ABSTRACT

Objective To investigate the effect of rh-resistin on lipid metabolism in HepG2 cells and to elucidate its relation to AMPK pathway. Methods We treated the HepG2 cells with 50 ng/ml rh-resistin and 0.5 mmol/L palmitic acid,used siRNA technique to inhibite α2 subunite expression of AMPK in HepG2 cells and quantitative RT-PCR to detect ACC1,ACC2,and HL mRNA expression levels of related lipid metabolism genes. The P-AMPK-Thr172 of AMPK and P-ACC-Ser79 of ACC were determined by Western blotting. The Lipid accumu-lation in cells was determined by images of Laser Scanning Confocal Microscope after Nile red staining. Results Rh-resistin decreased the AMPKα2,HL mRNA expressions and the phosphorylation level of AMPK and ACC in both basal and insulin-stimulated conditions (P 0.05),while it increased ACC2 mRNA expressions and cytoplasmic lipid droplets in the same conditions (P <0.05). Conclusion Rh-resistin may affect lipid metabolism via AMPK pathway with increase of fatty acid synthe-sis and inhibition of triglyceride catabolism,which leading to lipid accumulation in HepG2 cells.

ABSTRACT

Objective To investigate the correlation between hepatic triglyceride content and glucose lipid metabolism,insulin resistance and β cell function.Methods A total of 32 type 2 diabetic patients with nonalcoholic fatty liver disease was recruited in this study.Hepatic triglyceride content was measured with liver proton magnetic resonance spectroscopy.Oral glucose tolerance test (OGTT) was carried out in all participants,with measurements of plasma glucose and insulin levels.The homeostasis model assessment insulin resistance index (HOMA-IR),hepatic insulin resistance (HIR),and Matsuda Index (MSI) were used to assess insulin resistance.The homeostasis model assessment beta cell function (HOMA-βF),early insulin secretion index (EISI) and late insulin secretion index (LISI) were used to assess β cell function.Results Hepatic triglyceride contents had positive correlations with body mass index (BMI),waist circumference,Body fat,aspartate transaminase (AST),alanine transaminase (ALT),triglycerides (TG),HOMA-IR,HIR,and negative correlations with MSI.Stepwise regression analysis showed that body fat and HOMA-IR were independently risk factors for hepatic triglyceride contents.Conclusions Hepatic triglyceride content is closely correlated with obesity,liver function,blood lipid,and insulin resistance;especially obesity and insulin resistance are the most important factors.

ABSTRACT

Objective:To explore the effect of smart phone's application on health education in patients with periodontal disease,and to analyze its ethical significance.Methods:A sample of 160 patients with periodontal disease was selected from the department of stomatology in our hospital,from June 2015 to June 2016.They were divided into two groups.Patients in the experiment group (n =80) were received health education using smart phone's application,while those in the control group (n =80) were not.The effects of oral health education in both groups were analyzed.Results:Compared with the control group,patients in the experiment group had a better understanding of oral health knowledge and higher compliance of oral health maintenance,and the difference was statistically significant (P < 0.05).Conclusion:The smart phone's application can improve the awareness rate of health knowledge of periodontal disease and the compliance of oral health maintenance,meet the demands of patients with periodontal disease,and highlight the nursing ethical spirit of respecting for patients and wholeheartedly serve for patients in clinic.

ABSTRACT

Objective:To explore the function of risk management in outpatient blood collection work under the conditions of informationization. Method:This paper retrospectively reviewed the nursing risk management in out-patient blood collection work from January 2014 to January 2015 in our hospital, analyzed the causes of risk in out-patient blood collection work, evaluated the possible adverse outcomes, and put forward the measures to prevent and control risks. Results:Through the nursing risk management, the nurses′ risk prevention consciousness was enhanced, as well, both nurse and patient satisfaction was improved. Conclusion:Application of risk management in outpatient blood collection work could improve the quality of nursing, conform to the ethical requirements of guaranteeing patient safety, and effectively reduce the incidence of medical risks and accident.

