ABSTRACT

This paper summarized the clinical experience of CHEN Tongyun in the treatment of postinflammatory dyspigmentation with the method of unblocking and nourishing qi and blood. It is believed that the core pathogenesis of this disease is poor qi movement and skin blood stasis, for which the method of unblocking and nourishing qi and blood should be used. Postinflammatory pigmentation on the face is mainly caused by qi stagnation and blood stasis, and it is suggested to regulate liver and spleen, move qi and invigorate blood usually with modified Tonghua Decoction (通化汤). Postinflammatory hypopigmentation is mainly due to qi and blood depletion, for which the treatment should be fortifying the spleen and strengthening kidney, replenishing qi and generating blood, and modified Yangfu Decoction (养复汤) is commonly used. Simultaneously, medicinals of ascending and descending functions, moving qi and blood, warming yang and nourishing yin should be combined, and the results from modern pharmacological research should be considered.

ABSTRACT

Objective:To study the effect of graphene oxide (GO) on adipose-derived mesenchymal stem cells (Ad-MSCs) to promote diabetic skin wound healing.Methods:Ad-MSCs were cultured with GO at concentration of 100, 200, and 400 μg/mL and with 25 mmol/L of glucose for 48 h. The apoptosis of Ad-MSCs was detected by flow cytometry and Western blotting. Eighteen BALB/c thymus free female nude mice were selected to establish the skin defect model of diabetic nude mice. Then phosphate-buffered saline, Ad-MSCs, and GO+ Ad-MSCs were injected into the damaged skin of nude mice by intradermal injection for treatment, thereby created the control group, Ad-MSCs group, and GO+ Ad-MSCs group, 6 mice each group. The survival of wound cells and skin healing of mice were observed after surgery, and the wound healing was determined by HE staining and Masson staining.The level of epidermal growth factor (EGF), vascular endothelial growth factor (VEGF), tumor necrosis factor-α (TNF-α), and interleukin 6 (IL-6) were determined by enzyme-linked immunosorbent assay (ELISA).Results:Compared with the high glucose group without GO(control group), the apoptosis rate of Ad-MSCs co-culture with GO was significantly reduced( P<0.05), and the apoptosis rate decreased with the increase of GO concentration. The expression level of Bax protein decreased, and the expression level of Bcl-2 protein increased ( P<0.05). Diabetic skin defect model showed that the survival rate of Ad-MSCs and wound healing degree in GO+ Ad-MSCs group were significantly increased ( P<0.05), and the degree of epithelialization and thickness of collagen regeneration were better than those in other groups. ELISA results showed that the expressions of EGF and VEGF were significantly increased in GO+ Ad-MSCs group ( P<0.05), while the expressions of TNF-α and IL-6 were significantly decreased ( P<0.05). Conclusion:GO can inhibit the apoptosis of Ad-MSCs in vitro. In vivo, Ad-MSCs treated with GO have higher survival rate, faster wound healing, and better effect than Ad-MSCs treated alone.

