ABSTRACT

Objective To provide reference for the cold chain of stored RBCs on the sea by evaluating both blood containers containing phase-changed materials and the quality of stored RBCs during transportation.Methods In order to simulate blood supply on the sea,we transported the stored RBCs on land(100 min),on the sea(45 h)and stored them on the sea for another 7 days.The free hemoglobin(Hb), lactate dehydrogenase(LDH), and concentrations of K +and Na+were measured.Results The temperature of the blood container containing phase-changed materials rose from 4.1℃to 9.5℃.The contents of free Hb,K+and LDH were increased to(0.083 ±0.032)g/L,(15.097 ±1.791)mmol/L, and(106.00 ±17.83)U/L,respectively.During blood storage,the contents of the above three indices were increased to (0.111 ±0.035)g/L,(27.238 ±3.509)mmol/L and(227.00 ±111.94)U/L, while Na +decreased to(113.63 ± 4.012)mmol/L.Conclusion The temperature of the blood container containing phase-changed materials can be maintained at a constant temperature under more complicated environmental conditions,and the quality of the stored RBCs can be ensured.RBCs stored on the sea for more than 7 days are damaged more seriously than those stored on land.

ABSTRACT

Objective To establish a new dialysis washing method for human frozen platelets and to evaluate a new type of washing machine based on dialysis.Methods Twenty bags of platelets(1U)were divided into two groups:centrifugation group and dialysis group.The effect of the washing machine was evaluated by measuring platelet function in vitro.Results After the platelets were treated by the two methods, the recovery rate of dialysis group[(73.57 ± 12.49)%]was significantly better than that of centrifugal group[(62.21 ±7.07)%].The osmotic pressure of dialysis group was 305.8 ±5.27mOsm/kg, compared to 383.2 ±22.00 mOsm/kg in centrifugation group.There was significant differences in the recovery rate and osmotic pressure(P<0.05), but there was no significant difference between dialysis group and centrifugation group in the mean platelet volume[(9.44 ±0.93)vs(9.23 ±0.50)fl], ADP-induced aggregation rate[(20.7 ±8.00)%vs(27.7 ±11.83)%],thrombin-induced platelet aggregation rate[(84.1 ±7.88)%vs(92.9 ±5.28)%], hypotonic shock responses[(19.02 ±4.42)% vs(23.19 ±9.08)%], CD62p activation rate [(16.00 ±9.40)%vs(21.86 ±8.14)%]or apoptosis rate[(46.31 ±10.28)% vs(54.59 ±16.76)%](P>0.05). Conclusion Compared to centrifugation group,the frozen platelet washing machine can not only improve platelet recovery,but also solve the problem of excessive DMSO residues.

ABSTRACT

Objective To improve the buffer for washing frozen red blood cells(RBCs)and explore the feasibility of replacing the middle buffer of 2% NaCl with 0.9% NaCl solution in the protocol of deglycerolization and the effect was evaluated.Methods Two units of RBCs separated from whole blood of healthy donors were frozen at -80℃.The thawed frozen RBCs were treated with a washing machine.On the basis of the old protocol(protocol 1),the middle buffer of 2%NaCl was replaced,and the usage of washing buffer and the washing steps were adjusted to create a new protocol(protocol 2).The quality of RBCs washed by the two protocols was evaluated.Results According to the results of the detection of RBCs treated by protocol 1 and 2: the hemoglobin contents(g)were 42.18 ±3.35 and 44.98 ±1.68,respectively,free hemoglobin contents(g/L)were 0.53 ±0.06 and 0.45 ±0.05 respectively, the residual amount of white blood cells (×107)was 1.92 ±1.04 and 1.12 ±1.12,and the osmolarities(mOsm)were 327.0 ±9.06 and 331.8 ±10.62 respectively. Sterile experiments were negative for both bacteria and fungi.The hemolysis rates(%)were 12.02 ±5.78 and 14.30 ± 5.67 respectively.The deforming abilities(%)were 21.42 ±1.45 and 21.32 ±0.84 respectively.The RBC recoveries (%)were 77.18 ±5.58 and 79.63 ±2.06 respectively.The processing time(min)was 79.60 ±0.55 and 78.80 ±1.30 respectively.There was no significant difference in the quality of frozen RBCs washed by the old protocol(protocol 1)and the new protocol(protocol 2)(P>0.05).Conclusion The quality of frozen RBCs washed by the two protocols meet national standards.
