ABSTRACT

Objective: To analyze the clinical and imaging features of patients with sudden sensorineural deafness and acute cerebral infarction in order to provide evidence for early recognition of such diseases. Methods: This was a case series reporting study. A retrospective analysis was performed on the clinical and imaging data of 29 patients with sudden hearing loss (SHL) who admitted to the Otolaryngology Head and Neck Surgery Department of Beijing Tiantan Hospital from January 2017 to December 2021 and diagnosed with acute cerebral infarction using MRI-DWI. Results: The patients were aged 31-71 years, with an average age of 56±12 years, and 82.8% (24/29) were men. In total, 82.8% (24/29) of the patients had three or more atherosclerotic risk factors, and 24.1% (7/29) had a history of SHL. The hearing types were flat and total deafness: 86.2% (25/29) of the patients had severe hearing loss, 27.6% (8/29) had bilateral SHL, 17.2% (5/29) had further hearing loss during hospitalization, and 82.8% (24/29) had dizziness or vertigo at the onset. The signs of central nervous system involvement mainly included speech impairment, diplopia, dysphagia, central facial paralysis, facial and limb hypoesthesia, ataxia, and decreased muscle strength. Imaging evaluation showed that 21 cases were located in the posterior circulation supply area and 8 cases in the anterior circulation supply area. Additionally, 82.8% (24/29) patients had vertebrobasilar artery stenosis, and 58.6% (17/29) patients had severe vertebrobasilar artery stenosis or occlusion. Conclusion: Patients with SHL who progress to cerebral infarction often have multiple atherosclerotic risk factors and SHL. Most of the patients are middle-aged and older men who often complain of dizziness or dizziness accompanied by severe flat and total deafness with unilateral or bilateral SHL. Imaging findings suggest that most patients have posterior circulation infarction, often accompanied by severe stenosis or occlusion of the vertebrobasilar artery..

ABSTRACT

Lung cancer is one kind of malignant tumors with the highest morbidity and mortality, and lung cancer metastasis is the leading cause of death in patients. Lung cancer metastasis is a complex process. A large number of studies have confirmed that lung tumors can secrete soluble molecules, mobilize and recruit related bone marrow cells or immune cells, regulate the matrix remodeling in distant organs which leads to the formation of pre-metastatic niche, therefore, promote tumor cells metastasis and implantation in distant organs. Detection of key target factor molecules and cellular components involving in pre-metastatic niche is of great clinical significance for diagnosis and prognostic evaluation. This article will summarize the progression of lung cancer metastasis modulated by the pre-metastatic niches and its clinical application status.

ABSTRACT

The Lewis lung carcinoma (LLC) metastatic mouse model was used to investigate the effects of gefitinib and Sijunzi Tang (SJZ) on pre-metastatic niche. The experimental protocol was approved by the Ethics Committee which belongs to Cancer Hospital, Chinese Academy of Medical Sciences. To generate spontaneous lung metastatic models, 1×106 luciferase-labeled LLC cells were injected subcutaneously in the shaved right flank of mice. One day after LLC inoculation, the mice were randomly divided into model (saline), gefitinib (50 mg·kg-1) treatment, SJZ treatment (25.74 g·kg-1), and co-treatment gefitinib with SJZ groups, with intragastrical administration. After 14 days of continuous administration, tumor size was detected by IVIS® Spectrum system. The number of monocytes and neutrophils and the expression levels of chemokine receptors (CXCR1, CCR2) and carcinogenic gene (c-Kit), in peripheral blood, spleen and lung tissues of mice were determined by flow cytometry. The contents of interleukin-IL-1α (IL-1α) and interleukin-6 (IL-6) were detected by the enzyme linked immunosorbent assay (ELISA). After 21 days of treatment, tumors were surgically removed, weighed and the tumor volume was measured with vernier caliper and the antitumor effect of co-administration was evaluated. After 45 days of administration, the survival of mice was recorded. The results of flow cytometry showed that the percentage of neutrophils in gefitinib group, SJZ group, and co-treatment group was significantly decreased in the lung tissue compared to the model group (P<0.05 or P<0.01), but there was no significant difference between three treatment groups (P>0.05). In the mouse peripheral blood and lung tissue, compared with the model group, the expression levels of CXCR1, CCR2 and c-Kit on the surface of neutrophils and monocytes in SJZ group and co-treatment group decreased or decreased significantly (P<0.01 or P<0.05). However, there was a significant increase in the expression level of c-Kit on the surface of monocytes (P<0.05). In the mouse spleen tissue, the expression levels of CXCR1, CCR2 and c-Kit in the gefitinib group increased significantly (P<0.05), while decreased significantly in SJZ or co-treatment group (P<0.05). The results of ELISA showed that the content of IL-1α in SJZ group decreased significantly in the plasma of the mice compared with the model group (P<0.01) and the content of IL-6 in co-treatment group decreased significantly (P<0.05). Compared with the gefitinib group, the content of IL-1 in the co-treatment group decreased significantly (P<0.05). In the tumor tissues of mice, compared with the model group, the content of IL-1α in the co-treatment group decreased significantly (P<0.05). Furthermore, the content of IL-1α in co-administrated group and IL-6 in SJZ or co-treatment group decreased significantly compared with the gefitinib group (P<0.05). After 21 days of continuous administration, the tumor inhibition rates of gefitinib group, SJZ group and co-administrated group were 45.7%, 38.4%, and 84.8%, respectively. After 45 days of administration, the survival rate of the model group was 0%, whereas the gefitinib, SJZ or co-treatment group has a survival rate of 40%, 60%, or 60%, respectively. In summary, our study illustrated that Sijunzi Tang could improve the anti-tumor effect of gefitinib by regulating pre-metastatic niche.

