ABSTRACT

Objective:To investigate the characteristics and evolution of mild motor symptoms (MMS) in patients with prodromal Parkinson′s disease (pPD).Methods:Based on the pPD cohort screened by Parkinson′s Disease Prodromal Clinical Assessment Scale in Nanjing community from July 2018 to December 2020, the clinical data of 30 patients with pPD who completed the baseline assessment and were followed up for at least 1 year were analyzed. According to the Unified Parkinson Diease Rating Scale Ⅲ (UPDRS-Ⅲ) score, the patients were divided into MMS group (UPDRS-Ⅲ score>3) and non-MMS group (NMMS group, UPDRS-Ⅲ score≤3). The differences and evolution characteristics of clinical characteristics between the 2 groups were compared. Multivariate linear regression was used to analyze the risk factors of motor symptom progression in pPD patients.Results:Among the 30 patients with pPD, 7 of 23 patients in the MMS group were converted to PD at the end of follow-up, 1 of 7 patients in the NMMS group were converted to PD at the end of follow-up. The UPDRS-Ⅲ score [10.00 (7.00, 17.00)], Montreal Cognitive Assessment Scale (MoCA) score [25.50 (24.75, 28.00)] and the Hamilton Anxiety Scale (HAMA) score [9.00 (5.00, 13.00)] at the end of follow-up of pPD patients were significantly higher than those at baseline [7.00 (4.00, 12.00), 24.00 (22.75, 25.25) and 8.00 (2.00, 11.00)], and the differences were statistically significant ( Z=-3.505, P<0.001; Z=-2.956, P=0.003; Z=-2.427, P=0.015).Subgroup analysis showed that UPDRS-Ⅲ score [11.00 (7.00, 18.00)], MoCA score [25.00 (24.00, 27.00)] and HAMA score [ 9.00 (6.00, 15.00)] at the end of follow-up in the MMS group were higher than those at baseline [8.00 (6.00, 12.00), 24.00 (22.00, 25.00) and 9.00 (3.00, 11.00)], and the difference was statistically significant (Z=-2.768, P=0.006; Z=-2.457, P=0.014; Z=-2.250, P=0.024). The Non-Motor Symptoms Questionnaire score at the end of follow-up in the MMS group (8.96±5.20) was significantly lower than that in the baseline (11.04±4.41), and the difference was statistically significant ( t=2.441, P=0.023).There was no significant difference in Mini-Mental State Examination (MMSE), Hamilton Depression Scale (HAMD), Rapid Eyes Movement Sleep Behavior Disorder Questionnaire-Hong Kong (RBDQ-HK) and Sniffin′ sticks olfactory test score at the end of follow-up in the MMS group. Only UPDRS-Ⅲ score in the NMMS group was increased at the end of follow-up [7.00 (5.00, 8.00)] compared with the baseline [4.00 (1.00, 4.00)], and the difference was statistically significant ( Z=-2.375, P=0.018). There was no significant difference in MoCA, MMSE, HAMA, HAMD, RBDQ-HK, and Sniffin′ sticks olfactory test score between the NMMS group and the baseline at the end of follow-up. Conclusion:The clinical conversion rate of pPD patients with MMS is high,and screening of this population should be paid attention.

ABSTRACT

Objective To evaluate the change of right ventricular function before and after single hemodialysis by 3-dimensional speckle tracking imaging( 3D-STI) . Methods Seventy-six patients with end-stage renal disease ( ESRD ) and 35 controls were included .ESRD patients were divided into normal left ventricular ejection fraction group( LVEF ≥ 50% ,46 cases ,group A ) and decreased LVEF group( LVEF <50% ,30 cases ,group B) .Conventional echocardiography and 3D-STI parameters including right ventricular end diastolic volume( RVEDV ) ,right ventricular end systolic volume ( RVESV ) ,right ventricular ejection fraction( RVEF) ,right ventricular septal longitudinal strain [ RVLS ( Septum ) ] ,right ventricular free wall longitudinal strain [ RVLS( Freewall ) ] ,basal segment of right ventricular diameter[ RVDd( base ) ] ,right ventricular middle diameter [ RVDd ( mid ) ] ,right ventricular long diameter ( RVLD ) and the upper and lower right ventricular fractional area change ( FAC) were performed in patients and controls before and after a single hemodialysis . Results ① RVEDV ,RVESV ,RVDd( base) ,RVDd( mid) ,RVLD before a single hemodialysis in group A and B were significantly higher than those in control group ( all P < 0 .05) . RVEF , RVLS Freewall ,RVLS( Septum) and FAC in group A and B were significantly lower than those in control group ( all P < 0 .05) . ② RVEDV ,RVESV ,RVEF ,RVLD ,RVLS ( Freewall) were significantly lower in group A after a single hemodialysis ( all P < 0 .05 ) ;RVEDV ,RVESV ,RVEF ,RVLS ( Freewall) ,RVLS ( Septum) were significantly lower in group B after a single hemodialysis( all P < 0 .05) ;RVEDV ,RVESV , RVEF ,RVLS( Septum) and RVLS( Freewall) in group B before a single hemodialysis were lower than those in group A ( all P < 0 .05) . The difference of RVLS Septum decrease in group B after a single hemodialysis was higher than that in group A( P < 0 .05) ;The linear correlations analysis showed that the difference of body weight before and after a single hemodialysis was negatively related with the difference of RVLS ( Freewall) before and after a single hemodialysis( group A : r = 0 .351 , P = 0 .017 ;group B : r = 0 .696 ,P =0 .000) . Conclusions A single hemodialysis may have injurious effects on right ventricular systolic function and reduce strain value ,which is more significant in group B( LVEF < 50% ) . Cardiac dysfunction of patients may be remitted by reasonable control of water intake .

