ABSTRACT

The clinical utilization of doxorubicin (Dox) in various malignancies is restrained by its major adverse effect: irreversible cardiomyopathy. Extensive studies have been done to explore the prevention of Dox cardiomyopathy. Currently, ferroptosis has been shown to participate in the incidence and development of Dox cardiomyopathy. Sorting Nexin 3 (SNX3), the retromer-associated cargo binding protein with important physiological functions, was identified as a potent therapeutic target for cardiac hypertrophy in our previous study. However, few study has shown whether SNX3 plays a critical role in Dox-induced cardiomyopathy. In this study, a decreased level of SNX3 in Dox-induced cardiomyopathy was observed. Cardiac-specific Snx3 knockout (Snx3-cKO) significantly alleviated cardiomyopathy by downregulating Dox-induced ferroptosis significantly. SNX3 was further demonstrated to exacerbate Dox-induced cardiomyopathy via induction of ferroptosis in vivo and in vitro, and cardiac-specific Snx3 transgenic (Snx3-cTg) mice were more susceptible to Dox-induced ferroptosis and cardiomyopathy. Mechanistically, SNX3 facilitated the recycling of transferrin 1 receptor (TFRC) via direct interaction, disrupting iron homeostasis, increasing the accumulation of iron, triggering ferroptosis, and eventually exacerbating Dox-induced cardiomyopathy. Overall, these findings established a direct SNX3-TFRC-ferroptosis positive regulatory axis in Dox-induced cardiomyopathy and suggested that targeting SNX3 provided a new effective therapeutic strategy for Dox-induced cardiomyopathy through TFRC-dependent ferroptosis.

ABSTRACT

Objective To study the influence of volume change of tumor bed on radiotherapy dose volume distribution after breast conserving surgery for early breast cancer.Methods 13 early breast cancer patients received breast-conserving surgery were selected.For all 3 CT scanning in patients with tumors,row delineation of tumor and normal tissue target positioning,the tumor bed after volume as well as the planned radiation dose volume were compared in three times of scans.Results 13 cases of three CT scans of patients with tumor bed volume point to (32.85±9.30)cm3,(24.50±4.71)cm3,(16.48±4.46)cm3,there were statistically significant differences before and after(t=2.888,5.723,4.458,all P<0.05);3 planned radiation dose in patients with volume was stepped down,and the differences were statistically significant (F=20.28,96.32,14.21,16.75,26.29,all P<0.001).Conclusion Earlier the tumor bed following breast surgery volume decreased rapidly,tumor volume change will affect the target area of the bed covers and the exposure of normal tissue dose-volume.

ABSTRACT

Objective To discuss the application effect of lateral prone position in the left trigeminal nerve root microvascular decompression on the degree of compression on axillary nerve and arm discomfort,in order to supply evidence for selection of a suitable position during trigeminal nerve root microvascular decompression.Methods 44 cases who underwent microvascular decompression of trigeminal nerve roots were randomly divided into the observation group and the control group with 22 cases in each group.They were in lateral prone position with their arms placed in different positions.The influence on arm function was observed in two groups.Results The arm feeling,response to pain,the upper limb swelling and flexibility were statistically significant between two groups.Conclusions For lateral prone position arrangements during microvascular decompression of trigeminal nerve root,the patients' head should be moved ahead,the right arm should be out of bed and be fixed in a lower position,so that nerve and functional damage can be prevented by avoiding decompression of the right upper arm.

ABSTRACT

Objective To investigate a new method for detecting fetal cardiac arrhythmia.Methods Used two kinds of superposition:①left ventricular outflow tract color flow image with right atrium wall motion curves superposition,②left ventricular outflow tract color flow image with left ventricular inflow tract colour flow image superposition,167 cases of fetal arrhythmia were detected.Results Among them,84 fetuses with atrial premature heat,39 with ventricular premature beat,23 with temporal sinus bradycardia,12 with sinus tachycardia,4 with Ⅱ°atrioventricular block(AVB),2 with Ⅲ°AVB,3 with atrial fibrillation.Conclusions Using superposition of M-mode echocardiagraphy with color Doppler imaging to diagnose fetal cardiac arrhythmia is easier and more accurate.

ABSTRACT

The interaction of 5-fluorouracil (5-FU) with deoxyribonucleic acid (DNA) and serum albumin and the influence of some metal ions on the interaction have been studied by using spectral methods of UV-absorption,fluorescence and circular dichroism. It was shown that the interaction of 5-FU with DNA and serum albumin was effected by some metal ions. The binding costant of 5-FU with serum albumin was found to be KHSA = 4.38 × 103(mol/L)-1 and KBSA =4.95 × 103(mol/L)-1 and the distance between the bound 5-FU and the tryptophan residue was also determined to be rHSA = 3.53 nm, rBSA = 3.51 nm.
