ABSTRACT

Purpose@#To probe the effect and mechanism of androgen receptor antagonist MDV3100 on benign prostatic hyperplasia (BPH) of rats @*Methods@#BPH rat model was induced by testosterone propionate. Then antagomir-miR-21-3p or agomir-miR-21-3p was injected into rats before MDV3100 treatment. The prostate index was measured by weighing the wet weight of the rat prostate. The structural morphology of rat prostate was observed after hematoxylin & eosin staining. Immunohistochemistry was applied to evaluate the expression levels of Ki-6 and inflammatory cytokines (interleukin [IL]-6, IL-18, and tumor necrosis factor-α) in rat prostate tissues. Quantitative reverse transcription polymerase chain reaction was utilized for assessment of miR-21-3p expression, and Western blot for the performance of the phosphorylation levels of IKKα and p65. @*Results@#Injection of testosterone propionate caused increased prostate gland hyperplasia, heightened miR-21-3p level, and activated nuclear factor-kappa B (NF-κB) signaling pathway. Additionally, BPH was accompanied by inflammatory response, as evidenced by enhanced expressions of Ki-67 and inflammatory cytokines. MDV3100 exposure ameliorated BPH and suppressed miR-21-3p expression. Overexpression of miR-21-3p intensified BPH and inflammation level, while knockdown of miR-21-3p relieved BPH. The coeffect of miR-21-3p upregulation and MDV3100 subjection led to higher inflammatory response, elevated phosphorylation levels of IKKα and p65 than MDV3100 treatment alone. @*Conclusions@#Androgen receptor antagonist MDV3100 alleviates BPH and inflammatory response through miR-21-3p downregulation and NF-κB signaling pathway blockade.

ABSTRACT

The biocompatible hydrogel was fabricated under suitable conditions with natural dextran and polyethylene glycol (PEG) as the reaction materials. The oligomer (Dex-AI) was firstly synthesized with dextran and allylisocyanate (AI). This Dex-AI was then reacted with poly (ethyleneglycoldiacrylate) (PEGDA) under the mass ratio of 4∶6 to get hydrogel (DP) with the maximum water absorption of 810%. This hydrogel was grafted onto the surface of medical catheter via diphenyl ketone treatment under ultraviolet (UV) initiator. The surface contact angle became lower from (97 ± 6.1)° to (25 ± 4.2)° after the catheter surface was grafted with hydrogel DP, which suggests that the catheter possesses super hydrophilicity with hydrogel grafting. The evaluation after they were implanted into ICR rats subcutaneously verified that this catheter had less serious inflammation and possessed better histocompatibility comparing with the untreated medical catheter. Therefore, it could be concluded that hydrogel grafting is a good technology for patients to reduce inflammation due to catheter implantation, esp. for the case of retention in body for a relative long time.

ABSTRACT

<p><b>BACKGROUND</b>Currently, cystoscopy and urine cytology are standard modalities in therapy monitoring and follow-up of bladder carcinoma (BC). Cystoscopy is an invasive and uncomfortable procedure while cytology has a limited value because it is operator-dependent and has low sensitivity. This study was to assess the accuracy of ImmunoCyt in detecting BC by comparing it with cytology using systemic analyses of studies published in both English and Chinese.</p><p><b>METHODS</b>Cochrane systematic evaluation was used to search through MEDLINE, EMBASE, Cochrane Library, CMCC, and CNKI for studies regarding ImmunoCyt and cytology for detection of BC. Data were extracted and analyzed by the software MetaDiSc 1.4.</p><p><b>RESULTS</b>In total 42 relevant studies were searched, of which 15 were enrolled and 12 491 patients were included. Heterogeneity, except for threshold effects, was found within these studies. A meta-analysis was performed using the random effect model. Pooled accuracy indicators like sensitivity, specificity, and diagnostic odds ratio of ImmunoCyt™ and cytology were 0.75 (0.73-0.77) vs. 0.45 (0.43-0.48), 0.73 (0.72-0.74) vs. 0.97 (0.96-0.97), and 10.97 (7.53-15.99) vs. 16.40 (10.57-25.46), respectively. The sensitivity of both was increased with the increase of tumor grade and stage. The area under summary receiver operating characteristics curve was 0.834 4 and 0.853 4 and the Q index 0.766 7 and 0.785 3 for ImmunoCyt and cytology, respectively. Combination of both can obviously improve the accuracy of diagnosis.</p><p><b>CONCLUSIONS</b>ImmunoCyt has a high sensitivity in detecting BC, but its specificity is low. As an important adjunct, ImmunoCyt™ can not replace cytology, but combined with cytology it could improve sensitivity with high specificity in the detection and postoperative monitoring of BC.</p>