ABSTRACT

Objective:To explore the independent risk factors and early diagnosis of dermatomyositis (DM) with positive anti-melanoma differentiation associated gene 5 (MDA5) antibody.Methods:A total of 223 DM patients admitted to the Department of Rheumatology and Immunology, Affiliated Hospital of Xuzhou Medical University from January 2012 to June 2021 were retrospectively analyzed, according to whether the anti-MDA5 antibody was positive or not, the patients were divided into anti-MDA5 antibody positive group ( n= 34) and anti-MDA5 antibody negative group ( n=189). The demographic data, clinical manifestations and laboratory test results of the two groups were compared. The risk factors of DM patients with positive anti-MDA5 antibody were analyzed by binary Logistic regression, and the receiver operating characteristic curve (ROC) was drawn to evaluate the predictive value of various risk factors for anti-MDA5 antibody positive DM patients. Results:The incidence of skin ulcer [20.6%(7/34) vs 9.0%(17/189), χ2=4.03, P=0.045], the positive rate of anti-Ro52 antibody [61.8%(21/34) vs 21.2%(40/189), χ2=23.90, P<0.001], the incidence of interstitial lung disease (ILD) [91.2% (31/34) vs 38.6%(73/189), χ2=31.98, P<0.001] and rapidly progressive interstitial lung disease (RP-ILD) [67.6%(23/34) vs 5.3%(10/189), χ2=88.87, P<0.001], the levels of D-dimer [1.87(1.23, 2.56) μg/ml vs 1.15(0.59, 1.29) μg/ml, χ2=4.68, P<0.001] and serum ferritin (SF) [931.65(579.12, 1 160.43) ng/ml vs 507.40(200.40, 588.55) ng/ml, χ2=5.60, P<0.001] in the anti-MDA5 antibody positive group were higher than those in the anti-MDA5 antibody negative group, wherease the creatine kinase (CK) level in the anti-MDA5 anti-body positive group was significantly lower than that in the anti-MDA5 antibody negative group [85.50(61.25, 1 55.00) U/L vs 263.00(66.50, 1 111.14) U/L, χ2=3.08, P=0.002]. Multivariate Logistic regression analysis showed that positive anti-Ro52 antibody [ OR(95% CI)=5.027(1.632, 15.483), P=0.005], increased D-dimer level [ OR(95% CI)=1.665(1.124, 2.466), P=0.011], occurrence of ILD [ OR(95% CI)=10.071(2.061, 49.207), P=0.004] and RP-ILD[ OR(95% CI)=10.91(3.294, 36.134), P<0.001] were independent risk factors for anti-MDA5 antibody positive DM patients. The ROC curve showed that the areas under the curve (95% CI) of anti-Ro52 antibody, D-dimer, ILD and RP-ILD were 0.703(0.615, 0.791), 0.752(0.661, 0.843)], 0.763(0.703, 0.822), 0.812(0.730, 0.893) respectively. The P value of all these parameters were <0.001 and their value in predicting anti-MDA5 antibody positive DM was high. Conclusion:Positive anti-Ro52-Ab, increased D-dimer level, presence of ILD and RP-ILD are independent risk factors for anti-MDA5 antibody positive DM patients, and they have certain early diagnostic value for DM patients with positive anti-MDA5 antibody.

ABSTRACT

Objective:To study the clinicopathological features of thyroid micropapillary carcinoma and the risk factors of cervical lymph node metastasis.Methods:Two hundred and ninty-eight thyroid micropapillary carcinoma patients diagnosed by postoperative pathology at the Affiliated Hospital of Xuzhou Medical University from Sep 2018 to Jun 2021 were retrospectively divided into metastasis group and non metastasis group according to whether there were cervical lymph node metastasis. Multivariate logistic regression was used to analyze the independent risk factors of cervical lymph node metastasis in thyroid micropapillary carcinoma.Results:The cervical lymph node metastasis rate was 41.3%. Univariate analysis showed that there were significant differences between the two groups in patient's height, maximum tumor diameter, calcification, capsule infiltration and Hashimoto thyroiditis (all P<0.05). Multivariate logistic regression analysis showed that height ≥ 160.5cm, maximum tumor diameter ≥5.5 mm, capsule infiltration and Hashimoto thyroiditis were independent risk factors for cervical lymph node metastasis. Conclusion:Cervical lymph node metastasis in patients with thyroid micropapillary carcinoma is related to the maximum diameter, height, capsule infiltration and Hashimoto thyroiditis.

