ABSTRACT

Objective:To monitor the cerebral vascular blood flow parameters in the early stage of simulated acute exposure to high altitude hypoxia by transcranial color Doppler (TCCD), and to evaluate the change trend of cerebral hemodynamics and cerebrovascular reactivity.Methods:Sixty-four healthy volunteers were selected to observe the changes of peak systolic flow velocity(Vs), end diastolic flow velocity(Vd), mean flow velocity(Vm), resistance index (RI) and pulsatility index (PI) of middle cerebral artery (MCA) 30 minutes after they quickly entered the simulated altitude of 4 500 meters. Combined with breath holding test, breath holding index (BHI) was used to evaluate cerebrovascular reactivity (CVR), and subjects were divided into ≤30 years old group and >30 years old group, and the changes of CVR after hypoxia of the two groups were compared.Results:In the early stage of hypoxic environment, compared with baseline, SpO 2 decreased, heart rate increased, and blood flow velocity of middle cerebral artery(Vs, Vd, Vm) increased significantly, BHI showed a decreasing trend (all P<0.01). After hypoxia, the BHI rate of change in >30 years old was lower than that of the subjects ≤30 years old ( P<0.05). After breath holding, cerebral blood flow velocity increased significantly, PI and RI decreased significantly (all P<0.01). Conclusions:Cerebral blood flow is very sensitive to hypoxia. The application of TCCD technology can evaluate the trend of cerebral blood flow dynamics and cerebrovascular reserve capacity in real time and accurately, which is helpful to provide objective basis and research basis for the prevention and treatment of high altitude hypoxia.

ABSTRACT

Objective To investigate the expression and clinical significance of cysteine protease inhibitor A(CSTA) in esophageal squamous cell carcinoma. Methods A total of 59 patients with esophageal cancer who underwent esophagectomy or endoscopic submucosal tumor dissection were enrolled. The esophageal squamous cell carcinoma and normal esophageal tissues were collected and clinical pathological data were collected. The expression of CSTA mRNA and protein in cancer tissues and normal tissues was determined by real-time quantitative fluorescent polymerase chain reaction (RTFQ-PCR) and immunohistochemistry. The expressions of CSTA and Ki-67 mRNA and protein in cancer tissues and normal tissues were determined by RTFQ-PCR and Western Blot. Results Compared with normal, the expression of CSTA mRNA and protein in esophageal squamous cell carcinoma tissues was significantly lower, and the difference was statistically significant (all P<0.05). In squamous cell carcinoma, the CSTA-positive expression is often associated with Ki-67 expression, whereas normal esophageal tissue has CSTA expression but no Ki-67 expression. Squamous cell carcinoma with CSTA-positive expression had higher tumor pT stage and tumor grade (all P<0.05). Conclusions The expression of CSTA in cancer tissues of patients with esophageal squamous cell carcinoma is significantly lower than that in normal tissues. The CSTA-positive expression in esophageal squamous cell carcinoma is related to the pT clasification and tumor grade. The CSTA test for esophageal squamous cell carcinoma can provide a basis for clinical treatment.

ABSTRACT

BACKGROUND@#Non-small cell lung cancer (NSCLC) is a kind of lung cancer, because its high incidence has been concerned. Therefore, it has great significance to reveal the pathogenesis of NSCLC. As a transcriptional regulatory factor, MATF-A plays an important role in the development of multiple tumors, can regulate the migration process of a variety of tumor cells. HOTAIR is a long non-coding RNA (LncRNA) found in recent years, which expresses abnormally in multiple tumors and is involved in the proliferation and migration of multiple tumors. The aim of this study is to explore the role of MRTF-A through HOTAIR to regulate the proliferation and migration of NSCLC cell A549 cell.@*METHODS@#We constructed the overexpression plasmid and interfering plasmid of MRTF-A, and detected the effect of MRTF-A on the proliferation and migration of A549 cells by CCK8 and wound healing methods respectively. Then, we designed the siRNA of HOTAIR to detect its effect on the proliferation and migration of A549 cells. Through qRT-PCR, we detected the effect of MRTF-A on HOTAIR expression. Finally, we constructed HOTAIR's promoter, and detect the effect of MRTF-A on HOTAIR promoter activity by luciferase reporter gene test.@*RESULTS@#Overexpression of MRTF-A promotes the proliferation and migration of A549 cells, while silent MRTF-A inhibits its proliferation and migration. Next, we found that interfered HOTAIR expression inhibited the proliferation of A549 cells. We found that MRTF-A could influence the expression of HOTAIR and regulate the activity of HOTAIR promoter.@*CONCLUSIONS@#MRTF-A regulates the proliferation and migration of A549 cell through HOTAIR.

ABSTRACT

Objective To study and observe the clinical efficacy of using non skid plate by posteromedial approach for fixing splitting or compression tibial plateau fracture.Methods Twenty eight cases of patients who were treated for fixing splitting or compression tibial plateau fracture from January 2014 to January 2016 in people's hospital of Xinyu were retrospectively reviewed.All patients were treated byposterior medial approach with non skid plate for fixation,all patients were followed up by postopera tive imaging and clinical follow-up to observe its clinical curative efficacy.Results Twenty-eight patients were followed up,the mean follow-up time was 12 months.All patients had no internal fixation loosening fracture,send,vascular and nerve damage and other adverse reactions.All the patients were healed within 4 to 7 months,the average healing time was 18.5 weeks.After healing,knee joint function was evaluated,18 cases were excellent,5 cases were good,the excellent and good rate was 82.1 %.Conclusion The efficacy of using non skid plate by posteromedial approach for fixing splitting or compression tibial plateau fracture is good,it is worthy of clinical application.

