ABSTRACT

Objective To study the effect and mechanism of berberine (BBR) on the lung metastasis of mouse breast cancer via epithelial-mesenchymal transition (EMT). Methods CCK-8 and Transwell migration assays were utilized to investigate the proliferation and migration properties of breast cancer 4T1 cells after BBR treatment.Mouse 4T1-Luc cells were injected into mice under the fourth mammary fat pad, and the mice were then randomly divided into the control and BBR groups. The mice in the BBR group received daily intraperitoneal injections of BBR working solution and those in the control group were continuously intraperitoneally injected with the same volume of the solvent used to dissolve BBR powder.Tumor metastasis in the lungs of living mice was detected by using an in vivo imaging system. After 42 days of administration, lung metastasis was measured via microscopy and HE staining.Western blot analysis was used to examine the effects of BBR on the expression of EMT-related proteins (Vimentin and Snail) as well as the activation of the Akt and ERK signaling pathways. Results BBR significantly promoted 4T1 cell migration (P < 0.05).In vivo experiments showed that the number of lung metastases in the BBR group had significantly increased compared with that in control group (P < 0.05) as observed under microcopy and histological staining.Compared with the control group, BBR upregulated the expression levels of Vimentin and Snail as well as the phosphorylated levels of p-Akt and p-ERK (P < 0.05). Conclusion BBR may promote EMT and lung metastasis of breast cancer 4T1 cells by activating the expression of proteins in the p-Akt and p-ERK pathways.

ABSTRACT

In order to effectively treat respiratory diseases, a non-invasive positive pressure ventilator system is designed, the overall structure design of the system is proposed, and the hardware construction is completed. The breathing state of the patient is identified by the threshold triggering method of the flow rate of change, and the calculation of the flow rate of change is realized by the least squares method. At the same time, the breathing parameters are calculated in real time according to the flow-time and pressure-time characteristic curves. In addition, CMV, CPAP, BiPAP and PSV ventilation modes are also implemented. Finally, the parameter measurement accuracy and ventilation mode setting tests are carried out. The results show that the calculation of key breathing parameters provided by the system meets the relevant standards, and supports the stable output of 4 ventilation modes at the same time, provides breathing treatment for patients, and meets the basic functional requirements of the ventilator.

ABSTRACT

Objective To establish an HPLC method for simultaneous contents determination of paeoniflorin, protocatechuic acid and chlorogenic acid in Penyanjing Capsule. Methods The chromatographic separation was performed on a SHIMADZU VP-ODS-C18 column (4.6 mm × 250 mm, 5 μm); the column temperature was maintained at 30 ℃; A gradient elution of acetonitrile-0.15% phosphoric acid aqueous solution was adopted at the flow rate of 1.0 mL/min; The UV detection wavelengths were at 231, 255, and 326 nm; The injection volume was 20 μL. Results Paeoniflorin, protocatechuic acid and chlorogenic acid could be separated efficiently with this condition. The regression equation was Y=1546.4128X+127.0756 (r=0.9999), Y=2925.8468X+2204.1076 (r=0.9998), Y=893.9045X-261.7315 (r=0.9994), respectively. Paeoniflorin, protocatechuic acid and chlorogenic acid were linear in the range of 150.054–1254.50 ng, 162.33–1352.75 ng, 11.43–95.25 ng, and the average recoveries were 97.60%, 102.09% and 98.52%. Conclusion The method is simple, convenient and accurate, which can be used to the quality control of Penyanjing Capsule.

ABSTRACT

Objective To probe into the clinical apply value of sentinel lymph node detection in patients with cervical cancer. Methods 33 patients with cervical cancer underwent SLNS detection by using blue dye or methlene blue 4ml or lymphazurin was injected into the cervix at 4 points around the tumor at the time of radical hysterectomy and bilateral pelvic lymphadenectomy1 tumor characteristics surgical findings and specific locations of lymphatic dye uptake were recorded and correlated with final pathology results. Results Among 33 patients undergone this detection, dye uptake was not seen in 1 patient, dye uptake was seen in 32 patients, the SLNS were successfully detected in 81.25%(26/32);7 patients didn't discriminated SLN,the rate was 21.88%(7/32).There were 6 cases who were discovered pelvic cavity lymph nodes metastases in 26 patients discriminated SLN successfully,in all 12 piece metastases lymph nodes were SLN,sensitivity was 6/6,accuracy was 100%(26/26),fake negative rate was 0.Conclusion Sentinel lymph node dectection by the way of blue dye has more clinical apply value in patients with cervical cancer.