ABSTRACT

Objective:To investigate the diagnostic value of triglyceride/cystatin C (TG/Cys-C) ratio combined with diabetic retinopathy, diabetes course and systolic pressure in patients with diabetic kidney disease (DKD).Methods:Patients with type 2 diabetes and renal insufficiency, who underwent renal biopsy in four Grade A tertiary hospitals in Xuzhou from January 2013 to February 2021 were included in this retrospective study. Patients were divided into DKD group ( n=51) and non-DKD (NDKD) group ( n=49) based on renal biopsy results. Another 50 patients with type 2 diabetes mellitus without renal dysfunction were selected as control group. The first admission information and blood biochemical indexes were collected, and the TG/Cys-C ratio was calculated. The differences of clinical indexes between the DKD and NDKD groups were compared. The correlative factors of DKD in type 2 diabetes patients with renal dysfunction were analyzed by logistic regression. The ROC curve was used to evaluate the diagnostic value of TG/Cys-C ratio, diabetic retinopathy, duration of diabetes, systolic blood pressure and combined detection for DKD in patients with type 2 diabetes and renal dysfunction. Results:Among patients with type 2 diabetes with renal dysfunction, renal biopsy results showed that membranous nephropathy was the most common pathological type in NDKD group, accounting for 63.3% (31/49) of all NDKD patients. Compared with Type 2 diabetes mellitus without renal injury, type 2 diabetic patients with kidney injury had higher systolic blood pressure, higher total cholesterol, higher low-density lipoprotein, higher cystatin, higher creatinine, higher uric acid, higher fibrinogen level, more hypertension, more changes of urinary active sediment, lower hemoglobin, lower albumin, and the lower the glomerular filtration rate (all P<0.05). Compared with NDKD group, patients in DKD group had higher systolic and diastolic blood pressure, higher cystatin C and creatinine, more hypertension, more diabetic retinopathy, longer course of diabetes, lower concentration of hemoglobin, lower glomerular filtration rate, lower triglyceride and lower TG/Cys-C ratio (all P<0.05). Multivariate logistic regression analysis showed that TG/Cys-C ratio was associated with DKD in patients with type 2 diabetes mellitus and renal impairment ( OR=0.298, P=0.007), diabetic retinopathy ( OR=12.209, P=0.005), duration of diabetes ( OR=1.016, P=0.034) and systolic blood pressure ( OR=1.049, P=0.006) were independent risk factors for DKD in type 2 diabetic patients with renal dysfunction. The ROC curve showed that AUC of the TG/Cys-C ratio was 0.866 (the cot-off point was 2.06), and the sensitivity was 88.2% and the specificity was 71.4% for the diagnosis of DKD, and AUC of the TG/Cys-C ratio in combination with diabetic retinopathy, diabetes duration and systolic blood pressure was 0.952, and the sensitivity was 92.2% and the specificity was 89.8% for the diagnosis of DKD. Conclusion:TG/Cys-C ratio has significant clinical value in the diagnosis of DKD. Combined detection of TG/Cys-C ratio with diabetic retinopathy, course of diabetes and systolic blood pressure could further improve the diagnostic efficacy of DKD.

