ABSTRACT

2-Oxoglutarate/Fe(II)-dependent dioxygenases (2-ODD) play an important role in plant primary and secondary metabolism. Based on the high-throughput sequencing platform Illumina NovaSeq 6000, the transcriptome of Salvia apiana Jepson was sequenced, and the obtained reads were de novo assembled. A total of 38 534 unigenes were obtained from the transcriptome. The assembled unigenes were annotated and 29 982 unigenes were given functional annotations. The 2-ODD genes were identified from the assembled S. apiana transcriptome database by bioinformatics methods, and the genes were analyzed, including the homology of the sequences, physicochemical characteristics, signal peptides, transmembrane domains, subcellular localization, secondary structure and tertiary structure, etc. The evolutionary relationships and the expression patterns of the identified 2-ODD genes were also analyzed. 39 full-length 2-ODD genes were identified from the transcriptome of S. apiana. The average length of these 2-ODD encoding proteins was 320 amino acids, the molecular weight was about 36.00 kDa, and most of them were hydrophilic proteins. Phylogenetic analysis divided these 2-ODD genes into several subfamilies. Gene expression analysis indicated that the 2-ODD genes were expressed in different parts of S. apiana, and the expression level of most genes was much higher in roots than that in leaves. This study can lay a foundation for further study of 2-ODD genes in S. apiana.

ABSTRACT

Migraine is a neurological disorder characterized by attacks of moderate or severe headache and various neurological symptoms. Acupuncture, as a commonly used non-pharmacological therapy, has the advantage of obvious therapeutic effect and few side effects in the prevention and treatment of migraine. But the underlying mechanism of acupuncture on migraine remains unclear. Recently, advances in neuroimaging technology have helped to objectively assess the effect of acupuncture on treating migraine and offered new opportunities to explore the central mechanism of acupuncture on treating migraine. In order to better understand the current status of neuroimaging studies on the therapeutic mechanism of acupuncture on migraine and shed light on future research, this review aims to overview the neuroimaging studies in recent 10 years from two aspects: (1) Central mechanism of acupuncture on treating acute migraine attack; (2) Central mechanism of acupuncture on preventing migraine attack.

ABSTRACT

Objective To investigate the effect of Ginkgo biloba extract on behaviors in rat with adolescent sedentariness and its possible mechanism. Methods Twenty-four male SD rats (4-week-old) were randomly divided into normal control (NC), adolescent sedentarines (AS), and Ginkgo biloba extract (GBE) groups. After 4 weeks intervention with GBE, open-field test and elevated plus-maze test were performed to detect the behavioral changes. The content of malondialdehyde (MDA) and activity of superoxide dismutase (SOD) in brain were determined by colorimetric method. Protein levels of glycogen synthase kinase 3β(GSK-3β) and β-catenin in cerebral white matter were determined by Western blotting. Results In open-field test, it was shown that the autonomic activity of rats in AS group increased, while the central regional travel time was reduced. Duration and number of residence in the open arm of elevated plus-maze test decreased in AS group. These anxiety-like behaviors were ameliorated by GBE intervention. Compared with the NC group, GSK-3β/ β-catenin ratio and the content of MDA was upregulated in AS group while downregulated in GBE group. And activity of SOD were downregulated in AS group while significantly upregulated in GBE group (P<0. 05). Conclusion Ginkgo biloba extract ameliorate anxiety-like behavior in rat with adolescent sedentariness. Wnt/ β-catenin pathway and antioxidant regulation may play a cerebral protective role.

