ABSTRACT

Objective:To study the teaching effect of traditional teaching combined with virtual simulation teaching in pharmaceutical content analysis using high performance liquid chromatography (HPLC).Methods:The students in two parallel classes of grade 2016 were divided into experimental group (44 students) and control group (38 students). Taking the content analysis and its related analytical methodology of Metronidazole by HPLC as an example, the experimental group adopted the traditional offline teaching combines with virtual simulation teaching, and the control group adopted the traditional offline teaching. The teaching effects of the two groups were compared. SPSS 22.0 was used to conduct independent-samples t-test. Results:According to the mid-term test results, the experimental group was superior to the control group in practical operation assessment [(17.98±6.75) vs. (14.03±5.92)], and the difference was statistically significant ( P < 0.05). But there were no differences between the two groups in the scores of theory, discussion report and presentation. There was no significant difference in the final exam scores between the two groups. Conclusion:The ability of practical training and the teaching effect can be improved by the teaching method of traditional offline teaching combined with virtual simulation. Through the training of this project, students have mastered the operation of HPLC, the content determination of pharmaceutical preparations, the verification of analytical methods, data processing, and the writing of analytical reports, and have certain practical analysis ability.

ABSTRACT

【Objective】 To summarize a rapid and effective method to eliminate autoantibodies interference with blood group identification and evaluate its treatment effect. 【Methods】 Blood samples with suspicious results in initial blood group identification were collected, and those caused by autoantibodies were chosen, and their red blood cells were washed, dispersed or treated with sulfhydryl reagent. After allogeneic or autologous absorption of plasma, blood groups of those patients were re-detected to evaluate the effectiveness of the above method. 【Results】 Among 39 patients presenting suspicious ABO blood group, 9 were interfered by autoantibodies. After appropriate treatment, the ABO/RhD blood group of those patients could be identified. 【Conclusion】 Autoantibodies could interfere the identification of ABO/RhD blood group, and the efficiency and accuracy of blood group identification could be improved by analyzing the test results and selecting appropriate treatment methods.

ABSTRACT

OBJECTIVE:To identi fy chemical components of Actinidia chinensis root rapidly ,and to provide reference for further material basis and quality control study of the crude medicine. METHODS :UHPLC-Q-TOF-MS/MS technique was used to detect chemical components of A. chinensis root. The separation was performed on Waters XSelect HSS T 3 column with mobile phase consisted of 0.1% formic acid acetonitrile solution- 0.1% formic acid water solution (gradient elution )at the flow rate of 0.3 mL/min. The column temperature was set at 40 ℃,and sample size was 3 μL. Electrospray ion source was adopted,the data was collected under negative ion mode ;the scanning range was m/z 50-1 500;the drying gas temperature was 350 ℃,the atomizing air pressure was 45 psi,the capillary voltage was 3 500 V,and sheath gas temperature was 350 ℃. According to the information of excimer ion and secondary fragment ion ,the chemical components were identified by combining with the relevant literature ,the retention time of the reference substance and the law of mass spectrometry cracking. RESULTS & CONCLUSIONS :Totally 58 chemical components was identified ,which included 16 pentacyclic triterpenes (such as hydroxyasiatic acid ,asiatic acid ,maslinic acid,corosolic acid ,oleanic acid ,ursolic acid ,etc.),12 flavonoids(such as rutin ,quercitrin,cynaroside,astragalin,etc.),17 organic acids (such as cryptochlorogenic acid ,chlorogenic acid ,isochlorogenic acid A ,isochlorogenicacid C ,etc.). There were 9 components(such as procydanidin B 1,B2 and luteolin ,etc.)identified for the first time in A. chinensis root. UHPLC-Q-TOF-MS/ MS technique can be used for the rapid identification of chemical components in A. chinensis root.