ABSTRACT

OBJECTIVE: To observe the inhibitory effects and possible mechanism of new small molecular kinase inhibitors Ibr-7 [Irutinil(Ibr) derivatives] on human pancreatic cancer Capan-2 cells. METHODS: Taking Capan-2 cells as objects, CCK-8 method was used to determine the proliferation of cells after treated with 1, 2, 4, 8 μmol/L Ibr/Ibr-7 for 48 h. The survival rates of cells were calculated. Sensitization effects of 1 μmol/L Ibr/Ibr-7 on different doses of gemcitabine/paclitaxel (0.062 5, 0.125, 0.25, 0.5, 1 μmol/L) were detected. Clone formation test was used to detect the situation of cell clone formation after treated with 1, 2, 4 μmol/L Ibr/Ibr-7 for 48 h. The number of cell colony formation was recorded. Flow cytometry or JC-1 method was used to detect the apoptosis of cells after treated with 2, 4, 8 μmol/L Ibr-7 for 24 or 16 h and the changes of mitochondrial transmembrane potential; total apoptotic rate and the percentage of mitochondrial membrane potential decrease were calculated. Western blotting was used to detect the expression of related apoptotic protein (PARP, Noxa, Bcl-2, Bax, Mcl-1, Bcl-xL). RESULTS: After treated with 1, 2, 4, 8 μmol/L Ibr/Ibr-7 for 48 h, the survival rates of cells were decreased significantly; those of Ibr-7 groups were significantly lower than those of same-dose Ibr groups; IC50 of Ibr-7 was significantly lower than that of Ibr (P<0.05 or P<0.01). After combined with Ibr/Ibr-7, the survival rate of cells was significantly lower than that of same-dose gemcitabine/paclitaxel alone group, and the Ibr-7 combination group was significantly lower than same-dose Ibr combination group (P<0.05 or P<0.01). After treated with 2, 4 μmol/L Ibr and 1, 2, 4 μmol/L Ibr-7 for 48 h, the number of cell clone formation was decreased significantly, while Ibr-7 groups were significantly lower than same-dose Ibr groups (P<0.01). After treated with different doses of Ibr-7 for 24 or 16 h, total apoptosis rate of cells (2, 4, 8 μmol/L), the proportion of cell mitochondrial membrane potential decrease (8 μmol/L), the relative protein expression of Noxa (2, 4, 8 μmol/L) and Bax (8 μmol/L) were increased significantly, while the protein expression of PARP (8 μmol/L), Bcl-2 (4 μmol/L), Mcl-1 (2, 4, 8 μmol/L) were decreased significantly; above indexes (except for relative expression of PARP and Bcl-2) of 8 μmol/L Ibr-7 group were significantly better than same-dose Ibr group (P<0.05 or P<0.01). There was no statistical significance in protein expression of Bcl-xL among those groups (P>0.05). CONCLUSIONS: Compared with Ibr, Ibr-7 has better inhibitory and apoptotic effects on human pancreatic cancer Capan-2 cells in vitro, and has stronger chemotherapeutic drug sensitization activity, the mechanism of which may be associated with reducing mitochondrial transmembrane potential, down-regulating the protein expression of PARP, Bcl-2 and Mcl-1 and up-regulating the protein expression of Noxa and Bax.

ABSTRACT

Objective We aimed to identify key genes and pathways of airway epithelial cells involved in bronchial asthma by comparing genetic information in the databases for patients with bronchial asthma and normal people. Methods To find differentially expressed genes (DEGs), mRNA microarray dataset, GSE43696, of airway epithelial cells in asthma was analyzed by GE02R. Functional and pathway enrichment analyses were performed for DEGs using the DAVID database. The protein-protein interaction networks were established using STRING to identify key genes and important complexes. Results A total of 355 DEGs were identified; of which, 130 were up-regulated and 225, down-regulated. The genes identified were involved in cell movement, growth factor binding, and ion channel activity. Nine key genes were recognized, including BDNF, ERBB2 IL6, VEGFA, KIT, ADCY4, PRKAR2B, CCR6, and NMU. Conclusion All nine key genes identified play important roles in asthma and serve as potential targets for treatment of bronchial asthma.

ABSTRACT

Objective To evaluate the effect of duloxetine on the expression of Toll?like receptor 4 (TLR4) in the spinal dorsal horn in a rat model of diabetic neuropathic pain (DNP). Methods Type 2 di?abetes mellitus was induced by high?fat and high?sucrose diet and intraperitoneal streptozotocin ( STZ) 35 mg∕kg in male Sprague?Dawley rats, aged 2 months, weighing 180-220 g. Seventy?five rats with type 2 di?abetes mellitus were randomly divided into 5 groups ( n=15 each ) using a random number table: group DNP, DNP + normal saline group (group DNP+NS), and DNP + duloxetine 5, 10 and 20 mg∕kg groups (DNP+D5, DNP+D10, DNP+D20 groups). Another normal 15 rats were selected and served as control group ( group C) . Duloxetine 5, 10 and 20 mg∕kg were injected intraperitoneally once a day for 14 consecu?tive days starting from 15 days after administration of STZ in DNP+D5 , DNP+D10 and DNP+D20 groups, respectively. The equal volume of normal saline was given instead in group DNP+NS. The mechanical paw withdrawal threshold ( MWT) and thermal paw withdrawal latency ( TWL) were measured on the day before STZ administration and 14, 17, 21 and 28 days after STZ administration. After the last measurement of pain threshold, the L4?6 segments of the spinal cord were removed for determination of the expression of TLR4 by immuno?histochemistry and Western blot. Results Compared with group C, the MWT was signif?icantly decreased, TWL was shortened, and the expression of TLR4 was up?regulated in DNP, DNP+D5, DNP+D10 and DNP+D20 groups (P0?05) . Conclusion The mechanism by which duloxetine attenuates DNP is related to down?regulated expression of TLR4 in the spinal dorsal horn of rats.

