ABSTRACT

Objective To explore the expression and significance of peripheral blood T lymphocyte subgroups and natural killer cell in children with asthmatic pneumonia.Methods From 2013 to 2016,asthmatic children with pneumonia were erolled in the study as asthmatic pneumonia group(36 cases) and 36 cases medical health children as control group at the same time in Zhongshan City people′s hospital.Flow cytometry was used to test analysis of two groups of T lymphocyte subsets and natural killer cell in serum and the expression of CD28,CD56,using ROC curve analysis results.Results Asthmatic pneumonia group compared with control group,the difference of CD3+ and CD3+CD8+,CD28+,CD8+CD28+,CD8+CD28-,CD3-CD16+CD56+ and CD3+CD16+CD56+,CD3+CD4+/CD3+ CD8+ was statistically significant(P<0.05).According to the results of flow parameters have higher sensitivity by the ROC curve statistics.Conclusion Asthmatic pneumonia children have immune dysfunction,immune function changes associate with asthmatic,which is of great clinical significance to carry out immune function detection for children with asthmatic pneumonia.

ABSTRACT

Objective To investigate the changes and clinical significance of CD19 ,CD20 ,CD3-CD16+ CD56+ (NK cells) and CD3+CD16+CD56+ (NKT cells) in the children with epilepsy .Methods 125 cases of children with epilepsy children confirmed by Pediatric neurology specialists in Zhongshan People's Hospital from January 2013 to March 2016 were selected as epilepsy group , while 125 cases of healthy children were selected as control group ,and the levels of CD19 ,CD20 ,NK cells and NKT cells in periph-eral blood of two groups were detected by flow cytometry .Results CD19 ,CD20 ,NK cells and NKT cells levels in epilepsy group were higher than those in control group ,and the difference was statistically significant (P<0 .05) .For ROC curve analysis ,CD19 optimal cutoff value of 9 .4 ,the area under the curve was 90 .3% ,with 91 .7% sensitivity .CD20 optimal cutoff value of 7 .75 ,the ar-ea under the curve was 87 .3% ,with 91 .7% sensitivity .NK cells optimal cutoff value of 8 .55 ,the area under the curve was 80 .5% , with 95 .8% sensitivity .NKT cells optimal cutoff value of 0 .35 ,the area under the curve was 76 .9% ,with 91 .7% sensitivity .Con-clusion CD19 ,CD20 ,NK cells and NKT cells can be used as important indexes for monitoring immune conditions in children with epilepsy .

ABSTRACT

<p><b></b>To study the role and molecular mechanism of epigallocatechin gallate (EGCG) in reversing drug-resistance to 5-fluorouracil (5-FU) in gastric cancer drug-resistant cell line SGC-7901/5-FU.</p><p><b>METHODS</b>Drug-resistance gastric cancer cell line (SGC-7901/5-FU) was established by high doses of repeated impact joint drug concentration increment methods. The cell viability of the parent cell line and the drug-resistance cell line were determined by standard MTT assay. Cell survival rate of drug-resistance was calculated by the formula [(Aof the treatment group / Aof the control group) × 100%]. Cell half inhibitory concentration (IC) and resistance index (RI) were calculated by the Graphpad prime 6.0 software(RI=ICvalue of drug-resistance cells / ICvalue of parent cells). The apoptosis rate of SGC-7901/5-FU cells was quantified by flow cytometry after staining with annexin-V and PI. Western blot was used to detect the protein expression of drug-resistance-related proteins (ABCG2, P-gp, MDR-1 and GST-π) and apoptosis-related proteins (PARP, Survivin, Bax and bcl-2).</p><p><b>RESULTS</b>ICvalue was significantly increased in drug-resistant cells compared with parental cells [(64.7±3.9) mg/L and (4.1±0.3) mg/L, respectively, t=26.46, P=0.000], and the RI was 15.6. Proliferation activity in the drug-resistant cells was higher than that in parental cells at different 5-FU concentrations (all P<0.05). In drug-resistant cells, the ICvalue of 5-FU combined with EGCG group obviously decreased compared with 5-FU group [(7.3±0.1) mg/L and (63.1±1.4) mg/L respectively, t=40.84, P=0.000], and the RI was 0.12. Proliferation activity in drug-resistant cells was significantly decreased after EGCG treatment at different 5-FU concentrations (all P<0.05). Cell apoptosis rates in control group, 5-FU group, EGCG group and 5-FU combined with EGCG group were (3.0±1.0)%, (7.0±1.3)%, (6.0±1.2)% and (18.0±1.4)%, while apoptosis rate in 5-FU combined with EGCG group was significantly higher than those of other 3 groups(F=129.5, P=0.000). Western blot revealed that after EGCG treatment, the expression levels of drug-resistance-related proteins (ABCG2, P-gp, MDR-1 and GST-π) in the drug-resistant cell line SGC-7901/5-FU decreased significantly; the expression levels of apoptosis marker protein PARP and pro-apoptotic protein Bax increased significantly; and the expression levels of anti-apoptotic protein Survivin and Bcl-2 decreased significantly (all P<0.05).</p><p><b>CONCLUSION</b>EGCG can reduce the resistance of gastric cancer resistant cell line SGC-7901/5-FU, whose role may be via the inhibition of the expression of drug-resistance-related proteins, and the elevation of the protein expression ratio of PARP/Survivin and Bax/Bcl-2.</p>

