIP values of peak 7,11,10,17 and 16 were all greater than 1. CONCLUSIONS The content of 4 components such as α-pinene in Xihuang pills combined with GC fingerprint and chemical pattern recognition analysis can be used to evaluate the quality of Xihuang pills . The components corresponding to 5 common peaks such as peak 7 may be differential markers affecting the quality of the samples .

ABSTRACT

Liposomes have made remarkable achievements as drug delivery vehicles in the clinic. Liposomal products mostly benefited from remote drug loading techniques that succeeded in amphipathic and/or ionizable drugs, but seemed impracticable for nonionizable and poorly water-soluble therapeutic agents, thereby impeding extensive promising drugs to hitchhike liposomal vehicles for disease therapy. In this study, a series of weak acid drug derivatives were designed by a simplistic one step synthesis, which could be remotely loaded into liposomes by pH gradient method. Cabazitaxel (CTX) weak acid derivatives were selected to evaluate regarding its safety profiles, pharmacodynamics, and pharmacokinetics. CTX weak acid derivative liposomes were superior to Jevtana® in terms of safety profiles, including systemic toxicity, hematological toxicity, and potential central nerve toxicity. Specifically, it was demonstrated that liposomes had capacity to weaken potential toxicity of CTX on cortex and hippocampus neurons. Significant advantages of CTX weak acid derivative-loaded liposomes were achieved in prostate cancer and metastatic cancer therapy resulting from higher safety and elevated tolerated doses.

ABSTRACT

Objective To observe the expression and transcription of MART-1 in human uveal melanoma cell lines 92-1,92-2,Ocm3,Me1285,as well as the possible effect ofmethylation on its expression.Methods The cell lines 92-1,92-2,Ocm3 and Me1285 were cultured routinely and tested for MART-1 expression at protein and mRNA level by FACS analysis,Western blot and RT-PCR respectively.Methylation status of the MART-1 promoter region in all the cell lines were checked by Southern blots of DNA digested with methylation sensitive restriction enzymes.Results As observed in FACS analysis and Western blot,92-1,92-2 and Ocm3 were MART-1 positive cell lines while Me1285 was negative cell line.Consistent with protein analysis,92-1 and Ocm3 cell lines showed MART-1 specific PCR products and there was no product in Me1285 cell line in RT-PCR.The MART-1 positive cell lines,92-1,92-2,and Ocm3 show methylation at the MspⅠ/Hpa Ⅱ site,and the Nru Ⅰ sites of all positive cell lines are not methylated.The MART-1 negative cell line Me1285 shows hyperrnethylation at the Nru Ⅰ site and the Msp Ⅰ/Hpa Ⅱ site is not methylated.Conclusions MART-1 could be expressed in human uveal melanoma cell lines 92-1,92-2 and Ocm3.The change of methylation status of MART-1 promoter may correlate with the transcription of MART-1.

ABSTRACT

Objective To observe the preventive effects of puerarinon development of deep vein thrombosis (DVT) in elderly women with osteoporosis after hip replacement. Methods 100 elderly women with osteoporosis who scheduled for femoral head arthrolastybetween January 2012 and January 2014 in Cangzhou Central Hospital were selected and then randomly divided into a study group (n=50) and a control group (n=50). The control group received routine anti?inflammatory and anticoagulant therapy ,while the study group received intravenous drip of puerarinof 400 mg mixed with 5%glucose liquid 500 ml daily for 15 days as a treatment course. After the treatment, DVT incidence rate ,change of early diagnostic indexes and hemorheology were compared with their baselines. Results DVT incidence rate was significantly lowerin the study group than in the control group (8.0%vs. 24.0%, P<0.05). Ascompare with the control group,thromboxane A2/prostacyclin,plasma homocysteine and hemorheology?were significantly reduced (P < 0.05). Conclusions Puerarin has a preventive effect for deep vein thrombosis in elderly women with osteoporosis after hip replacement.

