ABSTRACT

Objective To investigate the relationship between gene expression of endothelin-3(ET-3) and inflammation of acute pancreatitis(AP) in rats.Methods Fifty-four rats were divided randomly into 4 groups:sham operation group,AP group,arterial injection group and vein injection group.AP was induced by reverse intra-bile duct infusion 4.5% sodium taurocholate,treated with low dose dopamine 〔5 ?g/(kg?min)〕 by injecting arterial or tail vein.Rats were sacrificed at 1,6 and 24 h after the induction of AP.The mRNA expression of ET-3 was evaluated by semi-quantitative reverse transcription-polymerase chain reaction(RT-PCR) and pathological changes was observed in rats.Results Expression of ET-3 mRNA could be detected from 1 up to 24 h after the induction of pancreatitis.Expression of ET-3 mRNA of sham operation group was decreased significantly compared with other three groups.Expression of ET-3 mRNA showed a significant decrease by arterial injection dopamine than that by tail vein(P

ABSTRACT

Objective To investigate the effects and mechanism of Panax Notoginseng saponins (PNGS) on rat liver during cold preservation. Methods Using isolated perfusion of rat liver model (IPRL), Fura 2 method was used to measur the concentration of calcium ion in hepatic cells which had been preserved in DMEM solution with different concentration of PNGS added and cold preserved for 2 hours. Liver function, metabolic products of oxygen free radicals, energy substance and aucount of biliary flow as well as morphological study were measured from liver tissue which had been cold preserved in lactate riuges’s solution containing different dosages of PNGS for 24 hours and 30 minutes reperfusion.Results The contents of intracellur calcium of the rat hepatocytes,MDA, AST, ALT, LDH were lower than those in the control group,but SOD, ATP, TAN, EC and bile production were higher than those in the control group( P 0.05). Conclusion PNGS relieve the injury of the rat liver during cold preservation. The mechanism might through inhibition calcium overload, improve the energy metabolism, play a role against free radical injury realize.

Objective To study the effect of hepatocyte growth factor (HGF) on hepatocytes after partial hepatectomy in chronic liver injury rats.Methods 30% hepatectomy was performed in rats with cirrhosis induced by CCL 4 and alcohol,and subcutaneously injection of cow HGF into intraabdomen was done with different dosages.Results Small dose (0.16mg?100g -1d -1)of HGF did not stimulate liver regeneration or enhance liver function after partial hepatectomy in cirrhotic rats.Large dose (1.3mg?100g -1d -1) of HGF stimulated liver regeneration and enhanced liver function after partial hepatectomy in cirrhotic rats(P

ABSTRACT

Objective Study the effect of induced differentiation of abscisic acid (ABA) on human HCC cell line SMMC-7221.Methods Cultured SMMC-7221 cells were treated separately with RPMI-1640 culture medium, HMBA and ABA with different concentrations. Firstly, the appropriate concentration of ABA which inhibits SMMC-7221 cell proliferation was selected with the modified MTT method. Then electron microscopy was performed to observe the changes of microstructure. The cell cycle was analyzed by flow cytometry. Results Apart from 4?10 -4mmol/L concentration of ABA, the others could inhibit the cell proliferation. The inhibition rate increased with the time prolonging and the concentration increasing. The effect was most obvious with 4?10 -3mmol/L ABA. At this concentration the cells were arrested in G0/ G1 phase (P
