ABSTRACT

Objective To investigate regeneration and repair effect after ChABC,GDNF and Nogo-A Ab combination treatment for experimental spinal cord injury model. Methods Rat (T7-8 )complete spinal cord injury crosscutting animal model was established. The SD rats were randomly divided into 6 groups:normal group, sham operation group, simple transection group,A (ChABC)group,G (GDNF)group,N (Nogo-A antibody) group, and AGN (ChABC+GDNF+Nogo-A antibody)group. At 24 w after spinal cord injury,BDA tracer,NF-200,GAP-43,and GFAP immunohistochemistry were evaluated. Results BDA tracer of A group,G group and N group showed dye light, the proximal end of damaged zone showed the blue tracer particles, while damaged zone showed few blue regenerated nerve fibers.AGN group showed visible blue nerve fibers through the damaged zone and the distal segment in the damaged zone;the central zone of injury vacuolar degeneration showed the blue dyed fibers.NF-200 immunohistochemical staining showed NF-positive staining in A group,AGN was stronger than that in control group and simple transection group (P 0.05 ).SEP wave was detected in control group and AGN group, while the latency time was longer in AGN group than in control group. Conclusion ChABC,GDNF,and anti-Nogo-A antibody used alone or in combination can improve spinal cord injury and nerve cell function, and the joint application could improve regeneration after spinal cord injury than any monotherapy.

ABSTRACT

This experimental study was intended to find the possibility of using PDLLA as scaffolds in tissue engineering and to assess the effect of PDLLA scaffolds with different porosity on chondrocytes. Chondrocytes were cultured when they were seeded onto PDLLA scaffolds with different porosity. Under scanning electron microscope, the scaffolds in hydrophila and adsorptivity to chondrocytes and the function of the cells were observed. The chondrocytes were noted to proliferate on PDLLA scaffolds with different porosity, those with 90%-96% porosity being significantly better. So PDLLA can be used as a scaffold in tissue engineering, and the proper porosity of PDLLA scaffolds should be 90% approximately 96%.

ABSTRACT

This study was designed to assess the effect of a home-bred poly-DL-lactide (PDLLA) rods on experimental fracture healing. An operation was performed to create bilateral ulnar and radial fractures of forearm in 15 rabbits. The right sides were fixed with 2 PDLLA (Mv = 34 x 10(4), 1.6 mm x 1.6 mm x 30 mm) rods, and the contralateral sides were fixed with 2 Kirschner wires to be used as controls. After operation, casts were used for external fixation, and the joint above and the joint below the fracture were immobilized. The casts were removed at the 6th week after operation. The specimens were removed at specific intervals between 2 and 12 weeks. Histologically, the light microscopy and scanning electron microscopy evaluation of the rabbit ulnar and radial fractures fixation revealed that, the fractured bone gradually healed and the PDLLA rods properly fixed on the fracture area during the implanted period. However, results suggest that internal fixation by absorbable material made the course of fracture healing slower than that in the control group, but the osteogenesis appeared to be normal. In the absorbable group, the collagenous fiber was characterized by order and orientation, and calcified normally. In conclusion, this kind of PDLLA rods have an effect on delaying the process of the normal union of fracture and should be applicable to fractures where the tissues are rich in blood supply.

ABSTRACT

Objective To study the role of HMGA2 in the occurrence and the progression of uterine leiomyomas. Methods Fifty cases of uterine leiomyomas who met the inclusion criteria(aged 30-50 years old, free from hormone at least for 6 months before surgery,no other diseases, postoperative pathological diagnosis as uterine leiomyoma) admitted to our hospital from January to July 2003 were studied. The expression of HMGA2 mRNA in uterine leiomyomas and neighbour normal myometrium was detected by ISH.Results HMGA2 mRNA was strongly expressed in 30 cases of uterine leiomyomas but not expressed in neighbour normal myometrium. There was no difference in HMGA2 mRNA between proliferative stage and progestational stage. Conclusion The increased expression of HMGA2 in uterine leiomyomas may be important in the occurrence and the progression of uterine leiomyomas.
