ABSTRACT

【Objective】 To prepare positron emission computed tomography (PET) molecular probes targeting macrophages using Nb119 and BCⅡ10 labeled with 68Ga nuclide. 【Methods】 To explore the labeling conditions of nanobodies with 68Ga nuclides, first, the anti-Vsig4 nanobody Nb119 and isotype control antibody BCⅡ10 were incubated with NOTA and then purified to obtain the chelating agent-modified NOTA-Nb119 and NOTA-BCⅡ10. The NOTA-Nb119 and NOTA-BCⅡ10 were further incubated with 68Ga. Finally, NOTA-Nb119 was identified by SDS-PAGE and MALDI-TOF methods, and the radiochemical purity was detected by radio-HPLC. 【Results】 The results of SDS-PAGE showed that the lanes of the successfully labeled NOTA-Nb119 had apparent hysteresis than the unlabeled nanobody band, which proved that the molecular weight of the labeled product was increased and NOTA successfully modified the nanobody. Subsequently, 68Ga nuclides could successfully label Nb119 and BCⅡ10. According to radio-HPLC detection, the radiochemical purity of NOTA-Nb119 and NOTA-BCⅡ10 labeled with 68Ga was greater than 90% and remained stable. 【Conclusion】 68Ga-labeled nanobodies have high radiochemical purity and high stability, indicating that this study can be applied to other nanobodies for modification and labeling and provides a practical and feasible method for the preparation of PET using nanobodies.

ABSTRACT

Hepatocellular carcinoma (HCC) most often develops in patients with liver disease characterized by chronic non-resolving inflammation. The inflammatory response is mainly derived from innate immune cells, and tumor-associated macrophages (TAMs) play an important role in the development of tumors.It is usually chemotactic from mononuclear progenitor cells in the blood to tumor tissue, then is induced by the tumor microenvironment and further develops into TAMs. They play an important role in promoting tumor growth,angiogenesis and tumor invasion and inducing tumor tissues to form an environment without inhibition mechanisms. As the relationship between TAMs and malignant tumors becomes clearer, TAMs are beginning to be seen as therapeutic targets. The heterogeneity of TAM subtypes and their origin and dynamic phenotype during the initiation and progression of HCC has been partially unraveled. It further forms the base for developing therapeutic agents by decreasing the population of TAMs via blocking recruitment of bone marrow-derived monocytes and functionally reprograms TAMs to anti-tumoral behavior. This review focuses on the preclinical evolution and hitherto clinical trials for TAM-targeted therapy in HCC.

ABSTRACT

BACKGROUND:How to get a lot of stable and dynamic human umbilical cord mesenchymal stem cels is stil a difficulty. OBJECTIVE:To investigate the best culture way of human umbilical cord mesenchymal stem cels by comparing tissue explant, enzyme digestion, enzymolysis methods. METHODS:Ten fresh umbilical cords were isolated to human umbilical cord mesenchymal stem cels by using tissue explant, enzyme digestion, and enzymolysis methods, respectively. Hereafter, we compared cels harvested using three culture methods in terms of the time for the primary cels to creep, successful rates of cel-culturing, and curves of cellgrowth. Surface markers of cels were detected by flow cytometry and multiple differentiations of cels were detected. RESULTS AND CONCLUSION: The time for the primary cels to creep out in the enzymolysis and enzyme digestion groups had no significant difference, but both of them were significantly shorter than that in the tissue explant group (P < 0.01). The successful rate of primary cellculture in the enzymolysis group was significantly higher than that in the other two groups. The cellproliferations of three groups had no significant difference. The cellsurface markers and differentiation ability of the third generation of human umbilical cord mesenchymal stem cels cultured by enzymolysis method met the characteristics of mesenchymal stem cels. In conclusion, the enzymolysis method can shorten the time for primary human umbilical cord mesenchymal stem cels to creep out and increase the successful rates of primary human umbilical cord mesenchymal stem cels.

