ABSTRACT

Objective:To investigate the effect of piceatannol (PIC) on the proliferation, apoptosis and cell cycle of MDA-MB-468 triple negative breast cancer cells and its mechanism. Method:The methylthiazolyldiphenyl-tetrazoliu bromide (MTT) colcorimetry method was used to investigate the effect of different concentrations of PIC (0, 2.5, 5.0, 10.0, 20.0, 40.0, 80.0, 160.0 μmol·L<sup>-1</sup>) on the cell viabilities of triple negative breast cancer MDA-MB-468 cells and calculate the half maximal inhibitory concentration (IC<sub>50</sub>) value, the effect of different concentrations of PIC (5.0, 10.0, 20.0 μmol·L<sup>-1</sup>) on the cell cycle of MDA-MB-468 were investigated by flow cytometry with propidium iodide (PI) staining. The apoptotic effect of PIC (5.0, 10.0, 20.0 μmol·L<sup>-1</sup>) on MDA-MB-468 cells in triple negative breast cancer was investigated by flow cytometry with cell apoptosis detection Annexin V-FITC and PI double staining. Western blot was used to investigate the effect of different concentrations of PIC (5.0, 10.0, 20.0 μmol·L<sup>-1</sup>) on the proliferation and apoptosis of MDA-MB-468 cells and detect the expressions ofsecreted glycoprotein Wnt/<italic>β</italic>-catenin pathway related proteins. Result:MTT results showed that compared with the blank group, PIC could inhibit the proliferation of MDA-MB-468 cells in a concentration-dependent manner (<italic>P</italic><0.05, <italic>P</italic><0.01), with IC<sub>50</sub> at(39.4±4.6)μmol·L<sup>-1</sup>. Compared with the blank group, PIC could increase the percentage of MDA-MB-468 cells in G<sub>0</sub>/G<sub>1</sub> phase about cell cycle in a concentration-dependent manner (<italic>P</italic><0.01). Compared with the blank group, 5.0, 10.0, 20.0 μmol·L<sup>-1</sup> PIC could induce apoptosis of MDA-MB-468 cells for 48 h(<italic>P</italic><0.01), and the apoptosis rate of MDA-MB-468 cells reached 49.87% when treated with 20.0 μmol·L<sup>-1</sup> for 48 h. Compared with the blank group, PIC could significantly reduce the expressions of <italic>β</italic>-catenin, proto-oncogene (C-myc) and adhesion factor (CD44) proteins in MDA-MB-468 cells, significantly inhibit the phosphorylation of<italic> </italic>protein kinase B (Akt) and p38 mitogen activated protein kinase (p38 MAPK) proteins and the protein expression of B lymphocyte tumor-2 (Bcl-2), and enhance cysteine aspartic acid protease-3 (Caspase-3), Bcl-2 related X protein (Bax) and phosphorylated <italic>β</italic>-catenin protein expression(<italic>P</italic><0.01). Conclusion:PIC may inhibit the proliferation of MDA-MB-468 cells by inhibiting the Wnt/<italic>β</italic>-catenin signaling pathway, block the cell cycle in G0/G1 phase, and induce its apoptosis.

Purpose@#The present study determined the effects of saphenous nerve stimulation (SNS) at different stimulation frequencies on bladder reflex and explored a possible action mechanism of tibial nerve stimulation (TNS) on bladder activity in cats. @*Methods@#Two bipolar nerve cuff electrodes were implanted on the saphenous nerve and the contralateral tibial nerve in 13 cats, respectively. Multiple cystometrograms were obtained to determine the effects of single SNS at different frequencies and that of combined SNS and TNS on the micturition reflex by infusing normal saline. @*Results@#SNS at 1 Hz significantly reduced the bladder capacity (BC) to 59.8%±7.7% and 59.3%±5.8% of the control level at the intensity threshold (T) and 2T, respectively (P0.05), while combined stimulation at 1T significantly increased the BC to 122.7%±1.9% of the control level and induced an inhibitory effect which was similar to that TNS at 2T. @*Conclusions@#The current study revealed that SNS reduced and increased BC depending on different stimulation frequencies. The combined SNS and TNS maximized the clinical efficacy at a low intensity. Also, SNS may be a potential therapeutic mechanism of TNS.

