ABSTRACT

Flagellum is a slender and wavy protein-attached filament on the surface of certain bacterial cells. It not only plays an important role in the movement and pathogenic ability of bacteria, but also participates in a variety of host immune regulation. Flagellin is a structural protein that forms the main part of flagellar filaments and can be recognized by TLR5 and other receptors in the host cell to induce the body′s immune response. At present, flagellin is widely used in the research of new immune adjuvants due to its immune activation, and its inflammation inhibitory effect also has good prospects against immune pathological damage. In this review, we summarized and analyzed the recent progress on the basic structure and function of flagellin, the host recognition mechanism, and its role in regulating the host immune system.

ABSTRACT

Natural product bufotenine (5) which could be isolated from Venenum Bufonis, has been widely used as a tool in central nervous system (CNS) studies. We present here its quaternary ammonium salt (6) which was synthesized with high yields using 5-benzyloxyindole as raw materials, and we firstly discover its analgesic effects in vivo. The analgesic evaluation showed that compounds 5 and 6 had stronger effects on the behavior of formalin induced pain in mice. Moreover, the combination of compound 6 and morphine has a synergistic effect. We intended to explain the molecular mechanism of this effect. Therefore, 36 analgesic-related targets (including 15 G protein-coupled receptors, 6 enzymes, 13 ion channels, and 2 others) were systemically evaluated using reverse docking. The results indicate that bufotenine and its derivatives are closely related to acetyl cholinesterase (AChE) or α

ABSTRACT

Objective:To analyze the exercise and rehabilitation training programs for children's balance ability using scoping review. Methods:The literatures about children's balance ability, exercise and rehabilitation training were retrieved from CNKI, VIP, PubMed and Web of Science, until 2021-03-17. The authors, published time, countries, objects and their ages, study tools, interventions and the time, and outcomes were extracted. Results:A total of 2679 literatures were returned, 34 of which were enrolled, which came from 13 countries, and were almost published in journals of medicine, sports science, and rehabilitation and disabilities. The literatures were mainly published after 2010, investigate with measurements and scales. The measurements mainly included foot pressure system, force platform, Biodex balancer, etc.; and the scales mainly included Berg Balance Scale, Pediatric Balance Scale, Gross Motor Function Measure, Pediatric Evaluation of Disability Inventory, and Bruininks-Oseretsky Test of Motor Proficiency, etc. There were four typies of program: balance training, interactive computer games, virtual reality and sports events. Interventions for sick children included strength training, whole-body vibration training, Wii-Fit balance games and virtual reality treadmill training, etc. Sports for healthy children mainly included skating, Taiji Quan and gymnastics, etc. Conclusion:There are differences in the researches for children's balance ability at home and abroad. The abroad researches focus on the children with various diseases, in a larger age range, using more evaluation tools and more intervention methods. However, Chinese researches are usually for a longer intervention time, more subjects, and focusing more on healthy children. It is needed to pay more attention on children with diseases before three years old, for early diagnosis and treatment. Long-term effects should be observed other than immediateness after intervention.

ABSTRACT

Although localized prostate cancer (PCa) can be cured by prostatectomy and radiotherapy, the development of effective therapeutic approaches for advanced prostate cancer, including castration-resistant PCa (CRPC) and neuroendocrine PCa (NEPC), is lagging far behind. Identifying a novel prognostic and diagnostic biomarker for early diagnosis and intervention is an urgent clinical need. Here, we report that apolipoprotein A-I (ApoA-I), the major component of high-density lipoprotein (HDL), is upregulated in PCa based on both bioinformatics and experimental evidence. The fact that advanced PCa shows strong ApoA-I expression reflects its potential role in driving therapeutic resistance and disease progression by reprogramming the lipid metabolic network of tumor cells. Molecularly, ApoA-I is regulated by MYC, a frequently amplified oncogene in late-stage PCa. Altogether, our findings have revealed a novel indicator to predict prognosis and recurrence, which would benefit patients who are prone to progress to metastasis or even NEPC, which is the lethal subtype of PCa.

