ABSTRACT

OBJECTIVE@#To explore the clinical features and genetic basis for a child with early-onset Isolated sulfite oxidase deficiency (ISOD).@*METHODS@#A child with ISOD who was admitted to Weihai Hospital Affiliated to Qingdao University on May 10, 2020 was selected as the study subject. Clinical data of the child was analyzed. The child and her parents were subjected to trio-whole exome sequencing, and candidate variants were verified by Sanger sequencing.@*RESULTS@#The female neonate was transferred to the intensive care unit due to "secondary pollution of amniotic fluid and laborious breathing for 11 minutes", and had developed frequent convulsions. Genetic testing revealed that she has harbored c.1200C>G and c.188G>A compound heterozygous variants of the SUOX gene, which were inherited from her mother and father, respectively. The c.1200C>G has been described previously and was rated as pathogenic based on guidelines from the American College of Medical Genetics and Genomics, whilst the c.188G>A variant was unreported previously and rated as variant of unknown significance.@*CONCLUSION@#The compound heterozygous variants of the SUOX gene probably underlay the ISOD in this child. Above finding has enriched the spectrum of SUOX gene variants and provided a basis for the clinical diagnosis and genetic counseling.

ABSTRACT

Tinnitus refers to the perception of abnormal sound in the absence of external sound stimulation. It can have an impact on a person's mood, memory, attention, and mental state, although the mechanism of tinnitus is still unclear. In recent years, the research on the central neural mechanism of tinnitus has attracted the attention of scholars.Functional magnetic resonance imaging (fMRI),as an effective imaging technology, has been actively employed in this field. This paper provides a systematic summary of studies on the central neural mechanism of tinnitus by fMRI in recent years, revealed the changes of functional connections among tinnitus-related neural networks, such as auditory network, limbic system, default mode network and salience network. The central neural mechanism of tinnitus involves multiple networks that interact with each other. By understanding this mechanism, we hope to develop more targeted prevention and treatment strategies to help patients alleviate long-term tinnitus.

ABSTRACT

A case of Usher syndrome with methylmalonic acidemia and homocysteine is reported. The patient was a two-month-old and small for gestational age male infant hospitalized for "feeding difficulties" during the neonatal period. The baby boy presented hypotonia, microcephaly, and hearing loss after birth. Genetic test found compound heterozygous mutations of c.482G>A and c.567dup in MMACHC, and both were pathogenic mutations inherited from his parents. Moreover, the patient also had compound heterozygous variants at c.2802T>G and c.14017T>C of USH2A gene. The former was suspected to be pathogenic, and the latter was of unknown clinical significance. Both were from the parents. Usher syndrome and methylmalonic acidemia with homocysteine were clinically diagnosed. Followed up to the age of two, the child was found with moderate mental retardation, while the physical development was comparable to that of the same age group.

ABSTRACT

Objective@#To investigate the influence of unitization encoding strategy and the moderating effect of unitization level on directed forgetting effect of associative recognition.@*Methods@#Association recognition paradigm combined with directed forgetting paradigm were employed in this study. The 39 participants acquired by simple random sampling were asked to remember or forget compound words or unrelated word pairs which were randomly presented according to cues.In the retrieval stage, they needed to distinguish " old" or " rearranged" word pairs regardless of the cues.@*Results@#(1) The reversed unitization effect reached significance. The discrimination of compound words (0.43±0.03) was lower than that of unrelated words (0.55±0.03) (F=27.27, P<0.001, ηp2=0.42). For the word pairs (to be remembered, TBR), the discrimination of compound words (0.43±0.03) was lower than that of unrelated words (0.59±0.03) (t=-6.05, P<0.001); for the word pairs (to be forgotten, TBF), the discrimination of compound words (0.43±0.03) was still lower than that of unrelated words (0.50±0.04) (t=-2.30, P=0.025). (2) The directed forgetting effect of associative recognition was significant. However, TBR (0.51±0.03) was more discriminative than TBF (0.46±0.03) (F=4.30, P=0.045, ηp2=0.10). But the difference was mainly reflected in the recognition of unrelated words.TBR (0.59±0.03) was more discriminative than TBF (0.50±0.04) (t=3.19, P=0.003). However, there was no significant difference between TBR (0.43±0.03) and TBF (0.43±0.03) in recognition of compound words.@*Conclusion@#The unitization encoding stratege can simultaneously promote hit rate and false alarm rate. The directed forgetting effect can be eliminated when unitization level is high enough.