ABSTRACT

Objective:To explore the clinicopathologic features of 112 patients with mantle cell lymphoma (MCL). Methods:Da-ta from 112 MCL cases were collected, and immunohistochemical assay was conducted. A break in the CCND1 gene was detected by fluorescence in situ hybridization (FISH). The t-test was used in the statistical analysis. Results:All tumor cells in the 112 cases ex-pressed B cell-related antigen, including 1 blastoid subtype and 1 polymorphic subtype. Among all the cases, 106 expressed CD5 and 104 expressed cyclinD1. A break in the CCND1 gene was not found in 3 cases with CD5-MCL. IgH/CCND1 polyploid was found in 2 classical cases. Conclusion:MCL is a type of special immunophenotypic B-cell lymphoma. The prognoses of blastoid and polymorphic subtypes are poor. Special subtypes should be classified during diagnosis.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the reversal effects of different concentrations of DNA methylation inhibitor, 5-aza-2-deoxycytidine, on the hypermethylation of maternally expressed gene 3 (MEG3) gene promoter, and then the inhibitory effect of restoration of MEG3 expression on the proliferation of ovarian cancer cells.</p><p><b>METHODS</b>Human ovarian cancer OVCAR3 cells were treated with various concentration of 5-aza-2-deoxycytidine (0, 1, 5, 10, 20 µmol/L, respectively) for 6 days. Then the methylation status of MEG3 promoter was detected by methylation specific PCR (MSP). The alteration of MEG3 gene expression was detected by RT-PCR. Cell proliferation was determined by MTT assay and EdU incorporation assay.</p><p><b>RESULTS</b>After treated with 5-aza-2-deoxycytidine, the methylation status of MEG3 in the 0, 1, 5, 10, 20 µmol/L 5-aza-2-deoxycytidine groups were 1.00 ± 0.00, 0.79 ± 0.00, 0.67 ± 0.00, 0.65 ± 0.03 and 0.61 ± 0.01 folds, respectively (P < 0.05 for all). The relative expressions of MEG3 mRNA in the 0, 1, 5, 10, 20 µmol/L 5-aza-2-deoxycytidine groups were 1.00 ± 0.00, 2.04 ± 0.16, 2.44 ± 0.17, 3.19 ± 0.34 and 5.34 ± 0.39, respectively (P < 0.05 for all). In contrast to the negative control, the inhibition rates of the OVCAR3 cell growth were increased significantly when treated with 1, 5, 10, 20 µmol/L 5-aza-2-deoxycytidine in 2, 4 and 6 days. There were (40.78 ± 0.80)%, (35.65 ± 0.33)%, (31.81 ± 0.66)%, (27.33 ± 1.27)% and (17.75 ± 1.85)% of EdU-positive cells in the 0, 1, 5, 10 and 20 µmol/L 5-aza-2-deoxycytidine groups (P < 0.01 for all).</p><p><b>CONCLUSIONS</b>Maternally expressed gene 3 promoter hypermethylation is reversed by 5-aza-2-deoxycytidine in ovarian cancer cells. The downregulation of MEG3 gene might be resulted from the methylation, and the re-expression of MEG3 partly contribute to the growth inhibition of epithelial ovarian cancer cells.</p>

