ABSTRACT

Objective: To explore the value of thyroid imaging report and data system (TI-RADS) recommended by American Radiology of Society (ACR) in 2017 for differentiating thyroid benign and malignant nodules. Methods: Ultrasonic images of 497 thyroid nodules diagnosed with pathology were retrospectively analyzed using TI-RADS classification and given score and classification. The optimal critical score was determined with ROC curve, and the sensitivity, specificity and accuracy were calculated. The efficacy and consistency of senior and junior physicians in diagnosis of benign and malignant thyroid nodules using this optimal critical score were evaluated. Results: The area under ROC curve was 0.883 (P<0.001), and the optimal critical score was 5, while the sensitivity and specificity was 86.22% and 78.68%, respectively. The sensitivity, specificity and accuracy of senior physicians in diagnosis of benign and malignant thyroid nodules was 78.22% (176/225), 76.47% (208/272) and 77.26% (384/497), of junior physicians was 77.33% (174/225), 74.26%(202/272) and 75.65% (376/497), respectively, and moderate consistency was found between senior and junior physicians (Kappa=0.581). Conclusion: TI-RADS recommended by ACR in 2017 has high clinical value, and the optimal critical score for diagnosing benign and malignant thyroid nodule is 5.

ABSTRACT

Gut microbial ecosystem is closely related to the immune system and influences human health. Fusobacterium nucleatum ( F. nucleatum) is one of the species of great concern. Increasing evidence shows that F. nucleatum promotes intestinal inflammation and influences the tumor-associated immune micro-environment. This review focused on the recent progress in the influence of F. nucleatum on intestinal immu-nity and the pathogenesis of intestinal diseases in order to discover the immune mechanism of F. nucleatum-associated intestinal diseases and find new therapeutic targets for immunotherapy.

ABSTRACT

Objective To study on the CDKN2/P16 gene in primary osteosarcoma.Method By using molecular biological methods that inclued genome DNA extraction from paraffined tissue and PCR SSCP analysis technique, we studied alternations of CDKN2/P16 gene in 25 primary osteosarcomas.Results (1)The deletions frequency in differentiation degree of osteosarcomas was① bone brood cell, 16.7% ;② cartilage brood cell,12.5% ;③ Fiber brood cell:20% ,(P >0.05).(2)The deletion frequency in male patients was 17.6% , female patients 12.5% ,(P >0.05).(3)In early metastatic osteosarcomas the deletion rate was 33.3% ,which was significantly higher than that of the control group with the rate of 10.5% (P< 0.05).(4)The deletion rate was 16% and the mutations were not found.Conclusion (1)The deletion rate was 16% and the mutations were not found.This suggests that the deletions of CDKN2/P16 gene were closely related to the genesis of primary osteosarcoma and that the main type of the alternation of CDKN2/P16 gene was deletion.(2)In early metastatic osteosaarcomas the deletion rate was 33.3% , which was significantly higher than that of the control group with the rate of 10.5% .This indicates to great extend that the deletions of CDKN2/P16 gene were closely related to the metastatic ability.(3)The deletions frequency had no significant relationship with differentiation degree of osteosarcomas, so was with the sex of the patient.

ABSTRACT

Human osteosarcoma cells (HOS-8603) cultured in vitro were injected into the medullary cavities of femur in 5 8-weeks aged NC nude mice and 5 6-weeks aged NC nude mice and in 7 of these animals human osteosarcoma grew. Osteolytic damage, parostium reaction and mass formation were observed on X-ray film. The pathologic findings in this study were similar to the characteristics of original tumor cells and AKP stain was positive.Anti-osteosarcoma monoclonal antibody labeled with 125I was used to carry out experimental guiding diagnostic study in these animal models with a high specificity.

ABSTRACT

For the purpose of improving the accordance rate with early diagnosis of osteosarcoma, we used the osteosarcoma cell line (HOS-8603) to establish 6 cell lines of anti-human osteosarcoma monoclonal antibodies (OSMAb) and sifted out 2 lines (OSMAb-H7, OSMAb-B4) through ELISA and immunofluorescent methods, which could secrete antibodies with high specificity. By antibodies prepared, purified and labeled with 125I we performed a series of experiments in NC nude mouse osteosarcoma models of soft tissue and bone tissue, and found the guiding localization showed stronger specificity. Then we chose OSMAb-B4 treated by bacterial filtration and labeled with 131I to do ?-scintigraphical localization examinations in 8 cases of bone tumor and obtained satisfactory results.