High-throughput metabolomics have developed very rapidly in recent years and been widely used in medicinal plants research .At present ,high-throughput metabolomics mainly applied in the following areas ,quality control of medicinal plants by fingerprints ,metabolites difference comparison before and after genetic engineering ,monitoring metabolites change in different environment and gene function study .High-throughput metabolomics have a great future ,but still have some challen-ges ,such as the requirements for more sophisticated equipment and complexity of data integration .With the advancement of science and technology ,high-throughput metabolomics will be an irreplaceable tool for the research of medicinal plants .

ABSTRACT

Baphicacanthus cusia ,widely distributed in the Southwest ,Southern and Eastern regions of China ,is an im-portant medicinal plant of Acanthaceae family .Indigo made from stem and leaf of Baphicacanthus cusia in Fujian has the best quality in China ,and is known as “Jian Indigo naturalis” ,which is the genuine medicinal of Fujian Province .The rhizoma of Baphicacanthus cusia could be used as medicine ,called“Nanbanlangen” ,which together with indigo were included in the“Chi-nese Pharmacopoeia” .Indigo and its original plant both contain indirubin ,which has anti-cancer activity .Indirubin is an active ingredient of Huangdai Pian and Danggui Longhui Wan ,two kinds of traditional Chinese medicine ,which have been successful-ly used in the treatment of malignancies such as chronic myelogenous leukemia .The international advances in the biological characteristics ,genetic diversity ,cultivation technology and molecular biology of Baphicacanthus cusia germplasm resources were summarized .The main problems in Baphicacanthus cusia germplasm resources research are indicated ,which could pro-vide references for the further study and application of Baphicacanthus cusia germplasm resources .

ABSTRACT

BACKGROUND:Ultrasound has been widely used in developmental hip dysplasia screening in infants and young children, but there are significant differences among different inspection findings, even in the reexamination by the same examiner. Therefore, the accuracy of ultrasound imaging in the diagnosis is of insufficient evidence. OBJECTIVE:Through virtual reality three-dimensional reconstruction images showing the structure of acetabulum and femoral head space, to obtain a new parameter in order to enrich measurement standards of Graf ultrasonic diagnosis technology and to further improve the credibility of the diagnosis of developmental dysplasia of the hip. METHODS:Ten fresh cadaver specimens of Chinese infants were selected, including five males and five females. Two-dimensional ultrasonic method was employed for hip screening, and at the same time, multi-slice spiral CT was used for three-dimensional reconstruction fol owed by volume reconstruction and fitting of surface reconstruction to display the anatomic structure around the hip and to simulate Graf’s two-dimensional ultrasound method to measure a andβangles on three-dimensional virtual reality images. RESULTS AND CONCLUSION:As confirmed by three-dimensional reconstruction, Graf’s two-dimensional ultrasound could be used to measure a and angles, truly reflecting the development of bony and cartilaginous acetabulum. Graf’s two-dimensional ultrasound contributed to the proper evaluation of beonatal hip dysplasia, which can be used as a routine examination for developmental dysplasia of the hip. These indicate that to master Graf’s ultrasound technology and strictly grasp the evaluation standards for developmental dysplasia of the hip is the important guarantee for diagnosis of developmental dysplasia of the hip.

ABSTRACT

Objective To study the effect of humic acid (HA) and soil on dissolution performance of depleted uranium (DU).Methods Using the static dissolve-adsorption experiment and inductively coupled plasma-mass spectrometry(ICP-MS) to determine uranium concentration and 235U/238U isotope ratios of the samples at different time and study the dissolution of DU in water.Results The solubility of DU in water was reduced by 90% by adding HA.Soil could increase the solubility of DU in water by nearly 25%,adding an appropriate amount of HA could play a supporting role on the dissolution of DU,in this experiment adding 5% of HA was best.Conclusions Soil and HA could produce positive and negative impact on the solubility of DU in water,and the combined effect of the two relied on the complex absorption and complexation reactions of soil,HA and dissolved uranium ions.

