ABSTRACT

Objective:This study aimed to evaluate the performance of breast magnetic resonance imaging (MRI) abbreviated protocol (AP) in diagnosing breast neoplasms.Methods:We retrospectively analyzed the data of 86 patients who had undergone breast MRI examinations and compared the images using an AP and full diagnostic protocol (FDP). The AP consisted of axial T2-weighted imaging (T2WI), diffusion-weighted imaging (DWI), and four-phase dynamic enhancement sequences. The FDP consisted of sagittal T2WI, axial T2WI, T1-weighted imaging, DWI, and seven-phase dynamic enhancement sequences. All the images were analyzed using the Breast Imaging Reporting and Data System (BI-RADS). The consistencies between the different protocols were then calculated. With the pathological diagnosis as the gold standard, the diagnostic capabilities of the two protocols were compared.Result:Two radiologists analyzed the AP and FDP images. The consistencies in the BI-RADS between the different protocols were 0.856 and 0.900, and those in time-signal intensity curves (TICs) were 0.822 and 0.922. Within the same protocol, the consistencies in the BI-RADS between different radiologists were 0.744 and 0.822, and those in TICs were 0.889 and 0.878. No significant differences were found ( P>0.05). In terms of diagnosing malignant neoplasms using the BI-RADS, the sensitivities of the AP and FDP were 89.8% (95 %CI: 0.785-0.958) and 91.5% (95 %CI: 0.806-0.968), respectively; their specificities were 71.0% (95 %CI: 0.518-0.851) and 77.4% (95 %CI: 0.585-0.897), respectively; and the areas under the curves (AUCs) were 0.804 (95 %CI: 0.698-0.910) and 0.845 (95 %CI: 0.748-0.941), respectively. Diagnosing malignant neoplasms using TICs, the sensitivities of the AP and FDP were 86.4% (95 %CI: 0.745-0.936) and 89.8% (95 %CI: 0.785-0.958), respectively; their specificities were 61.3% (95 %CI: 0.423-0.776) and 67.7% (95 %CI: 0.485-0.827), respectively, and the AUCs were 0.739 (95 %CI: 0.623-0.855) and 0.788 (95 %CI: 0.679-0.897), respectively. There was no significant difference between the AP and FDP ( P>0.05). The MRI acquisition times of the AP and FDP were 11.97±0.94 min and 21.25±1.12 min, respectively, with a significant difference ( P<0.001). The average reading time was reduced by 13.5% using the AP compared with that using the FDP. Conclusion:Compared with the FDP, the AP reduced the acquisition time and maintained the diagnostic accuracy, which can be used as an improved pattern for MRI screening in high-risk populations of breast neoplasms.

ABSTRACT

Objective@#To explore the role of serum microRNA (miRNAs) levels in the detection of pneumoconiosis, and to establish a combined application model of multiple serums miRNAs for pneumoconiosis diagnosis.@*Methods@#152 cases were selected in the case group and the control group respectively. The TaqMan Low Density Array method was used to screen out the candidate miRNAs for early screening of pneumoconiosis, and RT-qPCR was used to verify. According to the area under the curve (AUC) , the sensitivity and specificity of the candidate indicators were investigated. The logistic regression model was established by the two-class logistic regression model.@*Results@#The expression of 7 candidate miRNAs in the serum of pneumoconiosis patients was significantly different (P<0.05) . The receiver operating curve (ROC) of the above 7 miRNAs was analyzed, miRNA-21, miRNA-200c, miRNA-16, miRNA-206, miRNA-155, miRNA-29a had statistical significance, and their ROC-AUC is 0.629~0.932. Logistic regression model was: logitP=13.769+0.536×miRNA-21-0.878×miRNA-200C-0.012×miRNA-16-0.111×miRNA-206+0.117×miRNA-155-1.192×miRNA-29a.@*Conclusion@#Multiple serum miRNAs combined application models may be used for the diagnosis of pneumoconiosis patients.

