ABSTRACT

Objective To provide the feasible basis for the implementation of integration processing technology of origin of Moslae Herba by comparing the traditional cutting processing and integration processing of origin of Moslae Herba. Methods Taking the softening time, the length of cutting section and the drying temperature as the investigation factors of the traditional cutting processing, and the cutting length, drying temperature and drying time as the investigation factors of the integration processing of origin, L9(34) orthogonal test was used. Carvacrol, thymol, volatile oil content were setting as the evaluation indexes, the traditional cutting processing and the integration processing of origin were optimized, and the two selected process were compared. The contents of carvacrol and thymol were deterrmined by Agilent 1200 high performance liquid chromatography and ECOSIL-C18 column. The mobile phase was acetonitrile-0.2% acetic acid, gradient elution, with wavelength of 274 nm, flow rate of 1.0 mL/min, sample volume of 20 μL. The extraction of the volatile oil was in accordance with the volatile oil determination method in Chinese Pharmacopoeia 2015 edition. Results The optimized traditional cutting processing: softening 1 h with 3 times amount of water, cutting to 1.0 cm, drying at 50–60 ℃. The optimized integration processing of origin: cutting to 1.0 cm in fresh, drying at 50–60 ℃, drying to 36 h. The contents of carvacrol, thymol, and volatile oil in the optimum integration processing of origin were all higher than the traditional cutting processing. Conclusion The technology of integration processing of origin is simple and convenient, which can make up the defects of the complicated traditional cutting processing and the lessen of effective components, and is feasible.

ABSTRACT

The present study focused on the characterization and genomic sequence of phage PS2 that infects Serratia marcescens clinical isolates.The morphology of phage PS2 was observed with electron microscope.The one-step growth curve,host range,and stability of PS2 were investigated.In addition,Phage DNA was extracted from the purified phage particles using a MiniBEST Viral RNA/DNA Extraction Kit.DNA sample was analyzed by digesting with restriction enzymes.The phage DNA was used for constructing the sequencing library.The library was sequenced on a MiSeqTM platform.The whole genome sequence was obtained by Velvet (version:1.2.08) assembling.Phage PS2 belongs to the Myoviridae family.The linear,circularly permuted,167 266-bp double-stranded DNA genome of PS2 has high similarities to T4-1ike phages.The phage DNA contains 41.7% GC and 276 ORFs.PS2 exhibited a 21-minute latent period and 70 PFU per cell at burst size when the pathogenic S.marcescens strain S2 served as a host.Further investigation suggested that PS2 is stable in a wide pH range (pH5 to pH10) and at extreme temperatures (50 ℃ and 60 ℃) after incubation alone at different pHs and different temperatures,respectively.The paper focused on the isolation and identification of a novel lytic S.marcescens phage,the biological characteristics,the whole genome sequencing and the preliminary study of bioinformatics,which laid the foundation for deeply analysis to the phage therapy of multi-drug resistant bacteria and the phage biological information.

ABSTRACT

Objective To evaluate the effect of the prevention and control technique training of human important parasitic diseases for basic professional and technical personnel in Jiangsu Province in 2017. Methods A workshop including theory courses and practical operations was carried out for the basic professional and technical personnel. At the end of the workshop , the effects were evaluated through the theoretical and microscopic examinations. Results There were 132 trainees from 13 cit-ies,and all of them took part in the final examinations. The average score of total was 118.36. There were significant differences among different regions(χ2 =13.38,P<0.01). The pass rate of the theory test was 92.4%with the average score of 79.05. There were significant differences of theory scores among different regions(χ2=14.51,P<0.01). The theory score of Northern Jiangsu Province was significantly higher than the scores of Southern and Central Jiangsu Province(P<0.001,P=0.009). The pass rate of microscopic examinations was 89.4%with the average score of 39.32. There was no significant difference of micro-scopic examination scores among different regions(F=2.37,P=0.09). Among the total 7 species,the detection rate of Para-gonimus westermani was the highest(75%)and the rates of Schistosoma japonicum and Fasciolopsis buski were only 51.5%and 54.5%respectively. The detection rates of eggs of Trichuris trichiura,unfertilized Ascaris lumbricoides,Clonorchis sinensis and Taenia solium were 71.2%,65.9%,72.7%and 72.0%respectively. Conclusions The training course has promoted the capa-bility of prevention and control of parasitic diseases for basic technical personnel. Also it provides the technical supports for para-sitic disease monitoring,control and risk assessment in the next step.

ABSTRACT

Objective To examine the effects of application of ultrasonography guided water injection for inser-tion of naso-jejunal tubes. Methods Hospitalized patients in ICU who needed naso-jejunal tubes were recruited from one tertiary hospital in Beijing from November 2016 to April 2017. Ultrasonography guided water injection was used to assist insertion of naso-jejunal tubes. Meanwhile,we conducted semi-structured interviews to learn feel-ings and suggestions from the patients. Results A total of 40 patients were included in this study,37 patients (92.5%) were successfully inserted with the tubes at the first attempt. The duration of insertion of naso-jejunal tubes was 25 (20,38.75) min. Conclusion Ultrasonography guided water injection is a simple and convenient method to guide the placement of naso-jejunal tubes for critical ill patients,which provides guarantee for early en-teral nutrition.