Atorvastatin is proven to ameliorate cardiac hypertrophy induced by chronic intermittent hypoxia (CIH).However,little is known about the mechanism by which atorvastatin modulates CIH-induced cardiac hypertrophy,and whether specific hypertrophyrelated microRNAs are involved in the modulation.MiR-31 plays key roles in the development of cardiac hypertrophy induced by ischemia/hypoxia.This study examined whether miR-31 was involved in the protective role of atorvastatin against CIH-induced myocardial hypertrophy.H9c2 cells were subjected to 8-h intermittent hypoxia per day in the presence or absence of atorvastatin for 5 days.The size of cardiomyocytes,and the expression of caspase 3 and miR-31 were determined by Western blotting and RT-PCR,respectively.MiR-31 mimic or Ro 31-8220,a specific inhibitor of protein kinase C epsilon (PKCε),was used to determine the role of miR-31 in the anti-hypertrophic effect of atorvastatin on cardiomyocytes.PKCε in the cardiomyocytes with miR-31 upregulation or downregulation was detected using RT-PCR and Western blotting.The results showed that CIH induced obvious enlargement of cardiomyocytes,which was paralleled with increased atrial natriuretic peptide (ANP),brain natriuretic peptide (BNP),and slow/beta cardiac myosin heavy-chain (MYH7) mRNA levels.All these changes were reversed by the treatment with atorvastatin.Meanwhile,miR-31 was increased by CIH in vitro.Of note,the atorvastatin pretreatment significantly increased the mRNA and protein expression of PKCε and decreased that of miR-31.Moreover,overexpression of miR-31 abolished the anti-hypertrophic effect of atorvastatin on cardiomyocytes.Upregulation and downregulation of miR-31 respectively decreased and increased the mRNA and protein expression of PKCε.These results suggest that atorvastatin provides the cardioprotective effects against CIH probably via up-regulating PKCε and down-regulating miR-31.

ABSTRACT

Objective To explore the relationship of expression and the effect of preoperative radiotherapy of endothelin-1 (ET-1) and pyruvate kinase M-2 (PKM2) in rectal carcinoma.Methods Immunohistochemical method was used to detect the expression of ET-1 and PKM2 proteins of rectal cancer tissues in 96 cases.The expressions of ET-1 and PKM2 were analyzed with the effect of preoperative radiotherapy in rectal cancer tissue.Results The high expression of ET-1 protein was 59 cases (61.46%).The high expression of PKM2 proteins was 54 cases (56.25%).The high expressions of ET-1 and PKM2 protein were worsen the effect of tumor regressive grade (TRG) than lower expressions of those after preoperative radiotherapy of rectal cancer tissue (P < 0.05).The protein expression of ET-1 and PKM2 were positively correlated (P =0.006).Conclusions The high expressed ET-1 and PKM2 proteins in rectal cancer are closely related to preoperative radiotherapy resistance.ET-1 and PKM2 proteins are expected to become new targets of radiotherapy sensitivity and radiotherapy sensitization of rectal cancer.

ABSTRACT

<p><b>OBJECTIVE</b>To observe the effect of Hydroxy Safflower Yellow A (HSYA) on the expression of osteogenic markers, such as alkaline phosphatase, Cbf(alpha)l and type I collagen, and explore the mechanism of HSYA in the prevention and treatment of glucocorticoid-induced ischemic necrosis of femoral head.</p><p><b>METHODS</b>Fifteen healthy and adult New Zealand white rabbits were collected and weighted 0.9 to 1.3 kg. The rabbits were injected abdominally with anesthetic drugs, then received marrow cavity puncture of tibia and anterior superior iliac spine to get bone marrow blood. Rabbits bone marrow mesenchymal stem cells (BMSCs) were separated from the bone marrow blood, cultured in vitro and passaged. The 3rd generation of BMSCs which had good growth condition were randomly divided into blank group, model group and HSYA groups with different doses. The BMSCs in model group were treated with high dose of dexamethasone to induce adipogenic differentiation of cells cultured in vitro, and inhibit osteogenic differentiation. The BMSCs in HSYA groups received high dose of dexamethasone and different concentrations of HSYA simultaneously. The blank group received not any special handling. After a week,the expressions of alkaline phosphatase, Cbf(alpha)l and type I collagen mRNA were detected.</p><p><b>RESULTS</b>The alkaline phosphatase activity was significantly decreased in BMSCs of the model group as compared with the blank group (P < 0.01), and the expression of Cbf(alpha)l and type I collagen mRNA were also decreased significantly (P<0.01). The alkaline phosphatase activity was significantly increased in BMSCs of each HSYA group as compared with the model group (P < 0.05 or P < 0.01), and the expression of Cbf(alpha)l and type I collagen mRNA were also increased significantly (P < 0.05 or P < 0.01).</p><p><b>CONCLUSION</b>The mechanism of HSYA may be related to the effect of antagonism to the reduced osteogenic differentiation induced by glucocorticoid.</p>

