ABSTRACT

Objective@#Schizophrenia (SCZ) is one of the most common and severe mental disorders. Modified electroconvulsive therapy (MECT) is the most effective therapy for all kinds of SCZ, and the underlying molecular mechanism remains unclear. This study is aim to detect the molecule mechanism by constructing the transcriptome dataset from SCZ patients treated with MECT and health controls (HCs). @*Methods@#Transcriptome sequencing was performed on blood samples of 8 SCZ (BECT: before MECT; AECT: after MECT) and 8 HCs, weighted gene co-expression network analysis (WGCNA) was used to cluster the different expression genes, enrichment and protein-protein interaction (PPI) enrichment analysis were used to detect the related pathways. @*Results@#Three gene modules (black, blue and turquoise) were significantly associated with MECT, enrichment analysis found that the long-term potentiation pathway was associated with MECT. PPI enrichment p-value of black, blue, turquoise module are 0.00127, <1×10-16 and 1.09×10-13, respectively. At the same time, EP300 is a key node in the PPI for genes in black module, which got from the transcriptome sequencing data. @*Conclusion@#It is suggested that the long-term potentiation pathways were associated with biological mechanism of MECT.

ABSTRACT

Objective:To evaluate the effect of down-regulation of RACK1 expression on growth and radiosensitivity of oral squamous cell carcinoma cells.Methods:The shRNA vector for RACK1 gene was constructed and transfected into HSC-3 cells by lipofectamine. The stably-transfected cell line was obtained by constructing G418. The expression levels of RACK1 mRNA and protein were detected by RT-PCR and Western blot. The cell proliferation was detected by CCK8 assay. Cell apoptosis was examined by flow cytometry. The invasive and metastatic capabilities of cancer cells were assessed by cell invasion assay in vitro.The effect of X-ray irradiation combined with down-regulation of RACK1 expression upon cell proliferation was assessed by clone formation assay. The xenograft tumor nude mouse model was established to observe the inhibitory effect of down-regulating RACK1 gene expression combined with X-ray irradiation on oral squamous cell carcinoma. Results:RT-PCR revealed that the expression level of RACK1 mRNA of transfected HSC-3 cells was significantly down-regulated ( P<0.05). Western blot showed that the expression level of RACK1 protein was significantly down-regulated ( P<0.05). CCK8 assay demonstrated that down-regulation of RACK1 expression could remarkably inhibit the growth of HSC-3 cells ( P<0.05). RACK1 gene shRNA interference combined with X-ray irradiation significantly enhanced the apoptosis rate of HSC-3 cells ( P<0.05). The number of invasion cells in vitro in the RACK1 silencing group was evidently decreased ( P<0.05). Clone formation assay showed that the survival fraction in the shRACK1 group was significantly lower than that in the control group. The sensitization enhancement ratio was 1.37(ratio of D 0 value). Xenograft tumor experiment in nude mice showed that tumor growth was significantly inhibited in the shRACK1 group, the tumor volume was significantly decreased and the tumor mass was significantly lower than those in the control group (all P<0.05). Conclusion:Down-regulating RACK1 expression can enhance the radiosensitivity of oral squamous cell carcinoma cells, providing novel thinking to improve the radiosensitivity of oral squamous cell carcinoma.

ABSTRACT

Objective:To investigate the classification of patients with gout, and further analyze their clinical features.Methods:Outpatients with gout were enrolled from January 2018 to July 2019 in Depart-ment of Rheumatology, Zhongshan Hospital. Subjects were classified into four groups according to their 24-hour urinaryexcretion and fractional excretion of urate. Clinical features of different groups were analyzed using one-way Analysis of Variance (ANOVA), Kruskal-Wallis H test, or χ2 test. Results:Finally, 378 subjects were enrolled in this cross-sectional study. Among them, 186(49.2%) were renal underexcretion type, 100(26.5%) were combined type, 57(15.1%) were renal overload type, 35(9.3%) were the normal type. Renal underexcretion type was the main subtype in any age-stratified groups. With aging, the proportion of combined type decreased, while the normal type increased. Participants in the combined type were the youngest [(42±14) years of age] with the highest estimated glomerular filtration rate [(94±18) ml·min -1·1.73 m -2], while their serum urate levels were the highest [(554±104) μmol/L]. Subjects in the normal type were the oldest [(60±15) years of age] with the lowest estimated glomerular filtration rate [(71±19) mL·min -1·1.73 m -2], however, their serum urate concentrations were the lowest [(427±118) μmol/L], The difference was statistically significant (age, F=13.98; estimated glomerular filtration rate, F=16.11; urate, F=17.14; P<0.01). Prevalence of urolithiasis were similar among the four groups ( χ2 =2.00, P>0.05). Conclusion:The renal underexcretion type is the main type of gout. Young patients are more likely to suffer from combined type with the highest serum urate levels and the best renal function.