ABSTRACT

Accurate methods for identifying pelvic lymph node metastasis (LNM) of prostate cancer (PCa) prior to surgery are still lacking. We aimed to investigate the predictive value of peripheral monocyte count (PMC) for LNM of PCa in this study. Two hundred and ninety-eight patients from three centers were divided into a training set (n = 125) and a validation set (n = 173). In the training set, the independent predictors of LNM were analyzed using univariate and multivariate logistic regression analyses, and the optimal cutoff value was calculated by the receiver operating characteristic (ROC) curve. The sensitivity and specificity of the optimal cutoff were authenticated in the validation cohort. Finally, a nomogram based on the PMC was constructed for predicting LNM. Multivariate analyses of the training cohort demonstrated that clinical T stage, preoperative Gleason score, and PMC were independent risk factors for LNM. The subsequent ROC analysis showed that the optimal cutoff value of PMC for diagnosing LNM was 0.405 × 109 l

ABSTRACT

Intestinal microecology is an important defense system in the human body. The intestinal flora is the core micro-ecosystem in the human intestine. It has a symbiotic relationship with the overall functions of the body. It has strong metabolic activity to maintain the normal functioning of the body and resist the invasion of various viral antigens in the body. Playing a protective function, the imbalanced intestinal microecology can cause various diseases. Polysaccharides can be extracted from a wide range of sources and have low toxicity and side effects. They have attracted wide attention because of their anti-tumor, anti-oxidant, anti-inflammatory and other biological activities. Studies have demonstrated that polysaccharides can regulate intestinal microecological disorders. According to the studies in recent years, this review summarizes that polysaccharides mainly modulate intestinal microecological disorders through regulating the composition of intestinal flora, improving the metabolism of the flora, and repairing the intestinal tract barrier. On the basis of these mechanisms of action, this paper elaborates the anti-tumor, immunomodulatory, and anti-inflammatory activities of polysaccharides. This paper can provide reference for the future research on the intestinal microecology-regulating mechanism and biological activities of polysaccharides.

ABSTRACT

BACKGROUND: Foot strike patterns in adults during running have always been the focus of worldwide research, and the strike patterns of children are also something that cannot be ignored. OBJECTIVE: Using biomechanical methods to explore the differences in kinematics and kinetics of children in different strike patterns during the running process, and to provide a scientific basis for children’s correct way of running. METHODS: Seventy-four children were randomly selected from a public kindergarten in Haidian District, Beijing, and were divided into 3-year-old group, 4-year-old group and 5-year-old group. The kinematics and kinetics data of enrolled children in different strike patterns during running were acquired simultaneously using the BTS infrared motion capture system, the Kistler three-dimensional force table and the VIXTA video analysis system. The muscle strength index of the lower limbs was calculated using the Anybody 5.2 simulation modeling software. Before participation in the trial, children’s parents were fully informed of study protocol and signed the informed consent form. The trial protocol met the relevant ethical requirements of Beijing Normal University. RESULTS AND CONCLUSION: (1) In the 3-year-old group, the proportion of mid foot strike (MFS) was the highest, and the proportion of fore foot strike (FFS) was the lowest. In the 5-year-old group, the proportion of MFS was the lowest, and the proportion of FFS was the highest. The rear foot strike (RFS) extension time was longer than that of FFS (P < 0.01) and MFS (P < 0.05). (2) At the moment of landing, the flexion angle of FFS was greater than that of RFS (P < 0.01) and MFS (P < 0.05), and the flexion angle of MFS was greater than that of RFS (P < 0.05). The hip adduction-abduction angle of RFS was greater than that of FFS (P < 0.01) and MFS (P < 0.05). The maximum hip abduction angle of RFS was greater than that of FFS (P < 0.01) and MFS (P < 0.01). The amount of joint changes in the RFS hip adduction and abduction was greater than that of FFS (P < 0.01) and MFS (P < 0.05). The minimum flexion and extension of RFS was greater than that of FFS (P < 0.05). The maximal hip adduction-abduction angular velocity of RFS was greater than that of FFS (P < 0.05), and the maximal knee adduction-abduction angular velocity of RFS was greater than that of FFS (P < 0.01) and MFS (P < 0.05). (3) The muscle strength of the short bones of the tibia, the long tibia and the third metatarsal muscle of FFS and MFS was greater than that of RFS (P < 0.05). The bundle muscle strength of the medial femoral muscle, the lateral femoral muscle bundle, the lateral femoral muscle bundle, the medial femoral muscle bundle, the medial femoral muscle bundle, and the medial femoral muscle of RFS were greater than that of FFS (P < 0.01) and MFS (P < 0.05). (4) In the 3-6 years old, children often run in the heel or full-foot landing mode to meet their stability during the running process. As the age increases, the running pattern with the forefoot landing gradually appears. To keep the movement steady, RFS can trigger more hip and knee frontal motions, FFS and MFS can offer more muscle strength on the anterior and posterior sides of the calf, while RFS can offer more muscle strength on the anterior side of the thigh.