ABSTRACT

Objective To investigate the difference of clinical efficiency and safety in obsessive-compulsive disorders (OCD) treatment with escitalopram or paroxetine.Methods A total of 156 OCD patients were randomly divided into escitalopram group (ESC group) and paroxetine group (PAR group).Yale-Brown Obsessive Compulsive Scale (Y-BOCS),Hamilton Depression Scale (HAMD) and Treatment Emergent Symptom Scale (TESS)were used to evaluate the clinical efficiency and safety before and after1,2,4,6,8 weeks treatment.Results The cure rate(21.79% vs 17.95%) and effective rate(70.51% vs 71.79%) had no statistically difference between ESC group and PAR group,and incidence of side effect had no significant difference between two groups(x2 =1.99,P>0.05).Compared with the group before treatment,HAMD scores were significantly decreased from the first weekend in ESC group,but in PAR group HAMD scores did not decrease until the second weekend,and the differences were also significant(P<0.05).Conclusion Escitalopram is a safety,effective and well-tolerated drug in the treatment of obsessive-compulsive disorder.

ABSTRACT

Objective To examine the effect of family environment and family function on suicide behavior in schizophrenic patients.Methods 53 schizophrenic patients with suicidal behavior in the last month were selected for the schizophrenic suicide group.53 schizophrenic patients and 53 normal controls matched with the schizophrenic suicide group were selected for the schizophrenic no suicide group and normal control group.All of them completed family environment scale and family assessment device.Results The cohesion factor,the expressiveness factor,the active-recreational orientation factor and the control factor of family environment scale in the schizophrenic suicide group were significant lower than those in the schizophrenic without suicide group(separately 5.70 ± 1.29 vs 6.62± 1.23,4.77 ± 1.49 vs 5.96 ± 1.11,3.94 ± 1.63 vs 5.68 ± 1.98,2.68 ± 1.68 vs 4.40 ± 1.81,P< 0.05),but the conflict factor in the schizophrenic suicide group was higher than in the schizophrenic without suicide group(3.55 ± 1.60 vs 2.81 ± 1.52,P < 0.05).The affective responsiveness factor,the affective involvement factor and the behavior control factor of family assessment device in the schizophrenic suicide group were significant lower than those in the schizophrenic without suicide group(separately 16.05 ± 2.57 vs 14.53 ± 3.38,19.13 ± 4.09 vs 16.58 ± 2.82,23.35 ± 3.52 vs 21.60 ± 3.25,P < 0.05).Multiple liner regression analyses showed that the expressiveness factor,the active-recreational orientation factor,the control factor,the affective involvement factor and the behavior control factor were important factors for schizophrenic suicide(F=21.583,P=0.000,adjusted R 2 =0.519).Conclusion Attempted suicide patients with schizophrenia have some deficiencies at family function and family environment.

ABSTRACT

OBJECTIVE:To prepare and establish quality control for the compo und metronidazole hollow suppository.METHODS:The compound metronidazole hollow suppository was prepared with semisynthesis fatty acid glyceride as ground substance;The contents of metronidazole and ofloxacin were determined by dual-wave length K-ratio method and equivalent absorption dual-wave length method.RESULTS:The detecting concentration linear range of metronidazole and ofloxacin were6.0~18.0?g/ml and3.0~9.0?g/ml respectively,the average recovery were100.98%and99.56%(n=5)respectively,RSD were0.82%?0.80%respectively.CONCLUSION:This method is convenient,accurate and reproducible,which can be used for the quality control of compound metronidazole suppository.