ABSTRACT

Objective To explore the effect of avascular necrosis cystic degeneration on distribution of bone density. Methods Based on the bone reconstruction model of Weinans and Huiskes, bone density distribution in normal femoral head and in femoral head with avascular necrosis cystic degeneration were calculated by finite element analysis. Results (1) The medial system of lamellae, lateral system of lamellae, intertrochanteric arch and Ward's triangle were generated on the normal proximal femur under simulation. (2) If a primary cystic degeneration occurred, a secondary cystic area would appear below the primary one and its bone density would decrease dramatically with the primary cystic area increased. Cystic degeneration would also change the femoral bearing truss system. Conclusions (1) Wolff’s law on bone remodeling is consistent with modern topology optimization theory. (2) If cystic degeneration appeared due to avascular necrosis of the femoral head, it should be treated timely to prevent the secondary cystic degeneration and collapse of the femoral head.

<p><b>OBJECTIVE</b>To study the chemical constituents of Memorialis hirta.</p><p><b>METHOD</b>Compounds were isolated and purified by multiple methods, and their structures were identified based on physicochemical property and spectrum data.</p><p><b>RESULT</b>12 compounds were isolated from ethyl acetate from 95% ethanol extracts of M. hirta, they were isorhamnetin (1), kaempferol (2), quercetin (3), isorhamnetin-3-O-alpha-L-rhamnopyranoside (4), kaempferol-3-O-alpha-L-rhamnopyranoside (5), isorhamnetin-3-O-beta-D-glucopyranoside (6), kaempferol-3-O-beta-D-glucopyranoside (7), quercetin-3-O-alpha-L-rhamnopyranoside (8), quercetin-3-O-beta-D-glucopyranoside (9), isorhamnetin-3-O-rutinoside (10), kaempferol-3-O-rutinoside (11) and quercetin-3-O-rutinoside (12), respectively.</p><p><b>CONCLUSION</b>All compounds were obtained from the genus Memorialis for the first time.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore a method for the treatment of the skin defects at the distal phalanges of 2-5th fingers.</p><p><b>METHODS</b>The island flap at the dorsum of the middle phalange was designed with the pedicle of dorsal branches from the digital proper artery. When the flap was used to repair defect at finger pulp, the dorsal branch of the digital proper nerve in the flap was kept to be anastomosed to the digital proper nerve at the recipient finger. From Feb. 2005 to May. 2010, 54 cases with skin defects at the distal phalanges of 61 fingers were treated with the flap, including 35 defects at finger pulp and 26 defects at finger tip.</p><p><b>RESULTS</b>The maximum size of defects and flaps was 2.2 cm x 2.5 cm and 2.4 cm x 2.7 cm, respectively. 61 flaps survived completely. Blister was happened in 3 flaps 2 days after operation, which healed spontaneously without necrosis. 54 cases were followed up for 5 to 22 months (average, 11 months). The flaps had good texture and color match with normal sensation (grade S4). The 2-point discrimination distance was 6-9 mm. The interphalangeal joint had normal movement.</p><p><b>CONCLUSIONS</b>The island flap at the dorsum of the middle phalange is an ideal method for the skin defect at the distal phalange of finger.</p>