ABSTRACT

Objective@#To observe the effect of overexpression of miRNA200a (miR-200a) recombinant lentivirus on the expression of Wnt/β-catenin signaling pathway in mouse lung epithelial cell line MLE-12 induced by silica (SiO2) .@*Methods@#The mice were divided into SiO2 control group (SiO2) , virus control group (SiO2+Lv-NC) group and overexpressing miR-200a virus group (SiO2+Lv-miR-200a). The expression of β-catenin, MMP2, MMP9, TCF-4 and Cyclin D1 mRNA and protein were detected by realtime-PCR and western blot after incubating cells for 18 h stimulating at the final concentration of 200 μg/ml of SiO2.@*Results@#The expression of miR-200a in MLE-12 cells of SiO2+Lv-miR-200 a group was significantly higher than that in SiO2 group and SiO2+Lv-NC group. The mRNA and protein expression of β-catenin, MMP2, MMP9, TCF-4 and Cyclin D1 in MLE-12 cells of SiO2+Lv-miR-200a group were significantly lower than those in SiO2 group and SiO2+Lv-NC group (P<0.05) .@*Conclusion@#Overexpression of miR-200a can inhibit the expression of related genes of Wnt/β-catenin signaling pathway in silica-induced mouse lung epithelial cells.

ABSTRACT

OBJECTIVE: To investigate occupational stress and its influencing factors in workers of electronic manufacturing.METHODS: The judgment sampling method was used to select 2 251 workers as study subjects. The Simple Occupational Stress Questionnaire and the Effort-Reward Imbalance( ERI) Questionnaire were used to investigate job demand-control(JDC) and ERI occupational stress respectively. RESULTS: Among the 2 251 workers,the prevalence rates of high occupational stress in JDC and ERI models were 56. 4% and 10. 1%,respectively. The positive rate of JDC occupational stress was higher than that of ERI( P < 0. 01). The multivariate logistic analysis indicated that workers with assembly line had higher risk of JDC and ERI occupational stress than workers without assembly line( P < 0. 05). Workers with labor dispatching had higher risk of JDC and ERI occupational stress than workers with long-term contract( P < 0. 05). Workers exposed to occupational hazards had higher risk of JDC and ERI occupational stress than workers not exposed to occupational hazards( P < 0. 05). Workers with disease had higher risk of JDC and ERI occupational stress than healthy workers( P < 0. 05). CONCLUSION: JDC is the main occupational stress model in workers of electronics manufacturing factory. The main influencing factors are assembly line,labor dispatching,weekly work time,exposure to occupational hazards and illness.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the effect of smoking on the microRNAs (miRNAs) expression in pneumoconiosis patients.</p><p><b>METHODS</b>Real-time qPCR was used to measure the expression levels of miR-21, miR-200c, miR-16, miR-204, miR-206, miR-155, let-7g, miR-30b, and miR-192 in 36 non-smoking patients with pneumoconiosis and 38 smoking patients with pneumoconiosis, and the differences in expression levels between the two groups were evaluated by two-independent samples t-test.</p><p><b>RESULTS</b>The expression of miR-192 in serum showed a significant difference between non-smoking and smoking pneumoconiosis patients (P < 0.05), and it decreased gradually in smoking patients with stage I and II pneumoconiosis. In the serum of all pneumoconiosis patients, the expression level of miR-16 was the highest, while the expression level of miR-204 was the lowest.</p><p><b>CONCLUSION</b>Pneumoconiosis patients have differential expression of miRNAs in serum, and smoking has an effect on the miRNAs expression in pneumoconiosis patients.</p>

ABSTRACT

Objective: To study the effect of serum from maintenance hemodialysis(MHD) patients on the expression of intercellular adhesion molecule-1(ICAM-1) in human umbilical vein endothelial cells(HUVEC) cultured in vitro, and the mechanism of endothelial cells dysfunction caused by serum from MHD patients thereof. Methods: HUVEC were incubated for 1 h, 3 h, 6 h and 12 h in RPMI 1640 culture media containing 10 % fetal cattle serum(FCS )(group A), 10% normal human serum (group B) and 10% MHD patient serum (group C) respectively. The immunocytochemical method was used to determine the expression of ICAM-1. Results: There were significant differences in the expression of ICAM-1 in HUVEC among stimulation groups with different serums(P < 0.05). Compared with groups A and B, the expression of ICAM-1 was significantly increased in HUVEC of group C. There was significant difference in the expression of ICAM-1 at the different stimulation times(P < 0.05). There was significant interaction between the expression of ICAM-1 in HUVEC and stimulation groups with different serum and the stimulation times(P < 0.05). Conclusion: The serum from MHD patients with chronic renal failure can promote the expression of ICAM-1 in HUVEC cultured in vitro, thus causing the dysfunction of endothelial cell.
