ABSTRACT

<p><b>OBJECTIVE</b>To study the physiologic indexes, yield and the contents of alkaloids of Fritillaria cirrhosa D. Don under different temperature.</p><p><b>METHOD</b>The growth temperatures (15, 20, 25, 30 degrees C) of F. cirrhosa were controlled by using artificial climate, the growth was observed, the contents of chlorophyll a and b, soluble sugar, MAD, proline of the leaves of F. cirrhosa were tested, and the yield and the alkaloids content of the bulbs were analyzed.</p><p><b>RESULT</b>The growth period of F. cirrhosa under 15, 20 degrees C were appropriately extended. The difference of the content of leaves chlorophyll b under four temperatures and the contents of total chlorophyll and chlorophyll a under 15, 20, 30 degrees C were not significant. The contents of soluble sugar, MAD and proline of leaves and the growth ratio, dry weight and content of alkaloids of bulb increased with the temperature decrease.</p><p><b>CONCLUSION</b>Higher temperature is not suitable for the growth of F. cirrhosa. Under the relatively lower temperature, the growth period of F. cirrhosa extended, the bulb can grow properly, and the content of alkaloid increased. F. cirrhosa can improve its cold tolerance by increasing the content of proline and soluble sugar, and it also can maintain the normal content of chlorophyll under the lower temperature.</p>

ABSTRACT

Using SDS-PAGE analysis, the High-molecular-weight (HMW) glutenin subunits of two Crithopsis delileana accessions were detected. It was found that the two accessions had the same HMW glutenin subunits. Only one HMW glutenin subunit with the similar electrophoresis mobility to the y-type HMW glutenin subunit of hexaploid wheat was observed in C. delileana. However, It was indicated that this glutenin subunit was an x-type glutenin subunit Kx by gene sequence analysis. The full coding region of Kx gene is 2052bp and could encodes a mature protein with 661 amino acid residues. The Kx gene could be expressed in the bacterial expression system, and the expressed protein had the same electrophoresis mobility as that in the seed of C. delileana. The primary structure of Kx subunit was very similar to the x-type HMW glutenin subunits encoded by the A, B and D genomes of wheat, the C and U genomes of Aegilops, and the R genome of Secale cereale. In the phylogenetic analysis, Kx subunit was clustered together with Ax1 subunit by an interior parallelled branch. In conclsion, Kx gene is a novel x type glutenin subunit gene from C. delileana.

ABSTRACT

The alpha-amylase inhibitors have been proposed as possibly important weapons against pests. Thus, it is of importance to identify the specificity of them. Based on the EST data of alpha-amylase inhibitor genes that were retrieved from NCBI, BBSRC and GrainGenes, two PCR primers were designed. The coding sequences of 24 kD dimeric alpha-amylase inhibitors with resistance to insects in 17 wheat and Aegilops accessions were investigated and 17 new genes were obtained. Only one 24 kD alpha-amylase inhibitor gene was found in each diploid wheat and Aegilops accession, whereas 8 genes were characterized from one hexaploid wheat variety, indicating that the 24 kD alpha-amylase inhibitors in hexaploid wheat were encoded by multi-gene. The deduced amino acid sequences of 2 genes from common wheat and 1 gene from Ae. tauschii were the same as the sequence of the inhibitor 0.19, and the deduced amino acid sequence of another gene from common wheat was similar to the inhibitor 0.53 with only one amino acid difference. The amino acid sequences of 24 kD dimeric alpha-amylase inhibitors shared very high coherence (91.2%). These results suggest that the alpha-amylase inhibitors in 24 kD family were derived from common ancestral genes by phylogenesis.

ABSTRACT

<p><b>OBJECTIVE</b>To construct the fast propagation system of Houttuynia cordata Thunb. new line by selecting the proper type and concentration of plant growth regulators and proper explants.</p><p><b>METHOD</b>The different explants of Houttuynia cordata new line were cultured on MS media with different concentration of different plant grow regulators.</p><p><b>RESULT</b>Stem node over-ground and shoot tip were optimum explants for fast propagation. The stem which node grew on MS medium with KT at 0.5 mg x L(-1) could induce more leaves and grow higher. The shoot tip cultured on MS medium with 1.0 mg x L(-1) 6-BA could induce better callus. Adventitious shoots were better achieved from over-ground nodal explants cultured on MS medium supplemented with 6-BA at 1.0 mg x L(-1) and NAA at 0.2 mg x L(-1), or 6-BA at 0.5 mg x L(-1) and NAA at 0.2 mg x L(-1). MS medium with IAA at 0.2 mg x L(-1) was the best one on inducing roots.</p><p><b>CONCLUSION</b>The propagat coefficient can be highly improved by inducing adventitious shoots through stem node over-ground, and thus plentiful seeds for production can be provided.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the yield of different Houttuynia cordata and the changing regularity of their main officinal components in different periods, and provide a basis for harvesting in good time and breeding new varieties.</p><p><b>METHOD</b>The main officinal composition and wet weight of overground parts per acreage of different accessions were determined periodically. The plot yields were tested when harvested and analyzed by analysis of variance.</p><p><b>RESULT</b>The difference of the yields of different accessions was significant. The varying tendency of the content of main officinal component methyl nonyl ketone and its total content per acreage of different accessions were different.</p><p><b>CONCLUSION</b>The integrate characters of Qionglai and Wangyu wild accessions are relatively better, worthy to be widely applied in production.</p>