<p><b>OBJECTIVE</b>To study the chemical constituents of Androsace umbellata.</p><p><b>METHOD</b>Many chromatography means were used in separation and purification, and the structures of all compounds were identified by the means of spectroscopic analysis and physico-chemical properties.</p><p><b>RESULT</b>10 compounds were elucidated as kaempferol 3-O-(3-O-acetyl-)-alpha-L-rhamnopyranoside(1), kaempferol 3-O-(2-O-acetyl-)-alpha-L-rhamnopyranoside(2), kaempferol 7-O-alpha-L-rhamnopyranoside(3), kaempferol 3-O-alpha-L- rhamnopyranoside(4), kaempferol 3-O-beta-D-glucopyranoside(5), kaempferol 3-O-(3-O-acetyl-)-a-L-rhamnopyranosyl-7-O-alpha-L- rhamnopyranoside(6), kaempferml 3-O-(4-O-acetyl-)-alpha-L-rhamnopyranosyl-7-O-alpha-L-rhamnopyranoside(7), quercetin 3-O-alpha-L- rhamnopyranoside(8), quercetin 3-O-beta-D-glucopyranoside(9) and myricetin 3-O-beta-D-glucopyranoside (10), respectively.</p><p><b>CONCLUSION</b>All compounds were obtained from the title plant for the first time.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the pathogenesis of steroid-induced avascular necrosis of femoral head and the control mechanisms of Ground Beetle.</p><p><b>METHODS</b>The bone marrow-derived mesenchymal stem cells were cultured in vitro. The BMSCs in the 3rd generation were randomly divided into blank group, model group and Chinese medicine low, medium and high dose group. BMSCs of model group using high-dose steroid-induced in vitro were adipogenic differentiation to inhibit osteogenesis. Chinese medicine low, medium and high-dose groups at the same time were given the Ground Beetle intervention serum containing insects. The expression of osteocalcin, Alkaline phosphatase and Type I Collagen mRNA were detected and interfered 6 days later.</p><p><b>RESULTS</b>Serum containing Ground Beetle could reverse content of steroid-induced alkaline phosphatase of BMSCs, the expression of ostercalcin and Type I Collagen mRNA decreased.</p><p><b>CONCLUSION</b>The control mechanism of Ground Beetle on steroid-induced avascular necrosis is not only to improve the microcirculation, but also to inhibit steroid-induced osteogenic differentiation of BMSCs reduced.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To discuss the effectiveness of individualized strategy of surgical management on the great arteries (TGA).</p><p><b>METHODS</b>From March 1998 to October 2009, 127 cases (97 males and 30 females) with TGA were treated. There were 97 male and 30 female, aged from 4 hours old to 17 years old with a mean of (25 ± 37) months, weighted from 2.7 to 47.5 kg with a mean of (8 ± 8) kg. The palliative operations included Glenn operation in 14 cases (3 cases double Glenn procedure), Balalock-Taussing shunt in 14 cases, Banding operation in 8 cases, and atrial septal defect enlarge/Banding/Balalock-Taussing shunt in 15 cases. The end-stage operation included Senning procedure in 5 cases, Switch procedure in 32 cases, 2(nd)-stage Switch procedure in 11 cases, Switch procedure with VSD repairing in 20 cases, Switch procedure with Hybrid in 1 case, Nikaidoh procedure in 3 cases, Rastelli procedure in 13 cases, Fonton procedure in 18 cases, other procedure in 4 cases. Twenty-one cases underwent 2 operations, and 5 cases underwent 3 or more operations. Sixty-six cases underwent delayed sternal closure.</p><p><b>RESULTS</b>There were 12 cases of death operatively in 127 cases. The total operative mortality was 9.4%. There were 5 cases dying of low cardiac output during the operation, 2 of pulmonary hypertension crisis, 2 of hemorrhage, 1 of grafting problem of coronary artery deformation, 1 of renal failure after Fonton procedure and 1 case of newborn dying of spontaneous rupture of liver post-operatively. The patients were followed up for 1 month to 12 years. There were 10 patients with vary degrees complications such as pulmonary stenosis, residual shunt and narrow channel. Three cases underwent reoperation. The rest of survived cases had normal heart function, good growth and development state.</p><p><b>CONCLUSIONS</b>Individualized strategy of surgical management based on anatomical conditions of TGA can significantly improve the success rate of surgery and long-term survival.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To establish rat models of Steroid-avascular necrosis of femoral head, observe the effects of activating blood circulation of chinese herbal medicine on genetic expression of transforming growth factor-beta1 (TGF-beta1). To interpret the mechanism of the effect on Steroid-avascular necrosis of femoral head by activating blood circulation,and offer a effective method to clinical.