ABSTRACT

Objective To investigate the relationship between serum markers β amyloid (Aβ), tau and thyroid hormone levels and post-stroke cognitive impairment (PSCI) in the acute phase of cerebral infarction. Methods A total of 214 patients with acute cerebral infarction were enrolled. The baseline data and serological indicators were collected and the cognitive function of patients was evaluated. All patients were divided into cognitive impairment group and normal group based on follow-up results. The differences of Aβ1-42, tau protein and thyroxine levels between the two groups and their relationship with disease progression were analyzed. The Cox regression analysis and ROC curve were used to compare the above parameters to predict the development of PSCI. Results The total protein level of Tau (210.6 ±98.9 pg/mL) was higher and Aβ1-42 (426.1 ±123.5 pg/mL) and triiodothyronine (T3) (1.43 ±0.57 nmol/L), free thyroxine (FT4) (13.15±2.23 pmol/L) was significantly lower in the cognitive impairment group than in the normal group (P<0.05). Tau protein (r=-0.457), Aβ1-42 (r=0.348), T3 (r=0.211), and FT4 (r=0.306) were all associated with disease progression (P<0.05). Cox regression analysis showed that Aβ1-42 and T3 were important influencing factors in the occurrence of PSCI. The area under the curve of Aβ1-42 combined with T3 was 0.841. The specificity and the sensitivity were 74.8% and 85.3%, respectively, with a diagnostic cutoff value of 0.572. Conclusion Aβ1-42 and T3 levels in the acute phase of cerebral infarction may predict the progression of PSCI.

ABSTRACT

Objective To study the differences in the inclination of lower incisors among the female patients with beautiful profiles in different vertical skeletal types so as to provide the clinical treatment design for a certain guiding. Methods 137 cephalometric films of female patients with beautiful profiles were selected and divided into three groups based on the vertical facial types(∠SNMP):small degree(n = 31),average degree(n = 89) and larger degree groups(n=17). The angles of∠L1/MP and∠L1/A-Pog were measured and treated with covari-ance analysis for detecting the differences between the groups. Results The index of L1-MP(°)was the smallest in the larger degree group,the largest in the small degree group and average in the average group,with significant differences between the three groups(F = 17.38,P 0.05). Conclusion Lower incisors have compensatory inclined lingually in the group of higher mandible angle,but labially in the group of lower mandible angle.

ABSTRACT

Objective To explore the splenic macrophages phenotype and secretory function of mice with periodontitis, so as to explore effects of periodontitis on macrophages. Methods 22 mice were randomly divided into periodental ligation group (group P10d ) and pseudo periodental ligation control group (group C), with 11 mice in each group. The experimental periodental ligation on mice lasted for 10 days before they were sacrificed. Flow cytometry was applied to detect the expression of M1 and M2 in mononuclear macrophages. Real-time PCR was applied to detect the relative expression of pro-inflammatory cytokines IL-1β and anti inflammatory cytokines IL-10. Results Compared with the control group C, the proportion of M1 macrophages in the periodontitis group decreased, and the ratio of M1/M2 was also decreased significantly, and IL-1β mRNA also down-regulated. Conclusions Chronic periodontal infection could down regulate the proportion of M1 macrophages , decrease ratio of M1/M2 and the expression of inflammatory cytokines IL-1βmRNA.

ABSTRACT

Objective To investigate the expression level of antisense non-coding RNA in the INK4 locus (ANRIL) in hepatocellular carcinoma (HCC) tissues and to evaluate its relation to clinicopathological features and prognosis of HCC.Methods Quantitative real-time polymerase chain reaction (qRT-PCR) method was used to detect the expression of ANRIL in HCC tissues and adiacent tissues (n =90) and to analyze its relationship with clinicopathological data.Kaplan-Meier curves and multivariate Cox proportional models were used to study the impact on clinical outcome.Small interfering RNA (siRNA) was used to silence ANRIL and to explore the effects of reduced ANRIL expression on cell growth and metastasis.Results ANRIL expression in HCC tissues was significantly higher than in the adjacent non-tumor tissues (t =13.083,P < 0.05).The expression of ANRIL was remarkably associated with the histologic grade (x2 =40.724,P < 0.05) and TNM stage (x2 =43.245,P < 0.05).The mean survival time of the patients with high ANRIL was 18.2 months (95% CI:14.9-21.5 months),shorter than 39.4 months (95% CI:35.5-43.4 months) in low expression (x2 =47.590,P <0.05).Multivariate analysis suggested that high ANRIL expression was an independent predictor of poor prognosis (HR =2.143,95% CI:1.083-4.243,P < 0.05).Decreased expression of ANRIL could suppress the cell proliferation,migration and invasion of HCC cells.Conclusion Positive ANRIL expression is negatively correlated with the prognosis of HCC patients.