ABSTRACT

Macrolide antibiotics are a class of broad-spectrum antibiotics with the macrolide as core nucleus. Recently, antibiotic pollution has become an important environmental problem due to the irregular production and abuse of macrolide antibiotics. Microbial degradation is one of the most effective methods to deal with antibiotic pollution. This review summarizes the current status of environmental pollution caused by macrolide antibiotics, the degradation strains, the degradation enzymes, the degradation pathways and the microbial processes for degrading macrolide antibiotics. Moreover, the critical challenges on the biodegradation of macrolide antibiotics were also discussed.

ABSTRACT

Objective:To investigate the role of peroxisome proliferator-activated receptor γ (PPARγ)/CD36 signaling pathway in macrophage lipid metabolism after Mycobacterium tuberculosis ( Mtb) infection. Methods:THP-1-derived macrophages were infected with Mtb. Four groups were included in this study, which were control group, Mtb infection group, Mtb+ rosiglitazone (ROZ, PPARγ agonist) group and Mtb+ GW9662 (PPARγ antagonist) group. Western blot and RT-PCR were used to detect the expression of PPARγ in macrophages at protein and mRNA levels, respectively. The lipids in cells were detected by oil red O staining. The concentrations of total cholesterol (TC), triglycerides (TG), low density lipoprotein (LDL-C) and high density lipoprotein (HDL-C) in the supernatant of cell culture were detected by automatic biochemical analyzer. The expression of CD36 was detected by immunohistochemistry. CCK-8 was used to detect the proliferation rate of macrophages. Results:Mtb infection significantly increased the expression of PPARγ in macrophages ( P<0.001), promoted intracellular lipid aggregation and CD36 expression and decreased the levels of TC, TG, LDL-C and HDL-C in the supernatant of cell culture ( P<0.001) and cell proliferation rate ( P<0.001). PPARγ agonist significantly enhanced the intracellular lipid accumulation and CD36 expression that were induced by Mtb infection and down-regulated the lipid level in the supernatant of cell culture and cell proliferation rate, while PPARγ antagonist reversed the above effects. Conclusions:PPARγ played a role in lipid metabolism in Mtb-infected macrophages through affecting CD36 expression.

ABSTRACT

OBJECTIVE:To management recheck and sorting weight for finished intravenous solutions in PIVAS ,to provide reference for reducing dispensing error and improving the safety and quality of intravenous infusion therapy. METHODS :The weight analysis method was used to determine the weight of main drug and solvent in the finished intravenous solutions and infusion bottle . The weight maintenance information was added in PIVAS information management system ,and the marked weight of finished intravenous solutions was calculated for the verification of finished intravenous solutions. Average daily check quantity of finished product ,checking time ,average checking time of finished products per bag ,detection rate of dispensing error ,external error and timeliness of finished infusion batch were compared before (Mar.-May,2019,n=83 006)and after (Jun.-Aug.,2019, n=83 173)management. The effects of weighting recheck management were evaluated. RESULTS :Compared with before the implementation of weighting recheck management ,there were no significant differences in the average daily check quantity of finished products ,the detection rate of dispensing errors caused by wrong labeling of liquid ,or the times of delayed drug delivery batches after the implementation of weighting recheck management (P>0.05). The checking time of finished products ,average checking time of finished products per bag ,the number of bags added or subtracted error ,detection rate of dose dispensing error , total error detection rate prolonged or increased significantly (P<0.05),and the number of external error was decreased significantly(P<0.05). CONCLUSIONS :The weighting recheck management improves the accuracy and safety of PIVAS preparation,effectively improves error detection rate ,reduces the occurrence of external error ,but prolongs the time of checking , which are urgent to be solved by information and automation means.