ABSTRACT

Objective To investigate the effect of rh-resistin on glucose metabolism in HepG2 cells and to elucidate whether the underlying mechanisms are related to AMPK pathway. Methods Cells transfected with control siRNA or AMPKα2 siRNA were cultured in 6-well plates and then treated with 50 ng/mL rh-resistin for 24 hours , while untransfected cells were treated with or without 50 ng/mL rh-resistin on the same conditions , followed by serum-starving in glucose-free DMEM for 3 ~ 5 hours in the continued absence or presence of rh-re-sistin. Then the cells were treated with or without insulin for 2 hours. AMPKα2, G6Pase, PEPCK and Glut2 mRNA expression levels were determined by quantitative RT-PCR. The phosphorylation state of AMPK was deter-mined by Western blotting. Glycogen synthesis was measured by the incorporation of D-[U-14C] glucose to glycogen. Results Rh-resistin suppressed the AMPKα2, Glut2 mRNA expressions, and reduced the phosphory-lation level of AMPK and glycogen synthesis on both basal and insulin-stimulated conditions (P < 0.05), while it accelerated G6Pase and PEPCK mRNA expressions on the same conditions (P < 0.05). The mRNA expression levels of G6Pase , PEPCK , Glut2 and the phosphorylation level of AMPK and glycogen synthesis were signifi-cantly different between the rh-resistin group and the rh-resistin in conjunction with AMPKα2 siRNA-treated group. Conclusion Rh-resistin may affect glucose metabolism in HepG2 cell via AMPK pathway.

ABSTRACT

ABSTRACT:Objective To analyze the stress changes of tooth tissues in the medial-occlusal (Class Ⅱ)cavity restored with different kinds of inlay materials by the three-dimensional (3-D)finite element.Methods The inlay restored 3-D finite element model of medial-occlusal (Class Ⅱ)cavity was established by CBCT scanning method and reverse engineering software Mimics,Geomagic Studio,Pro/E5.0 software,and finite element analysis software. The Von-mises stress and change tendency of tooth tissues and four different kinds of inlay materials were compared after taking vertical loading and tongue 45°to loading.Results There were different stress levels of tooth tissues among different inlays of medial-occlusal (MO)Class Ⅱ cavity.The stress level of the cobalt chromium alloy inlay was the largest,the stress level of the gold alloy inlay was lower,and the stress level of the composite inlay was the lowest.Dental tissues stress distribution was similar for the four different restorative materials,and focused on hole bottom of dentin near the pulp in dentin.Conclusion Compared with ceramics,gold alloy and cobalt chromium inlay restoration,restoration with composite resin inlay can reduce the organization stress of the remaining tooth in the medial-occlusal (Class Ⅱ)cavity type.

ABSTRACT

Objective:To analyse the stress distribution of the tooth and inlay with the restoration of resin,porcelain,gold alloy,co-balt chromium materials respectively.Methods:3-D finite element models of mandibular first molar with meiso-occlusal (class II) cavity and different inlays were established.Von-mises stress distribution on the tooth and inlay with vertical load and lingual load at 45°were analyzed.Results:After restoration peak stress of high elastic module materials and dental inlays was higher than that of the low elastic module material inlays,however,the restorations of the different elastic module materials had the similar stress distribution trend.The stress under of lateral load(lingual at 45°)to the teeth inlays was significantly higher than that under vertical load.Peak stress concentration of tooth was on the bottom of the cavity near the pulp chamber dentin;inlay peak stress distribution is mainly in its corresponding to the gingival wall.In the tooth tissues stress level of the contact surface of inlay restoration,the strength was as the fol-lowing:Composite resin inlay >ceramics inlay >the gold alloy ceramics inlay >cobalt chromium alloy inlay.The stress level of the inlay of the four kinds of inlay restoration materials was just opponent with the tooth tissues.Conclusion:Gingival wall is the weakest part of meiso-occlusal(class II)cavity inlay restoration,while near the pulp chamber at the bottom of the cavity is the weakest part of the tooth.Among the 4 materials Under the same load condition,composite resin inlay restoration has minimal tooth stress and uniform stress distribution,and can reduce the posibility of odontoschism and microleakage.

ABSTRACT

The relationship of erectile dysfunction (ED) with serum levels of total testosterone (TT) and free testosterone (FT) was investigated in 53 men with type 2 diabetes under 50 years of age.The relevant clinical status was matched and no significant difference was noticed for TT [ ( 14.11 ±5.81 vs 14.97 ±4.93 ) nmol/L] and FT [ (9.31 ± 8.84 vs 8.72 ± 5.33 ) pg/ml ] levels in type 2 diabetic patients with and without ED.This study has demonstrated that testicular androgen may not contribute to ED in young and middle-aged men with type 2 diabetes.

