ABSTRACT

Objective To observe the effect of calcified lymph nodes on video-assisted thoracoscopic surgery (VATS) lobectomy in the chronic obstructive pulmonary disease (COPD) patients with lung cancer. Methods A retrospective analysis was conducted on the COPD patients with lung cancer who underwent VATS lobectomy in the Department of Thoracic Surgery in the First Affiliated Hospital of Hebei North University from May 2014 to May 2018.The patients were assigned into a calcified lymph node group and a control group according to the presence or absence of calcified lymph nodes in CT,and the size,morphology,and calcification degree of the lymph nodes were recorded.The operation duration,intraoperative blood loss,chest tube retention time,hospitalization days,and overall complication rate were compared between the two groups. Results The 30 patients in the calcified lymph node group included 17 patients with one calcified lymph node and 13 patients with two or more calcified lymph nodes,and a total of 65 calcified lymph nodes were recorded.The calcified lymph nodes with the size ≤5 mm were the most common (53.8%),and complete calcification was the most common form (55.4%) in lymph node calcification.The mean operation duration had no significant difference between the calcified lymph node group and the control group (t=-1.357,P=0.180).The intraoperative blood loss (t=-2.646,P=0.010),chest tube retention time (t=-2.302,P=0.025),and hospitalization days (t=-2.274,P=0.027) in the calcified lymph node group were higher than those in the control group. Conclusion Calcified lymph nodes increase the difficulty and risk of VATS lobectomy in the COPD patients with lung cancer.The findings of this study are conducive to predicting the perioperative process of VATS lobectomy.

ABSTRACT

Objective:To investigate the expression of LncRNA ANCR in human glioma tissues and its relationship with malignant proliferation of cells.Methods:The samples of 10 normal brain tissue,13 low-grade and 45 high-grade gliomas were regarded as normal group, low-grade group and high-grade group, which were collected from neurosurgery department in Linyi Central Hospital, and the expression of ANCR and potential interaction molecule eIF4B was detected by reverse transcription polymerase chain reaction (RT-PCR) in vitro. Lentivirus transfection in vitro was used to construct the U251 shRNA ANCR and control cell line in human high-grade gliomas as control, test 1 and test 2 group cells in the study. QPCR detect the expression level ANCR, eIF4B and Myc mRNA in cells. Western blot was used to detect the expression of eIF4B and c-Myc protein, CCK-8 assay was used to detect the relative proliferation ability of cells, and the colony formation assay was used to observe the change of cell clone formation. SPSS 21.0 was used for statistical analysis, analysis of variance was used for inter group comparison, and SNK-q pairwise comparison method was used for intra group comparison.Results:The expressions of ANCR mRNA in high-grade glioma tissues, low-grade gliomas and normal brain tissues were 0.710±0.125, 2.033±0.312 and 3.408±0.296. The expressions of eIF4B mRNA in high-grade glioma tissues, low-grade gliomas and normal brain tissues were 0.176±0.019, 0.268±0.022 and 0.426±0.028. The expression of ANCR and eIF4B in high-grade glioma tissues was higher than that in low-grade gliomas and normal brain tissues ( P<0.001). The expression of ANCR in low-grade glioma tissues was higher than that in normal brain tissues ( P=0.013). There was a significant positive correlation between the expression of ANCR and eIF4B in glioma tissues ( P<0.001) ; The expressions of ANCR mRNA in Control, test1 and test2 were 1.000±0.021, 0.202±0.057 and 0.300±0.016. The expressions of eIF4B mRNA were 1.000±0.078, 0.452±0.012 and 0.526±0.037, and the expressions of c-Myc mRNA were 1.000±0.053, 0.688±0.067 and 0.564±0.089. the expressions of ANCR, eIF4B and c-Myc mRNA and protein in test1 and test2 cells were significantly lower than those in the control group ( P<0.01) ; the proliferation of test1 and test2 groups were significantly decreased at 72h and 96h, and the ability of colony formation was significantly decreased ( P<0.001) . Conclusion:The expression of ANCR was significantly up-regulated in high-grade glioma tissues and positively correlated with the expression of eIF4B. Interference with ANCR in vitro could mediate the decrease of the expression of eIF4B and c-Myc mRNA and protein molecules, thereby inhibiting the proliferation of glioma cells.