ABSTRACT

In the post-genomic era, gene function analysis has attracted much attention. Transformation is often needed to investigate gene function. In this study, an easy, rapid, reliable, and cost-effective colony polymerase chain reaction (PCR) method for screening mushroom transformants was developed: picking up a suitable amount of transformant's tissue (1–10 μg) to 20 μl 0.25% Lywallzyme solution, and vortexing for 10 s followed by incubation at 34 °C for 15 min. Finally, 2 μl of the suspension was used as templates to perform PCR and single target bands were successfully amplified from respective transformants of Tremella fuciformis, Pleurotus ostreatus, and Pleurotus tuber-regium. This procedure could be widely employed for screening transformants in mushroom transformation experiments.

Objective To investigate the clinical value of the serum new molecular markers,soluble triggering receptor expressed on myeloid cells-1 (sTREM-1)and soluble hemoglobin scavenger receptor(sCD163),in the diagnosis of sepsis in elderly patients with burns.Methods A total of 58 inpatients with burns from Jun 2017 to June 2018 were enrolled in the study.Patients were divided into three groups:the sepsis group(n=12),the localized infection group(n=21)and the non-infection group (n=29).The levels of sTREM-1 and sCD163 were determined by enzyme-linked immunosorbent assays(ELISAs).The clinical diagnostic value of sTREM-1 and sCD163 was assessed by receiver operating characteristic(ROC)curve analysis.Results There was a statistically significant difference in thelevels of sTREM-1 and sCD163 at day 1 between the three groups(F =20.994 and 38.363,P<0.01).Serum levels of sTREM-1 and sCD163 were higher in the sepsis group than in the localized infection group and the non-infection group.Serum levels of sTREM-1 and sCD163 were higher in the localized infection group than in the non-infection group.Serum levels of sTREM-1 and sCD163 were lower at day 7 than those at day 1 in all groups(F=21.242 and 41.035,P<0.01).Serum sTREM-1 levels were positively correlated with serum sCD163 levels (r =0.609,P =0.000).The AUC of sTREM-1 and sCD163 for the diagnosis of sepsis was 0.880(95%CI:0.816~0.926).Conclusions Serum levels of sTREM-1 and sCD163 are elevated with increasing degrees of infection.Monitoring serum sTREM-1 and sCD163 levels is helpful for the diagnosis of sepsis in elderly patients with burns.

ABSTRACT

Objective: To evaluate the biomechanical effects of combined use of miniscrew and clear aligner in different kinds of loading condition on the en-mass retraction of maxillary anterior teeth. Methods: 3 D finite element models of the maxillary bone with miniscrews and clear aligner were reconstructed using the method of reverse engineering with CBCT data of an adult patient who had maxillary first premolars extracted. The orthodontic force was loaded by (1) clear aligner, (2) clear aligner and 1. 47 N force of retraction at appliance and (3) clear aligner and 1. 47 N force of retraction at canine, respectively. Results: Under the working condition of (1), (2) and (3), in sagittal direction, the displacement difference of crown and root of the maxillary central incisor was 1. 12 E-02 mm, 1. 29 E-02 mm and 9. 62 E-03 mm respectively, the displacement of the first molar crown was-2. 49 E-02 mm, -2. 09 E-02 mm and-2. 00 E-02 mm respectively; in vertical direction, extrusion of the maxillary central incisor was 1. 77 E-03 mm, 2. 93 E-03 mm and 6. 53 E-04 mm respectively. Conclusion: The working condition (3) is more advantageous to control the torque of incisors and to save the anchorage of posterior teeth, and more effective to control the extrusion of the incisors.

ABSTRACT

OBJECTIVE: The present study is to evaluate the biological functions of long non-coding RNA (lncRNA), X-inactive specific transcript, X-inactive specific transcript (XIST) in human epithelial ovarian cancer (EOC). METHODS: XIST was upregulated in EOC cell lines, CAOV3 and OVCAR3 cells by lentiviral transduction. The effects of XIST overexpression on cancer cell proliferation, invasion, chemosensitivity and in vivo tumor growth were investigated, respectively. Possible sponging interaction between XIST and human microRNA hsa-miR-214-3p was further evaluated. Furthermore, hsa-miR-214-3p was overexpressed in XIST-upregulated CAOV3 and OVCAR3 cells to evaluate its effect on XIST-mediated EOC regulation. RESULTS: Lentivirus-mediated XIST upregulation had significant anticancer effects in CAOV3 and OVCAR3 cells by suppressing cancer cell proliferation, invasion, increasing cisplatin chemosensitivity and inhibiting in vivo tumor growth. Hsa-miR-214-3p was confirmed to directly bind XIST, and inversely downregulated in XIST-upregulated EOC cells. In EOC cells with XIST upregulation, secondary lentiviral transduction successfully upregulated hsa-miR-214-3p expression. Subsequently, hsa-miR-214-3p upregulation functionally reversed the anticancer effects of XIST-upregulation in EOC. CONCLUSION: Upregulation of lncRNA XIST may suppress EOC development, possibly through sponging effect to induce hsa-miR-214-3p downregulation.