Insulin autoimmune syndrome (IAS) is rare.A patient with IAS was herewith reported and 50domestic cases reported in the last two decades were reviewed to discuss the approach to this syndrome.In hypoglycemic patients with high insulin level and with Graves' disease and taking tapazole,insulin autoantibody detection is mandatory to avoid misdiagnosis and unnecessary surgery.

ABSTRACT

Objective To explore the effect of glucose fluctuation on resistin. Methods The phorbol-12-myristate-13-acetate(PMA)-activated and differentiated U937 cells were exposed to experimental condition for 3 days, three groups of cells were formed, each one receiving the following fresh medium every 6 hours, respectively: (1) continuous 11.1 mmol/L glucose concentration medium (Con group), (2)continuous 22.2 mmol/L glucose concentration medium (CHG group), (3) alternating 11.1 mmol/L glucose concentration and 22. 2 mmol/L glucose concentration medium every 6 hours (IHG group). The supernatants of cell mediam at the last 6 hours were collected to test resistin concentration. Besides, 92 subjects were selected and classified into three groups according to the results of oral glucose tolerance test:normal glucose tolerance group ( NGT group, n =30), impaired glucose tolerance patients (IGT group, n =31) and newly diagnosed type 2 diabetes patients (T2DM group, n =31). Blood glucose and serum resistin levels were measured at 0 h and 1 h during oral glucose tolerance test ( OGTT) to compare the glucose fluctuation (△Glu1-0) and the change of serum resistin level (△lnRes1-0) among the three groups. Results Resistin concentration in the Con , CHG and IHG group was (73.62 ± 5.07)ng/L, (97.78 ±7.00)ng/L and(212.49 ± 28. 81 )ng/L respectively and in IHG group it was higher as compared with the other two groups (P<0.05). △Glu1-0 in NGT, IGT and T2DM group was(2.31 ±2.30)mmol/L,(5.70 ±2.08)mmol/L and (8.41 ±2.63)mmol/L respectively; △Glu1-0 increased gradually in all the three groups (P<0.05). Serum resistin level from 0 h to 1 h in the NGT group was 6.41 (1.52-15.76) μg/L to 6. 96( 1.52-22. 70) μg/L, in the IGT group 5.47( 1.49-24. 09)μg/L to 9. 12( 1.27-21.94)μg/L and in the T2DM group 5.77( 1.11-30.10) μg/L to 9. 27(1.02-48.15)μg/L In the IGT and T2DM group serum resistin level increased from 0 h to 1 h (P<0.05), but no difference was observed in the NGT group (P>0. 05).△lnRes1-0 in these 3 groups was (0.05 ± 0.05) μg/L, (0.25 ± 0.04) μg/L and (0.37 ± 0.03 )μg/L respectively and the change in the T2DM group was significant as compared with that in the NGT group,△lnRes1-0 was positively correlated with △Glu1-0 (r = 0.23, P = 0.02). Conclusion Glucose fluctuation induced monocyte/macrophage to secrete resistin, greater the glucose fluctuation, greater the change of amplitude of serum resistin.