ABSTRACT

Pediatric patients usually require intravenous infusion maintenance fluid. Currently, maintenance fluid with 1/5 tension, 1/2 tension, and isotonic contraction were commonly used and lfuid with 1/5 tension and even with no tension is still commonly used. In recent years, a large number of high quality randomized controlled trials and meta analyses showed that compared with the traditional low tension maintenance lfuid, 1/2 tension or isotonic maintenance lfuid can effectively reduce the incidence of acute hyponatremia and the adverse reactions such as hypernatremia while hypernatremia were not caused. A number of national guidelines also recommend the use of 1/2 tension or isotonic contraction as a standard for maintaining liquid. In this paper, the present status and recent progress in the use of intravenous maintenance lfuid in pediatric patients were reviewed.

Fragmented QRS (fQRS) complex drew a large number of research interests recent years.It said to be prevalent in patients with coronary artery disease and myocardial infarction.Researchers also found fQRS was a sign of myocardial scarring,myocardial focal necrosis and substandard perfusion in patients suffered acute myocardial infarction,and the relationship of fQRS with malignant arrhythmia and cardiovascular events in these patients was also investigated lately,patients with fQRS were said to have an unfavorable prognosis.We summarized the definition,pathogenetic mechanism of fQRS and the relationship with ventricular arrhythmia in this article to figure out the cardiovascular event's predictive value of fQRS in patients suffered with acute myocardial infarction.

ABSTRACT

Objective To explore the influence factors of poor myocardial perfusion in patients with ST-segment elevation myocardial infarction( STEMI) after primary percutaneous coronary intervention(PCI). Methods One hundred and forty-three patients with first STEMI who were on admission from April 2010 to May 2014 and underwent primary PCI within 12 hours were enrolled as our subjects. According to the sum-ST-segment resolution(sumSTR)and TIMI myocardial perfusion grade(TMP)after primary PCI,all patients were divided into well myocardial perfusion group( sumSTR ≥ 50% or TMP 2 - 3 grade)and poor myocardial perfusion group(sumSTR < 50% and TMP 0 - 1 grade). The influence factors between two groups were collected and analyzed,including sex,age,pain to balloon time,blood pressure on admission,left ventricular ejection fraction,leucocyte count,neutrophil ratio(NR),high-sensitivity C-reactive protein(hs-CRP),blood lipid,and the history of hypertension,diabetes mellitus. Results The leucocytes count,NR,hs-CRP in patients of poor myocardial perfusion group were(11. 60 ± 3. 57)× 109 / L,0. 84 ± 0. 06 and 9. 80 ± 11. 37 mg/ L,higher than those in well myocardial perfusion group((9. 51 ± 2. 59)× 109 / L,0. 77 ± 0. 11 and(3. 83 ± 5. 58)mg/ L),and the differences were significant(t = 3. 497,P = 0. 001;t = 3. 390,P = 0. 001;t = 3. 973,P < 0. 001). Multiple linear regression analysis showed that neutrophil ratio was independent risk factor of sumSTR in STEMI patient after primary PCI(P = 0. 000). Conclusion The increase of leucocyte count,NR and hs-CRP are related to the poor myocardial perfusion after primary PCI. The increase of neutrophil ratio is an independent risk factor of poor myocardial perfusion.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the detection rate and correlates of behavioral problems among eco-migrant children in Hui and Han ethnicities.</p><p><b>METHODS</b>Using multistage randomized sampling method, 2 653 eco-migrant teenagers, 3 174 indigenous and 2 334 homeland peers were selected. Their parents were asked to finish the Achenbach's Child Behavior Checklist (CBCL) and a self-developed correlates questionnaire.</p><p><b>RESULTS</b>765 eco-migrant children with behavioral problems were detected, with detection rate as 28.8%, higher than that of homeland group (24.2%) and native group (19.3%) (χ(2) = 73.547, P < 0.012 5). Ecological migration mainly influenced factors as bad contacts, depression, obsessive-compulsive, discipline, attack for boys aged 6-11, bad contacts for boys aged 12-16 and extraversion behavioral problems for girls aged 12-16, in Hui. These factors both associated to eco-migrant children's behavioral problems in Hui and Han:cesarean section (OR = 1.863, 95% CI:1.144-3.035, for Han) and (OR = 2.979, 95% CI:2.067-4.293, for Hui), physical defects (OR = 1.730, 95%CI:1.087-2.751, for Han) and (OR = 2.552, 95%CI:1.649-3.950, for Hui), arable as a main income way(OR = 1.272, 95%CI:1.066-1.518, for Han) and (OR = 1.212, 95%CI: 1.033-1.422, for Hui), parents' education level above middle school (OR = 0.709, 95% CI: 0.539-0.932, for Han) and (OR = 0.698, 95%CI: 0.542-0.898, for Hui), parents' frequently in good mood (OR = 0.710, 95%CI:0.613-0.821, for Han) and (OR = 0.826, 95%CI:0.718-0.949, for Hui). However, major diseases in the process of growth (OR = 1.794, 95% CI:1.163-2.767), and parents with chronic illness or disability (OR = 1.463, 95% CI:1.061-2.016) only associated with that in Han;single-parent or remarried families(OR = 1.583, 95% CI:1.078-2.325), parents often drinking (OR = 1.557, 95%CI:1.019- 2.379), the time of parents' work longer than half a year (OR = 1.494, 95% CI:1.197-1.864), parents' more content to life now(OR = 0.813, 95% CI:0.700-0.945), and children in older age (OR = 0.939, 95%CI:0.889-0.991) only related to that in Hui.</p><p><b>CONCLUSION</b>The eco-migrant children showed a low state in mental health, especially among Hui peers, and the correlated factors were different in Han and Hui, which was supposed to get great importance when taking effective intervention.</p>