ABSTRACT

<p><b>OBJECTIVE</b>To compare the immune function after laparoscopic surgery (LS) and conventional open surgery(OS) for colorectal cancer (CRC).</p><p><b>METHODS</b>PubMed, EMBASE, the Cochrane Library, China National Knowledge Infrastructure (CNKI), and Wanfang Database were systematically searched for randomized controlled trials published before August 2013 concerning the immunological difference between LS and OS. Data extraction was performed independently by two reviewers and data analysis was performed using Review Manager ver. 4.3.1.</p><p><b>RESULTS</b>Twelve studies including 638 patients (307 in LS group and 331 in OS group) were eligible for analysis. Overall analysis demonstrated that no significant differences were identified for blood C-reactive protein level on postoperative days (POD) 0-1 (P=0.40), plasma lymphocyte count on POD 1-3 (P=0.92) and POD 4-7 (P=0.64), plasma CD4⁺ T cell count on POD 1-7 (P=0.63), plasma CD8⁺ T cell count on POD 4-7 (P=0.09), and plasma NK cell count POD 1-3 (P=0.34) as well as POD 4-7 (P=0.46). Data analysis also showed that a significantly lower serum level of IL-6 on POD 0-1 after LS (WMD=-25.03, 95% CI:-34.06 to -15.99, P=0.000), and a significantly higher plasma level of CD8⁺ T cell count on POD 1-3 after LS(WMD=0.05, 95% CI:0.01 to 0.08, P=0.004).</p><p><b>CONCLUSIONS</b>Although postoperatively short-term humoral immune function trends to be better after LS for CRC compared to OS, there is no sufficient evidence to support superior preservation of global immune function after acute reactive phase.</p>

ABSTRACT

Objective To investigate the therapeutic effect of liraglutide on glycemic control in metformin-treated patients with type 2 diabetes mellitus (T2DM).Methods Twenty patients with T2DM by maximum tolerated dose of mefformin in this study.For at least 3 months,participants were treated by addingliraglutide.After 1 week,the dosage of liraglutide was increased to 1.2 mg for 3 months from 0.6 mg.Patients' blood glucose,glycosylated hemoglobin(HbA1c),islet function,blood lipid and body weight were observed.Results Compared with before treatment,there were significant change of fasting blood glucose ((8.9 ± 1.4) mmol/L vs.(6.9 ± 1.0) mmol/L,t =5.951,P <0.01),postprandial blood glucose ((14.4 ±1.5) mmol/L vs.(10.4 ± 1.4) mmol/L,t =8.878,P < 0.01)),HbA1 c ((8.7 ± 1.3) % vs.(7.6 ± 1.1) %,t =4.860,P <0.01),fasting C peptide ((1.7 ±0.6) mg/L vs.(2.9 ± 1.1) mg/L,t =5.752,P < 0.01) and postprandial2 h C peptide ((5.0 ±2.3) mg/L vs.(10.2 ±3.5) mg/L,t =5.667,P <0.01) after treatment.Significant change of blood liquid and body weight were also observed after treatment than before treatment (P < 0.01).Conclusion Liraglutide treatment resulted in persistent better glycaemic control,weight reduction,improvements of blood lipid and function of pancreatic island successfully.Liraglutide was also associated with low risk of hypoglycemic and should be chosen by patients with T2DM.
