ABSTRACT

OBJECTIVE: To identify an unknown tadalafil analogue illegally adulterated in health foods. METHODS: An unknown tadalafil analogue was observed in a routine screening for compounds illegally adulterated in heath foods by HPLC-DAD. The compound of interest was isolated by silica gel thin layer chromatography. Its molecular weight and structural fragments were obtained by UPLC-MS/MS, and its 1H-NMR spectum was also obtained using nuclear magnetic resonance. Its structure is elucidated by analysis of these data in combination of the literature. RESULTS: A new tadalafil analogue, nortadalafil, was detected in health foods. CONCLUSION: This is the first time to disclose that nortadalafil was illegally adulterated in health foods. It's necessary to supplement the current inspection standard, which lists 11 target compounds but not including nortadalafil, to punish the illegal producers in time.

ABSTRACT

<p><b>OBJECTIVE</b>To observe the effects of different ways in repairing scrotum of pigs with full-thickness burn on spermatogenesis of testis.</p><p><b>METHODS</b>Twenty male Guizhou miniature pigs were divided into normal control (NC), natural-healing (NH), flap-repairing (FR), and skin-grafting (SG) groups according to the random number table, with 5 pigs in each group. Pigs in NC group were not subjected to any injury. Scrotum of pigs in the latter three groups were inflicted with full-thickness burn. Wounds in NH group healed naturally. Wounds in FR group were repaired with inguinal region flap, and those in SG group with full-thickness skin from lower abdomen. Appearance of scrotum in the latter three groups was observed right after injury, and three months post injury or surgery (PIM or PSM). Specimens of testes of pigs in the latter three groups were obtained in PIM or PSM 3 to detect apoptosis of spermatogenic cells with TUNEL, and bcl-2 protein expression with immunohistochemistry. The same indexes were observed and determined in pigs of NC group. Data were processed with one-way analysis of variance and LSD test.</p><p><b>RESULTS</b>(1) Scrotum of pigs in NC group had skin folds with contraction function. Scrotum of pigs became hard with a leathery appearance right after burn in the other three groups. In PIM or PSM 3, wounds of pigs in NH group healed with scar, and the testes were squeezed into inguinal region. Scrotal skin of pigs in FR group was thick with testes in the scrotum, and that of pigs in SG group was thin with testes in the scrotum. (2) Spermatogenic cells in each level in NC group were arranged regularly, with few apoptotic spermatocytes and spermatoblasts. In NH, FR, and SG groups, seminiferous epithelium was thinner with most of the spermatogenic cells showing apoptosis, and they were mainly spermatogonia and spermatocytes. Apoptotic index of spermatogenic cells in NH, FR, SG, and NC groups was respectively (46.3 ± 3.3)%, (40.9 ± 3.5)%, (20.6 ± 2.3)%, (7.5 ± 1.9)%, and the difference among them was statistically significant (F = 405.65, P < 0.01). There were significant statistical differences among the former three groups (with P values below 0.01). (3) bcl-2 protein expression in NH, FR, SG, and NC groups was respectively (52 ± 5)%, (53 ± 4)%, (64 ± 5)%, (75 ± 5)%, and the difference among them was statistically significant (F = 56.63, P < 0.01). There was no significant statistical difference in bcl-2 expression between NH group and FR group (P = 0.66), and it was lower in both groups as compared with SG group (with P values below 0.01).</p><p><b>CONCLUSIONS</b>Either scar healing, flap transplantation, or SG in repairing scrotum with full-thickness burn in pigs inhibits spermatogenesis, but repair with SG produces less deleterious effect on the testis.</p>