ABSTRACT

Objective:To explore the effects of alantolactone on cell proliferation and apoptosis of acute myeloid leukemia cell line THP-1 and the related mechanisms.Methods:THP-1 cells in logarithmic growth phase were taken. The control group was added with an equal volume of culture medium containing 10% fetal bovine serum, and the experimental group was added with 0.5, 1.0, 2.0, 4.0, 6.0, 8.0, 10.0 μmol/L alantolactone. The cells in each group were cultured for 72 hours. The cell proliferation was detected by CCK-8 method. In addition, the cells of the control group were taken, and the experimental group was added with 2.0 and 4.0 μmol/L alantolactone, respectively. The cells in each group were cultured for 72 hours. The apoptosis of the cells was detected by flow cytometry, and the expressions of NF-κB pathway-related proteins were detected by Western blotting.Results:After THP-1 cells were treated with different concentrations of alantolactone (0.5, 1.0, 2.0, 4.0, 6.0, 8.0, 10.0 μmol/L) for 72 hours, the inhibitory rates were (9.4±1.4)%, (27.7±4.0)%, (45.1±2.5)%, (66.9±2.9)%, (87.0±1.2)%, (91.7±1.0)% and (94.4±0.8)%, respectively, which were in a dose-dependent manner, and the control group was (0.4±0.1)%, the difference was statistically significant ( F = 241.87, P < 0.01). After THP-1 cells were treated with different concentrations of alantolactone (2.0, 4.0 μmol/L) for 72 hours, the apoptotic rates were (26.1±4.2)% and (37.8±5.6)%, and the control group was (8.5±1.4)%, the difference was statistically significant ( F = 43.04, P < 0.05). Furthermore, alantolactone could down-regulate the expression levels of bcl-2 and p65, and up-regulate the expression levels of bax, cleaved-caspase-3 and cleaved-PARP. Conclusions:Alantolactone can induce the apoptosis of AML THP-1 cells via inhibiting the NF-κB pathway, thereby inhibiting its proliferation.

ABSTRACT

Objective To explore the expression of circ_0006692 in NSCLC and its relation with clinicopathological characteristics and related mechanism. Methods We collected 50 pairs of NSCLC tissues and adjacent tissues. The expression of circ_0006692 was detected by qRT-PCR and its relation with clinicopathological features was analyzed. We constructed lung cancer A549 cell line with circ_0006692 overexpression and knockdown. Cell proliferation, migration and invasion were detected by MTS, colony forming, wound healing and Transwell invasion assays. qRT-PCR and Western blot were used to detect the effect of circ_0006692 expression change on EMT-related gene expression. Results The expression of circ_0006692 in NSCLC was significantly higher than that in adjacent tissues (P < 0.05). The expression level of circ_0006692 was closely related to tumor size, TNM stage and pulmonary membrane invasion (P < 0.05). circ_0006692 knockdown inhibited cell proliferation, invasion and metastasis, while its overexpression promoted cell proliferation, invasion and metastasis. circ_0006692 knockdown inhibited the expression of EMT-related genes CDH2 and MMP7 in A549 cells and promoted the expression of CDH1. Conclusion The upregulated expression of circ_0006692 in NSCLC is closely related to tumor size, TNM stage and pulmonary membrane invasion. circ_0006692 expression could regulate the proliferation, invasion, metastasis and EMT progression of lung cancer A549 cells.

ABSTRACT

Objective To investigate the effect of piR-9994 on the biological behavior of gastric cancer cells and its possible mechanism. Methods The expression of piR-9994 in gastric cancer cell lines (MGC803 and AGS) and normal gastric epithelial cells (GES-1) were detected by qRT-PCR. MGC803 cell line with piR-9994 overexpression and knockdown were constructed. The effects of piR-9994 expression changes on cell proliferation were detected by MTT and clone formation assay. The scratch wound healing assay and Transwell invasion assay were used to detect cell migration and invasion abilities. qRT-PCR and Western blot were used to detect cell proliferation and EMT-related genes expression. Results The expression level of piR-9994 in MGC803 cells was significantly higher than that in normal gastric epithelial cell line GES-1 (P < 0.05). The overexpression of piR-9994 promoted the proliferation, invasion and metastasis of gastric cancer cells, while piR-9994 knockdown had the opposite effect. piR-9994 expression was closely related to cell cycle, especially in S phase. The overexpression of piR-9994 promoted the expression of proliferation-related genes PCNA, CCND1 and Bcl-2, inhibited the expression of apoptosis gene C-PARP, and promoted the expression of EMT-related genes N-cadherin, MMP7, Twist and Vimentin; while piR-9994 knockdown had the opposite effect. Conclusion Abnormal expression of piR-9994 affects the proliferation, invasion, metastasis and EMT process of gastric cancer cells. piR-9994 may be a new biomarker and therapeutic target for gastric cancer.

ABSTRACT

Aim To explore the regulatory effect of mitochondrial solute carrier protein SLC25A26 on senescence of hepatoma cells induced by methionine cycle metabolism. Methods HepG2 cell line was cultured in vitro. After hepatoma cell senescence was induced by toposide (2 [xmol • Ľ'), a positive drug for inducing cell senescence, methionine circulating metabolite SAM (0. 1 mmol • L " ') was treated. Western blot and Real-time PGR were used to detect the senescence indexes including pl6, p21, and HMGAl, and flow cytometry was used to detect cell cycle. Transfecting SLC25A26 overexpression plasmid, the effect of overexpressing SLC25A26 on the senescence indexes of hepatoma cells was detected by Western blot and immunofluorescence detection, and the level of SAM after overexpressing SLC25A26 was detected by the kit. The effect of overexpressing SLG25A26 on the senescence of hepatoma cells after SAM treatment was detected by Real-time PGR. Results Western blot and Real-time PGR showed that methionine cycle metabolism could weaken the senescence level of HepG2 cells induced by Etoposide, and flow cytometry showed that cell cycle was arrested in Gl phase; overexpression of SLG25A26 decreased the levels of SAM and SAH in cytoplasm of HepG2 cells, and exogenous SAM partially offset the aging effect of HepG2 cells induced by SLG25A26. Conclusions Promoting methionine cycle metabolism can inhibit hepatoma cells senescence; overexpression of SLG25A26 can induce hepatoma cells senescence; SLG25A26 can induce hepatoma cells senescence by regulating methionine cycle metabolism.

