ABSTRACT

Objective:To investigate the value of the ultrasonography in the diagnosis of the white matter injury of premature infants based on gray-scale ultrasonography radiomics. Methods:A total of 256 premature infants in Huazhong University of Science and Technology Union Shenzhen Hospital and Shenzhen Hospital of Southern Medical University from August 2018 to April 2022 were analyzed retrospectively. The computer-generated random numbers were assigned to the training set and the verification set according to 6∶4 ratio. On the basis of standardized collection of craniocerebral ultrasound images, the radiomics features were extracted from imaging by Pyradiomics 3.0.1 software package, the Mann-Whitney U test and the least absolute shrinkage and selection operator (LASSO) and stepwise regression were used to select the optimal features. Then the Logistic regression was used to build radiomics model. According to MRI, ROC curve was utilized to evaluate the performance of the model. The craniocerebral ultrasound images in the validation set were independently diagnosed by a senior physician and a junior physician, and then the above two physicians diagnosed again with the help of the radiomics, and the diagnostic abilities of this model were compared with those of the junior and senior physicians with and without radiomics assist. Results:A total of 5 optimal features were selected to develop radiomics model. The sensitivity, specificity, accuracy and the area under the ROC curve (AUC) in the training and validation sets were 0.861, 0.775, 0.799, 0.818; 0.929, 0.824, 0.853, 0.876, respectively. The sensitivity, specificity, accuracy and AUC in the senior sonographer, the junior sonographer, and both of them with radiomics assist for the dagnosis in the validation set were 0.929, 0.892, 0.902, 0.910; 0.714, 0.743, 0.735, 0.729; 0.929, 0.919, 0.922, 0.924; 0.857, 0.824, 0.833, 0.841, respectively. Performance of radiomics model reached the level of the senior sonographer (AUC: 0.876 vs 0.910, P=0.284), which was significantly better than the performance of the junior sonographer(AUC: 0.876 vs 0.729, P=0.001). Performance of the junior sonographer with radiomics assist was significantly better than the performance of the junior sonographer(AUC: 0.841 vs 0.729, P=0.003). Performance of the senior sonographer with radiomics assist was comparable to that of the senior sonographer(AUC: 0.924 vs 0.910, P=0.156). Conclusions:The ultrasound diagnosis method based on radiomics technology shows good diagnostic performance for the white matter injury of premature infants. ABSTRACT

Ischemic stroke is the second leading cause of human death and the third reason of disability. Meanwhile, the incidence is rising year after year worldwide. Ischemic stroke could cause ischemia-reperfusion injury after blood recanalization treat-ment, but the mechanism of ischemia-reperfusion injury is still not very clear, so it is necessary to build a preclinical model with specific characteristics. Up to now, animal experiments have been still complicated, and the culture of brain slices has some limitations. The cell model in vitro has become a simplified and valuable tool widely used by researchers. The paper systematically summarizes the common type of nerve cells, and further analyzes establishment methods and principle, relevant research progress on the in vitro model of ischemia-reperfusion, in order to provide reference for rationally selecting hypoxia and reoxygenation model for basic research on cerebral ischemia and reperfusion and drug screening.

ABSTRACT

Objective To investigate the epidemiological characteristics and identify the risk factors of Giardia lamblia infections among patients with colorectal cancer in Henan Province. Methods A cross-sectional study was performed for questionnaire surveys among colorectal cancer patients in Henan Cancer Hospital during the period from March to July, 2021. Patients’ stool samples were collected, and the triosephosphate isomerase (tpi) gene of G. lamblia was amplified in stool samples using nested PCR assay to characterize the parasite genotype. Univariate analysis and multivariate logistic regression analyses were employed to identify the risk factors of G. lamblia infections among colorectal cancer patients. Results A total of 307 colorectal cancer patients were investigated, including 176 males (57.3%) and 131 females (42.7%). PCR assay detected 8.1% [95% confidential interval (CI): (0.056, 0.117)] prevalence of G. lamblia infections among the study subjects, and there was no significant difference in the prevalence between men [9.1%, 95% CI: (0.057, 0.143)] and women [6.9%, 95% CI: (0.037, 0.125)] (χ2 = 0.495, P = 0.482). In addition, there was no age-specific prevalence of G. lamblia infections among the participants (χ2 = 1.534, P = 0.675). Multivariate logistic regression analysis identified use of septic tanks [odds ratio (OR) = 3.336, 95% CI: (1.201, 9.267)], daily use of well water [OR = 3.042, 95% CI: (1.093, 8.465)] and raising livestock [OR = 3.740, 95% CI: (1.154, 12.121)] as risk factors of G. lamblia infections among colorectal cancer patients, and the prevalence of abdominal pain was significantly greater in colorectal cancer patients with G. lamblia infections than in those without infections (P = 0.017). Among the 25 patients with G. lamblia infections, assemblage A was characterized in 24 (96.0%) cases and assemblage B in one case (4.0%). Conclusions The prevalence of G. lamblia is high among colorectal cancer patients in Henan Province, and assemblage A is the dominant genotype of G. lamblia. Use of septic tanks, daily use of well water and raising livestock are risk factors of G. lamblia infections among patients with colorectal cancer.

