ABSTRACT

Objective:To investigate the feasibility, effectiveness and safety of ultrasound-guided transperineal prostate biopsy (TPB) with coaxial needle technique, and to improve the pain perception of TPB patients by reducing the number of direct perineal needling.Methods:A total of 200 patients who underwent ultrasound-guided TPB at the first clinical college of Three Gorges University & Yichang Central People′s Hospital from January 2019 to December 2020 were randomly divided into coaxial needle group (coaxial needle positioning puncture, n=100) and traditional group (traditional puncture frame guided repeated transperineal puncture, n=100). Visual analog scale (VAS) was used to evaluate the pain of patients during puncture. The number of samples, time-consuming of puncture, cancer detection rate, VAS pain score and complications between the two groups were compared. Results:The success rate of puncture in the coaxial needle group and the traditional group was 100%, and there was no significant difference in the cancer detection rate between the two groups (48% vs 40%, P>0.05). The average number of samples in the coaxial needle group was larger than that in the traditional group, the average puncture time in the coaxial needle group was less than that in the traditional group, and the average intraoperative VAS score of the coaxial needle group was lower than that of the traditional group, the differences were statistically significant[(14.8±1.8) vs (12.1±1.1), (12.9±1.3)min vs (16.5±1.9)min, (2.6±1.2) vs (4.4±1.4); all P<0.001]. The complication rate of the coaxial needle group was lower than that of the traditional group, the difference was statistically significant (18% vs 39%, P<0.001), the incidences of perineal hematoma and perineal pain in the coaxial needle group were lower than that in the traditional group (1% vs 8%, 8% vs 19%; all P<0.05). Conclusions:Coaxial needle technology for ultrasound-guided TPB can ensure the number of samples and accurate sampling in different areas, significantly reduce the number of direct perineal puncture, improve the pain in the process of puncture, reduce the incidence of postoperative perineal pain, with shorter operation time and fewer complications, which is worthy of clinical promotion.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the change of T lymphocyte subsets, interleukin (IL)-2 and tumor necrosis factor (TNF)-alpha in the tumor- bearing mice and patients with oral cancer receiving thermo-chemotherapy, and investigate the correlation among them.</p><p><b>METHODS</b>After treatments, the expression of lymphocyte transformation index (LTI), IL-2 and TNF-alpha in the tumor-bearing mice were detected with methyl thiazolyl tetrazolium (MTT), the expression of IL-2 and TNF-alpha in the patients with oral cancer were detected with MTT, the expression of LTI, CD4+ and CD8+ were detected with 3H-TdR incorporation.</p><p><b>RESULTS</b>LTI, IL-2 and TNF-alpha of thermo-chemotherapy group (HP group) had no significant difference comparing with those of normal mice group (N group) (P>0.05), but which were significantly higher than those of chemotherapy group (P group) and no treatment group (NT group) (P<0.01). In clinical trials, the expression of LTI, CD4+, CD4+/CD8+, IL-2 and TNF-alpha on oral cancer patients after thermo-chemotherapy were significantly higher than those before thermo-chemotherapy (P<0.01).</p><p><b>CONCLUSION</b>After thermo-chemotherapy, the expression of LTI, IL-2 and TNF-alpha of tumor-bearing hosts are significantly improved, there is a significant correlation between IL-2 and T cell.</p>

ABSTRACT

<p><b>OBJECTIVE</b>The aim of this study was to reveal the relations between expression and tumor behavior such as invasion, metastasis and prognosis in oral squamous cell carcinoma (SCC) through analyzing the expression of urokinase-type plasminogen activator (UPA).</p><p><b>METHODS</b>With Labelled streptavidin biotin method (LsAB), the expression of UPA was analyzed in 80 cases of oral squamous cell carcinoma, 10 cases of oral normal mucosa and 10 cases of oral leukoplakia.</p><p><b>RESULTS</b>Compared to oral normal mucosa and oral leukoplakia, the expression of UPA was significantly higher in SCC. The expression of UPA in SCC cases with lymph node metastasis was significantly higher than in cases without metastasis. The expression in cases with good prognosis was significantly higher than in those with poor prognosis and the expression was significantly higher in cases with invasive growth than in those with ulcerative and papillary growth.</p><p><b>CONCLUSION</b>The results obtained indicated that high expression of UPA in SCC might be closely related to lymph node metastasis, invasive growth and poor prognosis.</p>

ABSTRACT

<p><b>OBJECTIVE</b>The aim of this study was to observe the expression of urokinase-type plasminogen activator (UPA) in human tongue squamous cell carcinoma cell line (Tca8113) after heat shock with different dosage.</p><p><b>METHODS</b>The expression of UPA protein of Tca8113 after different heating temperatures(37 degrees C, 40 degrees C, 43 degrees C and 45 degrees C) treatment was examined by immunohistochemical technique (IH) and flow cytometry (FCM).</p><p><b>RESULTS</b>Compared with 37 degrees C group, the UPA protein expression in 40 degrees C and 43 degrees C groups decreased significantly (P < 0.05); however, the UPA protein expression in 45 degrees C group decreased but no statistical difference was found.</p><p><b>CONCLUSION</b>Hyperthermia could inhibit invasion and metastasis of oral squamous cell carcinoma and could be considered as a safe method in curing the tumor.</p>

ABSTRACT

Objective:To observe the activity of urokinase-type plasminogen activator (uPA) in human tongue squamous cell carcinoma cell line Tca8113 under hypoxia in vitro. Methods:According to the different time of hypoxia, the test was divided into four groups as following: (1) 0 h; (2) 6 h;(3)12 h;(4)24 h. The expression of uPA was examined using immunochemistry. The expression of uPA mRNA was examined using in situ hybridization (ISH). The results of ISH staining of uPA mRNA were analysed using Spot and IPP image analysis system.Results:The uPA expression was found in cytoplasm and cytomembrane of Tca8113 cells.uPA mRNA expression was found in cytoplasm of the cells.The staining intensity of uPA mRNA in the groups of (1),(2),(3) and (4) was 0.175 251?(0.013) 863,0.263 191?(0.000 680),0.406 745?0.014 016 and 0.478 919?0.018 998 respectively(P

ABSTRACT

Objective: To observe the expression of urokinase-type plasminogen activator receptor (uPAR) in human tongue squamous cell carcinoma Tca 8113 cells at different dosages of heat shock. Methods:The expression of uPAR protein in Tca 8113 cells was examined using immunohistochemical technique (IH) and flow cytometry (FCM) after the cells had been treated at 37,40,43 or 45 ℃ for 40 min. Results:The fluorescent intensity of uPAR protein of Tca 8113 cells treated at 37,40,43 and 45 ℃ was 11.36?0.06,9.98?0.12, 10.37?0.05 and 11.31?0.10 respectively.Conclusion:Heat shock can reduce the expression of the uPAR protein of Tca 8113 cells.

ABSTRACT

Objective: To investigate the expression of heat shock response protein(HSP_ 70 ) in human squamous tongue cancer Tca8113 cells during the recovery periods of heat shock.Methods:Tca8113 cells were subjected to heat shock at 43 ℃ for 30 min, then the cells were cultured for 2,4,6,8,12,24 and 48 h respectively. The expression of HSP_ 70 in the cells was examined with immunohistochemical method, Quantitative analysis was performed by FCM and the cell vitality was detected by MTT method.Results:Heat shock induced HSP_ 70 expression in Tca8113 cells at 43 ℃ for 30 min and the maximum proportion of the positive cells were observed and HSP_ 70 reached the maximum value at 12 h after heat shock(P
