ABSTRACT

Aim To explore the inhibitory effect of isobavachalcone (IBC)on migration and invasion of tongue squamous cell carcinoma Tca81 1 3 cells and its possible mechanism.Methods Tca81 1 3 cells were treated in different concentrations of IBC in vitro.Cell proliferation was detected by MTT;Wound healing as-say and Transwell chamber assay were used to detect the ability of cell migration and invasion;Western blot was applied to detect the expression of Akt,p-Akt, MMP-2 and MMP-9 proteins.Results IBC could in-hibit the proliferation of Tca81 1 3 cells in a concentra-tion-and time-dependent manner.IBC can reduce cell migration and invasion.Western blot showed that IBC could an decrease the expression of p-Akt,MMP-2 and MMP-9 proteins in a concentration-dependent manner. However,the level of Akt was not affected by the con-centration of IBC treatment.Conclusion IBC could inhibit the proliferation, migration and invasion in Tca81 1 3 cells and its mechanism may be associated with the down-regulation of MMP-2 and MMP-9 pro-teins and the inhibition of phosphorylation of upstream Akt.

ABSTRACT

BACKGROUND:Autologous fat transplantation has been widely used in soft tissue defect repair and cosmetic surgery, and the 1-year transplant survival rate is 20%-80%. Therefore, the establishment of timely and adequate blood supply at early period after transplantation is very important for the survival of transplanted fat tissues. OBJECTIVE:To observe the proliferation of human adipose-derived mesenchymal stem cel s transfected with vascular endothelial growth factor 165. METHODS:Human adipose-derived mesenchymal stem cel s were subcultured in vitro. Recombinant adenovirus carrying vascular endothelial growth factor 165 and blank virus were respectively transferred into adipose-derived mesenchymal stem cel s. Cel s cultured normal y served as blank group. RESULTS AND CONCLUSION:Compared with the control and blank groups, the expressions of vascular endothelial growth factor 165 mRNA and protein were higher in the experimental group (P<0.05). Experimental findings suggest that the recombinant adenovirus carrying vascular endothelial growth factor 165 transferred into adipose-derived mesenchymal stem cel s cannot only maintain the expression of target protein but also obviously promote the proliferation of adipose-derived mesenchymal stem cel s.

ABSTRACT

Objective To prepare TF conjugated doxorubicin and Rg 3 loaded liposome and evaluate their properties and effect on the treatment of gastric cancer. Method The liposomes were prepared by thin iflm hydration method. The efifciency of cellular uptake on MKN-28 cells in vitro was evaluated. The anti-proliferation efifciency of TF-LP-DOX/Rg 3 was evaluated by MTT assay. Tumor spheroids were used to evaluate anti-tumor ability of TF-LP-DOX/Rg 3. Results The result demonstrated that TF-LP uptaken by MKN-28 were 2.9 times higher than that of LP(P<0.01). The MTT assay and the inhibition of tumor spheroids in vitro conifrmed strong inhibitory effect of TF-LP-DOX/Rg 3. Conclusion TF-LP-DOX/Rg 3 easily prepared and it is a potential delivery system for the treatment of gastric cancer.

ABSTRACT

Bufalin is one of the major active components of Chan Su,a traditional Chinese medicine.Studies at home and abroad have confirmed that bufalin can inhibit cell proliferation significantly in many human leukemia and solid cancer cell lines by inhibiting endothelial hyperplasia and angiogenesis,and inducing cell differentiation and apoptosis. With the deepening of study,it is showed that bufalin has extensive anticancer activities and it has potential clinical value with very low drug concentration.

ABSTRACT

Objective To detect the expression of BAG-1 and p53 in postoperative patients paraffin section of non-snmll cell lung cancer (NSCLC), and retrospectively analyze the relationships of clinical feature and platinum-based chemotherapeutic sensitivity in NSCLC, to further investigate the resistance mechanism of platinum. Methods The expression of BAG-1 and p53 was measured by immunohistochemistry in paraffin sections of 125 NSCLC postoperative patients. The existence and prognosis were analyzed after follow-up. Results BAG-1 and p53 showed high expression in NSCLC (positive rate 64.8 %, 46.7 %); BAG-1 positive or p53 negative expression had survival advantage(P<0.05); The group of BAG-1, p53 negative expression could benefit from platinum-based chemotherapy (P<0.05). Conclusion Expressions of BAG-1, p53 correlates with platinum-based chemotherapeutic sensitivity, and maybe become new types of clinical prognostic indicators and contribute to individualized treatment options for NSCLC.

Background and purpose:The development of breast cancer is a multi-step process associated with changes in host chromosome,and some of which correlate with the appearance and progression of the tumor. We studied the expression of BUBl,APC protein in normal breast,hyperplasia of mammary glands and breast cancer tissues and evaluated its difference and clinical significance. Methods:The inmmunohistochemical method was used to analyze the expression of BUB1 and APC protein in normal breast,hyperplasia of mammary glands and breast cancer tissues.