[ ABSTRACT] Receptor type protein tyrosine phosphatase Q ( PTPRQ) is an unusual protein tyrosine phosphatase that has intrinsic dephosphorylating activity for various phosphatidylinositiol and phospho-tyrosine substrates, especially the phosphatidylinositol activity.Recent data show that PTPRQ has an important role in various biological processes and is as-sociated with some diseases.In this article, the structure and function of PTPRQ and the relationship between PTPRQ and diseases were briefly summarized.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the role of transforming growth factor β1 (TGF-β1) in patients with colorectal cancer.</p><p><b>METHODS</b>Fresh peripheral blood were obtained from 50 patients (before surgery and at least one week after surgery) and 25 healthy donors in the morning. Fresh colorectal cancer tissues and the adjacent tissues (at least 5 cm from the tumor site) were obtained from patients undergoing tumor resection. The expression levels of TGF-β1 in the blood and tissue specimens were determined using ELISA.</p><p><b>RESULTS</b>The plasma levels of TGF-β1 in patients with colorectal cancer were significantly higher than those in the healthy donors, and decreased after the surgery (P<0.05). The tumor tissues expressed higher levels of TGF-β1 than the adjacent tissues from both CEA-negative and -positive patients. The plasma level of TGF-β1 in the patients were positively correlated with the tumor size and clinical tumor stage (P<0.05).</p><p><b>CONCLUSION</b>TGF-β1 combined with CEA can provide important information for the diagnosis, prognostic assessment and prediction of recurrence in patients with colorectal cancer, and may provide new insights for anti-TGF-β1-based tumor immune therapeutic strategies.</p>

ABSTRACT

Objective To observe the X-ray appearance and clinical value of galactography.Methods 600 cases of galactorrhea confirmed by pathologly underwent galactography,among them,298 cases were blood type(49.8%),150 cases were serum type(25%),106 were rinsing type(17.7%) and others were 46 cases(7.7%).Results On galactography,the ductsl were divided into 3 groups: 345 cases were foliar type(59%),127 were ramate type(26.2%),87 were trunk type(14.8%).The main radiographic characteristics of lesions were mammary ductal ectasia in 578(96.3%),ductal distortion in 561(93.5%),irregular filling defect in ducts in 349(58.2%),and ductal damage in 39(6.5%).42 cases were breast carcinomas(7%),312 were intraductal papillary tumors(52%),129 were mammary ductal ectasia(21.5%),26 were mammary ductal mastitis(4.3%),49 were proliferous disease(8.2%),25 were mammary cyst(4.2%),17 were negative anamnesis(2.8%).Conclusion Galactography is beneficial in diagnosis of the pathogenesis of galactorrhea.

ABSTRACT

Objective To decrease misdiagnosis and to improve the correct diagnostic rate of mammary tuberculosis .Methods 9 cases of mammary tuberculosis proved by operation and pathology misdiagnosed as other diseases were retrospectively analyzed.Results Among 9 cases,5 cases were misdiagnosed as mammary cancer,1 case as chronic mammary inflammation,1 case as mammary abcess,1 case as mammary hyperplasia,1 case as mammary fibroadenoma.Conclusion The reasons leading to misdiagnosis are unsuitable methods of examinations,lack of solid basic knowledge,and lack of experience.