ABSTRACT

Non-Camellia tea is a part of the colorful Chinese tea culture, and is also widely used as beverage and medicine in folk for disease prevention and treatment. In this study, 37 samples were collected, including 33 kinds of non-Camellia teas and 4 kinds of teas (Camellia). Traditional functions of non-Camellia teas were investigated. Furthermore, non-Camellia teas of original plants were characterized and identified by molecular methods. Four candidate regions (rbcL, matK, ITS2, psbA-trnH) were amplified by polymerase chain reaction. In addition, DNA barcodes were used for the first time to discriminate the commercial non-Camellia tea and their adulterants, and to evaluate their safety. This study showed that BLASTN and the relevant phylogenetic tree are efficient tools for identification of the commercial non-Camellia tea and their adulterants. However, some sequences from original plants have not been found and there is a limitation of sequence number of original plants in GenBank. Submitting more original plant sequences to the GenBank will be helpful for evaluating the safety of non-Camellia teas.

ABSTRACT

This study was aimed to optimize the best extraction technology of traditional Chinese medicine (TCM) Biminkang and establish the HPLC-ELSD method for determination of Astragaloside Ⅳ content. This test used heat-ing reflux which preferred ethanol as solvent extraction, extraction rate as an index to extract. By the single factor ex-periment, three factors which affect extraction rate greater were selected from the solvent concentration, extraction time, liquid ratio and extraction times. And then L9(34) orthogonal test was used to design the extraction technology of compound preparation Biminkang. HPLC-ELSD was performed on Diamonsil C18 column (250 mm í 4.6 mm, 5 μm) with H2O(A)-acetonitrile(B) (0~45 min: 22%B, 45~60 min: 22%~32%B) as mobile phase, flow rate was at 1.0 mL·min-1. The temperature of drift tube was 100℃ and the flow rate of N2 was 2.5 L·min-1. The column temperature was 30℃. The results showed that the best extraction technology of compound preparation Biminkang was liquid-solid ra-tio of 8 mL·g-1, ethanol concentration of 70%, 1.5 h for each extraction time, and extracted for three times. The re-sults showed that the presence of ethanol concentration and extraction times affected significantly. The ultimately de-termined optimal extraction conditions were as follows. The liquid-solid ratio of 8 mL/g, ethanol concentration of 70%, 1.5 h for each extraction time, and extracted for two times. The linear range of Astragaloside Ⅳ content was from 0.87 μg to 8.72 μg. And the regression equation was Y = 1.545 4X + 5.875 9, r = 0.999 7. The average re-covery rate was 95.05%. The RSD was 2.64%. It was concluded that the optimized extraction technology was stable, reasonably practicable, and suitable for industrial production.