ABSTRACT

BACKGROUND: The experimental results showed that insulin sensitivity and glucose uptake in skeletal muscle could be improved by activating adenosine monophosphate-activated protein kinase a2 (AMPKα2). AMPKa2 is expected to become a new physiological and pharmacological target for the prevention and treatment of type 2 diabetes mellitus. OBJECTIVE: To clone human AMPKa2 subunit gene and to construct its wild-type and mutant eukaryotic expression vectors. DESIGN: A single sample observation.TIME AND SETTING: The experiment was performed in the Clinical Molecular Biology Laboratory, the Second Affiliated Hospital of Sun Yet-sen University from April 2007 to January 2008.MATERIALS: QuikChange II Site-Directed Mutagenesis Kit was produced by Stratagene. Eukaryotic expression vector pcDNA3.1(+) and E. coll DH5a were provided by the laboratory. Human skeletal muscle tissue was from patients who received amputation surgery in the Second Affiliated Hospital of Sun Yat-sen University. Informed consent was obtained from the patients, and fresh samples were collected and frozen in liquid nitrogen.METHODS: The human AMPKo2 subunit gone was amplified from human skeletal muscle by RT-PCR, cloned into T vector, and the recombinant plasmid was confirmed by sequencing. In vitro site-directed mutagenesis was carded out with Quickchange site-directed mutagenesis kit. The wild-type and mutant coding genes were subcloned into eukaryotic expression vector pcDNA3.1, and the recombinant plasmids were validated by enzyme digestion and sequencing.MAIN OUTCOME MEASURES: ①The cloning of aim gone; ②site-directed mutagenesis; ③ eukaryotic expression plasmid. RESULTS: The human AMPKα2 subunit gene (about 1700 bp) was successfully cloned, with 99% homology to the reported AMPK α2 gene. A GenBank accession number was EF056019, The achieved mutation of the 45<'th> Lysine (AAA) was found to Arginine(AGA). The wild-type and mutant pcDNA-AMPKα2 recombinant plasmids were constructed successfully. CONCLUSIONS: The human AMPKα2 subunit gene was cloned successfully from human skeletal muscle and its wild-type and mutant eukeryotic expression vectors were constructed successfully in the experiment.

ABSTRACT

The effect of globular adiponectiin (gAd)on the function of NIT-1 cells under high glucose medium was investigated. The results showed that gad could completely block the increase of NADPH oxidase components p47phox expression and recover mRNA expression of pancreatic duodenal homeobox-I ,paired box gene 6,glucose transpoter 2,and glucokinase except neurogenic differentiation factor 1 (P<0.05 or P<0.01). Whereas,impaired insulin secretion and mRNA expression at high glucose concentration were not significantly improved by gAd.

BACKGROUND:Adiponectin possess functions of lowering blood glucose and blood lipids, and improve insulin sensitivity. But, controversy results about the effect of adiponectin on skeletal muscle have been reported.OBJECTIVE:To study the effects of eukaryon expressed adiponectin on the glycogen synthesis and glucose oxidation in skeletal muscle cell strain C2C12 myotubes by transfecting plasmids carrying mouse adiponectin.DESIGN: A controlled experiment.SETTING: The Second Affiliated Hospital of Sun Yat-sen University.MATERIALS: PcDNA3.0 plasmid with mouse adiponectin cDNA, pcDNA3.0-mad (generously presented from Dr. Gong,University of Maryland), C2C12 cell strain (purchased from ATCC, GRL-1722), DMEM high glucose (Gibco), MEM (Hyclone), fetal bovine serum (Hanagzhou Sijiqing), equine serum (Hyclone), lipofectamine 2000 (Invitrogene), G418 (Gibco), rabbit anti-mouse adiponectin IgG (ACRP303-A, Alpha Diagnostic International), chemiluminescence kit (ECL+PLUS,Amersham), SABC instant immunohistochemistry kit (Boster), D-[U-14C] glucose (specific activity 9.25-13.32 GBq/mmol,NEC), scintillation fluid POP, POPOP (SIGMA), liquid scintillation counter (LS3801, Beckman, USA).METHODS:This study was carried out in the Central Laboratory of the Second Affiliated Hospital of Sun Yat-sen University from March to August, 2003. ① After extraction of plasmid, double digest with Xho Ⅰ and Xba Ⅰ and identification with HindⅢ digest were carried out. ② Plasmid pcDNA3.0-mad and pcDNA3.0 blank vector were transfected using liposome to C2C12 cells, and the stably transfected cells were screened by 500 mg/L G418 for 3 weeks, G418 resistant C2C12 cells were thereafter harvested, therefore stable transfected pcDNA3.0-mad and pcDNA 3.0 C2C12 cell strains were established.③ Adiponectin protein expression was determined by Western blot analysis and immunohistochemistry. ④ Glucose oxidation and glycogen synthesis detections were divided into control, vector and pcDNA3.0-mad (mad)group. Each group was further divided into 4 subgroups with 0, 0.5, 5 and 100 nmol/L insulin (n =6), respectively. Detection of glucose oxidation and glycogen synthesis was carried out with 14C-labeled glucose by counting radioactivity of 14CO2 or 14C labeled glycogen with scintillation, respectively.MAIN OUTCOME MEASURES:Changes of glycogen synthesis and glucose oxidation in skeletal muscle cell strain C2C12myotubes.RESULTS: ① Results of plasmid transfection and restrict digest: After plasmid extraction, double digest with Xba Ⅰ and Xho Ⅰ was carried out along with HindⅢ digest identification.Digest fragments were in accordance with expectation.Length of adiponectin cDNA fragment was 781 bp, plasmid fragment was 5 446 bp, adiponectin cDNA was inserted between digest sites (Xho Ⅰ and Xba Ⅰ ) of eukaryotic expression vector pcDNA3.0. ② Plasmid transfection of C2C12 cell and positive clone screening: On the 10th day of G418 media culture screening after transfection, most C2C12 cells died.Positive clone appeared at the 2nd week. G418 resistant C2C12 colonies were harvested at the 3rd week. ③ Western blot and immunohistochemical identifications: Both confirmed that adipoenctin gene was stably transfected into cells in the Mad group, with successful adipoenctin expression. ④ Effect of stably transfected adiponectin gene to myocyte glucose metabolism:The myocyte glycogen synthesis and glucose oxidation increased along with the increasing of insulin concentration. The linear regression analyses of measured myocyte glucose oxidation amount showed that the regression coefficients of the control group, blank vector group and mad group were 23.34, 2;3.23 and 26.06 respectively. This result indicated that in C2C12 cell stably transfected with adiponectin gene, when insulin concentration increased, the acceleration rate of glucose oxidation increasing was higher than other 2 groups. However, no significant difference could be observed in glycogen synthesis and glucose oxidation of C2C12 cells under basic status without insulin stimulus and treatment status with different insulin concentrations between control group, blank vector group and mad group (P> 0.05).CONCLUSION: ① We have successfully established stably adiponectin gene transfected C2C12 cell strain with adiponectin protein expression ability. ② Transfection with adiponectin cDNA had no significant effect on the glucose oxidation and glycogen synthesis of C2C12 myotubes.③ The glucose oxidation and glycogen synthesis of C2C12 myotubes increased with the increasing of insulin concentration. ④ Adipoenctin may coordinate with insulin in improving myocyte glucose oxidation and increasing myocyte glucose uptake.

