ABSTRACT

Objective:To evaluate the effect and possible mechanism of B-cell-specific moloney murine leukemia virus insertion site 1(BMI-1) on the radiosensitivity of pancreatic cancer SW1990 cell line. Methods:According to our previous study results, 4 Gy X-ray and down-regulation of BMI-1 expression were adopted as intervention conditions. The effect on the proliferation of SW1990 cells was assessed by clony formation assay. The effect on cell apoptosis and cell cycle was evaluated by flow cytometry. The effect upon the epithelial-mesenchymal transition (EMT) was determined by Western blot. Results:Clony formation assay and flow cytometry found that the lowest cell proliferation ability and the highest apoptotic rate were obtained after BMI-1 down-regulation and 4 Gy X-ray compared with other groups (all P<0.05). Flow cytometry revealed that the proportion of G 0/G 1 phase was significantly decreased, whereas the G 2/M phase was remarkably increased in pancreatic cancer stem cells treated with 4 Gy X-ray compared with the control group (both P<0.05). Nevertheless, opposite results were obtained after down-regulation of BMI-1 expression. Western blot showed that the expression of E-cadherin was up-regulated, whereas that of Vimentin was down-regulated following 4 Gy X-ray (both P<0.05). Conclusion:Downregulation of BMI-1 expression can enhance the radiosensitivity of pancreatic cancer SW1990 cells, and its mechanism may be related to cell cycle and EMT.

ABSTRACT

To investigate the effects of thanatos-associated protein 11 (THAP11) on the proliferation and apoptosis of esophageal cancer cell and the underlying mechanism.
Methods: Expression of THAP11 in human esophageal epithelial cells (Het-1A) and esophageal cancer cells (Eca109, TE-1, Ec 9706) were detected by Western blotting. Esophageal cancer TE-1 cells were divided into 3 groups: a normal control (NC) group, a negative control (LV-NC) group and a THAP11 (LV-THAP11) group. Then the cell proliferation were detected by MTT assay, cell apoptosis were detected by flow cytometry, caspase-3 and caspase-9 levels were detected by caspases kits. Ubiquitination of p53 was determined in esophageal cancer TE-1 cells.
Results: Expression of THAP11 was reduced in esophageal cancer cells compared with human esophageal epithelial cells (P<0.05). After transfection with LV-THAP11 in TE-1 cells, cell viability was reduced (P<0.05), while apoptosis rate and caspase-3 and caspase-9 levels were increased (P<0.05), indicating that THAP11 inhibited growth of esophageal cancer cells. In addition, the THAP11 increased the levels of p53 (P<0.05) and inhibited the ubiquitination of p53 regulated by MDM2.
Conclusion: THAP11 may inhibit the proliferation of esophageal cancer cells by inhibiting ubiquitination of p53.

ABSTRACT

<p><b>OBJECTIVE</b>To understand that whether initiation of antiretroviral therapy (ART) would impact on the change and its reasons regarding the HIV-related high risk behaviors among HIV-positive clients who attending the methadone maintenance treatment (MMT).</p><p><b>METHODS</b>In-depth interviews were conducted among 34 MMT clients in Yunnan province who were under ART. The related contents would include information on general demographic charicteristics, HIV infection,MMT, number of sexual partners before and after ART, sexual behavior and frequency of condom use, drug use, needle sharing, changes in risk behaviors before and after the ART, reasons for high-risk behavior, of the clients.</p><p><b>RESULTS</b>The average age of the interviewees was 38.5 years, and most of them were male (70.6%). The clients under this study all admitted that the frequencies of unsafe sex and needle sharing did not increase after the ART initiation, with the main reasons as increasing HIV related awareness, the use of methadone, high accessibility of free condoms and access to clean needles etc. However, 12 of 34 reported being relapsed and 3 reported inconsistent condom use. The interaction of ART and dosage of methadone were connected to the episodes of relapsing and the ART optimism would result in inconsistent condom use.</p><p><b>CONCLUSION</b>No evidence supported that the ART initiation would increase the risk behaviors among the HIV-infected MMT clients. However, attention needs to be paid to the new challenges caused by high expectation of ART.</p>

ABSTRACT

Objective To evaluate two methods of selecting region of interest (ROI)to measure the apparent diffusion coefficient (ADC)of breast lesions on diffusion-weighted imaging (DWI).Methods MRI findings (including plain MRI,dynamic contrast-en-hanced MRI and DWI)of 5 3 cases with breast mass confirmed by pathology were analyzed retrospectively. Two observers independ-ently measured the ADC of breast lesions by the method of fixed ROI with the size of 10 mm2 ±2 mm2 and the routine method. The mean ADCs of benign and malignant breast lesions were compared between two methods and two observers by using paired samples T-test.The values of ADC diagnosing breast cancer were compared between the two methods by using the receiver operating charac-teristic (ROC)curve. Results Among 62 lesions of 53 cases,33 lesions were malignant, and 29 lesions were benign. For the first observer, the mean ADCs of benign and malignant lesions were (1.647±0.359)×10-3 mm2/s and (1.136±0.227)×10-3 mm2/s measured by the fixed ROI method, respectively. The area under the ROC was 0.897.The mean ADCs of benign and malignant le-sions were (1.603±0.415)×10-3mm2/s and (1.251±0.237)×10-3 mm2/s measured by the routine method, respectively. The area under the ROC was 0.768.For another observer, the mean ADCs of benign and malignant lesions were (1.647±0.357)× 10-3 mm2/s and (1.130±0.250)×10-3 mm2/s measured by the fixed ROI method, respectively. The mean ADCs of benign and malignant lesions were (1.590± 0.390)×10-3 mm2/s and (1.250±0.237)×10-3 mm2/s measured by the routine method,respec-tively.There was no significant difference for the mean ADC of benign lesions between the two methods. However, there was sig-nificant difference for the mean ADC of malignant lesions between the two methods. The diagnostic value of ADC measured by the fixing method was better than that of the routine method (P=0.008).There were no significant differences for ADCs of benign and malignant lesions between the two observers. Conclusion DWI has an important value for differential diagnosis of benign and malignant breast lesions. The measurement of ADC by the fixed and smaller ROI is better than that by the routine ROI for dis-tinguishing malignant from benign breast lesions.