Objective To study the variation of the dental and basal arch forms before and after orthodontic treatment of patients with skeletal class Ⅱ malocclusion by tooth extraction.Methods The mandibular dental casts of 25 skeletal class Ⅱ patients were laser scanned before and after treatment.The facial axis (FA) points,which were used to represent the dental arch,and the WALA point,which were used to represent the basal bone,were identified for each tooth from the right first molar to the left first molar.We constructed the curve with points of projection of the FA and WALA point on the reference plane,and compared the curve of dental arch and basal bone before and after treatment.Results The width of anterior middle and posterior of the basal arch increased (0.835± 1.259) mm,(1.700±1.280) mm and (2.170±1.227) mm,respectively.While the dental arch had a wider width (1.110±1.566) mm in posterior segment but a narrower one in anterior (3.345±2.907) mm.Conclusions The width of middle and posterior of basal bone represented by WALA ridge becomes wider after orthodontic treatment with tooth extraction in patients with skeletal class Ⅱ malocclusion,while the dental arch turns to a new form,which is more coordinated to the basal bone.

Objective@#To investigate the relationship between expressions of OCT-4, CD117 and clinicopathological features and prognosis of patients with ovarian cancer.@*Methods@#A total of 70 paraffin-embedded tissues of patients with ovarian cancer from January 2010 to February 2016 in Shanxi Provincial Cancer Hospital were collected. The expressions of OCT-4 and CD117 were detected by immunohistochemistry.@*Results@#OCT-4 was mainly expressed in cytoplasm, while CD117 was expressed in cell membrane and cytoplasm. The positive expression rate of OCT-4 was 74.3% (52/70), and the positive expression rate of CD117 was 68.6% (48/70). The positive expression rates of OCT-4 in ovarian cancer tissues with poorly differentiation and high CA125 levels (≥500 U/ml), no peritoneal effusion and sensitive to chemotherapy drugs were 92.1% (35/38), 87.5% (28/32), 88.9% (24/27), and 78.7% (48/61), respectively, which were higher than those in ovarian cancer tissues with well and moderately differentiation, low CA125 levels (<500 U/ml), peritoneal effusion and resistance to chemotherapy drugs, the differences were statistically significant (P values were 0.000, 0.020, 0.047, and 0.043). The positive expression rates of CD117 in ovarian cancer tissues with poorly differentiation and peritoneal effusion were 84.2% (32/38) and 79.1% (34/43), respectively, which were higher than those in ovarian cancer tissues with well and moderately differentiation and no peritoneal effusion, the differences were statistically significant (P values were 0.006 and 0.017). Patients with OCT-4-positive expression, peritoneal effusion, and poorly differentiation had a shorter overall survival time (all P < 0.05). The peritoneal effusion and differentiation were independent prognostic factors in patients with ovarian cancer (both P < 0.05).@*Conclusion@#OCT-4 can be used as an important reference for predicting drug sensitivity and evaluating prognosis in patients with ovarian cancer.

In the past decade, mounting evidence points to the possibility of targeting bone for treating, preventing, and predicting type 2 diabetes mellitus. Osteoblast-derived osteocalcin (OCN) can stimulate insulin secretion, enhance insulin sensitivity, and favor glucose and fatty acid uptake and utilization. Lipocalin 2 is another osteokine secreted by osteoblasts and acts in appetite suppression. Neuropeptide Y may function in browning of white adipose tissue and energy expenditure. Osteocytes are proposed to have impact on the browning of white adipose tissue and energy expenditure through the secretion of bone morphogenetic protein 7 and sclerostin. Active bone resorption is also implicated in glucose homeostasis. In addition, there is evidence indicating the involvement of bone-derived receptor activator of nuclear factor κ-B ligand in the regulation of energy metabolism. ABSTRACT

An improved method for extracting high quality and quantity RNA from a jelly mushroom and a dimorphic fungus—Tremella fuciformis which is especially rich in polysaccharides, is described. RNA was extracted from T. fuciformis mycelium M1332 and its parental monokaryotic yeast-like cells Y13 and Y32. The A260/280 and A260/230 ratios were both approximately 2, and the RNA integrity number was larger than 8.9. The yields of RNA were between 108 and 213 µg/g fresh wt. Downstream molecular applications including reverse transcriptional PCR and quantitative real-time PCR were also performed. This protocol is reliable and may be widely applicable for total RNA extraction from other jelly mushrooms or filamentous fungi rich in polysaccharides.

