ABSTRACT

@# Objective: The aim of this study was to investigate the effect of co-culture with AEC (amniotic epithelial cell) on the biological characteristics of AMSC (amniotic mesenchymal stem cell), and to investigate the roles of SDF-1/CXCR4 axis in the homing and migration of AMSC. Methods: AMSC andAEC were isolated from human amnion, and then cultured, amplified and identified, respectively. TheAMSC were divided into three groups:AEC co-cultured group, serum-free cultured group and serum cultured group.After culture for 24 h, 48 h, and 72 h, the proliferation viability ofAMSC was measured by CCK-8 assay and trypan blue staining; the expression of CXCR4 mRNAwas analyzed by flow cytometry and Real-time RT-PCR, and the migration ability ofAMSC in vitro was observed by migration assay. Results: Cell viability (48 h and 72 h) and survival rate in the co-culture and serum groups were higher than those in the serum-free group (all P<0.05). The mRNA and protein expressions of CXCR4 in AMSC of the co-culture and serum-free groups were significantly higher than those of the serum group (P<0.05). The migration ability of AMSC in the co-culture and serumfree groups, which increase with the SDF-1 (stromal cell derived factor-1) concentration gradient, were higher than that in the serum group (P<0.05). Conclusion: AMSC co-cultured with AEC still have the basic biological characteristics of MSC, and showed good growth activity. Co-culture withAEC can up-regulate CXCR4 onAMSC surfaces and enhance the migration ability ofAMSC in vitro.

ABSTRACT

Low birth weight (LBW) and preterm birth (PB) are associated with newborn mortality and diseases in adulthood.We explored factors related to LBW and PB by conducting a population-based case-control study from January 2011 to December 2013 in Wuhan,China.A total of 337 LBW newborn babies,472 PB babies,and 708 babies with normal birth weights and born from term pregnancies were included in this study.Information of newborns and their parents was collected by trained investigators using questionnaires and referring to medical records.Univariate and logistic regression analyses with the stepwise selection method were used to determine the associations of related factors with LBW and PB.Results showed that maternal hypertension (OR=6.78,95% CI:2.27-20.29,P=0.001),maternal high-risk pregnancy (OR=1.53,95% CI:1.06-2.21,P=0.022),and maternal fruit intake ≥300 g per day during the first trimester (OR=1.70,95% CI:1.17-2.45,P=0.005) were associated with LBW.BMI ≥24 kg/m2 of mother prior to delivery (OR=0.48,95% CI:0.32-0.74,P=0.001) and gestation ≥37 weeks (OR=0.01,95% CI:0.00-0.02,P<0.034) were protective factors for LBW.Maternal hypertension (OR=3.36,95% CI:1.26-8.98,P=0.016),maternal high-risk pregnancy (OR=4.38,95% CI:3.26-5.88,P<0.001),maternal meal intake of only twice per day (OR=1.88,95% CI:1.10-3.20,P=0.021),and mother liking food with lots of aginomoto and salt (OR=1.60,95% CI:1.02-2.51,P=0.040) were risk factors for PB.BMI ≥24 kg/m2 of mother prior to delivery (OR=0.66,95% CI:0.47-0.93,P=0.018),distance of house from road ≥36 meters (OR=0.72,95% CI:0.53-0.97,P=0.028),and living in rural area (OR=0.60,95% CI:0.37-0.99,P=0.047) were protective factors for PB.Our study demonstrated some risk factors and protective factors for LBW and PB,and provided valuable information for the prevention of the conditions among newborns.

ABSTRACT

Recent research based on various animal models has shown the neuroprotective effects of erythropoietin (EPO).However,few studies have examined such effects of EPO in the clinic.In this study we enrolled patients with spinal cord ischemia-reperfusion (I-R) injury to investigate the clinical application of EPO and methylprednisolone (MP) for the neuroprotection against spinal cord I-R injury.Retrospective analysis of 63 cases of spinal cord I-R injury was performed.The Frankel neurological performance scale was used to evaluate the neurological function after spinal cord injury (SCI),including 12 cases of scale B,30 cases of scale C,and 21 cases of scale D.These cases were divided into 2 groups:group A (27 cases) got treatment with both EPO and MP; group B (36 cases) got treatment with MP only.The neurological function of patients after treatment was evaluated by American Spinal Cord Injury Association (ASIA) index score,and activity of daily living (ADL) of the patients was also recorded.All patients got follow-up and the follow-up period ranged from 24 to 39 months (mean 26 months).There was no significance difference in neurological function between groups A and B before the treatment (P>0.05).However,the neurological function and ADL scores were significantly improved 1 week,1 year or 2 years after the treatment compared to those before the treatment (P<0.05),and the improvement was more significant in group A than in group B (P<0.05).It is suggested that the clinical application of EPO and MP provides the neuroprotection against spinal cord I-R injury.