ABSTRACT

According to the latest data, the annual mortality rate of liver cancer in China ranks the second among malignant tumors, and it has become one of the most fatal cancers in urban and rural areas. This article starts with the active ingredients of traditional Chinese medicines for liver cancer to summarize the relevant literature at home and abroad, with the hope to provide theoretical references for the development of traditional Chinese medicines against liver cancer. The results show that the active ingredients of traditional Chinese medicines such as Chinese herbal extracts, flavonoids, polysaccharides, alkaloids, saponins, volatile oils, terpenes, quinones, phenols, bioenzymes, and protein components are the entry points, mainly by inhibiting the proliferation of cancer cells, blocking the cancer cell cycle, inhibiting the invasion and metastasis of cancer cells, regulating signal pathways, regulating vascular endothelial growth factor(VEGF) and reactive oxygen species(ROS), etc. to play the effects against liver cancer. It can be seen that the active ingredients of traditional Chinese medicine with coordinated intervention effects at multiple levels and multiple targets are expected to become ideal medicines for the treatment of liver cancer.

ABSTRACT

Gastric cancer is a disease with high mortality, which threatens the health of people for a long time. At present, the main treatment methods are surgery and chemotherapy, but these methods have great harm to the human body. However, it is found that the active ingredients of traditional Chinese medicine have an obvious therapeutic effect in the adjuvant treatment of the tumor. Therefore, the active ingredients of traditional Chinese medicine have become a research hotspot in the anti-tumor field. In recent years, many related researchers have been particularly active in studying the in vitro activity and mechanism of active ingredients of traditional Chinese medicine on human gastric cancer cells. In this paper, the Chinese herbal medicine extracts, polysaccharides, alkaloids, saponins, flavones, terpenes, quinones, volatile oils, esters, phenols, protein components and other active ingredients of Chinese medicine were used as the starting points to investigate the anti-gastric cancer mechanism, such as inhibiting cell proliferation and inducing apoptosis of cancer cells, inhibiting cell invasion and migration; inhibiting over-expression of vascular endothelial growth factor(VEGF); interfering with cell mitosis; and regulating cell signaling pathways. Their in vitro inhibitory activity and mechanism for gastric cancer cells were described in this study, providing a theoretical reference for the development and application of anti-gastric cancer drugs.

ABSTRACT

OBJECTIVE@#To analyze the relationship between the distribution of lower limb alignment and short term clinical efficacy in patients with varus-type osteoarthritis after primary total knee arthroplasty (TKA).@*METHODS@#From December 2016 to March 2018, 87 patients (101 knees) with knee osteoarthritis were treated with the first total knee arthroplasty by the same medical group, including 21 males(25 knees) and 66 females(76 knees), ranging in age from 51 to 85 years old, with a mean of (67.6±7.0) years old. According to the difference of hip knee ankle angle (HKA) after total knee arthroplasty, the patients were divided into 4 groups:neutral position group (group A), -3°≤HKA≤3°, 50 knees;slight varus group (group B), 3°0.05). There was no significant difference in knee joint activity among the 4 groups. The score of femoral prosthesis force line within ±3°was better than that of the other group (0.05).@*CONCLUSION@#The short term clinical efficacy of patients with knee varus osteoarthritis after primary total knee arthroplasty is related to the distribution of lower limbs alignment. The short-term clinical efficacy of slight inversion position can be similar to that of neutral position. The force line distribution of femoral prosthesis is related to the short term clinical efficacy after primary knee arthroplasty.

ABSTRACT

The objective data from the three-dimensional gait analysis equipment test is complex and difficult to explain in the actual evaluation. In order to solve it, the gait index is proposed. The purpose of this review is to analyze the most commonly used gait evaluation index in clinical practice and discuss its calculation methods, advantages and limitations. The results showed that Gait Deviation Index (GDI) and Gait Profile Score (GPS) were the two most widely used indexes, but the operation of GDI was complex; the selection of parameters of Gillette Gait Index (GGI) was poor in objectivity, and the application was limited; the electromyography data is very important in the complete evaluation of gait mode, but the combination with gait indices was not close at present. The influence of gait speed has not been discussed in researches about indices except GGI.