ABSTRACT

Objective To study the measure and two-year effects of cognitive-behavior therapy on adolescent with internet addiction disorder(IAD). Methods 76 cases with IAD were divided to randomized controlled trial, then 38 cases in each group. But 36 cases ( lost 2 cases) were completed in follow-up study group and control group of 34 cases (lost 2 cases). There were various forms of cognitive behavioral intervention in two years.The Chinese Internet Addiction Scale( CIAS), Symptom Gheck List-90 (SCL-90) scale and Coping Style Questionnaire(CSQ) were adapted to evaluate the effects when the cognitive-behavior therapy was started, after treatment at 2 months, 12 months, 18 months, 24 months respectively assessment for 5 times. Results The scores of CIAS in the study group were significantly lower than that of the control group( (43.53 ± 10. 37 ), (53.06 ± 5.40),(47.56 ± 10.02) ,(53.50 ±5.28) ,(42.68 ±6.23), (54.02 ±6.49), P<0.05 or P<0. 01 ) and decreased obviously with the extension of time (F= 14. 464, P< 0. 01; F= 0. 537, P> 0.05 ). Problem-solving and help factor of CSQ in the study group were significantly higher than that of control group( (0.82 ± 0. 11 ), ( 0.62 ± 0. 48 ), (0.78 ±0.12),(0.65 ±0.38) ,(0.90±0.28),(0.62 ±0. 37)); (0. 68 ±0.41), (0.45 ±0.29),(0. 74 ±0. 30),(0.45 ± 0. 11 ), (0.79 ± 0. 33 ), (0.46 ± 0. 18 ) ) and self-blame, fantasy , withdrawal factors were significantly lower than it((0. 21 ±0. 13),(0.40 ±0.27), (0.24 ±0.06),(0. 38 ±0. 19), (0.25 ±0.16),(0.42 ±0.24) );( (0.27 ±0.02) ,(0.45 ±0.27) ,(0.31 ±0.09) ,(0.44 ±0.28) ,(0.30 ±0.24) ,(0.46 ±0.19) ;(0.42 ±0.05),(0.54 ±0.33),(0.34 ±0.17),(0.55 ±0.29),(0.31 ±0. 19),(0.50 ±0.30), P<0.05 or P<0. 01 ). There were significant differences of SCL-90 total score between the study group and the control group at 4 times for evaluation ( ( 142.25 ± 30.44), ( 182.56 ± 36.23 ), ( 175.36 ± 26.44 ), ( 191.56 ± 38.32 ), ( 163.36 ±29.21),(196.75±35.46),(143.33±21.35),(194.11 ±28.19), P<0. 05 orP<0.01). Conclusion Developed cognitive-behavior therapy has obvious effects on the adolescents with IAD in two years.