ABSTRACT

Objective To develop a candidate reference method for the determination of serum creatinine and to evaluate the ac-curacy of conventional detection systems though method comparison to achieve traceability .Methods The candidate reference method was established according to the sarcosine oxidase and the accuracy and reliability of the method was verified through par-ticipation in international reference laboratories EQA activities (IFCC-RELA) .20 fresh single human serum samples with different concentration and calibrator were simultaneously measured by using conventional detection system and candidate reference method . Results The calibration curve for serum creatinine was linear in the concentration range from 50-2 000 μmol/L with a correlation coefficient of 0 .999 9 under the optimum experimental conditions (the linear equation was Y=0 .000 884 2X-0 .000 325 3) and the imprecision was less than 1 .0% .The proposed method has been applied to the determination of RELA samples with satisfactory re-sults .The measured results with conventional detection systems were consistent with candidate reference method ,and the slope of the regression equation was 1 .005 6 .Conclusion The candidate reference method of serum creatinine is successfully established and which can be used for traceability and standardization .It may provide an effective way for conventional detection system traceable to the reference method or reference material .

ABSTRACT

Objective To investigate the feasibility of fine needle aspiration cytopathology in the diagosis of lymphoma.Methods To compare cytopathology with histopathology of 72 lymphoma cases and analyse immunochemistry stain of 3 cases.Results 61 of 72 cases were diagnosised as lymphoma in cytopathology,while 58 of 61 cases were diagnosised as lymphoma in histopathology.The sensitivity,specificity,accuracy,positive predictive value and negative predictive value rates of cytopathology were 89.2 % (58/65),57.1% (4/7),86.1% (62/72),95.1% (58/61),and 36.4 % (4/11) respectively.Conclusion Fine needle aspiration cytopathology is significant to the diagnosis of lymphoma.It should greatly improve the accuracy of lymphoma diagnosis and make it possible to subclassify lymphoma,combined cytopathology with other auxilary detection.

ABSTRACT

Objective To investigate the efficacy and safety of isosorbide mononitrate sustained release tables plus vinorelbine and cisplatin in patients with previously untreated advanced non-small-cell lung cancer (NSCLC).Methods One hundred and ten patients with stage ⅢB-Ⅳ NSCLC were randomly assigned to group A (57 cases) and group B (53 cases) by random mumber table.Patients in group A were treated with vinorelbine 25 mg/m2 on the first and eighth day and cisplatin 25 mg/m2 on day 2-4,with transdermally applied isosorbide mononitrate sustained release tables (40 mg,daily for 8 days),and patients in group B were treated with vinorelbine and cisplatin.Response to treatment was assessed by RECIST1.1 and adverse effect was assessed by NCI-CTC(3.0).Results The response rate in group A (58.2%,32/55 patients) was significandy higher than that for patients in group B (30.8%,16/52 patients; x2 =8.120,P =0.004).Median TTP and median OS in group A were longer than those in group B (8.2 vs 5.8 months,x2 =10.684,P =0.001 ; 11.6 vs 9.0 months,x2 =11.231,P =0.001).While,patients with squamous carcinoma showed better response to chemotherapy (RR =2.438,95% CI:1.136-5.231,P =0.022).Adverse effect difference was not significant between group A and group B,except headache.The rate of grade 1 to 2 headache in group A (34.5% ; 19 of 55 patients) was significantly higher than that in group B (3.8% ; 2 of 52 patients; P <0.001).Conclusion Use of isosorbide mononitrate sustained release tables combined with vinorelbine and cisplatin may improve overall response,TTP and OS in patients with advanced stage NSCLC.