Objective@#To analyze the trend of percentile changes in body composition among children and adolescents aged 7 to 18 by gender and age, and to provide a scientific basis for reference value.@*Methods@#A stratified cluster random sampling method was used to select 1 585 children and adolescents from September to December 2020. Use the GAMLSS to estimate the P 25 , P 50 and P 75 percentile value of gender, age specific body fat percentage (BF%), fat mass index (FMI) and fat free mass (FFM).@*Results@#In different age groups, body fat percentage of boys with normal nutritional status peaked at around 10-year old, among them, body fat percentage of 10-year old P 25 , P 50 and P 75 were 22.2%, 26.1% and 30.1%, respectively. Body fat percentage of obese boys was at a high level. Among girls, body fat percentage under different nutritional status did not change. With the increase of age, fat mass index plateaued slightly in normal and overweight boys. However, in obese boys, with the increase of age, FMI gradually increased with the increase of percentile, while the value of FMI gradually decreased in girls. Whether in boys or girls, with the increase of age, fat free mass index shows an increasing trend, and boys increase faster.@*Conclusion@#Under different nutritional status, there are differences in the percentile value of body fat percentage, fat mass index and fat free mass by gender and age, which can provide a scientific basis for reference value of body composition in children and adolescents.

ABSTRACT

At present, the mechanism of scar formation in filtering area after glaucoma filtering surgery has not been fully understood.It is believed that stimulation of cytokines and growth factors during postoperative wound healing causes excessive activation of fibroblasts in the Tenon capsule, resulting in the deposition of extracellular matrix and scar formation.Long non-coding RNA (lncRNA) is a kind of RNA sequence that does not encode proteins.It can regulate cell proliferation, apoptosis and become a precursor of microRNA, and its specific expression has been proved and plays an important regulatory role in various fibrotic tissue.With the deepening of research, lncRNA has also been found to be involved in the formation and development of filtering bleb scarring after glaucoma filtering surgery.Previous studies have shown that lncRNA is specifically expressed in Tenon capsule tissues after glaucoma filtering surgery, and different lncRNAs can affect the proliferation of fibroblasts in Tenon capsule or the synthesis of extracellular matrix through different ways, which can affect the formation of fibroblasts scarring.In this article, the role of different lncRNAs in the proliferation of Tenon fibroblasts and its potential in treating bleb scarring were summarized so as to provide a new direction for the treatment of postsurgical scarring.

Talent evaluation is the premise of development management and the use of human resources. The establishment of the talent classification and evaluation mechanism of university affiliated hospitals plays an important role in setting up the correct employment orientation, stimulating the career development of talents and promoting the high-quality development of public hospitals. The authors took the perspective of university affiliated hospitals, summed up the construction process of talent evaluation system in China, analyzed the existing problems in evaluation indicators, evaluation methods and operation mechanism of talent evaluation system, and came out with suggestions that the national level should strengthen the top-level design and collaborative management of talent evaluation, the hospital level should refine the evaluation indicators, improve the evaluation methods and establish the concept of developmental evaluation, at meanwhile, schools should improve the talent management mechanism and strengthen the function of audit and supervision.

ABSTRACT

Objective:To preliminarily observe the clinical efficacy of microwave hyperthermia combined with intensity-modulated radiotherapy (IMRT) and chemotherapy for patients with locally advanced gastric cancer.Methods:Forty patients who could not been operated or refused operation were enrolled in this clinical trial, who were confirmed as locally advanced proximal or distal gastric cancer by gastroscopy pathology and imaging. Radiotherapy was delivered by IMRT technology for 5 times per week with a total dose of 46 to 56 Gy (median dose of 50 Gy) in 25 to 28 fractions. Synchronous hyperthermia was given at 42 to 44℃ twice a week, 45 min/time. S-1 or capecitabine-based synchronous chemotherapy was performed, d1-14/3 weeks. The symptom remission rate, adverse reactions, objective remission rate (complete and partial remission) and survival were observed.Results:A total of 40 patients, aged between 56 and 83 years (median age of 71 years), were enrolled in this study. The male-to-female ratio was 7: 1. Among them, 38 cases (95%) showed symptom remission. The most common adverse reactions were grade 1-2 gastrointestinal reactions and leukopenia. The objective remission rate was 87.5%, the 2-year progression-free survival and overall survival rates were 68.6% and 70.5%, respectively.Conclusion:Preliminary findings demonstrate that microwave hyperthermia combined with chemoradiotherapy achieve satisfactory outcomes and yield tolerable toxicity in patients with locally advanced gastric cancer.

