ABSTRACT

Objective:To establish a qualitative and quantitative analysis method of ginsenoside Rb1, cinnamic acid, paeoniflorin and berberine in Wenxin formula based on UPLC-MS/MS MRM model so as to provide a rapid and accurate evaluation method for the research and development of new drugs. Methods:Adopting Waters ACQUITY UPLCTM HSS T3 (2.1 mm×100 mm, 1.8 μm) and gradient elution was performed by mobile methanol-0.03% formic acid water. chromatographic column was used with electrospray ionization source in the scanning mode of multiple reactive ion monitoring (MRM) for ion separation and screening. The retention time and the relative abundance ratio (qualitative ion pairs/quantitative ion pairs) was used for qualitative analysis, while quantitative ion pairs was used for quantitative analysis. Results:The four components tested in Wenxin Formula have been qualitatively detected and showed a good linear relationship, the method showed accuracy, precision, repeatability, and stability, the recovery rate was 96.24%-104.19% and RSD was 1.29%-3.30%. Conclusion:The established method is simple, accurate, rapid and sensitive, which is reliable for the qualitative and quantitative study of the four main components in Wenxin Formula.

ABSTRACT

[Objective] To optimize the conditions of the extracting process of Hujin Granules. [Methods] The orthogonal design was applied. With the total emodin and the total anthraquinone (TA) content as the parameters for the alcohol-extraction, the concentration of alcohol, the volume of solvent and the extracting time were used for optimization of alcohol-extraction. With the total polysaccharide (TP) as the parameters for water-extraction, the soaking time, the volume of water and the extracting time were used for optimization of water-extraction. [Results] The optimum conditions of alcohol-extraction were: extracting with 70% alcohol 245 mL for 2 hours and extracting twice. The optimum conditions of water-extraction were: extracting with 80 mL water (not for soaking) for 1.5 hours, extracting 3 times. [Conclusion] The results indicate that the extracting process is rational and feasible, and can provide evidence for the extracting process of Hujin Granules.

ABSTRACT

Objective To study the effect of Aloe polysaccharides (AP) on the thymocytic apoptosis and cell cycle in ? - ray irradiated mice. Methods Single- cell thymocytes suspension was sampled at different time points to observe the thymocytic apoptosis and cell cycle by flow cytometry. DNA ladders were tested by 1.8 % agarose gel electrophoresis. Transmission electron microscopy was used to examine the ultrastructure of thymocytes. Results Pre- treating with AP (50 mg/kg,ip) 30 min before irradiation could significantly decrease the percentages of apoptotic thymocytes in? - ray irradiated mice 4 h, 8 h and 12 h after irradiation, increase the percentage of thymocytes at G0/G1 phase and reduce the percentage of thymocytes at G2/M phase. It could also lessen the DNA ladders and reduce the number of apoptotic bodies. Conclusion The protective effects of AP on the thymocytes in ? - ray irradiated mice is related with the alleviation of the disorder of cell cycle and the inhibition of the apoptosis of thymocytes.

ABSTRACT

Having referred the Chinese academic publications on TCM during the last decade ,we summarized the researches in Angelica Decoction (AD), which included immunoregulatory, circulatory system, the basis of decoction composition, anti-oxidation, liver injury and compaibility. AD can improve the immunity action, meliorate abnormal circulation, and also has the effects of anti-oxidation and protection of liver injury. Futher study on AD should be focused on its mechanism and material basis in the future.

ABSTRACT

Objective: To study the pharmacodynamic effect of Reganling Oral Liquid(RGL) in treating wind heat epidemic syndrome. Methods: After RGL was administered ig to experiment animals, we observed its antifebrile effect by using pathologic hectic rabbit caused by typhoid bacterin and pathologic hectic rat caused by injection of 2,4 dinitro phenol, anti inflammation effect by weighing the tampon acestoma in rat, anti anaphylaxis effect by using delay anaphylactoid mice caused by injection of 2,4 dinitrochloro benzene. The Liquid Tube Observing Method was used to investigate the bacteriostatic activity of RGL on eight kinds of germina in vitro . Results: RGL could evidently inhibit the raising of animal heat caused by bacterin and chemical, and also could markedly restrain the weight of tampon acestoma in rat and delay anaphylactoid in mice. RGL also had remarkably bacteriostatic activity on eight kinds of germina in vitro which mainly caused wind heat epidemic syndrome. Conclusion: RGL had the evident effects of antifebrile, anti inflammation, anti anaphylaxis and bacteriostatic activity in vitro .