ABSTRACT

Objective To investigate the prevalence of love affairs and intimate partner violence,and to explore the relationship between intimate partner violence and other mental health and risk behavior in college students.Methods Three universities were selected using cluster sampling method in Hefei and Wuhu.Totally.2575 college students completed an anonymous questionnaire.Intimate partner violence,depression,satisfaction of school life,self-esteem,suicidal psychology and behavior were evaluated to estimate the relationship between intimate partner violence and mental health/risk behavior.Results There were 46.9%students reported that they had intimate partner currently or in the past.The rate of having intimate partner in male students was higher than that in female students(x2=44.13,P<0.001).And the rates were higher in sophomores and juniors than in freshmen(x2=161.84,P<0.05).There were 21.1% students had sexual behavior with their intimate partners.But only 21.8%(34/156)intimate partners reported that they used condom every time.There were 11.5%(18/156)intimate partners reported that they never took any contraception.There were 18.6%(29/156)students reported that they were pregnant or led to their girlfriend becoming pregnant,but only less than 50.0% adolescents induced abortion in a legal hospitals.The rates of being the victim of physical assault,emotional abuse,sexual coercion,the total intimate partner violence were 18.0%,33.6%,5.1%.37.1%.The rates of being the victim of physical assault,emotional abuse,total intimate partner violence in male adolescents were higher than those in female adolescents,but the rate of sexual coercion was on the contrary(x2=70.21,13.25,14.04,5.77,P<0.05).Among the intimate partners who had suffered from intimate partner,74.2%(345/446)students had underwent more than 3 times.and 47.1% had underwent more than 2 types of intimate partner violence.The score of depression was highest in the victims of intimate partner violence,but the scores of self-esteem and school life satisfaction were on the contrary(F=4.00,16.39,8.76,P<0.05).The rates of suicidal ideation,suicidal plan,suicidal preparative and attempted suicide were highest in the victims of intimate partner violence(x2=13.80,9.72,8.52,11.96,P<0.05).Conclusion The rate of having sexual behavior with their intimate partners was high in college students,but their self-protection awareness was insufficient.In the present study,a high prevalence of intimate partner violence among university students was observed,and highlighted a need for attention to the other mental health and risk behavior in adolescents with intimate partner violence.

ABSTRACT

Objective To investigate the episodes and influencing factors on self-harm and to explore the relationship between self-harm episodes and suicidal psychology and behaviors in college students. Methods Four universities were selected using cluster sampling method in Anqing city and Chaohu city. Totally, 2713 college students completed this survey. Data were analyzed by Pearson Chi-square and logistic regression. Results In the last six months, rates of highly lethal self-harm,less lethal self-harm with visible tissue damage, self-injury without visible tissue damage,self-harmful behaviors with latency damage, other self-harmful behaviors with menticide were 1.9%,5.5%, 15.3%, 21.2% and 17.0% respectively. The total rate of self-harm was 31.3%. 73.1% of the students with self harmful experiences had the above mentioned behaviors more than 3 times in the last six months. The top 3 reasons for taking self-harm actions were: having learning problems (43. 1% ), failed love affairs (25.0%) and having conflicts with others (23.9%). There were different influencing factors among different kinds of self-harm episodes. Depression was the risk factor of self-harm. The higher score of having high self-esteem was the protective factor of all kinds of self-harm actions except highly lethal ones. Higher score of difficulties in identifying feelings was one of the risk factors. The rates of suicidal psychology and behaviors in students with self-harm were significantly higher than those in students without those behaviors. Result from linear x2 test indicated that the graveness of tissue damage of self-harm was higher along with the rates of suicidal psychology and behaviors (P<0.01). Conclusion Among 2713 college students, about 1/3adolescents having experienced self-harm in the last 6 months, many with repeated ones. Depression and difficulties in identifying feelings were the two risk factors while self-esteem was the protective factor related to most of the self-harm cases.

ABSTRACT

Objective To examine the epidemiological features and relative risk factors of repeated injuries among middle, high and college students in Ma' anshan city of Anhui province. Methods A prospective study on repeated injuries for 1-year follow-up period was carried out among 1494 students from 3 middle schools or colleges. Risk factors for repeated injuries were analyzed with multivariate logistic regression model. Results 799 students suffered 1639 episodes of injuries, with an incidence rate of 53.48%. The repeated group consisted of 200 students who suffered 854 injuries, accounted for 52.10% among the total injuries. Results from Multi-normial logistic regression analysis showed that male, number of family members of 4 or over, extraversion of character trait, and upper quartile of self-rating anxiety symptoms were the risk factors for the occurrence of repeated injuries. Moderate or well-of family income, mothers with senior high school education, and upper quartile of self-rating depression symptoms were protective factors for repeated injuries. Conclusion Socio-demographic factors and emotional status may predict the occurrence of repeated injuries that called for further study.