ABSTRACT

Collect and analyze recent studies on the reversal of the mechanism of tumor drug resistance by traditional Chinese medicines, and provide ideas for research on the intervention of acquired tumor resistance with traditional Chinese medicines. CNKI, Wanfang, Pubmed and other databases were searched for literature on the reversal of the mechanism of tumor drug resistance by traditional Chinese medicines and Chinese herbal compound in the past 5 years. The search terms included ‘traditional Chinese medicine’, ‘traditional Chinese medicine formula’, ‘traditional Chinese medicine monomer’, ‘traditional Chinese medicine patent prescription’, ‘traditional Chinese medicine injection’, ‘drug resistance, multidrug resistance’, and ‘tumor’. The literatures were collated and analyzed. The interventions of traditional Chinese medicine on drug resistance of tumors mainly were regulating the expression of transporter proteins on the cell surface, inducing apoptosis, and regulating the expression of related enzymes in cells. Traditional Chinese medicine monomers and their compound preparations can reverse the multidrug resistance of tumors. The combined use of Chinese medicine preparations and chemotherapeutic drugs can significantly enhance the clinical treatment effect and reverse the drug resistance of tumors. Traditional Chinese medicines can reverse acquired tumor resistance through multiple targets, but the precise reversal mechanism remains to be further studied.

ABSTRACT

Objective To investigate the expression profiles and chromosome distribution characteristics of miRNA in bone marrow samples between the patients with adult B-cell acute lymphoblastic leukemia (B-ALL) and their controls. Methods Illumina sequencing technology was applied to perform global miRNA expression analysis and chromosome distribution analysis in the bone marrow samples of 15 adult B-ALL patients relative to a control group of 10 patients without hematologic malignancies. Then, qPCR was carried out to validate the reliability of the se-quencing data. Results In total,291 differentially expressed miRNAs including 168 up-regulated and 123 down-regulated miRNAs were indentified in adult B-ALL patients as compared with their controls. Despite the distribution of miRNA genes in chromosome was similar in both samples, the distribution of miRNA expression abundance in chromosome show obvious differences. B-ALL group was mainly located in the chromosome 1, 2, 5 and 9, while the control group was mainly located in the chromosome 3,7,9,17 and X. Conclusions The altered expression profile of miRNA in bone marrow samples of adult B-ALL patients implies that aberrantly expressed miRNAs maybe plays roles in the occurrence and development of B-ALL.

ABSTRACT

The Direct Amplification of Length Polymorphisms was applied to assess genetic diversity and structure of 7 populations of Dendrobium nobile, comparing one population of D. lituflorum. The five primer combinations were amplified to produce 140 clear bands, and 102 polymorphic bands had been detected with each pair of primer producing 20.4 polymorphic bands on average. At species level, the percentage of polymorphic bands (PPB) was 72.86%, the Nei's gene diversity index (H) was 0.288 9, and the Shannon's information index (I) was 0.424 2. At population level, the average PPB was 47.96%, H was 0.1861, and I was 0.273 9 in 7 populations. The coefficient of gene differentiation (Gst) was 0.338 6 among populations of D. nobile. It showed that 33.86% of the total genetic diversity was attributable to genetic differentiation among populations, while the rest 66.14% was resided between individuals within population.