ABSTRACT

Objective Diabetic cardiomyopathy (DCM) is one of the complications of diabetes, which is closely related to the change of miRNA. In this study, we observed the characteristic expression of miR-26b in the tissues of the C57BL/6J mouse and in the heart, adipose tissue and liver of the ob/ob mouse, and investigated the effect of high glucose (Glu) on the expression of miR-26b in H9C2 cardiomyocytes. Methods Using RT-PCR, we measured the levels of miR-26b in the heart, adipose tissue, liver and other tissues of C57BL/6J and ob/ob mice. H9C2 cardiomyocytes were treated with Glu at 5.5, 15, 25 and 35 mmol/L for 0, 24, 48, 72, 96 and 120 hours, followed by detection of the proliferation of cardiomyocytes by CCK-8 and determination of thelevels miR-26b. Results The expression of miR-26b was the highest in the heart of the C57BL/6J mice, significantly higher than in the cardiac and white adipose tissues of the ob/ob mice (P < 0.05). The proliferation of cardiomyocytes was markedly increased in the 15, 25 and 35 mmol/L Glu groups at 24, 48, 72, 96 and 120 hours as compared with that in the 5.5 mmol/L Glu group (P < 0.05), higher in the 25 than in the 15 mmol/L Glu group at 24 hours (0.74±0.02 vs 0.72±0.01, P<0.05), but lower in the 35 than in the 15 mmol/L Glu group at 48 hours (0.92±0.01 vs 0.94±0.01, P<0.05), in the 25 and 35 mmol/L Glu groups at 96 hours (P < 0.05), in the 35 mmol/L Glu group at 120 hours (1.12±0.02 vs 1.19±0.05, P<0.05), in the 35 than in the 25 mmol/L Glu group at 24 and 48 hours (P<0.05). The expression of miR-26b in the H9C2 cardiomyocytes was significantly down-regulated in the 25 and 35 mmol/L Glu groups in comparison with that in the 5.5 mmol/L Glu group (P<0.05), remarkably lower in the 25 mmol/L Glu group at 96 and 120 hours than at 0 hour (P<0.05). Conclusion High glucose can down-regulate the expression of miR-26b in H9C2 cardiomyocytes, which suggests that miR-26b may participate in the pathogenesis of DCM.

ABSTRACT

Objective To investigate the acceptance of HPV self-sampling mode in cervical cancer screening population and explore its feasibility. Methods From March 5 to 15, 2018, researchers investigated women who participated in cervical cancer screening organized by Beijing Shunyi Women's and Children's Hospital in the form of questionnaires. Questionnaires were conducted on their acceptance status and the factors that affect the self-sampling experience. The specific contents of the questionnaires were as follows: (1) the experience of using self-sampling included operability, comfortable, sample time-consuming, bleeding or not after sampling; (2) psychological changes after self-sampling, including the willingness to accept self-sampling again, the worrying problems during self-sampling process. According to whether or not have operating video guidance, the self-sampling experience and psychological changes after self-sampling were compared. Results (1) There were 1 375 women participated in the questionnaire survey, and 86.55% (1 190/1 375) of them thought the self-sampling was convenient, 78.40% (1 078/1 375) thought it was not uncomfortable, 88.58% (1 218/1 375) thought the sampling time was fast (less than 5 minutes), 94.04% (1 293/1 375) self-sampling without bleeding; and 83.27% (1 145/1 375) were willing to self-sampling for cervical cancer screening again, 85.82% (1 180/1 375) were afraid of inaccurate sampling. (2) Among the 1 375 women, 1 202 were in the video guidance group and 173 were in the non-guidance group. The self-sampling experience of women in video guidance group was better than those of non-guidance group in operability, comfortable, sampling time-consuming and bleeding after sampling. The proportion of women who willing to self-sampling again was higher than that of non-guidance group (86.69% vs 59.54%, respectively). The proportion of women who worried operating incorrectly was lower than that of non-guidance group (11.23% vs 32.37%, respectively). The differences were significant (all P<0.05). Conclusions Self-sampling for HPV testing in cervical cancer screening is easy to operate and has little discomfort complaint. It is feasible in cervical cancer screening. Operational video guidance during the screening process could effectively improve the women's experience and willingness to self-sampling again in the future.
