ABSTRACT

Objective:By summarizing and analyzing the data of the National Natural Science Foundation of China (NSFC) for distinguished young scholars in the department of health science, this paper provides support and reference for the organization and management of talent projects in related institutions to better serve the growth of young talents in medical field.Methods:Based on the data of distinguished young scholars projects approved from 2011 to 2020, this paper explored the characteristics of the projects in the department of health science. Information about the research field, institution distribution, age and gender of the principle investigator, and the situation of hosted projects were analyzed, main challenges were summarized to propose possible measurements for improvement.Results:The overall funding rate of distinguished young scholars projects in the department of health science is relatively low and more competitive. The growth of medical talents takes longer, more support and further attention is needed.Conclusions:The organization of the distinguished young scholars project in health science needs to respect the characteristics and development nature of medicine, at the same time, strengthen multi-dimensional configuration and management, and create a soft environment conducive to the growth of outstanding medical talents.

ABSTRACT

Objective To establish a prostate urethral re-epithelialization model with Chinese rural canine by 2 μm laser vaporization resection.Methods We used 2 μm laser to vaporiz prostate of 15 uncastrated male Chinese rural canines from March to April in 2016.These canines mean age was (6.3 ± 0.6) years(ranging 5-7 years),and weight was (20.5 ± 1.3) kg(ranging 18-22 kg).We began to surgery in which we saw the protruding part of the prostate in urethra,and narrow prostate urethra after a successful anesthesia by intraperitoneal injection of chloral hydrate.The operation time,anesthesia time,survival rate,first time to drink water,first time to feed,first time to stand,first time to defecate,the time when canine bladders rinse became clear,wound healing time were recorded.After 3 days,1 week,2 weeks,3 weeks and 4 weeks,we randomly select 3 canines to observe regeneration of prostate urethra wound under cystoscope.After surgery,the bladder,prostate and prostate distale urethra were removed to make specimen and measure the diameter size of prostate.The HE staining and immunohistochemistry was performed in each sample.Results The experimental operation time was (70.5 ± 18.3) min (ramging 50-90 min).The average anesthesia time was (120.1 ± 21.1) min (ranging 95-145 min).The survival rate was 100%.In post surgery duration first standing time,first eating time,first drinking water time,first defecation time were (6.5 ± 1.8) h,(10.3 ± 2.1) h,(23.7 ± 5.6) h,(26.3 ± 3.1) h,respectively.The time when canine bladders rinse became clear and wound healing time were (5.2 ± 1.6) d,(8.7 ± 1.5) d respectively.Cystoscopy observated that the wound was covered by pale necrotic tissue 3 d and 1 week after operation,covered by epithelium 2 weeks after operation,covered by more thicker epithelium 3 weeks after operation,covered by epithelium which color was close to normal urothelium 4 weeks after operation.HE staining observated that the wound wasn't covered by epithelium 3 d after operation,partial wound was covered by flaky single or 2-3 cubic regenerated epithelial 1 week after operation,all wound was covered by epithelial which was lack of polar 2 weeks after operation,wound was covered by polarity epithelium which was thicken to 5-6 layer and observated a little umbrella cells on the surface 3 weeks after operation,wound was covered by polarity epithelium which was thicken to 5-6 layer and observated much umbrella cells on the surface 4 weeks after operation.Immunohistochemical staining observated that urinary spot protein from the wound or epithelium was negative 3 d,1 and 2 weeks after operation,urinary spot protein from the part of epithelium was positive 3 weeks after operation,and urinary spot protein from all epithelium was positive 4 weeks after operation.Conclusion It is feasible to establish prostate urethral re-epithelialization model in the Chinese rural canine by 2 μm laser vaporization resection of the prostate.

ABSTRACT

Aim To investigate the effect of 96 h paradoxical sleep deprivation(PSD) on learning & memory ability of rats and the concomitant gene expression change in the rat hippocampus.Methods PSD model was established by Modified Multiple Platform Method(MMPM).Learning and memory ability of the PSD model was assessed through hexagonal maze compared with the control, and the gene expression in hippocampus of each group was detected by gene chip technology.After that, GO and KEGG enrichment analysis were applied to determine the changes of the gene expression in PSD model compared with the control.Results PSD affected the memory consolidation process in rats.The gene chips showed that 100 genes expressed differently in the hippocampus of PSD model compared with the control, and among them, 25 genes were up-regulated and 75 genes down-regulated.The up-regulated genes mainly involved in cell growth and differentiation, learning and memory, inflammation, oxidative stress, and biological processes such as synaptic transmission, while the down-regulated genes mainly involved in immune response, central nervous system development, synaptic transmission, metabolism, signal transduction, neurotransmitter secretion, cell apoptosis, and so on.KEGG analysis showed that genes differentially expressed mainly involved in cGMP-PKG, metabolism, NF-kappa B and Pathways in cancer, and so on.Conclusion In rats, PSD leads to memory deficit, as well as a variety of gene expression changes in hippocampus of the PSD model.