ABSTRACT

Objective This study was undertaken to examine possible relationship between attempted suicide and underachievement,bullying,low life satisfaction and low self-concept at school.Methods An anonymous self-report survey assessing demographic characteristics and the major risk factors of teenage attempted suicide was completed by students from 16 middle schools in grades seven to twelve in 4 counties of Anhui province (age 10 to 21 years).An anonymous questionnaire was used to rate attempted suicide,bullying involvement and learning performance.Attempted suicide was defined as: experiencing specific suicide actions at least one time during the 12 months preceding the survey.Multidimensional Students' Life Satisfaction Scale and Children' s Self-concept Scale were used to evaluate satisfaction and self-conscience on and at respectively.In total,10 894 respondents substantially completed the survey.Multiple logistic-regression analyses,controlling for socio-demographic variables,was used to analyze if underachievement,bullying,low school life satisfaction and low children' s self-conscience at school had been risk factors.Results 629 participants (5.8%) reported having made at least one attempted suicide within the last 12 months.Students being underachieved were significantly having more attempted suicide events than those excellent students (X2=11.39,P=0.023).Students being both bully-victims and practiced bully were significantly more than those being only practiced bully (28.7% vs.15.8%,P<0.001),being victims (28.7% vs.10.6%,P<0.001) or having neither of them (28.7% vs.4.8%,P<0.001).Results from multiple logistic regression analyses showed that underachievement,bullying,lower school life satisfaction and low self-conscience were risk factors for attempted suicide.Conclusion Data from this study confirmed that school bullying and children' s self-conscience at school were significantly associated with attempted suicide among rural middle school students in Anhui province.It is of importance to improve the school' s environments to reduce the risk of attempted suicide among this group.

ABSTRACT

Objective To examine life satisfaction as a moderator or mediator of accommodation category and loneliness for elementary and middle school students in rural areas so as to provide evidence for psychological health intervention among said students. Methods All participants were asked to complete an anonymous questionnaire battery which including an 18-item General Health Questionnaire, Children' s Loneliness Scale and Multidimensional Students' Life Satisfaction. Results The average score of loneliness scale (35.0±9.5) among boarding-school students was significantly higher than those students living at home (33.1±10.1 ) (P<0.01). The average scores on school-satisfaction (33.5±6.3), friend-satisfaction (42.1±6.2) and self-satisfaction (32.4±5.0) of boarding-school students were significantly higher than those students living at home (35.9±6.6, 42.9±6.6,32.9±5.3 ) ( P< 0.05 ). The association between accommodation category and loneliness was fully mediated by school-satisfaction (the standardized coefficients of loneliness was reduced from 0.043 (P<0.05) to 0.021 (P>0.05) and partly mediated by self-satisfaction and friend-satisfaction. Data through Moderation analyses indicated that self-satisfaction, school-satisfaction and friend-satisfaction did not serve as moderators. Conclusion Accommodation category, life satisfaction seemed to be good predictors on loneliness among elementary and middle school students and the fully mediated effect of school-satisfaction between accommodation category and loneliness was significant, suggesting that intervention of loneliness should focus on these variables.

ABSTRACT

<p><b>OBJECTIVE</b>To construct chimerical DNA vaccine plasmid of human papillomavirus type 11 (HPV11) L1-E7, and to evaluate its immunogenicity.</p><p><b>METHODS</b>Molecular cloning techniques were used to construct recombinant plasmid pcDNA3 L1-E7 as a DNA vaccine. BALB/c mice were vaccinated with DNA recombinants through muscle injection.IL-2 and gamma-INF secreted by immunized spleens lymphocyte and HPV 11 L1 or E7 specific antibodies were assayed by ELISA method. Spleens lymphocyte proliferation was measured by MTT assay.</p><p><b>RESULTS</b>The chimerical DNA plasmid of pcDNA3 L1-E7 was constructed correctly. Specific anti-HPV11 E7 and L1 antibodies, specific lymphocyte proliferation and secretions of IL-2 and gamma-INF were detected in vaccinated mice.</p><p><b>CONCLUSION</b>Specific immune response, including cellular immunity and humoral immunity, could been detected in mice vaccinated with chimerical DNA vaccine of pcDNA3 L1-E7.</p>