ABSTRACT

Objective To investigate the iodine nutritional status of pregnant women in Jinan City,which may provide a basis for scientific supplementation of iodine.Methods Ten counties (districts) in Jinan City were divided into five areas as east,west,south,north and middle in 2018.At least one township (street) was selected from each area,and 20 pregnant women were selected from each township (street).Personal information was collected through standardized questionnaires,and household salt samples and urine samples of pregnant women were collected.Salt iodine and urinary iodine were detected by the "General Test Method in Salt IndustryDetermination of Iodine" (GB/T 13025.7-2012) and "Method for Determination of Iodine in Urine by As3+-Ce4+ Catalytic Spectrophotometry" (WS/T 107-2006),respectively.Results A total of 1 169 pregnant women's household salt samples were collected,the median of salt iodine was 22.6 mg/kg.The coverage rate of iodized salt was 90.5% (1 058/1 169),the qualified rate of iodized salt was 75.8% (802/1 058),and the consumption rate of qualified iodized salt was 68.6% (802/1 169).A total of 1 169 urine samples of pregnant women were tested,the median of urinary iodine was 141.9 μg/L,indicating the urinary iodine of pregnant women was insufficient (< 150 μg/L).Conclusions The iodine nutrition of pregnant women is slightly inadequate in Jinan City.It is necessary to strengthen the measures in prevention and control of iodine deficiency,and promote the health education measures in pregnant women to improve their iodine deficiency status.

ABSTRACT

Objective@#To study the clinical characteristics, image findings, therapeutic method and prognosis of metanephric adenoma.@*Method@#The clinical characteristic, image findings, operation methods and prognosis of 16 metanephric adenoma patients treated at Department of Urology, Peking University First Hospital from January 2004 to March 2016 were analyzed retrospectively.@*Results@#There were 6 male and 10 female patients in the study. The mean age of patients was 33.7 years (ranging from 14 to 83 years). Two patients came to the hospital because of fever, while other 14 patients had no symptoms and found renal tumor by medical examination. One case was found polythemia vera and another 1 case showed mild anemia. Serum creatine of all the cases were in normal range. The tumor of 11 cases were at left side and 5 cases were at right. All patients took urinary tract ultrasound. Fifteen patients took CT examination. Among them, 14 cases were solid mass and 1 case was cystosolid.CT value was (41±4) HU. CT scan showed that the tumor was slight enhanced and CT value increased to (77±9) HU. Six patients took MRI examination. The MRI showed high or low signal of T1WI or T2WI scans.Tumor size was (4.7±3.9)cm (ranging from 1.7 to 17.5 cm). All 16 patients took operation and 11 of them took laparoscopic surgery while the other 5 cases took open surgery. Eleven cases took partial nephrectomy, 4 cases took nephrectomy and 1 case took nephroureterectomy. The surgical procedures were all successful and no complications occured during perioperative period. All cases were all confirmed metanephric adenoma by postoperative pathology and surgery cut edge were all negative. Immunohistochemical study showed that the positive rate of Vimentin, CD57, AE1/AE3, WT1, CK7 and AMACR respectively were 16/16, 15/16, 12/16, 10/16, 3/16 and 2/16. The median follow-up time of 16 cases was 44 months (ranging from 8 to 125 months) and none had recurrence or metastasis.One case died 125 months after surgery because of advanced age(83 years old).@*Conclusions@#Metanephric adenoma is difficult to be diagnosed relying on clinical characteristics and image features. Pathology can help confirm the diagnosis. Partial nephrectomy is the first choice for operation and can achieve good prognosis. But it still needs a regular follow-up.

ABSTRACT

Objective@#To investigate the clinical features and prognosis of rare subtypes of renal cell carcinoma.@*Methods@#This retrospective study collected the data of 52 rare subtypes of renal cell carcinoma of patients who underwent surgery from January 2002 to December 2014 at Department of Urology, Peking University First Hospital. There were 12 patients with collecting duct carcinoma, 5 patients with Xp11.2 translocation renal cell carcinoma, 5 patients with mucinous tubular and spindle cell carcinoma, 30 patients with unclassified renal cell carcinoma. The study group included 25 male and 27 female patients, with mean age of 52 years. The mean tumour size was (6.5±3.9) cm (range: 1.5 to 21.0 cm). The basic clinical features, gross appearance, Fuhrman nuclear grade, TNM staging and prognosis of rare subtypes of RCC were studied. The OS curves were obtained for rare subtypes of renal cell carcinoma using the Kaplan-Meier method and compared using a Log-rank test.@*Results@#The rate of lymph node and distant metastasis were 34.6% (18/52) and 17.3% (9/52). Malignancies were screened and detected by color Doppler ultrasonography or CT scan, however, no case was diagnosed before operation or aspiration, all cases were confirmed by the pathological examination. The average period of postoperative follow-up process was 65 months, and the mean survival time was (34±23) months.@*Conclusion@#The clinical features of rare subtypes of renal cell carcinoma are similar to those of clear cell renal cell carcinoma, while the imaging changes will be helpful for diagnosis before operation.

