ABSTRACT

AIM: To explore the protection of Hyperoside (Hyp) on heart and thoracic aorta in mice with myocardial infarction (MI) as well as its potential mechanism. METHODS: The MI model was generated by a ligation on the left anterior descending coronary artery. The mice were then randomly divided into sham group (saline, 0.1 mg/10 g), model group (saline, 0.1 mg/10 g), Hyp-low, moderate and high concentration groups (Hyp, 9, 18 and 36 mg/kg), Fosinopril group (Fosinopril, 15 mg/kg), and Hyp-high concentration + 3-MA group (Hyp, 36 mg/kg; 3-MA, 30 mg/kg). The mice were treated with Hyp and Fosinopril for two weeks, and then the changes of heart weight versus body weight (HW/BW), electrocardiogram (ECG) remodeling, cardiac function, oxidative stress level in serum, thoracic aorta remodeling and endothelial function were investigated. RESULTS: In the model group, the HW/BW was elevated (P<0.01). The width of QRS in ECG was elevated, as a company with the reduction of height of QRS (P<0.01). The echocardiography assay showed that the cardiac cavity was enlarged (P<0.01). The oxidative stress level in serum was enhanced (P<0.01). The thoracic aorta remodeling and endothelial dysfunction became more serious (P<0.01). After being treated with different concentrations of Hyp and fosinopril for two weeks, the changes above were reserved (P<0.05, P<0.01). Co-treatment with 3-MA, an autophagy inhibitor, suppressed the protective effects of Hyp on impaired hearts and vessels (P<0.05, P<0.01). CONCLUSION: Hyp has protective prospects on injured heart and thoracic aorta remodeling, as well as endothelial dysfunction in MI mice; 3-MA, an autophagy inhibitor, can reverse the cardiovascular protection of Hyp. The mechanism may be explained that Hyp can elevate autophagy level in heart, which further weaken oxidative injury in MI mice.

ABSTRACT

Objective:To investigate the mechanism of Salvia miltiorrhiza extract regulating nuclear factor-E2-related factor 2/antioxidant response element(Nrf2/ARE) pathway in improving lung injury in rats with severe acute pancreatitis (SAP).Methods:The SAP lung injury rat model was established by retrograde injection of 5% sodium taurocholate (1 ml/kg) into biliopancreatic duct. The rats were randomly divided into model group, Salvia miltiorrhiza extract (1.5 g/kg) group, ML385 (Nrf2/ARE pathway inhibitor, dose: 20 mg/kg) group and Salvia miltiorrhiza extract + ML385 group (dose: 1.5 g/kg, 20 mg/kg). The ascites volume was measured, and W/D ratio (W/D=wet weight/dry weight) was calculated. Pathological symptoms of lung tissues were detected by hematoxylin-eosin (HE) staining, and Holfbauer score was performed. Arterial blood gas indexes were detected by automatic blood gas analyzer. Serum superoxide dismutase (SOD), malonaldehyde (MDA) levels and lung reactive oxygen species (ROS) levels were detected by enzyme-linked immunosorbent assay (ELISA) kit. And the expression of Nrf2 protein was detected by Western blot.Results:Compared with the control group, the lung tissue of the model group showed obvious dilation and thickening of alveolar wall capillaries, enlargement of alveolar septum, hyperemia, edema and infiltration of inflammatory cells in the pulmonary interstitium. The ascites volume, ROS level in lung tissue, W/D and Holfbauer score, arterialpartial pressure of carbon dioxide (PaCO 2) and serum MDA levels were significantly increased ( P<0.05), while arterial partial pressure of oxygen (PaO 2), oxygenation index (OI) and serum SOD levels were significantly decreased ( P<0.05). Compared with the model group, the above pathological damage symptoms were alleviated in Salvia miltiorrhiza extract group, the ascites volume, ROS level of lung tissue, W/D, Holfbauer score, PaCO 2 and serum MDA level were decreased ( P<0.05); while the PaO 2, OI, serum SOD level and Nrf2 protein expression in lung tissue were increased ( P<0.05). Compared with the model group, the above pathological damage symptoms were aggravated in ML385 group, and the ascites volume, ROS level of lung tissue, W/D, Holfbauer score, PaCO 2 and serum MDA level were significantly increased ( P<0.05), while PaO 2, OI, serum SOD level, and Nrf2 protein expression in lung tissue were significantly decreased ( P<0.05). Compared with the Salvia miltiorrhiza extract group, the above pathological damage symptoms were aggravated in the Salvia miltiorrhiza extract + ML385 group; the ascites volume, ROS level in lung tissue, W/D, Holfbauer score, PaCO 2 and MDA level in serum were increased ( P<0.05), while PaO 2, OI, SOD level in serum and Nrf2 protein expression in lung tissue were decreased ( P<0.05). Conclusions:Salvia miltiorrhiza extract can protect lung tissue of rats with acute pancreatitis and lung injury by activating Nrf2/ARE signal.