ABSTRACT

OBJECTIVE:To analyze the resistance of the main pathogens in our hospital to antimicrobials for clinical reference of rational use of antimicrobial agents. METHODS:The antibiotic resistance of the clinical pathogens in our hospital in 2008 was retrospectively analyzed. RESULTS:Of the total 938 clinical pathogenic bacteria isolated in our hospital,the most common pathogens(leading the first 10 places)were E. coli,staphylococcus aureus,enterobacter spp,staphylococcus epidermidis,pseudomonas aeruginosa,Klebsiella,acinetobacter,enterococcus,hemolytic streptococcus and bacillus proteus. CONCLUSION:Clinicians should use antimicrobial agents rationally to reduce the antibiotic resistance.

Objective To investigate the changes of nuclear factor?B activity in peripheral blood mononuclear cell(PMNC)in newly diagnosed type 2 diabetic patients.Methods Thirty normal individuals and 30 newly diagnosed type 2 diabetics were selected.Phosphorylation status of NF-?B P65 of PMNC was determined by immunoblotting with phosphor-specific antibodies to NF-?B P65(Ser 536 ).Serum hs-CRP was assayed by enzyme linked immunosorbent assay(ELISA).Results The level of NF-?B P65 phosphorylation was increased in newly diagnosed type 2 diabetic patients compared with control subjects[0.85(0.49,1.02)vs 0.47(0.26,0.67),P

ABSTRACT

Objective: To explore the psychological status and therapeutic efficacy of peri-operative psychological intervention among patients with oral carcinoma.Method: psychological statuses of 60 patients with oral carcinoma were investigated,and specific peri-operative psychological interventions were performed according to features and initiating factors of their psychological crisis,in order to provide references for the therapy of oral carcinoma.Result: Common psychological crises among patients with oral carcinoma such as anxiety and fear have been dramatically decreased by proper psychological investigation and peri-operative psychological intervention.Conclusion: Psychological investigation contributes to a great improvement of psychological status of patients with oral carcinoma.Therefore,in addition to routine peri-operative medical treatment,an effective psychological investigation is also crucial for patients with oral carcinoma to cure the disease and regain health.