ABSTRACT

OBJECTIVE:To simultaneo usly determine the contents of atractylenolide Ⅱ ,β-eudesmol,atractyloxin and atractylone in Atractylodes chinensis ,and to evaluate the quality of A. chinensis with different growth years combined with color difference principle. METHODS :HPLC method was adopted. The determination performed on Agilent Eclipse XDB-C 18 column with mobile phase consisted of acetonitrile- 0.2% phosphoric acid (gradient elution )at the flow rate of 1.0 mL/min;the detection wavelengths were set as 208 nm(atractylenolide Ⅱ,β-eudesmol),340 nm(atractyloxin)and 220 nm(atractylone);the sample size was 15 μ L. Using atractyloxin as reference,QAMS was adopted to establish relative correction factors (RCFs) of atractylenolideⅡ,β-eudesmol and atractylone ;the content of each component in A. chinensis with different growth years were calculated. The contents of above 4 components were determined by external standard method and then compared with the results of QAMS. The color difference values of A. chinensis powder were measured based on color difference principle. The correlation analysis of above 4 components content with color was carried out by Pearson correlation analysis. RESULTS :The separation degree of atractylenolide Ⅱ,β-eudesmol,atractyloxin and atractylone in A. chinensis was higher than 1.5. The linear range were 1.01-10.10,3.30-33.00,4.40-44.00,5.34-53.40 μg/mL,respectively. RSDs of precision ,reproducibility and stability tests were all lower than 2%,while the average recovery rates were 101.34%-104.67%(RSD<1.5%,n=6). Using atractyloxin as reference , RCFs of atractylenolide Ⅱ,β-eudesmol and atractylone were 3.896 7,5.928 2,9.727 9,with RSD of 0.35%,2.89%,0.36% (n=6),respectively. Relative deviation of 3 components (except for atractyloxin ) in 24 batches of A. chinensis ranged 0.03%-1.45% between QAMS and external standard method ,which indicated that the results of two methods were consistent ,and the content of each component increased with the increase of growth years. Atractylenolide Ⅱ,β-eudesmol,atractyloxin and atractylone in A. chinensis had significant negative correlation with its color shade (L*),total color difference (E*ab)(P<0.01), and significant positive correlation with color red-green direction (a*), color yellow-blue direction (b*)(P<0.01). CONCLUSIONS:The established HPLC-QAMS method can be used for the determination of atractylenolide Ⅱ,β-eudesmol, atractyloxin and atractylone in A. chinensis . The longer the growth period is ,the higher each component content is. The color of A. chinensis is closely related to the content of each component ,and the content of effective components is higher in A. chinensis with dark yellowish brown color.

@#Objective To evaluate the prevalence and prognosis of hepatitis E virus ( HEV) su- perinfection in patients with chronic hepatitis B ( CHB) . Methods Electronic databases of Pubmed, CNKI,and Wanfang were searched,and references of the relevant articles for literatures associated with HEV superinfection in CHB patients in China published during 2008-2019 were screened. Studies which met the inclusion and exclusion criterias were selected and related data were pooled for analysis. The R 3. 5. 2 and Revman 5. 3 were used to measure heterogeneity with the I2 statistic and pooled incidence and odds ratio ( OR) . Results 28 literatures were eligible for inclusion. The results showed the rate of HEV superinfection in hospitalized CHB ( CHB-HEV) patients was 13. 6% ( 95%CI: 12. 5% -14. 9%) . The pooled incidence of liver failure in CHB-HEV patients was 34. 7% ( 95%CI: 29. 6%-40. 1%) ,which was higher than CHB patients ( OR= 10. 9,95%CI: 6. 8-17. 4) ,CHB patients superinfected with hepatitis A virus ( CHB-HAV) ( OR= 3. 8,95%CI: 2. 4-6. 2) ,and hepatitis E patients ( OR= 5. 1,95%CI: 3. 8- 6. 8) . The pooled mortality of CHB-HEV patients was 13. 8% ( 95%CI: 10. 3-17. 8) ,which was higher than CHB patients ( OR = 8. 5,95%CI: 4. 0-18. 3) ,CHB-HAV patients ( OR = 14. 4,95%CI: 4. 8- 42. 9) ,and hepatitis E patients ( OR= 4. 4,95%CI: 3. 0-6. 4) . Conclusion In China,HEV sueperinfection on chronic hepatitis B patients are common,and is an important cause of liver failure and death.

ABSTRACT

Objective:To identify the influencing factors of maternal anxiety, and explore the moderating role of co-parenting in the effect of marriage quality on maternal anxiety.Methods:Totally 522 mothers with children aged 0-12 were enrolled in the surveys and completed the Generalized Anxiety Disorder (GAD-7), Marital Adjustment Test and Co-parenting Relationship Scale.The chi-square test, the spearman correlation analysis and non-parametric percentile Bootstrap method were processed by SPSS 22.0.Results:①The detection rate of anxiety in the mothers was 48.7%, and the rate for mild, moderate, and severe anxiety was 37.4%, 8.4%, 2.9%, respectively.② The chi-square test showed that the family factors, such as mother's age, level of workload, family economic conditions, husband's level of workload, the child's age and health condition, as well as the conflicts between parents and grandparents and the grandparents’ willingness of involving in raising children, significantly affected mother’s anxiety(χ 2=4.292-23.170, P<0.05). ②The maternal anxiety was negatively correlated with marital quality ( r=-0.419, P<0.01). ③ Both positive co-parenting (PCP) and negative co-parenting(NCP) played moderating roles in the relationship between marriage quality and maternal anxiety( BPCP=0.013, P<0.01; BNCP=-0.010, P<0.05). Conclusion:Marital quality is an important factor which affects maternal anxiety, and father’s performance in co-parenting plays a moderating role in it.

