ABSTRACT

Objective To preliminarily explore the early process of hepatitis B virus (HBV) infection in HepG2 cells induced by dimethyl sulfoxide(DMSO) ,and provide cytological bases for mechanism study of HBV infection in vitro .Methods HepG cells were divided into the DMSO inducing group and control group ,and were cultured 4 days by DMEM containing 1 .5% DMSO and normal DMEM respectively ;changes of cellular morphology were observed .In addition ,selected ECV304 cells as the negative con‐trol group and treated with DMSO .Cells in the three groups were incubated 2 hours with HBV positive serum after culturing 24 hours ,then trypsin digestive solution ,HepG2 cells and ECV304 cells were collected and polymerase chain reaction (PCR) was used for the determination of HBV DNA .Simultaneously ,the blank control group was set ,and the position of HBsAg on HepG2 cells were detected by using indirect immunofluorescence .Results HepG2 cell volume in the DMSO inducing group was obviously in‐creased .HBV DNA was found in HepG2 cells both in DMSO inducing group and control group ,and DMSO inducing group ex‐pressed much stronger .The results of IIF shown that green fluorescent signals of cell membrane and cytoplasm of HepG 2 cells in the DMSO inducing group were increased obviously ,while the results of HBV DNA and IIF both were negative in the negative con‐trol group .Conclusion DMSO could facilitate adsorption of HBsAg in some extent ,which are benefit for completing the process of early infection .

ABSTRACT

OBJECTIVE@#To investigate the antioxidant effect of different solvent extracts from persimmon leaves (PL) in diabetic mice induced by streptozotocin (STZ).@*METHODS@#The total ethanol-extracted fraction of PL was further extracted with chloroform, ethyl acetate and n-butanol, in that order, the residues after ethanol extraction were water-extracted and alcohol-precipitated, and concentrated. The hypoglycemic effects of different solvents extracts from PL were evaluated in diabetic mice induced by STZ. The experimental mice were randomly divided into groups: control group, model group, glibenclamide group, low and high dosage groups of the various solvent extracts. The drugs were administrated to mice in every morning for 15 days. During this time period, the contents of malondialdehyde (MDA) and superoxide dismutase (SOD) were determined.@*RESULTS@#The water-extracted and ethanol-precipitated fractions and the ethyl acetate-extracted fraction markedly reduced the content of MDA and increased the activity of SOD in the livers of STZ-induced diabetic mice (P0.05).@*CONCLUSION@#The ethyl acetate-extracted fraction, water-extracted and ethanol-precipitated fraction of persimmon leaves have potential value in the treatment of diabetes. The mechanism of action of the antioxidant is related to the hypoglycemic effects of extracts from persimmon leaves.

ABSTRACT

AIM:To study the effect of Ketangte2 (KTT2)on reducing blood glucose and its mechanism.METHODS:Adrenalin and streptozocin were used to induce hyperglycemia in two mice models, and then treated them with glybenzcyclamide (50 mg/kg),three different dose of KTT2 and normal saline(NS)(0.1 ml/10 g weight)for 15 days. And other healthy mice were set up as control group,During the experiment, the fast blood glucose(FBG),insulin was measured in different time. The pancreas and islets of diabetic mice induced by STZ were studied by pathologic section.RESULTS:In the adrenalin-induced model group,KIT2 could lighten hyperglycemic reaction, which Was significant different from that of model group(P<0.05 or 0.01),while KTT2 high, middle dose group could decrease blood glucose in streptozocin-induced hypoglycemic mice obviously(P<0.01).Histological examination showed that pancreatic island number in pancreas in KTT2 groups increased in comparison with the model group.CONCLUSION:KTT2 can obviously reduce the blood glucose in the two kinds of animal models of hyperglycemia (P<0.05 or 0.01).Its mechanism may be related to the protective effect on islet β cells, thereby increase the insulin secretion.

ABSTRACT

OBJECTIVE:To evaluate the status quo and tendency of the use of nootropics in our hospital.METHODS:The nootropics used in our hospital from 2004 to 2006 were analyzed statistically.RESULTS:The consumption sum of nootropics in our hospital increased year by year,2.03 times in 2005 and 1.46 times in 2006 respectively over their previous year. The consumption sum of Ginkgo biloba extract for nootropics took the lead from 2005 to 2006,over 60% of which were medical insurance drugs.CONCLUSION:Leading the list of nootropics in our hospital is plant amedica, with its consumption sum and DDDs showing the equal importance in drug control, suggesting the great market demand for nootropics.

ABSTRACT

Objective To observe the effects of Ketangte 2 on experimental diabetes in mice. Methods Adrenalin and streptozocin(STZ) were used to induce hyperglycemia mice model and diabetic mice model respectively. The mice were divided into five groups randomly:model group, glibenclamide control group,high-,middium-,low-dosage Ketangte 2 groups. The administration lasted consecutive 15 days. The blank control group and model group were treated with the same volume of saline. After treatment, the levels of blood sugar were measured in hyperglycemia mice induced by adrenalin, and the fasting blood-glucose,2-hour blood-glucose,insulin levels in diabetic mice induced by STZ were determined.Meanwhile the histopathological changes of pancreas were observed under light microscope. Results For STZ-induced diabetic mice, the hypoglycemic effect of Ketangte 2 was obvious(P