ABSTRACT

Objective To investigate the protective effects of intra-peritoneal fluid resuscitation on small intestinal mucosa in rats with hemorrhagic shock. Method Fifty Sprague-Dawley (SD) rats were randomly (random number) divided into five groups, namely sham operation group (group I ), hemorrhagic shock group (group Ⅱ ), intra-venous fluid resuscitation group (group Ⅲ ) . intravenous fluid resuscitation plus intra-peritoneal saline resuscitation (group Ⅳ ) and intravenous fluid resuscitation plus intra-peritoneal PD-2 solution resuscitation group (group Ⅴ ). The rats of 5 groups were processed with cannulations of right common carotid artery, right femoral vein and left femoral artery with systemic heparinization. The rat models of hemorrhagic shock were established with modified Wigger' s method by which the blood exsanguinated from left femoral artery. The rats of group Ⅲ were resuscitated with shed blood plus twice equal volume of Ringer's solution after modeling of hemorrhagic shock.The rats of group Ⅳ and group Ⅴ were administered intra-peritoneally with 30 mL saline and 30 mL of 2.5% PD-2 solution, respectively as adjuncts to those used in the group Ⅲ . The specimens of blood and small intestine of rats of all groups were collected 60-120 minutes after modeling and resuscitation. The activity of plasma diamine oxidase (DAO) was determined with chromatometry, the level of plasma D-lactic acid (D-LA) with spectorophotometry and the level of plasma lipopolysaccharide (LPS) with nephelometry. The histopathological and ultrastructure changes of small intestine tissue of rats were observed under light microscope and electronic microscope. Results There were remarkable differences in activity of DAO, and the levels of D-LA and IPS in rats between those ingroup Ⅱ and group I (P <0.01), and between those in group V and groups Ⅱ , Ⅲ or Ⅳ (P <0.05 or P < 0.01) The pathomorphology and ultra-structure of small intestine tissues were severely damaged in group Ⅱ compared with those in group Ⅰ , and those markedly lessened in group V compared with groups Ⅱ , Ⅲ and Ⅳ . Conclusions Intraperitoneal fluid resuscitation with PD-2 solution can significantly protect the integrity of intestinal mucosa and the normal permeability of intestinal wall, and blunts the histopathological changes, and restrains bacterial translocation from gut and reduces the level of plasma endotoxin.

ABSTRACT

Based on thermodynamic principle, the critical relative humidity of electrolytes is closely related to their solubility. The authors explored the relationship theoretically and calculated critical relative humidity of 21 electrolytes from their solubility in the light of Raoult's law and extended Wilson model. The results indicate that the critical relative humidity values calculated by Raoult's law can not accord with the reported ones and there is a systematic error in the high concentration range; while these calculated by extended Wilson model are comparable to the reported ones.

Background Researches have demonstrated that ocular hypertension induces the ischemia-reperfusion of retina and further leads to the degeneration of retinal ganglion cells,but its mechanism is beyond understanding.Objective The present study aims to observe the effects of static pressure on the morphology,proliferation activity and viability of cultured retinal microvascular endothelial cells (RMECs) and evaluate the expression of ET-1 and NO in these cells under variant static pressure.Methods RMECs were isolated from 30 healthy Wistar rats and cultured using explant culture method by Ⅷ factor antibody and PECAM-1 antibody.The static pressure of 1.33kPa,2.67kPa,5.33kPa and 10.67kPa was used in culture bottle respectively.The RMECs without static pressure were used as normal control group.The morphology of RMECs under the different static pressure was observed by inverted phase contrast microscopy,and the number of RMECs was counted using the counting plate.Cellular viability was studied by trypan blue staining.The changes of ET-1 and NO_2~-/NO_3~-,two metabolic products of NO,in the medium were detected by radioimmunoassay and Griess's nitrate reductase method.The expression of ET-1,eNOS and iNOS mRNA in RMECs was analyzed by semi-quantitative RT-PCR 24 hours after treatment of variant static pressure.Results Cultured RMECs sticked well at 24 hours and reached to confluence at 48 hours and showed the red fluorescence for Ⅷ factor antibody and PECAM-1 antibody.Enlargement of nuclei,extenders of cell bodies and suspension of RMECs in medium were observed.The number of RMECs was gradually increased.The cell viability was reduced with the raise of static pressure among these four groups(F=12.205,P<0.01;F=11.180,P<0.01).The static pressure increased the content of ET-1 released by RMECs in 2.67kPa,5.33kPa and 10.67kPa of static pressure groups,and concentrations of NO_2~-/NO_3~- in the medium showed a significant increase in 5.33kPa and 10.67kPa of static pressure groups compared with normal and 1.33kPa of static pressure groups(P<0.01).The expressions of ET-1 mRNA,eNOS mRNA and iNOS mRNA were considerably enhanced in 5.33kPa and 10.67kPa of static pressure groups compared with normal control group(P<0.01).Conclusion Raised static pressure causes the alteration of RMGCs structure and morphology.Static pressure could upregulate the expressions of ET-1,eNOS and iNOS mRNA in RMECs and increase the release of ET-1 and NO.This pathway might be one of pathologic mechanisms of retinal injury induced by high intraocular pressure.