ABSTRACT

Objective: To investigate the association of DNA X-ray repair cross complementary protein 1 (XRCC1) Arg399Gln polymorphism with the clinical outcome of gastric cancer patients treated with platinum-based chemotherapy. Methods: Eight-four patients with gastric cancer confirmed by pathological examination received postoperative combined chemotherapy (including platinum). Transcription of XRCC1 Arg399Gln was determined by polymerase chain reaction-ligation detection reaction (PCR-LDR). Results: Of the 84 patients, the frequencies of XRCC1 codon 399 Arg/Arg, Arg/Gln, and Gln/Gln genotype were 58.3% (49/84), 35.7% (30/84), and 6.0% (5/84), respectively; and the relapse and death rate were 63.1% (53/ 84) and 55.2% (43/84), respectively. The relapse and death rates in patients with Arg/Arg genotype were significantly lower than those in patients with Arg/Gln or Gln/Gln genotypes (P < 0.05). COX multivariate analysis showed that XRCC1 Arg399Arg polymorphism was associated with better recurrence-free and overall survival in gastric cancer patients compared with other genotypes (P < 0.05). Conclusion: Polymorphism of XRCC1 Arg399Gln correlated with the prognosis of gastric cancer patients treated with postoperative platinum-based chemotherapy. It may be helpful to a certain extent for evaluating the prognosis.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the expression of the T cell-specific transcription factors T-bet/GATA-3 in recurrent aphthous ulcerations (RAU).</p><p><b>METHODS</b>Peripheral blood mononuclear cells (PBMC) were isolated by density gradient centrifugation from RAU patients and healthy controls. Expressions of T-bet and GATA-3 mRNA and protein in the PBMCs were detected by reverse transcription-polymerase chain reaction (RT-PCR) and Western blot respectively. The concentrations of gamma-interferon (gamma-IFN) and interleukin-4 (IL-4) in the serum were determined by enzyme-linked immunosorbent assay (ELISA).</p><p><b>RESULTS</b>The expressions of T-bet mRNA were 0.73 +/- 0.14 in the control group and 0.12 +/- 0.04 in the recurrent aphthous ulcerations group, the levels of T-bet protein expressions were 0.73 +/- 0.18 in the control group and 0.24 +/- 0.09 in the recurrent aphthous ulcerations group, there was a significant difference between the two groups (P < 0.01). The levels of GATA-3 mRNA expressions were 0.89 +/- 0.11 in the control group and 1.30 +/- 0.13 in the recurrent aphthous ulcerations group; GATA-3 protein expressions were 1.09 +/- 0.16 in the control group and 1.80 +/- 0.16 in the recurrent aphthous ulcerations group, there was a significant difference between the two groups (P < 0.01). In the control group, the concentration of gamma-interferon in the serum was (22.15 +/- 1.52) ng/L, which was significantly elevated compared with that in the recurrent aphthous ulcerations group (16.32 +/- 0.22) ng/L (P < 0.01), in the control group, the concentration of IL-4 in the serum was (25.58 +/- 2.59) ng/L, which was significantly lower than that of the recurrent aphthous ulcerations group (43.60 +/- 3.15) ng/L (P < 0.01). The ratio of gamma-IFN and IL-4 was positively correlated with the ratio of T-bet and GATA-3 mRNA and protein expression (r = 0.96, 0.94, P < 0.01).</p><p><b>CONCLUSIONS</b>Imbalance of transcription factors T-bet and GATA-3 may be one of the key factors in immune dysregulation of recurrent aphthous ulcerations.</p>

ABSTRACT

<p><b>AIM</b>To study the chemical constituents of Polygala aureocauda Dunn..</p><p><b>METHODS</b>Chemical compounds were isolated by column chromatography and their structures were determined mainly by spectroscopic means (UV, IR, MS, 1HNMR, 13CNMR, HMQC, HMBC).</p><p><b>RESULTS</b>Three compounds were isolated and identified as 3-hydroxy-1,4-dimethoxyxanthone (I), 1, 7-dihydroxy-2, 3-methylendioxyxanthone (II), 7-hydroxy-1-methoxy-2, 3-methylendioxyxanthone (III).</p><p><b>CONCLUSION</b>Compounds I-III were isolated from Polygala aureocauda Dunn. for the first time, whereas compound I is a new xanthone.</p>

ABSTRACT

Objective To evaluate the possibility of methylation analysis of secreted frizzled-related protein gene 2 (SFRP2),hyperplastic polyposis protein gene (HPP1) and O~6-methylguanine-DNA methyhransferase gene (MGMT) in feces for screening colorectal cancer (CRC).Methods Total DNA was isolated from fecal samples of 52 patients with colorectal cancer and 35 patients with benign colorectal diseases and 24 normal volunteers,and the methylation status of SFRP2,HPP1 and MGMT was analyzed with methylation-specific polymerase chain reaction.Results SFRP2,HPP1 and MGMT were methylated in 94.2%,71.2%,48.1% of CRCs,respectively.At least one of the three genes was methylated in 96.2% of CRCs and 81.8% of precancerous lesions,respectively.In contrast,of the 24 normal controls,only one had methylated SFRP2.These results indicated 93.7% sensitivity,77.1% specificity,84.3% positive predicative value and 90.2% negative predictive value of the test for detecting CRC and precancerous lesions.Conclusions Methylation analysis of SFRP2,HPP1 and MGMT gene in stool is a high sensitive screening tool for CRC and precancerous lesions.Analysis of fecal DNA methylation is expected to serve as a new detection way for CRC or a new screening tool for individuals at risk of developing colorectal neoplasm.