</p><p><b>METHODS</b>Cleaner-40 SD rats,half males and half females, weight (200 +/- 20) g, were randomly divided into 2 groups: 4 rats were in common group and 36 rats were in medel group. The rats in medel group were administered with 24.5 mg/kg hydroxyprednisone twice a week peritoneal injection for 6 weeks induced to femur head necrosis. The rats in common group were through gluteus injection as control. There were 4 rats were killed in each group after 6 weeks, to be assure that the model were succed. All surplus rats were divided into treatment group and control group:the treatment group were administrated with activating blood circulation of Chinese herbal medicine 12.3 ml/kg per day,the control group were administrated with sodium chloride 12.3 ml/kg per day. Then, after 6 and 8 weeks, killed the animal and detected all indexes.</p><p><b>RESULTS</b>(1) The expression of TGF-beta by immunohistochemistry and image analysis: the expression of femoral head TGF-beta increased significantly in treatment group than in control group and two group had significant differences. (2) Serum levels of TGF-beta1: in the treatment group serum expression of TGF-beta1 increased,compared with the control group had significant difference (P < 0.01). (3) The femoral head local TGF-beta1 mRNA transcription: treatment group in the first six weeks, expressed that the increase in the first eight weeks, expressed also reduced,and the control group in the first six weeks, expressed a decrease in the first eight weeks,was not detected to TGF-beta1 mRNA expression of difference between the two groups was significant (P < 0.01). TGF-beta1 mRNA in serum and the femoral head with local expression of TGF-beta1, in consistency.</p><p><b>CONCLUSION</b>Rat abdominal cavity prednisolone acetate injection plus intermittent standing avascular necrosis modeling stability, good repeatability. Taohongsiwu through the promotion of hormone-ischemic necrosis of the femoral head rat model of TGF-beta1 mRNA transcription, and promote expression of TGF-beta1.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the increasing incidence and the characteristics of Tsutsugamushi disease in the areas of Nan Peng Lie islands, Nan Ao island, Wan Shan archipelago, Nao Zhou island and Lei Zhou peninsula, located in the southern part of China and to develop strategies for preventive measures.</p><p><b>METHODS</b>Both epidemiological investigation, isolation and gene identification of Orientia tsutsugamushi, as well as pilot preventive measures were carried out.</p><p><b>RESULTS</b>These islands belonged to the epidemic area of south subtropical zone of Tsutsugamushi disease. The main host was Rattus norvegicu and the overall rates of infection on Orientia tsutsugamushi were 22.78%-33.75%. The main biological vector was Leptotrombidium (Leptotrombidium) deliens and the rates of infection on Orientia tsutsugamushi were 40.00%-75.00%. 25 strains of Orientia tsutsugamushi had been isolated from Rattus norvegicu and Leptotrombidium (Leptotrombidium) deliens. Results showed that the isolated strains of Orientia tsutsugamushi were 15 Karp, 8 Kato, 2 Yonchon. Results from serological studies showed that the positive rate of anti-Orientia tsutsugamushi antibodies was high, in both residents and soldiers stationed in these islands. On these islands, rats and biological vectors were killed. Results showed that these measures had positive impact in reducing the incidence.</p><p><b>CONCLUSION</b>Islands from the southern part of the country belonged to the epidemic area of Tsutsugamushi disease. People visiting this areas should be under protection.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the chemical constituents in root of Phlomis medicinalis.</p><p><b>METHOD</b>The compounds were isolated and repeatedly purified on macroporous resin, silica gel column chromatography, TLC and Prep-HPLC and the structures were elucidated by physico-chemical properties and NMR spectra.</p><p><b>RESULT</b>Eight compounds were obtained and elucidated as 5-Hydroxy-7-methoxy-4, 6-dimethylphthalide (1), 4-hydroxymethyl-2-furaldehyde (2) and six iridoid glucosides: 6-O-acetyl-shanzhiside methyl ester (3), 8-O-acetyl-shanzhiside methyl ester (4), shanzhiside methyl ester (5), sesamoside (6), phloyoside II (7) and dehydropentstemoside (8).</p><p><b>CONCLUSION</b>All the compounds were isolated from the plant for the first time and 1 and 3 were obtained from the plants of Phlomis for the first time.</p>