OBJECTIVE@#To observe the short-term efficacy of autogenous bone pate and Palva graft for obliterating huge remnant mastoid cavity in canal wall down approach.@*METHOD@#Retrospective analysis clinical data of twenty-one cholesteatomatous cases operated by one surgeon from 2004 to 2007. In twelve cases, simultaneous III type tympanoplasty (Sheehy, P. O. P) was performed. Other 9 cases had undergone mastoidectomy elsewhere before the admission. Six of them were still draining with huge remnant mastoid cavity, and the rest three patients had relapsed cholesteatomas with intermittent draining and huge mastoid cavity. Normal saline solution perfusion was used to measure the volume of remnant mastoid cavity. The criterion of huge remnant mastoid cavity is more than 8 ml.@*RESULT@#Of twelve primary cases with III type tympanoplasty, 11 patients maintained a small, dry, and healthy mastoid cavity after twenty-seven days. The average increase of hearing level of them was 17.5dB, and the air-bone gap is less than 20 dB. Of one patient, bone pate was infected and was discharged. A dry mastoid cavity was achieved until fifty-five days after surgery. The patient is keeping a big air-bone gap caused by displacement of ossicle chain prosthesis. Just eighteen days later, other nine cases of revision mastoidectomy achieved a small, dry, and healthy mastoid cavity, with lightly improved hearing level.@*CONCLUSION@#Obliteration of a canal wall down huge mastoid cavity by Palva graft with autologous bone pate is a reliable and effective technique that results in a small, dry, low-maintenance mastoid cavity. The short-term efficacy of simultaneous III tympanoplasty is satisfactory if patient selection is suitable.

ABSTRACT

Purpose The aim is to investigate the effects of BSP on immunological function in normal and immunosuppressed mices.Methods Thymus and spleen indexes, peripheral blood WBC number,the lymphocyte proliferation response, IL-2 production and serum and splenocyte hemolysin contents were measured after intraperitoneal injection of BSP in normal and immunosuppressed mices.Results (1)BSP 100 mg/(kg*d)×10d significantly increased the thymus and spleen indexes and peripheral blood WBC number in immunosuppressed mice.The thymus and spleen indexes in normal mice was also increased. In addition,BSP markedly improved T,B lymphocyte proliferation responses and IL-2 production in normal and immunosupressed mices.(2) BSP improved the serum and splenocyte hemolysin contents in normal and immunosuppressed mice. Conclusion It was suggested that BSP was a kind of immunomodulator, and could improve the immunological function of normal and immunosuppressed mices.

ABSTRACT

Objective:To observe the anti-carcinoma and immuno-regulatory effects of shikonin derivatives.Methods:A water-soluble preparation of shikonin derivatives was prepared (designated as LE)and given by lavage (2.5~10 mg/kg daily for 10 days) to the mice inoculated with either HepA22 or S180 sarcoma. Their survival duration and the in situ tumor mass were observed. Thymus and spleen indexes of the mice were measured. The parameters for immuno-functions were detected by the routine activity assays, which included NK cytotoxicity, ConA-induced lymphocyte transformation and IL-2 production by the splenocytes of the mice. Thymic and splenic morphology of the experimental animals were microscopically examined with HE staining.Results:Both thymic and splenic indexes in the tumor-bearing mice diminished extremely compared to those of the normal control, and the immunological functions analyzed were also found obviously lowered when loaded with the transplantable carcinomas. Under light microscopy, it was surprisingly exhibited that thymus cortex was almost disappeared in the organs of tumor-bearing mice, and the germinal centers of their spleens were visibly shrunk. LE inhibited propagation of the inoculated tumors and at the same time, it amended the immunosuppresive impacts by tumor-bearing, including both structures of immune organs and the bioactivities of spleen cells.Conclusion:LE can reverse the immune damages mediated by carcinomas.