ABSTRACT

Objective@#To study the impact to operation safety of preoperative renal artery embolization for management of ≥10 cm renal cell carcinoma.@*Methods@#The clinical data of 239 cases with ≥10 cm renal cell carcinoma which all had underwent operation in Department of Urology, Peking University First Hospital from January 2002 to December 2014 were retrospectively analyzed. Fifty-three patients underwent preoperative renal artery embolization (therapeutic group) and 186 patients did not (control group). The effect of embolization on operative time, transfusion requirements, hospitalization, ICU stay and perioperative complications were analyzed by comparing the two groups using rank sum test and χ2 test or Fisher exact test.@*Results@#Comparing the therapeutic group and control group, there was significant difference in tumor location (on the left or right). The mean age, sex, mean primary tumor size, and TNM stage were similar in both groups. Comparing the therapeutic group and control group, there were more open surgeries in therapeutic group (96.2% vs. 82.3%, χ2=6.438, P=0.013). There were no significant differences in mean operative time (238 (525) minutes vs. 208 (583) minutes, Z=-2.182, P=0.062). The mean blood transfusion (700 (1 900) ml vs. 925 (8 800) ml, Z=-1.064, P=0.006) had significant difference. The therapeutic group had a longer mean hospitalization (21 (50) days vs. 15 (79) days, Z=-4.322, P=0.000) and higher rate of intensive care unit stay (54.7% vs. 34.4%, χ2=6.103, P=0.027). There was no significant difference in perioperative complications between two groups (0 vs.3.2%, P=0.408).@*Conclusion@#Preoperative renal artery embolization in ≥10 cm renal cell carcinoma patients undergoing operation provides benefit in increasing operation safety and reducing perioperative death.

ABSTRACT

Objective:To discuss the therapy for ureteral endometriosis.Methods:The clinical data of 25 cases of histopathologically confirmed ureteral endometriosis during 2001 -2015 were retrospectively analyxed.Results:In the 25 cases,all the patients took urinary ultrasound for examination before sur-gery,of whom 21 (84%)were examined by CT and 5 (20%)by MRI.Three (12%)cases underwent preoperative KUB and intravenous pyelogram (IVP)examination.Four (16%)cases were examined by retrograde pyelography before surgery.Eight (32%)of them took cystoscope for examination and 11 (44%)took preoperative radionuclide renal dynamic imaging examination.All of these cases were affec-ted with unilateral ureter,13 (52%)in the right and 12 (48%)in the left.The ureter lesions were at the upper part in 21 (84%)cases and at the middle part in 4 (16%)cases.In these cases,11 (44%) received partial ureteral resection and end-to-end ureteral anastomosis,10 (40%)received partial ure-teral resection and ureterocystoneostomy,3 (12%)received retroperitoneal laparoscopic nephroureterec-tomy,and 1 (4%)received endoscopic resection of ureteral endometriosis lesion.All of these cases were confirmed with ureteral endometriosis by post-surgery pathology results,with the expression of CA125 and ER in the glandular tissue and expression of PR in the mesenchymal tissue inside the ureteral muscle de-tected by immunohistochemistry.Four (16%)cases took postoperative adjuvant hormonal therapy.And no recurrence was found among 16 (64%)cases with the successful follow-up which ranged from 3 to 76 months and the median follow up was 28 months.Fifteen cases were submitted to the follow-up by urinary ultrasound (93.8%)and 5 (31.3%)underwent CT for examination.All the cases relieved from hydro-nephrosis,and symptoms of 10 (63.5 )cases disappeared.The cases with double-J stent all had the stent removed within 3 to 6 weeks.Conclusion:Surgical procedures should be considered as the main therapy for ureteral endometriosis.We recommend ureterolysis for patients with mild ureteral obstruction and hydronephrosis.As for those with moderate and severe ureteral obstruction and hydronephrosis,we recommend partial ureteral resection.When the situation comes to patients with little renal function of the affected side,the recommended management is nephroureterectomy.

ABSTRACT

To investigate the effects of carbamazepine (CBZ) on the plasma concentrations of valproic acid (VPA) and its toxic metabolite 2-propyl-4-pentenoic acid (4-ene VPA) in epileptic patients, the plasma concentrations of VPA and 4-ene VPA were determined, and the effect of CBZ on pharmacokinetics of VPA was evaluated. All patients had been divided into two groups (VPA group, n = 87; and VPA+CBZ group, n = 19). As compared to VPA group, the combination of CBZ significantly (P < 0.01) decreased the trough concentration of VPA [VPA group, (69.5 +/- 28.8) microg x mL(-1); VPA+CBZ group, (46.3 +/- 25.6) microg x mL(-1)] and does-adjusted VPA trough concentration [VPA group, (4.89 +/- 2.21) microg x mL(-1) x mg(-1) x kg(-1); VPA+CBZ group, (3.14 +/- 1.74) microg x mL(-1) x mg(-1) x kg(-1)]. However, the addition of CBZ did not influence the concentration of 4-ene VPA. The present study revealed that coadministration of CBZ can reduce VPA plasma concentration and may impact VPA clinical effect, therefore therapeutic drug mornitoring of VPA should be used when combined use of CBZ and VPA.