AIM: To investigate the expression level of transcriptional factor Elf-1 in mononuclear cells, CD4~+ and CD8~+ T cells from umbilical cord blood (UCB). METHODS: Real-time PCR with SYBR green I technique was used for detecting the Elf-1 expression in mononuclear cells, sorted CD4~+ and CD8~+ T cells from 12 cases of umbilical cord blood. The relative mRNA expression level of Elf-1 was analyzed by a formula of 2~(-△Ct)×100%. The expression level of β_2-microglobulin gene (β_2M) was used as an endogenous reference. The peripheral blood from 10 cases of healthy adults was served as control. RESULTS: Elf-1 mRNA expressed in all blood samples collected from both UCB and healthy adults. The expression level showed apparent diversity in different individuals. The relative mRNA expression of Elf-1 in both mononuclear cells (18.55%±2.48%) and CD8~+ T cells (3.52%±0.45%) from UCB were significantly higher than those from healthy adults (9.16%±1.92%, 2.02%±0.27%, respectively, P<0.01, P<0.05). CONCLUSION: The results of Elf-1 expression level from umbilical cord blood indicate that the over-expression of Elf-1 gene in mononuclear cells and CD8~+ T cells might be one of the features of T cell immune state in umbilical cord blood.

ABSTRACT

Objective To summarize our clinical experience of prophylaxis and treatment for complications associated with catheterization of brachial artery. Methods 87 patients underwent endovascular treatment via brachial artery. Complications associated with catheterization of brachial artery were retrospectively analyzed. Results Under ultrasonic guidance the catheterization procedure of brachial artery was successful in all 87 patients. The success rate of cannulation was 100%. In 53 patients(61% ) ultrasonic guided cannulation was successful at the first attempt, the other 35 (39% ) with 2 or more than 2 times puncture. Complications associated with catheterization of brachial artery were detected in 16 cases. The complication rate was 18. 4%. Guidewire insertion into peri vascular compartment took place in 4 cases (4. 6% ). Episodes of local hematomas were noted in 11 cases( 12. 6% ) and in three of the 11 cases nerve injuries were detected (one of which was of late-onset type). Brachial artery pseudoaneurysm was found in 1 case(1.2%). Conclusion Familiarity with the anatomical features of the brachial access, skillful application of the techniques for the catheterization, full understanding of complications and sufficient preparation of treatment strategies can prevent those complications effectively.

ABSTRACT

Objective To develop a portable sleep monitoring system with low burden. Methods Information from signals related with sleep were used and dug. Electrodes from patients' body were reduced to decrease patient's physiological and psychological burden. Results Polysomnography(PSG) is the gold standard in detecting the Sleep Apnea Hypopnea Syndrome disease at present. While this method has its problems, such as complicated operation, large expenditure and big physiology and psychology burden for the patient. A sleep monitor system was introduced which has the advantages of convenience, practicality, small physiology and psychology burden for the patient. Conclusion The result indicates that this device is warmly welcomed by doctors and patients, and it is a new device for the diagnosis of the SAHS disease.

Objective To design a wireless monitoring system which can obtain patients' multi-parameters (such as ECG, respiratory, blood pressure, SPO2, and heart rate). Methods The wireless communication system, based on CDMA, receives data through internet and analyses ECG by dealing and measuring signals. Results A wireless monitoring system is designed, which can analyse ECG, blood pressure, and respiratory. Doctors can give diagnostic suggestion based on the monitoring system's analyses and send it to patients by short messages. Conclusion This system can realize long -distance medical monitoring and report the diagnosis to patients timely.

ABSTRACT

Objective To evaluate clinical effects of endovascular angioplasty and stenting for treating lower extremity arteriosclerotic occlusion(ASO).Methods After the identification of the stenosis or occlusion by angiography via femoral artery access by percutaneous puncture or incision puncture,recanalization of the occlusion was done by ultrasound ablation or guildewire clearance.Then the percutaneous transluminal angioplasty(PTA) was performed and an endovascular stent was introduced.Results A total of 45 cases(53 legs) had undergone endovascular angioplasty and stenting.The procedure was technically successful in all the patients.The ankle brachial index(ABI) increased from 0.36?0.14 preoperatively to 0.77?0.21 on the 7 postoperative day((t=2.397),P=0.021).Follow-up checkups in all the cases for 6~54 months(mean,23 months) revealed a primary patency rate of 90.6%(48/53).Conclusions Endovascular angioplasty and stenting is a safe and effective method for treating lower extremity arteriosclerotic occlusion.

ABSTRACT

AIM: To investigate the effect of tea polyphenols (TP) on alcohol-induced liver injury in mice.METHODS: 40% alcohol(0 5 mL) and TP (5 mg) were administered intragastrically, the liver MDA level and serum alanine transaminase (ALT) activity were determined. The effect of TP on MDA level in alcohol-treated liver in vitro was also examined.RESULTS: Pretreatment with TP significantly inhibited alcohol-induced liver MDA increase in mice in vivo and in vitro , the increase in serum ALT induced by alcohol was also reduced by pretreatment with TP (0 5 mg). TP at a dose of 5 mg, administered 1 h after alcohol treatment, also suppressed increase in liver MDA level stimulated by alcohol. CONCLUSION: These results demonstrated that TP has a protective effect on alcohol-induced liver injury in mice.