ABSTRACT

Potassium 2-(1-hydroxypentyl)-benzoate (d,l-PHPB), a new drug candidate for ischemic stroke at the phase II clinic trial, has been shown to protect neurons by inhibiting oxidative injury and reducing neuron apoptosis in previous studies. But the mechanisms of d,l-PHPB remain to be studied. In this study, a neuron-astrocytes co-culture system was used to elucidate the roles of astrocytes in neuroprotection of d,l-PHPB under oxygen-glucose deprivation/reoxygenation (OGD/R) condition. Our data showed that d,l-PHPB reduced neuronal apoptosis in mono-culture system and this effect was enhanced in neuron-astrocyte co-culture system under the OGD/R condition. Meanwhile, d,l-PHPB obviously increased the levels of brain-derived neurotrophic factor (BDNF) and nerve growth factor (NGF), which were mainly secreted from astrocytes, in the co-culture system after OGD/R. The PI3K/AKT and ERK signaling pathways as well as the p-TRKA/B receptors were involved in the process. In addition, the levels of TNF-and IL-1secreted from astrocytes after OGD/R were markedly reduced after d,l-PHPB treatment, which was mainly due to the suppression of phosphorylated p38. In conclusion, the present study demonstrates that the neuroprotective effects of d,l-PHPB were improved by astrocytes, mainly mediated by increasing the release of BDNF/NGF and attenuating inflammatory cytokines.

T he police assigned forensic experts to judicially judge the w om an's injuries. A nd the identification results of X-ray and C T show ed that the transverse fracture of patella had been caused by m uscular violence, and that both direct and indirect force resulted in the injuries of left tibia, and the fracture of left m edial orbital w all m ight have been form ed during the violence. T his article expounded the identification betw een the injuries of fam ily violence and traffic accident by the applications of im aging, w hich em phasized the application value of im aging techniques in clinic forensic science.

ABSTRACT

OBJECTIVE:To optimize the extraction technology of polyphenols from Juglans regia branch and evaluate its anti-oxidant activity in vitro. METHODS:Using extraction amount of polyphenols from J. regia branch as response value,solvent-solid ratio,extraction temperature and ethanol volume fraction as response factors,based on single factor test,response surface method was used to optimize the extraction technology of polyphenols from J. regia branch. Using vitamin C as positive control,scaveng-ing on hydroxyl radicals and 1,1-diphenyl-2-picrylhydrazyl(DPPH)radicals and total reducing activities of polyphenols from J. re-gia branch were investigated. And its antioxidant activity in vitro was evaluated. RESULTS:Optimized extraction conditions for polyphenols from J. regia branch was as follow as solid-liquid ratio of 1:25(g/mL),extraction temperature of 50 ℃,ethanol vol-ume fraction of 70%. The extraction amount of polyphenols from J. regia branch was 9.30 mg/g(RSD=0.57%,n=3)in the veri-fication test. Clearance rate on hydroxyl radicals and DPPH radicals and total reducing activity of polyphenols from J. regia branch were respectively 50.24%,95.42%,1.118 when it was under the mass concentration of 12.0,3.0,3.0μg/mL;and the related data of vitamin C was 93.71%,46.17%,0.628 under the same mass concentration(P<0.05). CONCLUSIONS:Extraction technology of polyphenols from J. regia branch optimized by response surface method is stable and feasible;polyphenols from J. regia branch shows certain antioxidant activity in vitro.

ABSTRACT

Skeletal muscle extracellular matrix (ECM) is the microenvironment for muscle cells to survive and function. Changes in ECM components and structure directly affect the activity and function of muscle cells. Pathological remodeling occurs to skeletal muscle ECM in insulin resistance, including collagen deposition, hyaluronan accumulation, activation of membrane protein integrin signaling path-way, and the imbalance of matrix metalloproteinases and tissue inhibitors of metalloproteinases, while all those would impair the tissue insu-lin sensitivity.