ABSTRACT

Objective:To evaluate the clinical effect of modified Shaoyao-Gancao Decoction combined with oral morphine sulfate sustained release tablets in the treatment of cancer pain with qi and blood deficiency. Methods:A total of 102 cancer pain patients with qi and blood deficiency from March 2017 to May 2018 were divided into two groups according to the random number table method, 51 in each group. The control group was treated with morphine sulfate sustained release tablets on the basis of conventional treatment, and the treatment group was treated with modified Shaoyao-Gancao Decoction on the basis of the control group. Both groups were treated for 4 weeks. Numerical rating scale (NRS) was used to assess the severity of cancer pain, and Karnofsky performance status (KPS) scale was used to assess the quality of life. The level of Endothelin-1 (ET-1) was measured by radioimmunoassay, The levels of 5-hydroxytryptamine (5-HT), Prostaglandin E2 (PGE2) and substance P (SP), TNF-α, IL-1β, IL-6, IL-2 were detected by ELISA, the clinical effects were evaluated, and the adverse reactions were recorded. Results:The total effective rate was 94.1% (48/51) in the treatment group and 78.4% (40/51) in the control group, the difference between two groups was statistically significant ( χ2=5.299, P=0.021). After the treatment, the NRS score (2.84 ± 1.32 vs. 4.22 ± 1.44, t=5.045) in the treatment group was significantly lower than that of the control group ( P<0.01), and the KPS score (73.13 ± 10.19 vs. 59.93 ± 8.81, t=6.998) was significantly higher than that of the control group ( P<0.01). After the treatment, serum 5-HT (0.34 ± 0.09 pg/ml vs. 0.53 ± 0.14 pg/ml, t=8.153), PGE2 (65.03 ± 15.79 pg/ml vs. 94.85 ± 20.88 pg/ml, t=8.135), ET-1 (99.96 ± 21.57 pg/ml vs. 176.78 ± 26.01 pg/ml, t=16.236), SP (60.27 ± 12.71 pg/ml vs. 65.63 ± 12.09 pg/ml, t=2.182) in the treatment group were significantly lower than those in the control group ( P<0.01 or P<0.05). Serum TNF-α, IL-1β, IL-6 level were significantly lower than that of the control group ( t value were 35.071, 12.472, 8.122, all Ps<0.01), and IL-2 level was significantly higher than that of the control group ( t=6.579, P<0.01). During the treatment, the incidence of adverse reactions was 13.7% (7/51) in the control group and 2.0% (1/51) in the treatment group. The difference between the two groups was statistically significant ( χ2=4.630, P=0.031). Conclusions:Modified Shaoyao-Gancao Decoction combined with morphine sulfate sustained release tablets can relieve the pain of cancer pain patients with qi and blood deficiency, reduce the level of inflammatory cytokines, and improve the clinical symptoms and quality of life of patients.

ABSTRACT

BACKGROUND@#Early neurologic deterioration (END) may occur in patients with anterior circulation ischemic stroke (ACIS) after receiving endovascular treatment (EVT). Hemodynamic insufficiency, re-occlusion, and post-re-canalization hyper-perfusion are likely to play a critical role in END. We hypothesized that hemodynamic changes can predict END in patients with ACIS post-successful EVT using trans-cranial Doppler (TCD).@*METHODS@#We utilized a prospectively maintained database of ACIS patients treated with EVT between September 2016 and June 2018 in the Xuanwu Hospital, Capital Medical University. TCD parameters including peak systolic velocity (PSV), bilateral mean flow velocity (MFV), and pulse index (PI) were determined via the middle cerebral arteries within 72 h post-EVT. A logistic regression model was applied to detect independent predictors for END.@*RESULTS@#Totally, 112 EVT patients were included in this study and 80/112 patients experienced successful re-canalization with <50% residual stenosis, while 17/80 (21.3%) patients suffered END, for which vasogenic cerebral edema (11/17) was considered as a leading role and followed by symptomatic intra-cranial hemorrhage (4/17) and ischemia progression (2/17). For the 80 patients, the PSV (median: 127 cm/s vs. 116 cm/s, P = 0.039), the ratio of ipsilateral-MFV/contra-lateral-MFV (iMFV/cMFV) (median: 1.29 vs. 1.02, P = 0.036) and iMFV/mean blood pressure (MBP) (median: 0.97 vs. 0.79, P = 0.008) in END patients were higher than those of non-END. Using the receiver-operating characteristic curve to obtain cut-off values for PSV, PI, iMFV/cMFV, and iMFV/MBP for END, we found that PI ≥0.85 (odds ratio: 11.03, 95% confidence interval: 1.92-63.46, P = 0.007) and iMFV/MBP ≥0.84 (odds ratio: 9.20, 95% confidence interval: 2.07-40.84, P = 0.004) were independent predictors of END in a multivariate logistic regression model, with a sensitivity of 82.4% and 76.5% and a specificity of 42.9% and 66.7%, respectively, and had the positive predictive values of 29.0% and 38.2%, and negative predictive values of 90.0% and 91.3%, with an area under the receiver-operating characteristic curve of 0.57 and 0.71, respectively.@*CONCLUSION@#TCD examination of EVT patients may be used as a real-time tool to detect END predictors, such as the higher PI and iMFV/MBP, allowing for better post-thrombectomy management in ACIS patients.