ABSTRACT

This study compared a new type of polysaccharide-coated magnetic nanoparticles (in which the polysaccharide is derived from Angelica sinensis) with the dextran magnetic nanoparticles in terms of preparation, biocompatibility and tissue distribution in vivo and in vitro in order to examine the potential application of Angelica polysaccharide as a novel carrier in magnetic drug targeting (MDT). Magnetic nanoparticles were prepared by chemical co-precipitation. Their physical and chemical properties were determined by using the transmission electron microscope (TEM), laser particle size analyzer (DLS) and vibrating sample magnetometer (VSM), and their purity and structure by using X-ray diffractometer (XRD) and Fourier transform infrared spectroscopy (FTIR). The atomic absorption spectrometric method was performed for quantification of the iron content in different tissues. Histological sections were stained by Prussian blue staining to observe the disposition of magnetic nanoparticles in the liver and kidney. The results showed that both kinds of magnetic nanoparticles possessed small particle size, good dispersion and good magnetic properties. XRD showed the main component of the two magnetic nanoparticles was Fe(3)O(4) crystals, and FTIR proved Fe(3)O(4) was successfully coated by each polysaccharide, respectively. In vivo, Fe(3)O(4)-dextran accumulated in the liver, spleen and lung and Fe(3)O(4)-Angelica polysaccharide only in the spleen and lung.

It was concluded that Angelica polysaccharide may be applied as a novel carrier in the preparation of magnetic nanoparticles.

ABSTRACT

Objective To study serum albumin levels in children with severe sepsis and to correlate serum albumin levels with patient outcome and to identify the causes inducing hypoalbuminemia and its effective countermeasures. Methods Seventy-five children admitted to PICU of Provincial Hospital Affiliated to Shandong University for severe sepsis were included in the study from Aug 2010 to Sep 2011.According to their serum albumin levels within 24 hours and on the third and the seventh day of admission to PICU,75 children were divided into hypoalbuminemia group and normal serum albumin group. Then hypoalbuminemia group was divided into instant hypoalbuminemia group and continuous hypoalbuminemia group according to the duration of hypoalbuminemia. The correlation between the occurring and duration of hypoalbuminemia with patients' prognosis, the etiopathogenisis of hypoalbuminemia and its effective countermeasures were analyzed. Results (1) Sixty-three cases (84.0%) proceeded hypoalbuminemia and their mortality was 33.3% (21/63),while 12 cases (16.0%) showed normal serum albumin level and their mortality was 0.(2) In 63 patients with hypoalbuminemia,26 cases showed continuous hypoalbuminemia and their mortality was 46.0%,while 37 cases proceeded instant hypoalbuminemia and their mortality was 15.4%.There was significant difference (x2 =5.116,P < 0.05) between their mortality.(3) In the 63 cases with hypoalbuminemia,32 cases presented with hepatic injury and their mortality was 37.5%,13 cases proceeded capillary leakage and their mortality was 23.1%,and 18 cases displayed hepatic injury complicated with capillary leakage and their mortality was 33.3%.There was significant difference (x2 =7.812,P < 0.05) between the mortality with different causes. Conclusion Hypoalbuminemia influenced the prognosis of children with severe sepsis, the longer duration correlated with the worse prognosis.Hepatic injury and capillary leakage were two main causes inducing hypoalbuminemia.Active treatment of hypoproteinemia aimed directly at different causes could improve their prognosis significantly.
