ABSTRACT

Objective:To investigate the safety, efficacy and survival of brentuximab vedotin (BV) monotherapy and BV combined with chemotherapy for relapsed or refractory lymphoma. Methods:A total of 47 patients with relapsed or refractory Hodgkin's lymphoma (HL) in First Medical Center of PLA general Hospital and Fourth Medical Center of PLA General Hospital from October 2011 to December 2018 were admitted, including 35 cases (BV monotherapy group) and 12 cases (BV combined with chemotherapy group); there were 8 cases of relapsed or refractory anaplastic large cell lymphoma (ALCL), 4 cases in BV monotherapy group and 4 cases in BV combined with chemotherapy group. The safety, clinical efficacy and survival of two neoplasms in different groups were compared. Results:For relapsed or refractory HL, the objective remission rate (ORR) and complete remission rate (CRR) was 67.7% (21/31) and 16.1% (5/31), and the median progressive-free survival (PFS) time was 3.5 months (1.5-24.0 months) in BV monotherapy group; ORR and CRR was 81.8% (9/11) and 27.3% (3/11), and median PFS time was 5.5 months (2.0 - 24.0 months) in BV combined with chemotherapy group; there was no statistical difference in ORR and CRR between the both groups (χ 2 = 0.788, P = 0.375; χ 2 = 0.654, P = 0.419). There were 4 cases in BV monotherapy group for ALCL, of which 3 could be evaluated for efficacy, including 1 case of complete remission (CR) and 1 case of partial remission (PR); there were 4 cases in BV combined with chemotherapy group for ALCL, of which 4 could be evaluated for efficacy, including 2 cases of CR and 2 cases of PR. The common adverse events in BV monotherapy group were anemia, leukopenia, thrombocytopenia, fever, elevated transaminase, fatigue, nausea, peripheral neuritis and cough. Grade ≥3 adverse events were mainly anemia, thrombocytopenia and leukopenia. The common adverse events of BV combined with chemotherapy group were similar to those of BV monotherapy group, and there were significant differences in bone marrow suppression (thrombocytopenia, leukopenia) between the two groups (all P < 0.05). Conclusions:The clinical efficacy of BV combined with chemotherapy is better than that of BV monotherapy in treatment of relapsed or refractory lymphoma, and the survival time is prolonged. The adverse reaction of BV combined with chemotherapy is mainly manifested in bone marrow suppression, and the safety and tolerability of patients are acceptable.

ABSTRACT

AIM:To observe the effects of Egr-1 gene transfection on the expression of tumor necrosis factor-α( TNF-α) and intercellular adhesion molecule-1 ( ICAM-1) , and to investigate the role of Egr-1 in the pathogenesis of dia-betic nephropathy .METHODS:The diabetic mouse model was established .Ten mice were randomly selected as the dia-betic group .The remaining 40 mice were injected with empty plasmid , Egr-1 expression plasmid or Egr-1 siRNA plasmid via the tail vein once a week. The normal control group was also set up .The animals were sacrificed at the end of the 4th week. The renal tissues were harvested .The expressions of Egr-1, TNF-αand ICAM-1 were detected by immunohisto-chemistry and Western blot .The pathological changes were observed under electron microscope .RESULTS: In diabetic mouse kidney, the expression of Egr-1, TNF-αand ICAM-1 was increased, and irregular thickening of glomerular basement membrane , mesangial expansion and fusion of foot were observed .The change trend was more significant in Egr-1 gene transfection group , and these changes in siRNA plasmid transfection group were obviously reduced compared with diabetes group.CONCLUSION:Egr-1 up-regulates the expression of TNF-αand ICAM-1, and induces mesangial cell proliferation and mesangial extracellular matrix accumulation , which is probably one of the mechanisms of accelerating glomerulosclerosis .

ABSTRACT

Objective To investigate the correlation between drug-resistant genes and mobile genetic elements in multi-drug resistant Escherichia coli, and to explore phylogeny among the strains.MethodsTotally 20 strains of multi-drug resistant Escherichia coli were collected from Pan' an Hospital,Zhejiang Province during June 2009 and June 2010.Beta-lactam-resistance genes,aminoglycoside-resistance genes, genetic markers of mobile genetic elements were analyzed by PCR.Index and sample cluster analysis were performed on above results. Results In 20 strains of Escherichia coli,4 kinds of beta-lactamresistance genes,4 kinds of aminoglycoside-resistance genes, and 5 kinds of genetic markers of mobile genetic elements were detected.Index cluster analysis showed that correlation existed between resistance genes TEM,CTX-M-1,aadA5 and mobile genetic elements traA,IS26,ISEcpl; and correlation also existed between resistance genes OXA-1,aac(6′)-Ⅰ b,ant(3)-Ⅰ,rmtB and mobile genetic elements trbC,IS903.Sample cluster analysis showed that this group of Escherichia coli could be divided into 2 groups which were genetically different.ConclusionsDrug-resistant genes in multi-drug resistant Escherichia coli are correlated with mobile genetic elements.Sample cluster analysis can reveal phylogeny among the strains, which is important for hospital infection control.

ABSTRACT

OBJECTIVEA novel Eu-PB composite mercury film electrode (GC/Eu-PB/MFE) based on chemical modified was developed for the determination of baicalin and the investigation of its the electrochemical behavior.

METHODCyclic voltammetry and differential pulse voltammetry were used.

RESULTThe adsorption characteristic of baicalin was observed in Britton-Robinson buffer solution (pH 6.0) at GC/Eu-PB/MFE. The reduction peak of baicalin was observed at -1.22 V (vs SCE) and peak current was linear to the baicalin from 8.0 x 10(-9) to 1.0 x 10(-6) mol x L(-1) (r = 0.9992, n=8). The limit of detection was 1.2 x 10(-9) mol x L(-1) (S/N = 3). The average recovery was 99.83%, with RSD of 2.3% (n=8). The baicalin in Shuanghuanglian oral can be directly determined without pretreatment.

CONCLUSIONThe GC/Eu-PB/MFE was used to determine baicalin in Shuanghuanglian oral with good repeatability and high sensitivity. The method can be used for the quality control of Chinese traditional medicines containing Scutellaria baicalensis.

ABSTRACT

Objective To observe the effects of different doses of Yinxingdamo injection on changes of nerve-specific enolase (NSE) of patients with medium-sized brain injury and discusse the mechanism of Yinxingdamo injection in the treatment of brain injury. Methods The contents of serum NSE in 33 cases of craniocerebral injury were dectected by ELISA test and the results were analyzed. Results NSE levels in patients with brain injury were significantly higher than normal people (p <0.05). Yinxingdamo injection can reduce the content of NSE for brain injury patients. This reduction related with the dosage of the medicine, showing the higher the dosage is used, the greater reduction may be.Conclusion Yinxingdamo injection can reduce NSE in serum of patients with brain injury, and this may be the mechanism of Yinxingdamo injection in treating craniocerebral injury.