ABSTRACT

PURPOSE: To study the efficacy of capecitabine or S-1 plus oxaliplatin (CAPOX or SOX) for treating thymidine phosphorylase (TP)- or dihydropyrimidine dehydrogenase (DPD)-positive advanced gastric cancer.MATERIALS AND METHODS: Eighty-six patients with stage IIIC to IV gastric cancer were assessed for TP and DPD expression by immunohistochemistry. The association between CAPOX or SOX efficacy and TP/DPD expression was retrospectively analyzed.RESULTS: There were no significant differences in the objective remission rate (ORR, 52.27% vs. 47.62%; P>0.05), disease control rate (72.73% vs. 73.81%, P>0.05), progression-free survival (hazard ratio [HR], 1.119; 95% confidence interval [CI], 0.739–1.741; P=0.586), and overall survival (OS; HR, 0.855; 95% CI, 0.481–1.511; P=0.588) between CAPOX and SOX. A higher number of stage IV patients showed TP positivity, while DPD-positive patients predominantly showed intestinal type of gastric cancer. In TP-positive patients, the ORRs associated with CAPOX and SOX treatments were 57.14% and 38.10%, respectively; OS was better with CAPOX than with SOX (HR, 0.447; 95% CI, 0.179–0.978; P=0.046). Among DPD-positive patients, the SOX treatment-associated ORR (60.87%) was significantly higher than the CAPOX treatment-associated ORR (43.48%). Furthermore, SOX treatment resulted in better OS than did CAPOX treatment (HR, 2.020; 95% CI, 1.019–4.837; P=0.049).CONCLUSIONS: No significant difference in clinical efficacy was found between CAPOX and SOX. TP-positive patients might respond better to CAPOX while DPD-positive patients may respond better to SOX. Our findings might serve as a guide for personalized chemotherapy for gastric cancer.

ABSTRACT

Objectives: To analyze the cardiac T2* value, liver iron concentration (LIC) , and related laboratory parameters in myelodysplastic syndrome (MDS) with iron overload and evaluate the changes of organ functions after iron chelation therapy. To explore the value of magnetic resonance imaging (MRI) T2* in making early diagnosis and assessing organs iron overload. Methods: Retrospective investigation was used to observe the cardiac T2* value, LIC, iron metabolism parameters and related laboratory parameters of 85 MDS patients from Nov 2014 to Jan 2018. Among them, 7 MDS patients with Low/Int-1 have received iron chelation therapy for 6 months during two MRI examinations. The above parameters were collected before and after iron chelation therapy for comparison. Results: Correlations were found between heart T2* value and age (rs=-0.290, P=0.007) and left ventricular ejection fraction (LVEF) (rs=0.265, P=0.009) . There was a significant negative correlation between heart T2* value and blood transfusion units (rs=-0.701, P<0.001) . There was a significant positive correlation between LIC and serum ferritin (SF) (rs=0.577, P<0.001) . There was also a correlation between LIC and ALT (rs=0.268, P=0.014) and blood transfusion units (rs=0.244, P=0.034) . There was no correlation between heart T2* and pro-BNP, SF (all P>0.05) , and no correlation between LIC and age (P>0.05) . The increase of heart T2* between the normal and abnormal groups was statistically significant (P=0.005) , but the iron overload ratio of the heart T2*<20 ms was not significant between the two groups. There was statistical significance in the proportion of severe liver iron overload (LIC>15 mg/g DW) (P=0.045) . After iron chelation therapy, the values of SF, transferrin saturation, ALT, AST, pro-BNP and LIC of 7 patients were decreased compared with values before iron chelation therapy, and the peripheral blood cell level was increased. However, the changes of LVEF and T2* values after iron chelation were not obvious. Conclusion: MRI T2* may be a predictor of iron overload in patients with MDS in early stage, and may be more valuable compare with LVEF, SF and other laboratory indicators. The safety and repeatability of MRI cardiac T2* examination are recognized, and it can be used as an ideal detection for patients with iron overload.