ABSTRACT

BACKGROUND:Studies have found that combination of two of chitosan (CS), nano-hydroxyapatite (nHA) and poly(lactide-co-glycolide) (PLGA) can improve the mechanical properties and biocompatibility of the composite stent in certain extent as wel as improve osteogenic differentiation of the cels, but there is a certain distance from the ideal bone tissue engineering scaffolds. OBJECTIVE:To study biocompatibility and osteoinductive activity of nHA/CS/PLGA scaffolds with different proportions in vitro. METHODS: nHA/CS/PLGA scaffolds were prepared at mass ratio of 10:10:80, 10:20:70, 20:10:70 respectively by particle leaching method. And human bone marrow stem cels (hBMSCs) were co-cultured with these scaffolds in vitro. Adhesion, proliferation, and osteoinductive activity of these scaffolds were examined qualitatively and quantitatively by growth curve of hBMSCs on scaffolds. Gene expression of alkaline phosphatase activity and osteocalcin was detected by RT-PCR. RESULTS AND CONCLUSION: hBMSCs could be attached, proliferated, and osteoinduced better on the nHA/CS/PLGA scaffold with the mass ratio of 20:10:70, compared to the other two groups of scaffolds. The differences were significant statisticaly (P< 0.05). Alkaline phosphatase and osteocalcin expressions were respectively higher in the scaffold with the mass ratio of 20:10:70 after 9-27 days of co-culture and 15-27 days of co-culture, in comparison with the other two groups of scaffolds. These findings indicate that the nHA/CS/PLGA scaffolds with the mass ratio of 20:10:70 demonstrated preferable biocompatibility and osteogenic inductivity, which is expected to be a promising scaffold material for bone tissue engineering.

ABSTRACT

Objective To investigate the sleep self-adaptation for Chinese military personnel and the correlations with sleep quality. Methods 857 soldiers completed the Sleep Quality Scale of Chinese Military Personnel (SQSCMP) and the Soldier Sleep Self-Adaptation Scale (SSSAS). Results The average level of sleep selfadaptation of Chinese military personnel was 2.89, below the theoretical midpoint score 3. There were significant positive correlations between the sleep quality and the global level of sleep self-adaptation and its 5 factors ( r =0. 16 ~ 0. 533, P < 0.001 ), except the factor of perceived importance of sleep ( r = -0.027 ). Multiple stepwise regression analysis showed that the major determinants of the sleep quality were the factors of positive attitudes about sleep,the positive self-suggastions on sleep and the ability of anti-disturbance of sleep. Conclusion The positive and appropriate attitudes and beliefs about sleep are of benefit to sleep quality.

ABSTRACT

Objective To sum up the experiences of successful long-distance transportation of 35 burned casualties as a result of a mass fire catastrophe.Methods Thirty-five burned victims of a mass fire catastrophe were transported by air from a northern part of Heilongjiang province to Beijing,a voyage of 1600km.The successful experiences of organization and medical instructions during the transportation were summed up and analyzed.Results The transportation was launched 20 hrs after the arrival of specialists group.Under a well-knitted organization and meticulous care,all the casualties were transported to the destination safely and smoothly in 4 hrs.Conclusions Before transportation the condition of the patients to be transported and their tolerance to the ordeal of transportation must be evaluated or judged according to the general condition of each patient,and due precaution should be carefully taken.Medical staff,drugs and equipments for circulatory and respiratory care must be provided for immediate care of any life-threatening complications which might occur during the air-lift and road transportation.Before transportation tracheostomy tube and venous line should be secured,especially for those with head and face burns,the airway must be kept free.The patients were tagged with number and with their conditions documented on wound tags to ensure the patients to receive prompt and proper care in accordance to their condition in the accepting hospital.During airlift,the stretchers should be arranged close to the exit of the plane and perpendicular to the longitudinal axis of the cabin.Ample space should be kept between stretchers to allow emergent care.The hospital of destination must check the wound tags carefully and try its best to treat the patients as earlier as possible.All these efforts were contributory to successful transportation of these mass burn casualties.