ABSTRACT

Objective To explore the immune characteristic of a patient with hypogammaglobulinaemia and low natural killer(NK)cell numbers. Methods Case histories were analyzed repeatedly to grasp important characteristics of the disease. A routine blood examination and peripheral blood immunological detection was done. Complete neutrophil function test, lymphocyte proliferation ability and cytokine profile were evaluated. Results A 3-year-old boy was referred to the hospital because of fever and abdominal pain as well as partial intestinal obstruction. Laboratory examination revealed low immunoglobulin, reduced absolute numbers of B and NK cells and deficient B cell function of lower IFN-γ and IL-12 production. The patient also had mesenteric lymphadenopathy and a caecal mass with histology suggestive of ileocecal junctional tuberculosis. The patient was infected with cytomegalovirus(CMV)and fungus, which are not common in simple antibody deficiencies. The patient was followed for four years and during this period the patient had recurrent periodontitis resulting in the loss of one tooth. Conclusions Although the precise mechanism of the defect is unclear,we speculate that the case may cast new light on primary immunodeficiencies predominantly with antibody deficiency.(J Clin Pediatr,2011,29(7):601-608)

ABSTRACT

Objective To investigate the effects of Epstein-Barr virus (EBV) infection on apoptosis of cord blood monocytes-derived dendritic cells (DC) in neonates. Methods Cord blood monocytes were induced into DC by 50 ng/mL recombinant human granulocyte macrophage colony-stimulating factor(rhGM-CSF)and 10 ng/mL recombinant human interleukin-4 (rhIL-4),and DC were divided into three groups:①4G group:rhGM-CSF and rhIL-4 were added separately on the day of cell separation;②4G+0d EBV group:rhGM-CSF,rhIL-4 and B95.8 cell supernatants were added simultaneously on the day of cell separation;③4G+5d EBV group:hGM-CSF and rhIL-4 were added on the day of cell separation, and B95.8 cell supernatants were added on the 5th day.Percents of DC apoptosis were measured using Annexin V-FITC and PI staining by flow cytometry. The expression of X-linked inhibitor of apoptosis protein (XIAP) was detected by Western Blot. Results Percent of DC apoptosis in 4G+0d EBV group was significantly higher than that in 4G group on the 6th to 14th day(P<0.05),and percent of DC apoptosis in 4G+5d EBV group was significantly higher than that in 4G group on the 7th to 14th day(P<0.05).The effects of EBV infection on percent of DC apoptosis had relations with infection time points. The expression of XIAP in DC decreased significantly after EBV infection. Conclusion EBV promotes apoptosis of cord blood monocytes-derived DC,which is associated with the differentiation and maturation status of DC.

ABSTRACT

Objective To observe the differential gene expression in human nasopharyngeal carcinoma (NPC) cell line with different radiosensitivity by cDNA microarray analysis. Methods A radioresistant cell line, CNE-2R, was established from a human nasopharyngeal carcinoma cells line CNE-2 by repeated X-ray irradiation. The differential gene expression of CNE-2 and CNE-2R was screened with cDNA microarray by BioStarH-141s profile gene chip. Results There were expressed 308 genes to be screened out between CNE-2R cell line with different radiosensitivity and its parental CNE-2 cell line, while 176 up-regulated genes in CNE-2R cells and 132 down-regulated genes were found. In them, there were 40 up-regulated ones and 36 down-regulated ones whose ratios were higher than 6.0 or lower than 0.1. The different genes included DNA damage- and repair-related genes; cell cycle-related and cytoskeleton-related proteins; and apoptosis-related genes. Conclusion The radioresistant CNE-2R cells were isolated from the CNE-2 cell line by repeated X-ray irradiation. The stable radioresistance is the result of co-effect by polygene and multiple factors, which provide several gene targets to sensitize the radioresistant cells for improving the radiocurability of NPC.

ABSTRACT

Objective To explore the expression of livin and survivin and its correlation with caspase-3 and clinicopathology in breast cancer. Methods We examined paraffin-embedded archival specimens from 76 cases of malignant breast tumor and 17 cases of benign breast disease for the expression of livin, survivin and caspase-3 with immunohistochemistry. The relationship of livin and survivin expression with caspase-3 and clinicopathological characteristics was analyzed. Results The positive rates of livin and survivin protein expression were increased gradually in normal breast tissues, livin and survivin proliferating disease, preinvasive carcinoma and invasive breast carcinoma, while the caspase-3 protein expression was significantly decreased. The expression of livin and survivin was negatively correlated with that of caspase-3 in the four groups (P0.05). The expression of survivin was related to lymph node metastasis and tumor size (P0.05). Conclusion Livin and survivin may play an important role in accelerated tumor proliferation by inducing caspase-3 to inhibit apoptosis in breast carcinoma. Livin may be more correlated with carcinogenesis in young patients while survivin may promote metastasis and invasion in breast carcinoma.