Objective@#To investigate the effect of human glutathione peroxidase 4 (GPX4) on the proliferation and metastasis of renal clear cell carcinoma and its relationship with the expression of IGF-1R and COX-2.@*Methods@#Culture of human normal tubular cell line HK-2 and human renal clear cell carcinoma Caki-1, A498, Caki-2, 786-o in vitro. Detection of GPX4 mRNA and protein expression in different cell lines by quantitative real-time PCR (RT-PCR) and Western blot assay. Overexpression of GPX4 cell lines, including blank carrier (Vector) and overexpress GPX4 (oeGPX4) group, and interference with GPX4 renal clear cell carcinoma cell lines, including random sequence (shControl), interference GPX4#1 (shGPX4#1) and interference GPX4#2 (shGPX4#2) group by lentiviral transfection. RT-PCR technology and Western blot were used to detect the expression of GPX4, IGF-1R and COX-2 mRNA and protein. CCK-8 assay was used to detect the relative proliferation of cells at 0, 24, 48, 72 and 96 h in each group. Transwell invasion and migration assay to detect the invasion and migration ability of cells of each group.@*Results@#GPX4 is highly expressed in renal clear cell carcinoma cell lines compared to human normal tubular cell lines; The expression of GPX4, IGF-1R and COX-2 mRNA was significantly increased in oeGPX4 cells compared with Vector cells, the expression of GPX4,IGF-1R and COX-2 mRNA was significantly decreased in shGPX4#1 and shGPX4#2 compared with shControl cells; oeGPX4 cells significantly increased proliferative capacity compared to Vector cells at 72 and 96 h, the proliferation of shGPX4#1 and shGPX4#2 cells was significantly lower than that of shControl cells at 72 and 96 h; The number of invading and migrating cells of oeGPX4 cells was significantly higher than that of Vector cells, the number of invasive and migrating cells in shGPX4#1 and shGPX4#2 cells was significantly lower than that in shControl cells.@*Conclusion@#GPX4 is highly expressed in renal clear cell carcinoma cells, which is positively correlated with the expression of IGF-1R and COX-2, and can promote cell proliferation and metastasis in vitro.

ABSTRACT

BACKGROUND@#Although the rehabilitation of aphasia has been extensively studied, the prediction of language outcome still has not received sufficient attention. The aim of this study was to predict the language outcome using mismatch negativity (MMN) in patients with large left-hemispheric infarction.@*METHODS@#MMN was elicited by an oddball paradigm in which a standard tone (1000 Hz) and deviant tone (1500 Hz) were presented at 90% and 10% of the number of tones, respectively. The mean amplitudes and laterality indexes (LIs) of MMN were measured over the prefrontal, frontal, central, parietal, temporal, and perisylvian electrodes and both hemispheres during the first 7 days (session 1) and 10 to 20 days (session 2) post-onset. Mixed three-way analysis of variance (ANOVA) was used to investigate differences in these factors between two aphasia groups (the good recovery group and poor recovery group). The predictive value of the most significant LI was also compared with the score of National Institutes of Health Stroke Scale score and low-density volume on computed tomography.@*RESULTS@#A total of 18 patients were enrolled in this study. Mixed three-way ANOVA showed no interaction effect of session × region of interest (ROI) × group (F [3.59, 57.38] = 1.301, P = 0.282) and no interaction effect of ROI × group (F [1.81, 29.01] = 0.71, P = 0.487) and session × group (F [1.00, 16.00] = 0.084, P = 0.776) for MMN amplitude. No interaction effect of session × ROI × group (F [1.79, 28.58] = 0.62, P = 0.530), but an interaction effect of session × group (F [1.00, 16.00] = 5.21, P = 0.036) was found for LIs. In the poor recovery group, the LIs of MMN over all the ROIs, except the parietal area, became more negative at session 2 than those at session 1 (P -0.36 over the perisylvian area suggested good recovery, but a score <-0.36 suggested poor recovery. The LI cut-off value of -0.36 had the highest sensitivity (90.0%) and specificity (87.5%) for predicting a good language outcome at 3 months post-stroke.@*CONCLUSION@#LIs of MMN amplitudes at approximately 2 weeks post left-hemisphere stroke serve as more sensitive predictors of language outcome, among which the LI over the perisylvian area exhibits the best predictive value.