@#To investigate the effect of block of AGEs-RAGE pathway on the migration of aortic vascular smooth muscle in diabetic rats and its possible mechanisms, vascular smooth muscle cells(VSMCs)cells were pre-stimulated by antibody of RAGE, and then stimulated by AGEs. Transwell assay was adopted to assay migration of VSMCs. Proliferation of VSMCs and expression of p27 were analyzed by MTT and ELISA, respectively. The change of ROS level in VSMCs was defermined by DCFH assay, the expression of NOX1 mRNA was determined by RT-PCR assay. Results indicated that the AGEs induction for migration of VSMCs was significantly inhibited after treatment by RAGE antibody(P< 0. 01), which blocked the AGEs-RAGE pathway, and the inhibition of migration was stronger than that of proliferation. The ROS level was decreased(P< 0. 01), and the expression of NOX1 mRNA was decreased, yet the expression of P27 protein was not changed greatly. Block of AGEs-RAGE pathway by antibody of RAGE can inhibit the migration of VSMCs, and the mechanism may be related with the decrease of NOX1 mRNA and then down to the level of intracellular oxidative stress in VSMCs.

ABSTRACT

<p><b>OBJECTIVE</b>Optical coherence tomography images may be distorted by motion artifacts in dynamic imaging, so it may lead to misdiagnosis in clinical diagnosis. Motion artifacts correction has become an urgent issue in optical coherence tomography imaging.</p><p><b>METHODS</b>Firstly, using the improved complex nonlinear diffusion preprocessing filtering reduced the noise of the sequence images, then using the image mass center aligned the distortion data in the Y direction, finally, using the method of deviation average corrected motion artifacts along the Y direction.</p><p><b>RESULTS</b>After correction, the motion artifacts in the longitudinal 2D images and the 3D image disappeared, the surface of the 2D and the 3D image became more smooth, the structure between layers of the images got clear and distinct, retinal en-face single slice image was sharp, and the fundus tissue structure could be observed.</p><p><b>CONCLUSION</b>The algorithm of correction makes the physical structure of the retinal display truly after motion artifacts correction.</p>

ABSTRACT

Objective To investigate the effects of bortezomib on cellular migration and invasion ability in cuhured human prostatic cancer cells DU145 and its mechanism.Methods Transwell method was used to detect cell migration.Invasion was assayed by Matrigel-coated invasion chambers.Expressions of FAK and Tyr397 proteins were analyzed by Western blot.Results After treated by bortezomib at the concentration of 10,20 nmol/L for 24 h,the invasion index of DU145 cells were (69.05±10.56) and (52.55±6.98),they were gradually reduced compared with untreated group (81.55±10.56) (P < 0.05).The migration index were (39.35±6.45),(32.05±4.22),they were also reduced compared with untreated group (58.75±5.41) (P < 0.05).The group of treated by bortezomib showed Tyr397 protein expression had been suppressed.However FAK protein had not marked change.Conclusions FAK is involved in the regulation of cellular migration and invasion function.Bortezomib might inhibit cells migration and invasion function by down regulation of Tyr397 expression.

ABSTRACT

The present paper aims to investigate the relation between characteristic parameters of transesophageal photoelectric pulse wave in descending aorta and ambulatory artery blood pressure. The chests of ten adult experimental dogs were performed to take the photoelectric pulse wave of descending aorta transesophageally. The concurrent femoral artery invasive blood pressure was recorded simultaneously. Stepwise regression analysis method was used to study the correlation efficient between characteristic parameters of descending aorta pulse wave (H, h, h/H, g/H, At, s, H(1 + ts/td), k)and invasive artery blood pressure. The characteristic parameters, k and h/H (ratio: 90% and 80%) was proved that they had good correlation with systolic pressure; and k, H and s (ratio: 90%, 80% and 70%), had good correlation with diastolic pressure; while k and H (ratio: 90% for both) had good correlation with mean pressure. The mean values of multiple correlation coefficients of the selected characteristic parameters of descending aorta pulse wave with systolic pressure, diastolic pressure and mean pressure of femoral artery were 0.871, 0.900 and 0.856, respectively. The characteristic parameters of descending aorta pulse wave had specific correlation with systolic pressure, diastolic pressure and mean pressure.

ABSTRACT

OBJECTIVE: To investigate the effects of compound preparation of Cordyceps sinensis and Tripterygium hypoglaucum (CSTHC) on survival time of grafted pigskin after allogeneic transplantation and its mechanism. METHODS: The pigskin was treated with CSTHC solution before allogeneic transplantation, and CSTHC ointment was applied for external use on the grafted pigskin after skin transplantation. Cyclosporine A (CsA) and normal saline were served as control. The survival time, the appearance and the histomorphological changes of the grafted pigskin were observed. The histomorphological changes of testicles in pigs were also examined. The CD4 and CD8 expressions in the grafted pigskins were measured by immunohistochemical method. The white blood cell count in peripheral blood and the liver and renal functions were also examined. RESULTS: The survival time of the grafted pigskin in the CSTHC-treated group was (28.50+/-3.26)d, which was much longer as compared with (10.60+/-1.52)d in the untreated group (P<0.01). The survival time of the grafted pigskin in the CsA-treated group was (28.33+/-3.50)d, and there was no remarkable difference in the survival time of the grafted pigskin between the CsA-treated group and the CSTHC-treated group. The expressions of CD4 and CD8 were lower in the CSTHC-treated group than those in the untreated group on the 7th and 14th day after skin graft (P<0.05), while there was no significant difference in the indices between the CSTHC-treated group and the CsA-treated group. The WBC count was higher in the untreated group than that in the CSTHC-treated group or CsA-treated group on the 7th day after skin graft (P<0.05). CONCLUSION: CSTHC can prolong the survival time of allogeneic grafted pigskin. Its mechanism of inhibiting the immunological rejection may relate to decreasing the expressions of CD4(+) and CD8(+) in the grafted pigskin and reducing the local inflammatory reaction.