ABSTRACT

Background Whether negative suction during excimer laser in situ keratomileusis(LASIK) affect the structure and function of retina or not is in controversy,but it seems that temporary hypertention induced by negative suction is a key factor of impairment of retina in LASIK.ObjectiveThis study attempts to study the influence of transient high intraocular pressure (IOP) during LASIK on the contents of retinal amino acid.MethodsThe both eyes of 45 New Zealand white rabbits were suctioned for different periods (20s,45s,3min) with negative pressure generator during the LASIK to make the instantaneous high IOP models,and LASIK without negative suction was performed in the both eyes of 15 rabbits in the control group.The changes of the contents of retinal amino acids were evaluated with High Performance Liquid Chromatography(HPLC) at 0,7,10,14 and 28 days postoperatively and compared with those of control group.ResultsThere were no statistically significant differences in the contents of retinal amino acids among different time points after operation in negative suction for 20s group and 45s group,respectively(P>0.05).At negative suction for 3minuts,the content of glutamic acid in retina was significantly increased in comparison with control group in 7,10,14 and 28 days (P<0.05),and no statistically significant difference was seen in the contents of glutamic acid at postoperative instant group compared with control group(P>0.05).A statistically significant difference in the contents of glutamine,tryptophan,phenylalanine was revealed among postoperative 10 days,14 days and 28 days groups comparison with control group (P<0.05).ConclusionThe acute IOP elevation caused by negative suction during LASIK results in the reversible increase of retinal amino acids.The duration of negative suction time influent the reconstruction of retinal structure.

ABSTRACT

Objective To establish the mice model of lung injury induced by radon,and to observe the changes of pulmonary lesion at different doses and to analyze the influence of radon exposure on P53 and Bcl-2、Bax expression in lung tissue.Methods Mice were exposed to radon of 30 and 60 WLM,respectively.Apoptosis was detected by terrainal deoxynucleotidy transferse-mediated dUTP-biotin nick end labeling(TUNEL).The expressions of P53,Bcl-2 and Bax protein were observed by immunohistochemistry and Western blot.Results Compared with those in the control group,the apoptotic indexes increased significantly in the 30 and 60 WLM groups(t=18.11,-10.30,P<0.05).The protein expression of P53 was significantly increased(t=-11.08,P<0.05).The expression levels of P53 and Bax were remarkably inereased(t=-7.00,-2.52,P<0.05),while the expression of Bcl-2 protein was significantly decreased in the 60 WLM group(t=4.36,P<0.05).But Bcl-2,Bax expression was decreased in both 30 WLM and 60 WLM groups(t=2.78,4.07,P<0.05).Conclusions Radon could induce pulmonary lesion of mice.It may be involved in the regulation of apoptosis of pulmonary lesion by the P53,Bcl-2、Bax pathway.

ABSTRACT

Objective To study the expression of JNK/SAPK(c-Jun NH2-terminal kinase/stress activated protein kinase)in lung and bronchus of radon-exposed mice.Methods Male BALB/c mice were exposed to radon and its progeny with the cumulative dose of 0.02,30 or 60 working level month(WLM),respectively.The expression levels of JNK/SAPK in lung and bronchus were determined with Real-Time PCR,Western blot and immunohistochemistry methods.Results The JNK mRNA levels in lung tissues of mice exposed to radon of 30 and 60 WLM were higher than those of the control by 3.56 and 2.96 times,respectively.The relative expression levels of JNK and phospho-JNK proteins were higher than those of the control by using Western blot and immunohistochemistry methods.Condusiom Expose to the radon and its progeny might activate the JNK/SAPK intracellular signaling pathway.

ABSTRACT

<p><b>BACKGROUND</b>To construct the Bifidobacterium Infantis/CD tumor targeting gene therapy system.</p><p><b>METHODS</b>PCR expanded CD gene and pGEX-1LambdaT plasmid was transferred into E.Coli. JM109. with TSS solution. Then a dual restriction enzyme (EcoR I and BamH I) digestion was carried out to cut CD gene and pGEX-1LambdaT plasmid. Two segments (4.9 kb and 1.3 kb) were selected and extracted from the gel. T4 DNA Ligase was added to these two segments' mixture. Finally, reconstruction fragment was transferred into Bifidobacterium Infantis by electroporation. The plasmid was extracted from the positive clone selected and testified by an agarose eletrophoresis after enzymed.</p><p><b>RESULTS</b>A positive transferred Bifidobacterium Infantis with 6.2 kb long reconstruction plasmid fragment with CD gene was established which was in accordance with theoretical calculation.</p><p><b>CONCLUSIONS</b>The Bifidobacterium Infantis/CD tumor targeting gene therapy system could be successfully constructed.</p>