ABSTRACT

Objective To evaluate the chemopreventive effects of 8-allyl garcinol on oral squamous cell carcinoma(OSCC).Methods OSCC cell line CAL27 were cultured and treated with different concentrations of garcinol or 8-allyl garcinol. Their effects on the biological behaviors of OSCC cell line CAL27 were measured by MTT assay,clony formation assay,scratch migration assay,and flow cytometry with Annexin V-FITC/PI staining assay. We established DMBA-induced hamster cheek pouch models of dysplasia. While the negative control group was not treated,the positive group was treated with 0.5% DMBA solution tropically to the left cheek pouch three times per week for three consecutive weeks. The other four groups received 0.5 mmol/L or 1.0 mmol/L garcinol or 8-allyl garcinol respectively three times within the following two weeks after DMBA treatment. Hamsters were sacrificed at the fifth week to obtain tissue samples of the left cheek pouch. The samples were examined by histopathology and BrdU immunohistochemisty.Results MTT assay showed that both garcinol and 8-allyl garcinol inhibited the proliferation of CAL27 cells in a concentration-and time-dependent manner. The half maximal inhibitory concentration(IC)of 8-allyl garcinol[(13.13±2.55)μmol/L] was significantly lower than garcinol[(32.20±3.24)μmol/L;t=8.008,P=0.001]. Comparing the two grougs of medicine in the same concentration,the inhibiting proliferation effects 8-allyl garcinol had significantly stronger effect in inhibiting proliferation than garcinol when the same dose was applied,and the difference was largest at the concentrations of 10(24 h:t=8.012,P=0.001;48 h:t=5.939,P=0.001;72 h:t=12.551,P=0.001)and 20 μmol/L(24 h:t=8.887,P=0.001;48 h:t=9.324,P=0.002;72 h:t=5.361,P=0.002). The clone formation assay showed the clone formation rates after the treatment with 20 μmol/L garcinol and 20 μmol/L 8-allyl garcinol were(44.1±0.4)% and(23.6±0.6)%,respectively,which were significantly lower than those after treatment with 10 μmol/L garcinol[(55.6±2.8)%;t=6.894,P=0.019] and 10 μmol/L 8-allyl garcinol[(31.0±0.6)%;t=15.556,P=0.001]. The inhibiting effects of 8-allyl garcinol at the concentrations of 10 μmol/L(t=14.682,P=0.003)and 20 μmol/L(t=51.514,P=0.001)were significantly stronger than garcinol.Scratch test showed the relative cell migration rates after treatment with 10 and 20 μmol/L garcinol for 12 hours were(16.00±4.55)%(t=3.139,P=0.026)and(3.00±3.16)%(t=6.608,P=0.001),respectively,which were lower than negative control [(30.33±7.64)%]. The relative cell migration rates after treatment with 10 and 20 μmol/L 8-allyl garcinol for 12 hours were(16.25±3.86)%(t=3.245,P=0.023)and(6.00±2.65)%(t=5.214,P=0.006),respectively,which were also lower than negative control[(30.33±7.64)%]. In addition,the relative cell migration rates after treatment with 10 and 20 μmol/L garcinol for 24 hours were(23.75±4.57)%(t=4.718,P=0.005)and(5.75±1.50)%(t=10.432,P=0.001),respectively,which were lower than negative control[(45.33±7.64)%]. The relative cell migration rates after treatment with 10 and 20 μmol/L 8-allyl garcinol for 24 hours were(23.50±2.38)%(t=5.529,P=0.003)and(11.67±2.31)%(t=7.308,P=0.002),respectively,which were also lower than negative control[(45.33±7.64)%]. Furthermore,the relative cell migration rate after treatment with 20 μmol/L garcinol for 24 hours was significantly lower than after treatment with 8-allyl garcinol(t=4.151,P=0.009). The apoptosis experiments showed that the early apoptosis rate of CAL27 cells was(5.00±0.10)% after treatment with 10 μmol/L garcinol,which was significantly higher than negative control[(1.57±0.21)%;F=70.950,P=0.001]. The early and late apoptosis rates of CAL27 cells were(5.90±0.78)%(t=39.384,P=0.001)and(9.73±1.67)%(t=10.101,P=0.001),respectively,after treatment with 20 μmol/L garcinol,which were also significantly higher than negative control. The early apoptosis rate of CAL27 cells was(4.63±1.16)% after treatment with 8-allyl garcinol,which was significantly higher than negative control(t=4.511,P=0.041). The effects of 8-allyl garcinol in promoting cell apoptosis were weaker than garcinol(10 μmol/L:t=5.982,P=0.004;20 μmol/L:t=8.578,P=0.001). The histopathological test also showed that the hyperplastic areas of oral mucosal epithelium in hamsters after treatment with 0.5 mmol/L garcinol(t=2.546,P=0.031),0.5 mmol/L 8-allyl garcinol(t=3.485,P=0.008),1.0 mmol/L garcinol(t=4.556,P=0.001),and 1.0 mmol/L 8-allyl garcinol(t=5.393,P=0.001)were significantly smaller than positive control. The dysplasia areas of oral mucosal epithelium in hamsters after treatment with 0.5 mmol/L 8-allyl garcinol(t=2.130,P=0.046),1.0 mmol/L garcinol(t=3.434,P=0.010),and 1.0 mmol/L 8-allyl garcinol(t=4.518,P=0.004)were also smaller than positive control;1.0 mmol/L garcinol group(t=2.793,P=0.023)and 1.0 mmol/L 8-allyl garcinol group(t=4.997,P=0.001)were smaller than 0.5 mmol/L garcinol treatment group. Immunohistochemical staining of BrdU showed that the BrdU-labeled indicators were significantly lower in negative control group(t=7.563,P=0.001),0.5 mmol/L garcinol(t=2.862,P=0.029),0.5 mmol/L 8-allyl garcinol(t=4.693,P=0.002),1.0 mmol/L garcinol(t=5.071,P=0.002),and 1.0 mmol/L 8-allyl garcinol(t=5.133,P=0.001)when compared with the positive control. The BrdU-labeled indicators in 0.5 mmol/L 8-allyl garcinol(t=3.724,P=0.007),1.0 mmol/L garcinol(t=7.000,P=0.001),and 1.0 mmol/L 8-allyl garcinol(t=4.413,P=0.003)were also significantly lower than in 0.5 mmol/L garcinol group.Conclusions 8-allyl garcinol could inhibit the proliferation and migration of OSCC cell line CAL27 and promotes apoptosis. It also has prominent inhibitory effects on DMBA-induced hamster cheek pouch dysplasia. However,the specific effects are slightly different from garcinol.