ABSTRACT

<p><b>AIM</b>To study the stability of insulin-loaded polybutylcyanoacrylate nanoparticles (IPN) in an oily medium (soybean oil containing 0.5% (v/v) Tween-20 and 5% (v/v) Vitamin E) along with the hypoglycemic effect following their oral administration to streptozotocin (STZ) induced diabetic rats.</p><p><b>METHODS</b>The stability of IPN in the process was appraised by measurement of the amount of undegraded insulin associated to nanoparticles, the average size and the span of IPN, as well as the release of insulin from IPN. IPN in an aqueous medium (containing 0.5% (v/v) Tween-20) at pH 2.0 was also investigated as control.</p><p><b>RESULTS</b>The study showed that IPN in the oily medium was more stable than that in the aqueous medium over one year of storage in the dark at (25 +/- 2) degrees C and the in vitro stability of IPN in the oily medium against degradation by proteolytic enzymes was much better than that in the aqueous medium. The apparent bioavailability of an oral administration of IPN (50 u x kg(-1)) in the oily medium versus an (sc) injection of insulin (2 u x kg(-1)) was 22.4%, much higher than that of IPN in the aqueous medium (15.5%), based on decreased areas above curve (AAC) determination for the blood glucose depression from time zero to 144 h of a single oral administration of IPN to STZ-diabetic rats.</p><p><b>CONCLUSION</b>IPN in soybean oil containing Tween-20 (0.5% v/v) and Vitamin E (5% v/v) could be considered as an effective and stable delivery system for oral insulin.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the spatial distribution character of dengue fever and the change of Aedes' population, so as to provide macroscopical decision-making evidences of prevention and supervision on dengue fever.</p><p><b>METHODS</b>(1) Collecting data on morbidity of dengue and supervision on vector's population in the corresponding period. (2) Drawing digitized map of Chaozhou in scale of 1:50,000, including elements of boundary, residential areas, road and traffic, altitude, water systems etc. (3) Measuring the latitude and longitude of center position of surveillance safes on the scene. (4) Processing spatial analysis by the ArcGIS 8.5 software.</p><p><b>RESULTS</b>Distribution of Aedes showed spatial cluster in Chaozhou, while its density was related to the distance to the watersides. The closer to the watersides, the higher the density was. Map on spatial distribution showed that although the Aedes epidemic situation changed yearly, but primarily be kept in high, middle, low regions. Cross-validation effects of the distribution maps were satisfactory.</p><p><b>CONCLUSION</b>Geographic information system was promising in analyzing data on dengue fever, and better than other routine research methods.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the spatial distribution of malaria in Hainan province.</p><p><b>METHODS</b>Maps of spatial distribution on malaria incidence in Hainan province from 1995 to 1999 were produced, using a spatial local interpolation technique in the ArcGIS 8.1 software. Cross-validation techniques were used to evaluate the fitness of the distribution maps.</p><p><b>RESULTS</b>The spatial distribution of malaria in Hainan province from 1995 to 1999 showed that the incidence of malaria in the southern part of Hainan province were higher than that in the north. In the south, the incidence of malaria in east coastal areas were higher than that of other parts, especially in Wuzhi and Yingge mountains. The prediction error of the cross-validation indicated that the spatial distribution maps of malaria in Hainan had good fitness of malaria incidence of Hainan province.</p><p><b>CONCLUSION</b>Spatial local interpolation technique seemed to be promising in describing the spatial distribution of malaria.</p>