Objective To explore the relationship between the prothrombotic state and blood coagulation-fibrinolysis system changes with deep venous thrombosis(DVT)in aged patients after total joint arthroplasty,and to propose preventive measures.Methods 400 patients who underwent total hip or knee replacement from January 2003 to June 2011 were classified into suspected DVT(n=200 cases)and non-suspected DVT(n=200 cases)according to Well's clinical scoring system.The patients were divided into 4 subgroups based on the measures to prevent DVT:low molecular weigh heparin group,intermittent pneumatic bag compression group,combined above measures group,control group who refused any preventive measures.Plasma thrombin-antithrombin complex(TAT),plasmin-α2 antiplasmin complex(PAP)were determined preoperatively.Venous Doppler ultrasound was performed before surgery,4 d and 10 d after surgery to detect the presence of DVT.Results Totally 71 cases(17.8%)were diagnosed as DVT and 5 cases(1.3%)as pulmonary thromboembolism(PTE).The incidence of DVT in suspected DVT group(28.5%,57 cases)was lower than in non-suspected DVT group(7.0 %,14 cases)(x2 =31.66,P< 0.01).Among patients with suspected DVT,the DVT prevalence in combined measures group(2.0 %,1 cases)was decreased than in low molecular weigh heparin group(14.0%,7 cases),intermittent pneumatic bag compression (34.0%,17 cases)and control groups(64.0%,32 cases)(x2=4.89,17.34,18.01,all P<0.05).In the patients with non-suspected DVT,the DVT prevalence in combined measures group(0.0%)was lower than in control group(24.0 %,12 cases)(x2 =13.64,P<0.01).The average preoperative level of TAT[(9.63±3.06)μg/L]in patients with DVT was higher than without DVT[(2.59±0.87)μg/L](t=35.70,P<0.01),while PAP level in patients with DVT[(38.52± 21.13)μg/L]was reduced than without DVT[69.75±30.26)μg/L](t=8.27,P<0.01).Conclusions The levels of TAT and PAP before total hip or knee replacement are predictive for lower extremity DVT.The clinical effectiveness of Well's scoring on DVT prevention is dependent on the different TAT and PAP state.

ABSTRACT

Objective To investigate expression of VEGF and the in vitro angiogenesis ability induced by ovarian cancer cells after RNA interference on expression of matrix metalloproteinase-2 (MMP-2)gene.Methods One specific target sequence of MMP-2 and one non-specific sequence (NC group) were chosen,the DMEM as blank group.After transfection of ovarian cancer OVCAR-3 cells,mRNA and protein expression of MMP-2 and VEGF genes were examined by RT-PCR and Western blot analysis,and the angiogenesis ability was detected by in vitro angiogenic assay.Results When compared with the NC group,the mRNA expression of MMP-2 and VEGF were decreased by 78.8 % and 75.5 % (P < 0.05) in 48 h after transfected,respectively,and protein expression was decreased by 81.2 % and 78.3 % (P < 0.05) at the same time point.In vitro angiogenic assay suggested that the ability of angiogenesis was inhibited when down-regulated of MMP-2 gene (P < 0.05).Conclusion Down-regulation of MMP-2 gene in ovarian cancer cells by RNA interference could inhibit its VEGF expression and in vitro angiogenesis induced by ovarian cancer cells,which suggestes that the inhibition of MMP-2 gene has an anti-angiogenesis effect,and MMP-2 gene could be a potential target for ovarian cancer gene-therapy.

ABSTRACT

Objective To study the effect of BRMS1 on the invasion and metastasis of ovarian cancer cells. Methods BRMS1 small interfering RNA (BRMS1-siRNA) was transfected into human ovarian cancer cell-line OVCAR3 by liposome transfection method. The cells were divided into 3 groups: experimental group with BRMSl-siRNA, negative control group with siRNA that did not impact any gene, blank control group without any transfection. Changes of invasion and migration in the cells were per-formed using transwell invasion and migration assay. Results BRMS1 gene was silenced in OVCAR3 cell line successfully detecting by quantitative real-time RT-PCR and immunoblotting.In transwell invasion assay,the numbers of cells in lower chamber passing through the membrane in transfected group were more than negative control group and blank control group (190±8.5, 144±7.8 and 146±6.8 respectively) (t=8.747, t=8.869, P=0.000), while in transwell migration assay, the numbers of cells in lower chamber passing through the membrane in transfected group were more than negative control group and blank control group were (231 ±8.9, 177±9.7 and 182±7.9 respectively) (t=9.314, t=9.224, P=0.000), both with significant differences among the 3 groups. Conclusion BRMS1 gene could suppress the invasion and metastasis of ovarian cancer cells.