ABSTRACT

Objective: To explore the biological mechanisms of tibial transverse transport (TTT) for promoting microcirculation and tissue repair. Methods: The clinical application and animal model study of TTT were reviewed. Results: The possible biological mechanisms of TTT for promoting microvascular network formation and tissue repair: ① Tibial corticotomy reduces intramedullary pressure and improves microcirculation; ② Tension stress stimulation promotes microvascular regeneration and accelerates the formation of new "transcortical vessels" network; ③ Systemic mobilization of stem cells, mediating local inflammation, etc. Conclusion: TTT has been clinically proven to be effective for the management of lower limb ischemia and diabetic foot ulcers. The surgical procedure is relatively easy with little adverse effects on bone structures. The TTT has good application prospects despite the biological mechanisms of which still need further exploration.

ABSTRACT

Objective:To research the clinical effect of abdominal total hysterectomy on the patients with uterine fibroids and the serum vascular endothelial growth factor (VEGF) and interleukin 6 (IL-6),interleukin 8 (IL-8) levels.Methods:106 cases of patients with uterine fibroids from February 2014 to February 2016 were divided into the control group and the research group according to the lottery method,the control group was treated with total abdominal hysterectomy,while the research group was given abdominal total hysterectomy treatment,the operation time,blood loss,anal exhaust time,length of hospital stay,changes of serum VEGF,IL-6,IL-8,Follicle stimulating hormone (FSH),estradiol (E2) levels,resistance to seedling le tube hormone (AMH) before and after the surgery as well as the occurrence of complications were compared between two groups.Results:The operation time,blood loss,length of hospital stay,anal exhaust time of research group were significantly shorter than those of the control group (P<0.05).Before surgery,the serum levels of VEGF,IL-6,IL-8,FSH,E2,AMH showed no difference between two groups (P>0.05).After the operation,the serum levels of IL-6,IL-8,E2,AMH of research group were lower than those of the control group (P<0.05),the serum level of FSH in research group was higher than that of the control group (P<0.05).The serum level of VEGF showed no difference between the two groups (P>0.05).The incidence rate of complications in research group was lower than that of the control group (P<0.05).Conclusion:Abdominal total hysterectomy showed similar effect with the abdominal hysterectomy in the treatment of uterine fibroids patients,but the abdominal full hysterectomy induced less trauma,promoted the recovery of ovarian function after surgery,which might be related to the decrease of serum IL-6,IL-8 and VEGF levels.

ABSTRACT

Objective To investigate whether clinical,endocrine,and sonographic characteristics of infertile women with polycystic ovary syndrome (PCOS) could predict the ovarian response to clomiphene citrate (CC) medication.Methods A prospective study was conducted,85 women with PCOS were given CC 50 mg per day for 5 days.In the case of an absent response,doses were increased to 100 daily in subsequent cycles.Clinical,endocrine,and sonographic characteristics were compared between responders and nonresponders.Results The cycle history (oligomenorrhea or amenorrhea) and antral follicle count (AFC) were significantly different in responders from those in nonresponders (P < 0.05).There were no differences in age,serum testosterone,follicle stimulating hormone (FSH),luteotropic hormone (LH),body mass index (BMI),insulin resistant index (HOMA-IR),and glucose and insulin levels between the two groups (P > 0.05).In addition,multivariate logistic regression analysis indicated that cycle history and AFC were the predictors of ovarian response to CC treatment.Conclusions The cycle history and AFC can be used as the simple and effective parameters to predict ovarian response to CC treatment in PCOS patients.

ABSTRACT

ObjectiveTo evaluate the breast-conserving surgery for early breast cancer.MethodFrom 1993 to 2000, 86 female cases of stage ⅠⅡa breast cancer received breast conserving comprehensive therapy. Clinical data were analyzed retrospectively. ResultsThe 5 year survival rate of this group was 96.5% and the 5 year recurrence rate was 5.2%.The postoperative contour of the breast was satisfactory.ConclusionBreast-conserving surgery is safe and effective therapy for early breast cancer, when strict indication and necessary safe margin of resection is obeyed by. Postoperative radiotherapy and chemotherapy should be properly arranged and the patients should be closely followed-up.