ABSTRACT

Objective To investigate the effect of echinacoside on the protection of acute liver injury induced by concanavalin A (ConA) in mice and on the extracellular histones. Methods Male C57BL/6 (B6) mice were randomly divided into three groups: control group (n=6), which received saline only; ConA-treated group (n=8), which were given ConA (30mg/kg) via tail vein; echinacoside plus ConA-treated group (n=8), which were treated with echinacoside solutions by gavage at a dose of 50mg/ kg for 3 days before ConA injection. At 16 h after ConA treatment, all mice were sacrificed to collect blood and tissue samples for analysis of hepatic function, pathological indexes, as well as extracellular histones and the relevant inflammatory cytokines. Results Acute liver injury in mice was induced by ConA administration as demonstrated by elevated serum ALT, AST levels and severe pathological changes. Echinacoside plus ConA-treated mice showed significantly lower levels of ALT and AST and milder liver injury than that in ConA-treated mice (P<0.01). The number of mice survived in echinacoside plus ConA-treated group was more than that in ConA-treated group. Moreover, the levels of extracellular histones and IL-1, IL-6, IL-10 and TNF-α were markedly decreased in echinacoside plus ConA-treated mice as compared to those in ConA-treated mice (P<0.01). Conclusions Echinacoside may act an obvious protective effect against acute liver injury induced by ConA, and it might be due to its inhibitory effect on pro-inflammatory mediators including cytokines and lowering of extracellular histones.

ABSTRACT

Purpose To establish rabbit VX2 tumor model and to explore the relation between perfusion parameters and the expression of the VEGF in the portal vein VX2 implanting tumor emboli. Materials and Methods VX2 tumor was implanted in the portal vein of eight experimental rabbits. Multi-slice CT (MSCT) perfusion scan was performed after tumor formation to measure and compare portal vein tumor thrombus, hepatic blood lfow (HBF) near tumor foci and far away from tumor foci, hepatic blood volume (HBV), probability of surface area product (PS) and mean transit time (MTT). The VX2 tumor emboli were then resected to analyze the relationship between the liver perfusion parameters and VEGF expression using immunohistochemical method. Results MSCT liver perfusion parameters were not statistically signiifcant between foci close to or far away from the tumor (P>0.05). The HBF, HBV and PS within the tumor emboli were higher than that in hepatic parenchyma (P<0.05) and the MTT was higher (P<0.05). There was positive correlation (r=0.711, 0.646 and 0.626, P<0.05) between the HBF, HBV and PS of portal vein VX2 tumor emboli and VEGF expression, and there was negative correlation between MTT and VEGF expression (r=-0.565, P<0.05). Conclusion MSCT perfusion parameters in the portal vein VX2 implanting tumor emboli and the expression of VEGF are positively related. MSCT can evaluate the angiogenesis of portal vein VX2 implanting tumor emboli.

ABSTRACT

Objective To study the effects and current mechanism of low concentration of lidocaine on evoked-bursting (EB) firing of dorsal root ganglion (DRG) neurons in rat model of chronic compression (CCD) of DRG . Methods Twenty-four SD rats were divided into normal control group (n=12) and CCD model group (n=12). CCD group was treated with chronic oppression on L4 and L5 DRG with L shape bar. Normal control group received no treatment. In vivo intracellu?lar recording was used to record the incidence of EB and the effect of lidocaine on subthreshold membrane potential oscilla?tion (SMPO). Patch clamp recording was used to record the effect of lidocaine on persistent sodium current (INaP). Results The incidence of EB increased in CCD group( 45.97%, 57/124), which was significantly different when compared with nor?mal group (χ2=26.810, P<0.01). The magnitude of SMPO, INaP and EB were inhibited in a reversible way by lidocaine (50μmol/L). Conclusion The low concentration of lidocaine might play an analgesic effect in peripheral nervous system by se?lectively inhibiting INap, which participates in SMPO formation.

ABSTRACT

Objective To analyze the correlation of perfusion parameters obtained by functional multi-slice computed tomography (fMSCT) with vascular endothelial growth factor (VEGF) detected with immunohistochemistry on rabbit model with VX2 breast cancer and noninvasively evaluate the vascularization of untreated VX2 breast cancer in vivo. Methods Sixteen New Zealand femal rabbits were selected and suspension (1 ml) of mass was injected around the breast areola. CT perfusion was performed after two weeks and perfusion parameters including blood flow (BF), blood volume (BV), mean transit time (MTT)and permeability surface (PS) were assessed. Expression of VEGF in neoplasm was detected with immunohistochemistry. Paried t test was used for the comparison of perfusion parameters between the tumor and muscle and Pearson correlation analysis was used to assess the correlation of VEGF with perfusion parameters. Results The mean value of BF, BV, MTT and PS were (228.21 ± 13. 13 ) ml · min-1100 mg-1 ,(13.45 ± 1.01) ml · 100 mg-1 ,(3. 50 ±0. 20) sand (7.85 ±1.18) ml · min-1 · 100 mg-1 in tumor, respectively. They were (66. 10 ±22. 11 ) ml · min-1 · 100 mg-1 , ( 1.88 ± 1.80) ml · 100 mg-1,(23. 87 ±0. 63)s,(1.55 ±0. 38)ml · min-1 · 100 mg-1 in muscular tissue, respectively. The mean value of BF, BV and PS in tumor were obviously higher than those in muscle, and the mean value of MTT in tumor was lower than that in muscle. There were significant differences in CT perfusion parameters between tumor and muscle (t = 61.83,13.63,27.72,20. 54, P < 0. 01 ). The mean value of VEGF in tumor was 7. 33 ±0. 27 and there were positive correlation with BF ( r = 0. 712, P < 0. 01 ), BV ( r = 0. 647, P < 0. 01 ), PS ( r =0. 627 ,P < 0. 01 ), and negative correlation with MTT ( r = - 0. 564, P < 0. 05 ). Conclusion MSCT perfusion imaging can be used to noninvasively evaluate the vascularization in rabbits with untreated VX2 breast cancer in vivo.
