ABSTRACT

OBJECTIVE: To prepare standard decoction of fried Perilla frutescens seed and study the quality standard. METHODS: According to the preparation requirements of standard decoction, 17 batches of standard decoction of fried P. frutescens seed were prepared, and the yield of paste was calculated. HPLC method was used for quantitative analysis of rosmarinic acid in standard decoction of fried P. frutescens seed. The determination was performed on Agilent 5 TC-C18(2) column with mobile phase consisted of methanol-0.1% formic acid solution (40 ∶ 60, V/V) at the flow rate of 1.0 mL/min. The detection wavelength was set at 330 nm, and column temperature was 30 ℃. The sample size was 5 μL. The transfer rate was calculated. HPLC fingerprint was established for 17 batches of standard decoction of fried P. frutescens seed, and analyzed by using TCM Chromatogram Fingerprint Similarity Evaluation System (2012 edition). The chromatogram peaks were identified by comparing retention time of common peak. RESULTS: The yield of paste were 5.55%-9.75% in 17 batches of standard decoction of fried P. frutescens seed. The contents of rosmarinic acid were 0.44%-1.58%, and average content was 1.08%. The transfer rates of rosmarinic acid were 18.31%-34.32%, and average transfer rate was 25.42%. In HPLC fingerprints for 17 batches of standard decoction of P. frutescens seed, a total of 11 common peaks were identified, and the similarity was higher than 0.95. Five common peaks were identified, namely caffeic acid (peak 3),luteolin (peak 5), rosmarinic acid (peak 8) , luteolin (peak 9), apigenin (peak 10). CONCLUSIONS: The quality control method of standard decoction of fried P. frutescens seed is established, which can provide reference for the formulation of the quality standard of fried P. frutescens seed granules and related preparations.

ABSTRACT

OBJECTIVE:To establish a method for the simultaneous content determination of berberine hydrochloride,phello-dendrine hydrochloride and magnoflorine in Phellodendron amurense decoction piece, and to campare the contents of the 3 ingredi-ents in different grades of P. amurense decoction piece. METHODS:HPLC was performed on the column of Phenomenex Luna C18 with mobile phase of acetonitrile-0.05 mol/L KH2PO4 (gradient elution) at a flow rate of 1 ml/min,the detection wavelength was 280 nm,the column temperature was 30 ℃,and injection volume was 5 μl. RESULTS:The linear ranges were 0.387 0-7.740 μg for berberine hydrochloride(r=0.999 9),0.044 4-0.888 0 μg for phellodendrine hydrochloride(r=0.999 8)and 0.048 0-0.960 0 μg for magnoflorine(r=0.999 9);RSDs of precision, stability and reproducibility tests were lower than 3%, recoveries were 95.61%-103.22%(RSD=2.80%,n=6),96.18%-102.80%(RSD=1.84%,n=6) and 97.93%-102.78%(RSD=1.84%,n=6). CONCLUSIONS:The method is simple and accurate, and can be used for the contents determination of berberine hydrochloride, phellodendrine hydrochloride and magnoflonine in P. amurense. The contents of berbenine hydrochloride and phellodendrine hydro-chloride in the first-grade decation piece are higher than those in the second-grade decoction piece, and the content of magnoflorine in both decoction pieces shows no discernible differences.

ABSTRACT

To study the pharmacokinetics of ginsenosides Rg1 and its metabolites after iv and oral administration in Wistar rats, the LC-MS/MS method was selected to determine ginsenosides Rg1 and its metabolites in plasma and their pharmacokinetic parameters were calculated. After oral administration of ginsenosides Rg1 to rats, ginsenosides Rg1, Rh1, F1 and protopanaxatriol (Ppt) could be detected in plasma. Their Tmax were 0.92, 3.64, 5.17, and 7.30 h, respectively; MRT were 2.68, 5.06, 6.65, and 5.33 h, respectively; AUC(o-t), were 2 363.5, 4 185.5, 3 774.3, and 396.2 ng x mL(-1) x h, respectively. After iv administration of ginsenosides Rg1 to rats, ginsenosides Rg1, Rh1 and FI could be detected in plasma. Their T1/2betaS were 3.12, 5.87, and 6.87 h, respectively; MRTs were 1.92, 5.99, and 7.13 h, respectively; AUCo-tS were 1 454.7, 597.5, and 805.6 ng x mL(-1) x h, respectively. So, it can be concluded that after oral administration, the amounts of metabolites were higher than the prototype in vivo, and the distribution and elimination of the metabolites were relatively slow. After iv administration, the amount of prototype were higher than that of the metabolites in vivo, and the distribution and elimination of the metabolites were relatively slow.

ABSTRACT

AIM: To compare the components of crude and processed Dryopteris Crassirhizoma Nakai by different kinds of cutting and carbonizing. METHODS: UV-spectrometry was utilized to analyze the yield and extract content. RESULTS: Dryopteris Crassirhizoma Nakai after being curshed has the maximum in the yield, the total phenol of carbonized products, the water extract and alcohol extract. CONCLUSION: There are significant differences in intrinsic quality among various processed products of Dryopteris Crassirhizoma nakai, among them the crushed one has the highest quality, which conforms to China pharmacopeia 2000 VolⅠ.