Objective Through comparision of Her-2 gene in invasive breast carcinoma tissues assayed by IHC, CISH and FISH respectively, to explore an optimal detective methods. MethodsIHC, CISH and FISH were used in detection for Her-2 gene of 100 cases, respectively.ResultsCISH identified no Her-2 gene amplification in IHC score negative rumors (6/6) and gene amplication in 7.1%(1/14) of IHC score (+) and in 32 % of IHC score (++) tumors(l6/50) and in 80 % of IHC score (+++) (24/80), Concordant rate between CISH and FISH was 100 %(20/20). ConclusionThere are advantage and disadvantage among IHC,CISH and FISH. CISH is an accurate and dependable methods and would be easy to apply for detection of Her-2 gene.

ABSTRACT

Objective To evaluate the efficacy and safety of brachytherapy by percutaneous paracentesis implantation of iodine-125 seeds for elderly non-small cell lung cancer.Methods 18 patients with peripheral lung cancer were undergone brachytherapy by percutaneous paracentesis implantation of iodine-125 seeds.The effect was evaluated by periodically chest CT.The complications were analyzed.Results 18 patients were observed one,two and six months after operation.CR,PR,SD were seen in 27.8%(3/18),66.7%(14/18),5.6%(1/18)respectively of the patients.Pneumothorax,hemoptysis were seen in 16.67%(3/18),95.37%(17/18),no pleural cavity bleeding.Conclusion Brachytherapy by percutaneous paracentesis implantation of iodine-125 seeds for lung cancer was effecitve.The complications were easy to treat.The methods of implantation of iodine-125 seeds could fully play the advantages of CT,and reduce the damage to the body when raising the tumorous control rate.

ABSTRACT

<p><b>BACKGROUND</b>To investigate the clinical value of serum total MMP-9 detection in lung malignancies.</p><p><b>METHODS</b>Serum total MMP-9 levels were detected in 63 patients with lung malignancies by sandwich ELISA with monoclonal antibody against human total MMP-9.</p><p><b>RESULTS</b>Serum total MMP-9 was (92.2± 74.39) μg/L in lung malignancy group, which was significantly higher than that of healthy control [(9.5± 6.74) μg/L](P < 0.05). In lung cancer group, there were significant differences in serum total MMP-9 level between patients with and without lymph node metastasis, and between progressive disease+death group and complete response+partial response group. Significantly negative correlation was observed between serum CA242 and serum total MMP-9 levels (P=0.021).</p><p><b>CONCLUSIONS</b>Detection of serum total MMP-9 may be helpful to predict metastasis and treatment response of lung cancer.</p>

ABSTRACT

Purpose:To explore the effect of anti-FasL on ADM resistance of K562/ADM cell subline and its possible mechanism.Methods:Control group,untreated with VER or anti-FasL ,VER-treated group,anti-FasL-treated group and anti-FasL +VER-treated group were set up. MTT、FCM、TUNEL、S-P immunechemistry and RT-PCR assay were used for detection.Results:Anti-FasL or VER treatment was capable of strengthening sensitivity of K562/ADM cells to ADM,ID 50 of anti-FasL and VER was 200ng/ml and 6?g/ml,respectively;DNA content of apoptosis from cycling analysis curve of K562/ADM cell was 7.42%,32.10%,49.29%,56.26%,respectively,and positive rate in situ apoptosis was 4.5%,36%,45%,58%,respectively,which represented a tendency of gradual increase. Anti-FasL was capable of effecting FasL and bcl-2 expression,but not Pgp.Conclusions:Anti-FasL alone or synergically with VER was capable of reversing ADM resistance of K562/ADM cell,which may be used as a novel reversor of multidrug resistance.
